Abstract:Due to an increase in the consumption of food, feed, and fuel and to meet global food security needs for the rapidly growing human population, there is a necessity to obtain high-yielding crops that can adapt to future climate changes. Currently, the main feed source used for ruminant livestock production is forage grasses. In temperate climate zones, perennial grasses grown for feed are widely distributed and tend to suffer under unfavorable environmental conditions. Genome editing has been shown to be an effective tool for the development of abiotic stress-resistant plants. The highly versatile CRISPR-Cas system enables increasingly complex modifications in genomes while maintaining precision and low off-target frequency mutations. In this review, we provide an overview of forage grass species that have been subjected to genome editing. We offer a perspective view on the generation of plants resilient to abiotic stresses. Due to the broad factors contributing to these stresses the review focuses on drought, salt, heat, and cold stresses. The application of new genomic techniques (e.g., CRISPR-Cas) allows addressing several challenges caused by climate change and abiotic stresses for developing forage grass cultivars with improved adaptation to the future climatic conditions. Genome editing will contribute towards developing safe and sustainable food systems.

Abstract:Abiotic stress, brought about by salinity, drought, extreme temperatures and oxidative stress are serious threats to agriculture and result in a huge reduction of production.Drought and salinity are becoming major threats throughout the world.Abiotic stress leads to a series of morphological, physiological, biochemical and molecular changes that adversely affect plant growth and development (Wang et al., 2001).In order to survive from these harsh stresses, forage plants have developed precise and complicated tolerance mechanisms at the morphological, physiological and molecular levels.Under serious threat, forage plants can change the shape of the leaves and roots to decrease the water loss.Some plants have evolved special structures, such as salt glands to excrete salt.At the physiological level, respiratory, photosynthesis metabolism and osmotic adjustments etc. all change in order to resist stress (Chaves, 1991; Sheng, 2010;Yang et al., 2007).In fact, all of these changes are related to gene expression.The complex plant response to abiotic stress involves many genes and molecular mechanisms.In the past several decades, multiple genes responding to drought, salt, low-temperature and oxidative stress have been identified.These genes are divided into two groups (Shinozaki et al., 2003).The first group functions to directly protect the plant against stress, involving key enzymes for osmolyte biosynthesis, LEA (late embryogenesis abundant) proteins, detoxification enzymes and enzymes involved in many metabolic processes.The other group consists of contained protein factors involved in the regulation of signal transduction, including various transcription factors, proteins kinases, protein phosphatases, enzymes involved in phospholipid metabolism, and other signaling molecules (Yamaguchi-Shinozaki & Shinozaki, 2006).Genes have been used extensively to improve the stress-tolerance in crop and forage crops.The investigation of stress-tolerance genes will increase our knowledge of tolerance mechanisms, which could in turn be used to promote improvements in forage crop plants tolerance. Abstract:As plants are sessile, they cannot escape exposure to multiple environmental stresses. Over the course of evolution, a range of morphological and physiological adaptations has been developed in order to maximize their chances of survival. Alfalfa (Medicago sativa L.) is the most extensively cultivated forage legume in the world, while its yield and growing range are limited by abiotic stress. Physiology, genetic and molecular level studies have revealed complex regulatory processes that coordinate stress adaptation and tolerance, which are integrated at various levels. This review summarizes the current body of knowledge regarding the physiological, transcriptional, metabolic and genetic/epigenetic basis of the alfalfa stress response, along with the key signal transduction processes involved. Gathering the knowledge together will contribute to a more systematic understanding of the adaption to stress response in alfalfa, as well as provide guidance towards improving the crop through breeding.

Abstract:To adapt to fluctuations in light intensity, temperatures, and water/nutrient availability, plants have to dynamically and flexibly adjust their metabolism and growth processes. Specifically, hormone synthesis and signal transduction promptly respond to environmental stimuli. Understanding the mechanisms by which plants adapt to their environment is a hot spot of plant biology research. In this editorial, we provide a summary of recent advances in the roles and underlying mechanisms of abscisic acid (ABA) in coping with various stresses. Additionally, we highlight the functions of newly discovered hormones such as strigolactone. Furthermore, we emphasize future research directions aimed at gaining a deeper understanding of how plants maintain cellular homeostasis with the goal of improving crop yield in the background of global change.

Abstract:Climate change poses significant challenges to global agriculture, with rising temperatures, altered precipitation patterns, and increased frequency of extreme weather events threatening crop yields. These changes exceed the adaptability thresholds of many crops, decreasing their yield and threatening food security. At plant physiological levels, climate change‐induced stressors disrupt photosynthesis, growth, and reproductive processes, contributing to a reduced productivity. Furthermore, the negative impacts of climate change on agriculture are exacerbated by anthropogenic factors, with agriculture itself contributing significantly to greenhouse gas emissions. To mitigate these challenges, various approaches have been explored. This work reviews the most important physical, chemical, and biological strategies most commonly used in a broad range of agricultural crops. Among physical strategies, increasing water use efficiency without yield reduction through different irrigation strategies, and the use of foliar treatments with reflective properties to mitigate the negative effects of different stresses have been proven to be effective. Concerning chemical approaches, the exogenous treatment of plants with chemicals induces existing molecular and physiological plant defense mechanisms, enhancing abiotic stress tolerance. Regarding biological treatments, plant inoculation with mycorrhiza and plant growth‐promoting rhizobacteria (PGPR) can improve enzymatic antioxidant capacity and mineral solubilization, favoring root and plant growth and enhance plant performance under stressful conditions. While these strategies provide valuable short‐ to medium‐term solutions, there is a pressing need for new biotechnological approaches aimed at developing genotypes resistant to stressful conditions. Collaborative efforts among researchers, policymakers, and agricultural stakeholders are essential to ensure global food security in the face of ongoing climate challenges.

Abstract:Sustaining grassland production in a changing climate requires an understanding of plant adaptation strategies, including trait plasticity under warmer and drier conditions. However, our knowledge to date disproportionately relies on aboveground responses, despite the importance of belowground traits in maintaining aboveground growth, especially in grazed systems. We subjected a perennial pasture grass, Festuca arundinacea, to year-round warming (+3 °C) and cool-season drought (60% rainfall reduction) in a factorial field experiment to test the hypotheses that: (i) drought and warming increase carbon allocation belowground and shift root traits towards greater resource acquisition and (ii) increased belowground carbon reserves support post-drought aboveground recovery. Drought and warming reduced plant production and biomass allocation belowground. Drought increased specific root length and reduced root diameter in warmed plots but increased root starch concentrations under ambient temperature. Higher diameter and soluble sugar concentrations of roots and starch storage in crowns explained aboveground production under climate extremes. However, the lack of association between post-drought aboveground biomass and belowground carbon and nitrogen reserves contrasted with our predictions. These findings demonstrate that root trait plasticity and belowground carbon reserves play a key role in aboveground production during climate stress, helping predict pasture responses and inform management decisions under future climates.
Phyllosphere Community Assembly and Response to Drought Stress on Common Tropical and Temperate Forage Grasses

Abstract:Grasslands represent a critical ecosystem important for global food production, soil carbon storage, and water regulation. Current intensification and expansion practices add to the degradation of grasslands and dramatically increase greenhouse gas emissions and pollution. Thus, new ways to sustain and improve their productivity are needed. Research efforts focus on the plant-leaf microbiome, or phyllosphere, because its microbial members impact ecosystem function by influencing pathogen resistance, plant hormone production, and nutrient availability through processes including nitrogen fixation. However, little is known about grassland phyllospheres and their response to environmental stress. In this study, globally dominant temperate and tropical forage grass species were grown in a greenhouse under current climate conditions and drought conditions that mimic future climate predictions to understand if (i) plant host taxa influence microbial community assembly, (ii) microbial communities respond to drought stress, and (iii) phyllosphere community changes correlate to changes in plant host traits and stress-response strategies. Community analysis using high-resolution sequencing revealed Gammaproteobacteria as the dominant bacterial class, which increased under severe drought stress on both temperate and tropical grasses while overall bacterial community diversity declined. Bacterial community diversity, structure, and response to drought were significantly different between grass species. This community dependence on plant host species correlated with differences in grass species traits, which became more defined under drought stress conditions, suggesting symbiotic evolutionary relationships between plant hosts and their associated microbial community. Further understanding these strategies and the functions microbes provide to plants will help us utilize microbes to promote agricultural and ecosystem productivity in the future. IMPORTANCE Globally important grassland ecosystems are at risk of degradation due to poor management practices compounded by predicted increases in severity and duration of drought over the next century. Finding new ways to support grassland productivity is critical to maintaining their ecological and agricultural benefits. Discerning how grassland microbial communities change in response to climate stress will help us understand how plant-microbe relationships may be useful to sustainably support grasslands in the future. In this study, phyllosphere community diversity and composition were significantly altered under drought conditions. The significance of our research is demonstrating how severe climate stress reduces bacterial community diversity, which previously was directly associated with decreased plant productivity. These findings guide future questions about functional plant-microbe interactions under stress conditions, greatly enhancing our understanding of how bacteria can increase food security by promoting grassland growth and resilience.

Abstract:Drought stress is one of the most critical abiotic factors which negatively impacts on growth, productivity, and survival of plants. Grass species have an important role in the sustainable intensification of cropping systems. This review focus on the specific drought tolerance characteristics in grass species and application of prevalent classical and molecular methods for genetic improvement of them to drought stress. Generally, grass species adapt to drought stress by utilizing more than one strategy including of changes in the root growth, photosynthetic pigments, activation of antioxidant enzymes, and accumulation of compatible osmolytes. They also have other specific characteristics consisted of summer dormancy, drought recovery, and persistence, which lead to drought adaptation after prolonged drought. Studies on different grasses, indicated that most of above mentioned traits usually have positive correlation with drought tolerance. Also, high heritability has been reported for most of them in different grasses. Therefore, an effective index might be considering in identification of drought tolerance genotypes. Recently, high-throughput imaging phenotyping and advanced molecular techniques such as genotyping-by-sequencing (GBS), RNA sequencing, genome-wide association study, and genome editing help conventional breeding methods to increase the accuracy, selection efficiency, genetic gains, and speed of breeding programs for developing drought tolerant cultivars.

Abstract:Long-term climate change and periodic environmental extremes threaten food and fuel security1 and global crop productivity2-4. Although molecular and adaptive breeding strategies can buffer the effects of climatic stress and improve crop resilience5, these approaches require sufficient knowledge of the genes that underlie productivity and adaptation6-knowledge that has been limited to a small number of well-studied model systems. Here we present the assembly and annotation of the large and complex genome of the polyploid bioenergy crop switchgrass (Panicum virgatum). Analysis of biomass and survival among 732 resequenced genotypes, which were grown across 10 common gardens that span 1,800 km of latitude, jointly revealed extensive genomic evidence of climate adaptation. Climate-gene-biomass associations were abundant but varied considerably among deeply diverged gene pools. Furthermore, we found that gene flow accelerated climate adaptation during the postglacial colonization of northern habitats through introgression of alleles from a pre-adapted northern gene pool. The polyploid nature of switchgrass also enhanced adaptive potential through the fractionation of gene function, as there was an increased level of heritable genetic diversity on the nondominant subgenome. In addition to investigating patterns of climate adaptation, the genome resources and gene-trait associations developed here provide breeders with the necessary tools to increase switchgrass yield for the sustainable production of bioenergy.

Abstract:Abiotic stresses, such as low or high temperature, deficient or excessive water, high salinity, heavy metals, and ultraviolet radiation, are hostile to plant growth and development, leading to great crop yield penalty worldwide. It is getting imperative to equip crops with multistress tolerance to relieve the pressure of environmental changes and to meet the demand of population growth, as different abiotic stresses usually arise together in the field. The feasibility is raised as land plants actually have established more generalized defenses against abiotic stresses, including the cuticle outside plants, together with unsaturated fatty acids, reactive species scavengers, molecular chaperones, and compatible solutes inside cells. In stress response, they are orchestrated by a complex regulatory network involving upstream signaling molecules including stress hormones, reactive oxygen species, gasotransmitters, polyamines, phytochromes, and calcium, as well as downstream gene regulation factors, particularly transcription factors. In this review, we aimed at presenting an overview of these defensive systems and the regulatory network, with an eye to their practical potential via genetic engineering and/or exogenous application.

Abstract:Plant responses to abiotic stresses during vegetative growth have been extensively studied for many years. Daily environmental fluctuations can have dramatic effects on plant vegetative growth at multiple levels, resulting in molecular, cellular, physiological, and morphological changes. Plants are even more sensitive to environmental changes during reproductive stages. However, much less is known about how plants respond to abiotic stresses during reproduction. Fortunately, recent advances in this field have begun to provide clues about these important processes, which promise further understanding and a potential contribution to maximize crop yield under adverse environments. Here we summarize information from several plants, focusing on the possible mechanisms that plants use to cope with different types of abiotic stresses during reproductive development, and present a tentative molecular portrait of plant acclimation during reproductive stages. Additionally, we discuss strategies that plants use to balance between survival and productivity, with some comparison among different plants that have adapted to distinct environments.
Growth-Defense Trade-Offs Induced by Long-term Overgrazing Could Act as a Stress Memory

Abstract:Long-term overgrazing (OG) is one of the key drivers of global grassland degradation with severe loss of productivity and ecosystem functions, which may result in stress memory such as smaller stature of grassland plants. However, how the OG-induced stress memory could be regulated by phytohormones is unknown. In this study, we investigated the changes of four phytohormones of cloned offspring of Leymus chinensis that were developed from no-grazing (NG) plants and OG plants with a grazing history of 30 years. The concentrations of auxin (IAA) and gibberellic acid (GA) in OG plant leaves were 45% and 20% lower than control, respectively. Meanwhile, the level of abscisic acid (ABA) in OG leaves nearly doubled compared with that in NG leaves. The situation was quite similar in roots. Unexpectedly, no significant changes in the jasmonic acid (JA) level were observed between OG and NG plants. The changes in gene expression patterns between OG and NG plants were also investigated by transcriptomic analysis. In total, 302 differentially expressed genes (DEGs) were identified between OG and NG plants, which were mainly classified into the functions of synthesis, receptor, and signal transduction processes of phytohormones. The expression of 24 key genes related to the biosynthesis and signal transduction of IAA and GA was downregulated in OG plants. Among them, OASA1 and AO1 (regulating the biosynthesis of IAA and ABA, respectively) were reduced significantly by 88 and 92%, respectively. In addition, the content of secondary metabolites related to plant defense such as flavonoids and phenols was also increased in leaves. Taken together, the decrease of positive plant growth-related hormones (IAA and GA) together with the increase of plant stress-related hormones or factors (ABA, flavonoids, and phenols) induced the growth-defense trade-offs for L. chinensis adaptation to long-term OG stress. The findings reported in this study shed new light on the mechanism of plant-animal interaction in the grassland ecosystem and provide a deeper insight into optimizing grazing management and sustainable utilization of grassland.

Abstract:Abiotic stress is the adverse effect of any abiotic factor on a plant in a given environment, impacting plants’ growth and development. These stress factors, such as drought, salinity, and extreme temperatures, are often interrelated or in conjunction with each other. Plants have evolved mechanisms to sense these environmental challenges and make adjustments to their growth in order to survive and reproduce. In this review, we summarized recent studies on plant stress sensing and its regulatory mechanism, emphasizing signal transduction and regulation at multiple levels. Then we presented several strategies to improve plant growth under stress based on current progress. Finally, we discussed the implications of research on plant response to abiotic stresses for high-yielding crops and agricultural sustainability. Studying stress signaling and regulation is critical to understand abiotic stress responses in plants to generate stress-resistant crops and improve agricultural sustainability.

Abstract:As global climate change intensifies, the occurrence and severity of various abiotic stresses will significantly threaten plant health and productivity. Drought stress (DS) is a formidable obstacle, disrupting normal plant functions through specific morphological, physiological, biochemical, and molecular mechanisms. Understanding how plants navigate DS is paramount to mitigating its adverse effects. In response to DS, plants synthesize or accumulate various plant growth regulators (PGRs), including phytohormones, neurotransmitters, gasotransmitters, and polyamines, which present promising sustainable green chemical strategies to adapt or tolerate stress conditions. These PGRs orchestrate crucial plant structure and function adjustments, activating defense systems and modulating cellular‐level responses, transcript levels, transcription factors, metabolic genes, and stress‐responsive candidate proteins. However, the efficacy of these molecules in mitigating DS depends on the plant species, applied PGR dose, treatment type, duration of DS exposure, and growth stages. Thus, exploring the integrated impact of PGRs on enhancing plant fitness and DS tolerance is crucial for global food security and sustainable agriculture. This review investigates plant responses to DS, explains the potential of exogenously applied diverse PGRs, dissects the complex chemistry among PGRs, and sheds light on omics approaches for harnessing the molecular basis of DS tolerance. This updated review delivers comprehensive mechanistic insights for leveraging various PGRs to enhance overall plant fitness under DS conditions.

Abstract:Abiotic stresses such as drought, salinity, and extreme temperatures pose significant challenges to plant growth, development, and productivity. Understanding the physiological mechanisms underlying plant adaptation to these stresses is crucial for developing strategies to enhance crop resilience and ensure food security. This review explores the key physiological responses of plants to abiotic stress, including stress perception and signaling, osmotic adjustment, antioxidant defense, and hormonal regulation. We highlight recent advances in our understanding of these mechanisms and discuss their implications for crop improvement. Additionally, we examine the role of genetic and epigenetic modifications in enhancing stress tolerance and the potential of biotechnological approaches to develop stress-resistant crop varieties. By integrating physiological insights with modern breeding and biotechnological techniques, we aim to provide a comprehensive overview of the current state of knowledge and future directions in plant adaptation to abiotic stress.
Stoichiometric ratios support plant adaption to grazing moderated by soil nutrients and root enzymes

Abstract:Background: Vegetation succession is one of the major driving processes of grassland degradation. Stoichiometry significantly contributes to vegetation dynamics. However, a knowledge gap exists in how soil nutrients and root enzymes influence the stoichiometric ratio to affect vegetation dynamics. Methods: To address these questions, we selected a dominant species (Leymus chinensis (Trin.) Tzvel.) and a degraded-dominant species (Artemisia frigida Willd.) under different management regimes (enclosure and grazing) on the Inner Mongolia steppe. We measured (i) plant nutrient concentrations, (ii) root enzymes and (iii) soil nutrients to investigate how the selected plant species responded to grazing. Results: The results show that: (i) N and P concentrations and the C:N:P ratio in different organs are significantly affected by grazing, and there is variation in the plant species' response. Grazing significantly increased N and P in the leaves and stems of L. chinensis and the stems and roots of A. frigida. (ii) Grazing significantly increased the activities of glutamine synthase but decreased the activities of acid phosphatase in L. chinensis. The nitrate reductase and acid phosphatase activities significantly increased in A. frigida under grazing conditions. (iii) Grazing decreased the total nitrogen, total phosphorus, and available nitrogen, but increased the available phosphorus in the soil. Conclusion: We conclude that A. frigida is better adapted to grazing than L. chinensis, possibly because of its relatively increased stem and root growth, which enhance population expansion following grazing. Conversely, L. chinensis showed increased leaf and stem growth, but suffered nutrient and biomass loss as a result of excessive foraging by livestock, which severely affected its ability to colonize. Root enzymes coupled with soil nutrients can regulate plant nutrients and stoichiometric ratios as an adaptive response to grazing. Thus, we demonstrated that stoichiometric ratios allow species to better withstand grazing disturbances. This study provides a new understanding of the mechanisms involved in grazing-resistance within a plant-soil system.

Abstract:<p>The frequency and severity of global abiotic stresses such as heat, drought, and salt stress are increasing due to climate changes. Objectives of this study were to investigate effects of γ-aminobutyric acid (GABA) priming on inducing plants' acclimation to abiotic stress associated with alterations of endogenous polyamines (PAs), amino acids, and sugars in creeping bentgrass (<em>Agrostis stolonifera</em>). The pretreatment with GABA fertigation significantly alleviated heat-, drought-, and salt-induced declines in leaf relative water content, chlorophyll content, cell membrane stability, photochemical efficiency (Fv/Fm), and performance index on absorption basis (PIABS), and also further decreased stress-caused decline in osmotic potential in leaves. The GABA priming uniformly increased total PAs, spermidine, amino acids involved in GABA shunt (GABA, glutamic acid, and alanine), and other amino acids (phenylalanine, aspartic acid, and glycine) accumulation under heat, drought, and salt stress. The GABA priming also significantly improved methionine content under heat and drought stress, maltose, galactose, and talose content under heat and salt stress, or cysteine, serine, and threonine content under drought and salt stress. Interestingly, the GABA priming uniquely led to significant accumulation of spermine, fructose, and glucose under heat stress, putrescine, proline, and mannose under drought stress, or arginine, trehalose and xylose under salt stress, respectively. These particular PAs, sugars, and amino acids differentially or commonly regulated by GABA could play critical roles in osmotic adjustment, osmoprotection, antioxidant, energy source, and signal molecular for creeping bentgrass to acclimate diverse abiotic stresses.</p>
