Abstract:Objective. X-ray phase contrast imaging is a promising technique for future clinical diagnostic as it can provide enhanced contrast in soft tissues compared to traditional x-ray attenuation-contrast imaging. However, the strict requirements on the x-ray coherence and the precise alignment of optical elements limit its applications towards clinical use. To solve this problem, mesh-based x-ray phase contrast imaging method with one hexagonal mesh is proposed for easy alignment and better image visualization.Approach. The mesh produces structured illuminations and the detector captures its distortions to reconstruct the absorption, differential phase contrast (DPC) and dark-field (DF) images of the sample. In this work, we fabricated a hexagonal mesh to simultaneously retrieve DPC and DF signals in three different directions with single shot. A phase retrieval algorithm to obtain artifacts-free phase from DPC images with three different directions is put forward and false color dark-field image is also reconstructed with tri-directional images. Mesh-shifting method based on this hexagonal mesh modulator is also proposed to reconstruct images with better image quality at the expense of increased dose.Main results. In numerical simulations, the proposed hexagonal mesh outperforms the traditional square mesh in image evaluation metrics performance and false color visualization with the same radiation dose. The experimental results demonstrate its feasiblity in real imaging systems and its advantages in quantitive imaging and better visualization. The proposed hexagonal mesh is easy to fabricate and can be successfully applied to x-ray source with it spot size up to 300μm.Significance. This work opens new possibilities for quantitative x-ray non-destructive imaging and may also be instructive for research fields such as x-ray structured illumination microscopy (SIM), x-ray spectral imaging and x-ray phase contrast and dark-field computed tomography (CT).

Abstract:In the last several decades, there has been a tremendous wave of interest in the relationship between the behavior of soft material and their structure. In this research, we demonstrate a new proposal to enhance the contrast of soft material in their native state, named hollow cone dark field (HCDF) environmental electron microscope. HCDF imaging is in fact the reciprocal analogue of HAADF in which case the object is illuminated with a plane wave that is tilted over a fixed angle to image the thermal diffuse scattering electron as the Z- contrast image where Z stands for atomic number. In addition, to understand the native state of soft material, a special craft holder was made for this purpose, which is comprised of two parts: 1. self-aligned wet cell chips with spacer to control liquid thickness, and also provide appropriate space for soft material, and 2. liquid holder to exchange metabolic wastes for culture solution 1 . We select the magnetotactic bacteria with magnetosomes. Figure a, b show the bright field and HCDF imaging mode of magnetotactic bacteria in native state, respectively. There is a dramatic increase in contrast of the HCDF image; the feature of magnetotactic bacteria is more clearly visible. We also quantitatively analyze the contrast of both imaging mode, and the HCDF shows the 4 times higher than bright field mode. In light of this concerns, the HCDF environmental EM can become a much needed tool in future research of soft material.
Full-fieldin Vivoimaging of Nanoparticles Using Benchtop Cone-Beam XFCT System with Pixelated Photon Counting Detector.

Abstract:Objective.X-ray fluorescence computed tomography (XFCT) is a promising noninvasive technique forin vivoimaging of high-Z elements (e.g. gadolinium (Gd) or gold (Au)). In this study we upgraded our experimental XFCT system using a flat panel photon counting detector with redesigned pinhole collimation in order to achieve 3D XFCT images during one scan.Approach.Aiming at the characteristics of pinhole-collimated cone-beam XFCT imaging, a new scatter correction algorithm was proposed to estimate the normalized spectrum of scatter background based on K-N formula and realize correction by a weighted least squares method. Then, images were quantitatively reconstructed by a maximum likelihood iterative algorithm with the attenuation correction.Main results.The potential on full-fieldin vivoXFCT imaging of this new system was investigated. An imaging experiment of a PMMA phantom with the diameter of 35 mm was carried out for quantitative evaluation of the system performance. Results show that 2 mg ml-1Gd solutions can be successfully reconstructed with a 45 min cone-beam XFCT scan.In vivoXFCT imaging experiments of mice with injection of Gd nanoparticles (GdNPs) were also performed and demonstrated in this paper. A mouse was injected through the tail vein with 20 mg ml-1NaGdF4 solution and then anesthetized with isoflurane during the cone-beam XFCT scan.Significance.The distribution of the GdNPs inside the mouse can be well reconstructed so that the deposition of NPsin vivocan be clearly observed, which indicates the feasibility of the proposed system for full-field XFCT of small animals and further potential in relevantin vivoresearch.

Abstract:The contradiction between high resolution and large depth of field (DOF) of microscopic imaging limits its application in imaging 3D distributed samples. Here, we propose a new technique to achieve extended depth of field reconstruction in bright-field microscopy through complex field estimation. The complex field is estimated by a phase retrieval algorithm with the four illumination modulation patterns and the corresponding captured Fourier intensities as dual constraints. Then, the digital refocusing strategy based on Fresnel propagator is integrated into reconstruction framework to achieve the reconstruction of high-resolution complex-valued information for each part of the sample, which ensures an effective extension of the DOF. The feasibility of the proposed method to maintain high resolution over extended DOF was verified in actual experiments with a tilted USAF target, biological sample and microlens array (MLA). The extended DOF reconstruction provides a robust method for imaging in both industrial inspection and biomedical research.

Abstract:A theoretical model of digital inline holography system reveals that the local focus metric curves (FMCs) of different parts of an irregular micro-object present spatial shift in the depth direction which is resulted from the depth shift. Thus, the 3D morphology of an irregular micro-object can be accurately measured using the cross correlation of the local FMCs. This method retrieves the 3D depth information directly, avoiding the uncertainty inherited from the depth position determination. Typical 3D morphology measurements, including the 3D boundary lines of tilted carbon fibers and irregular coal particles, and the 3D swimming gesture of a live Caenorhabdities elegans, are presented.
High-resolution Dark-Field Confocal Microscopy Based on Radially Polarized Illumination.

Abstract:Dark-field confocal microscopy (DFCM) facilitates the 3D detection and localization of surface and subsurface defects in high-precision optical components. The spatial resolution of conventional DFCM is commonly undermined owing to complementary aperture detection. We employed a radially polarized (RP) beam for illumination in DFCM. The RP beam creates a sub-diffraction-sized longitudinal optical component after being focused and effectively enhances the lateral resolution by 30.33% from 610 nm to 425 nm. The resolution improvement was verified by imaging a 2D sample containing sparsely distributed gold nanorods along with a 3D neodymium glass containing surface and subsurface defects.

Abstract:A new method for dark field imaging is introduced which uses scanned electron diffraction (or 4DSTEM - 4-dimensional scanning transmission electron microscopy) datasets as its input. Instead of working on simple summation of intensity, it works on a sparse representation of the diffraction patterns in terms of a list of their diffraction peaks. This is tested on a thin perovskite film containing structural ordering resulting in additional superlattice spots that reveal details of domain structures, and is shown to give much better selectivity and contrast than conventional virtual dark field imaging. It is also shown to work well in polycrystalline aggregates of CuO nanoparticles. In view of the higher contrast and selectivity, and the complete exclusion of diffuse scattering from the image formation, it is expected to be of significant benefit for characterisation of a wide variety of crystalline materials.

Abstract:Recent developments in x-ray science provide methods to probe deeply embedded mesoscale grain structures and spatially resolve them using dark field x-ray microscopy (DFXM). Extending this technique to investigate weak diffraction signals such as magnetic systems, quantum materials and thin films proves challenging due to available detection methods and incident x-ray flux at the sample. We present a direct detection method focusing on DFXM studies in the hard x-ray range of 10s of keV and above capable of approaching nanoscale resolution. Additionally, we compare this direct detection scheme with routinely used scintillator based optical detection and achieve an order of magnitude improvement in exposure times allowing for imaging of weakly diffracting ordered systems.

Compact Dark-Field Confocal Microscopy Based on an Annular Beam with Orbital Angular Momentum

Abstract:This study proposes a dark-field confocal microscopy (DFCM) based on fiber-mode excitation-assisted orbital angular momentum annular beam generation with a two-mode fiber to avoid diffraction distortion. The application of optical fibers compresses the DFCM volume, providing new ideas for system miniaturization. System adjustment difficulty is reduced and feasibility is verified by imaging 2D and 3D samples. High imaging contrast is achieved by fully blocking the reflected light and annular illumination with a steady central dark-spot within a propagation distance of 3 m. The application of our scheme can be further extended to detect subsurface defects in optical components and high-contrast biological imaging.

Abstract:We demonstrate the capability of laboratory-based x-ray microscopes, using intensity-modulation masks, to access the sub-micron length scale in the dark field contrast channel while maintaining micron resolution in the resolved (refraction and attenuation) channels. The dark field contrast channel reveals the presence of ensembles of samples' features below the system resolution. Resolved refraction and attenuation channels provide multi-modal high-resolution imaging down to the micron scale. We investigate the regimes of modulated and un-modulated dark field as well as refraction, quantifying their dependence on the relationship between feature size in the imaged object and aperture size in the intensity-modulation mask. We propose an analytical model to link the measured signal with the sample's microscopic properties. Finally, we demonstrate the relevance of the microscopic dark field contrast channel in applications from both the life and physical sciences, providing proof of concept results for imaging collagen bundles in cartilage and dendritic growth in lithium batteries.

Abstract:This paper describes a new approach of quantification of annular-dark-field or Z-contrast image intensity as a function of inner acceptance angle of the detector in a scanning transmission electron microscope. By using size-selected nanoclusters of Pd (Z = 46) and Au (Z = 79), it is shown experimentally that the exponent in the power law I similar to Z(alpha) varies strongly between 1.2 and 1.8 as the collection angle changes from 14 to 103 mrad. The result is discussed in line with existing theoretical models. Factors, such as cluster size, structure, and orientation as well as the detector geometry, are also discussed for potential use of the work.
Spatially Resolved Spectroscopic Characterization of Nanostructured Films by Hyperspectral Dark-Field Microscopy

Abstract:Nanostructured films have been widely used for preparing various advanced thin-film devices because of their unique electrical, optical, and plasmonic characteristics associated with the nano-size effect. In situ, nondestructive and high-resolution characterization of nanostructured films is essential for optimizing thin-film device performance. In this work, such thin-film characterization was achieved using a hyperspectral dark-field microscope (HSDFM) that was constructed in our laboratory by integrating a hyperspectral imager with a commercial microscope. The HSDFM allows for high-resolution (Δλ = 0.4 nm) spectral analysis of nanostructured samples in the visible-near-infrared region with a spatial resolution as high as 45 nm × 45 nm (corresponding to a single pixel). Four typical samples were investigated with the HSDFM, including the gold nanoplate array, the self-assembled gold nanoparticle (GNP) sub-monolayer, the sol-gel nanoporous titanium dioxide (TiO2) film, and the layer-stacked molybdenum disulfide (MoS2) sheet. According to the experimental results, the plasmon resonance scattering bands for nanoplate clusters are identical with those for individual gold nanoplates, indicating that the gap between adjacent nanoplates is too large to allow plasmonic coupling between them. A different case was observed with the self-assembled GNP sub-monolayer in which the aggregated clusters with the internal plasmonic interaction show a considerable red-shift of the plasmon resonance band relative to the isolated single GNP. In addition, the protein adsorption on the nanoporous TiO2 film was observed to be inhomogeneous on the microscale, and the stepped boundaries of the MoS2 sheet were clearly observed. A quasi-linear dependence of the single-pixel light intensity on the step height was obtained by combining the HSDFM with atomic force microscopy. The minimum thickness detectable by the present HSDFM is 6.5 nm, corresponding to the 10-layer MoS2 film. The work demonstrated the outstanding applicability of the HSDFM for nanostructured film characterization.

Abstract:We report hollow-cone imaging and small-angle electron diffraction (SmAED) of nanoscale magnetictextures such as magnetic-striped domains and magnetic bubbles of M-type hexagonal ferrite BaFe10.35 Sc 1.6 Mg 0.05 O 19 . The advantage of the hollow-cone Foucault (HCF) method is that magneticdomains with various directions of magnetization can be visualized under an infocus condition.Moreover, the contrast of magnetic domain walls in magnetic bubbles depends on the inclination angleof the illumination beam. The combination of SmAED and HCF imaging proves that magnetization atdomain walls exhibits in-plane directions in the magnetic-striped domains and magnetic bubbles.

Abstract:Common path DHM systems are the most robust DHM systems as they are based on self-interference and are thus less prone to external fluctuations. A common issue amongst these DHM systems is that the two replicas of the sample's information overlay due to self-interference, making them only suitable for imaging sparse samples. This overlay has restricted the use of common-path DHM systems in material science. The overlay can be overcome by limiting the sample's field of view to occupy only half of the imaging field of view or by using an optical spatial filter. In this work, we have implemented optical spatial filtering in a common-path DHM system using a Fresnel biprism. We have analyzed the optimal pinhole size by evaluating the frequency content of the reconstructed phase images of a star target. We have also measured the accuracy of the system and the sensitivity to noise for different pinhole sizes. Finally, we have proposed the first dual-mode common-path DHM system using a Fresnel biprism. The performance of the dual-model DHM system has been evaluated experimentally using transmissive and reflective microscopic samples.
