Abstract:Reactions of ketenes generated from alpha-diazoketones with a series of acyclic and cyclic imines were investigated under both microwave and photoirradiation conditions. The results indicate that the zwitterionic azabutadiene-type intermediates yielded from imines and ketenes undergo a conrotatory ring closure exclusively to produce beta-lactams. It is notable that no Woodward-Hoffmann product was found under the ultraviolet irradiation. The photoirradiation-induced Staudinger reaction shows a different stereo selectivity from the electrocyclic reaction of substituted 1,3-butadiene.

Abstract:Under regular heating or microwave irradiation, 3-alkyl substituted imidazo[1,5-a]pyridines were synthesized from 2,2'-pyridil or di-2-pyridyl ketone and aliphatic aldehydes in the presence of ammonium acetate and acetic acid. Compared to the traditional heating condition, 3-alkyl imidazo[1,5-a]pyridines were obtained with higher yields and shorter reaction time under microwave irradiation. All products were characterized by their H-1 NMR, MS spectra and elemental analysis.

Abstract:Controlling diastereoselectivity is a challenging issue in the Staudinger reaction. The influence of ultraviolet irradiation on the stereoselectivity in the Staudinger reaction has been investigated. The results indicate that ultraviolet irradiation is one of the most important means to regulate the diastereoselectivity of the products in the Staudinger reaction, whatever ketenes were generated from diazomethyl ketones or from substituted acetyl chlorides in the presence of triethylamine. However, the regulation is dependent on the ketene substituents obviously. Ultraviolet irradiation can even reverse the diastereoselectivity for the reactions involving ketenes without strong electron-donating substituents.

Abstract:The effect of microwave irradiation on the selectivity, especially stereoselectivity, is one of the most important issues in microwave-assisted organic reactions. The diastereoselectivity in Staudinger reactions involving the representative ketenes and the corresponding matched imines has been used as a probe to investigate carefully the existence of the specific nonthermal microwave effects. The results indicate that the microwave irradiation-controlled stereoselectivity in the Staudinger reaction is in fact the contribution of temperature. No specific nonthermal microwave effect was found in the Staudinger reaction.
Photo-irradiated E/Z Isomerization Reaction of Star-Shaped Isomers Containing Two Cyanostilbene Arms with Charge Transfer Excited States.

Abstract:The E/Z isomerization reaction of the multi-cyanostilbene molecule is still not clear. Herein, we have designed and synthesized three star-shaped molecular isomers with a triphenylamine core linked to two cyanostilbene groups with E/Z isomerization, Z,Z-TPDCF, Z,E-TPDCF and E,E-TPDCF, possessing three different isomeric molecular configurations, to investigate the specific E/Z isomerization reaction of the cyanostilbene groups in the two molecular arms. The in situ UV, 1H NMR and HPLC spectra under UV-irradiation clearly showed that the E/Z isomerization reactions of both E,E-TPDCF and Z,Z-TPDCF firstly turned them into Z,E-TPDCF, and the Z,E-TPDCF was almost simultaneously turned into more E,E-TPDCF and less Z,Z-TPDCF due to the calculated lowest unoccupied molecular orbitals of Z,E-TPDCF on the cyanostilbene arm with the Z-configuration. In general, Z,E-TPDCF exhibited a relatively better configurational stability than Z,Z-TPDCF or E,E-TPDCF under the photo-irradiation conditions. Further research demonstrated that all three isomers exhibited excellent aggregation-induced emission (AIE) properties.
Effect of Microwave Irradiation on the Syn/Trans Selectivity in the Formation of Β- Lactams from Ketenes and Imines

Abstract:To study the effect of the microwave irradiation on the syn/trans selectivity in the formation of β-lactams from ketenes and imines,the syn/trans ratios of β-lactams generated from imines and ketenes yielded from S-phenyl 2-diazoethanethioate under classical heating and microwave irradiation conditions at the same temperature were determined via NMR.The effect of the microwave irradiation on the syn/trans selectivity in this reaction was investigated.The results indicate that the microwave irradiation affects the selectivity slightly.Cis products increase in nonpolar toluene and trans products increase in polar acetonitrile.

Abstract:The stereoselectivity of the Staudinger reactions involving monosubstituted ketenes with electron acceptor substituents was investigated experimentally by determination of the product stereochemistry and theoretically via DFT calculations. The results indicate that imines preferentially attack the less sterically hindered exo-side of the ketenes to generate zwitterionic intermediates. Subsequently, for cyclic imines, the intermediates undergo a conrotatory ring closure directly to produce beta-lactams, while for linear imines, the imine moiety of the intermediates isomerizes to more stable intermediates, which further undergo a conrotatory ring closure to afford trans-beta-lactams. The steric hindrance and the isomerization, rather than the torquoelectronic effect, play crucial roles in controlling the stereoselectivity in the practical Staudinger reactions involving monosubstituted ketenes with electron acceptor substituents, although the unaccessible borylketene with a powerful electron acceptor group controls the stereoselectivity torquoelectronically, in theory.

Abstract:A novel structural reorganization of cycloketoxime esters beyond the traditional Beckmann rearrangement process has been established to build cyano-containing ketones in the presence of photocatalyst. This novel transformation is remarkable with selective CC bond cleavage and an oxidation process enabled by DMSO used as the solvent, oxidant, and oxygen source avoiding acid, base and toxic cyanide salts as the cyano source. Further applications in late-stage modification of complex and chiral molecules have also been reported.

Abstract:A number of cyclic mono- and di-ketones underwent regioselective olefination with (carbethoxyethylidene)triphenylphospharane under controlled microwave heating. The Wittig reaction of 4-substituted cyclohexanones or 1,2- and 1,4-cyclohexanediones with the ylide at 190°C for 20min in MeCN afforded the exocyclic olefins in >94:6 isomer ratios. On the other hand, the same reactions carried out at 230°C for 20min in the presence of 20mol% DBU furnished the endocyclic olefins in >83:17 isomer ratios. The base-mediated isomerization of the exocyclic olefins into the endocyclic isomers was primarily driven by thermodynamic stability of the products and the effect of ring structures on deconjugation was examined.

Abstract:Photocycloaddition, along with subsequent transformation of the photocycloadducts, provides expeditious ways to construct various structures. The photo-induced reactions of α-diketones have been reported to proceed via different reaction pathways with the involvement of one or two of the carbonyl groups. Photoinduced reactions of cyclic α-diketones including N-acetylisatin, phenanthrenequinone and isoquinolinetrione with different C=C containing compounds could take place via [2 + 2], [4 + 2] or [4 + 4] photocycloaddition pathways. We have investigated the photoreactions of these cyclic α-diketones with different types of alkenes and alkynes, with a focus on the unusual cascade reactions initiated by the photocycloaddition reactions of these cyclic α-diketones and the applications of these photocycloaddition reactions along with the transformation of the photocycloadducts. In this paper, we discuss the diverse photo-cycloaddition pathways found in the photocycloaddition of o-diones leading to various photocycloadducts and the potential applications of these reactions via further transformation reactions of the adducts.

Abstract:Chemoselective annulations of phosphoryl carbenes generated from diazo(aryl)methyl(diaryl)phosphine oxides with pyridinium 1,4-zwitterionic thiolates were performed under microwave irradiation, affording 1-diarylphosphoryl-1H-benzo[c]thiopyran derivatives via [3+3] annulation and indolizine derivatives via ([1+5]-1) annulation with P-Cope elimination as the key step. The annuloselectivity was controlled by the steric hindrance of pyridiniums in pyridinium 1,4-zwitterionic thiolates.

Abstract:Photoinduced reactions of isatin and N-methyl-1,3,4-isoquinolinetrione with bicycloalkylidenes such as bicyclopropylidene, cyclopropylidenecyclobutane, cyclopropylidenecyclohexane and bicyclohexylidene were investigated. The reactions gave spirooxetanes as the major products derived from the [2 + 2] photocycloaddition pathway via 1,4-biradical recombination. Unusual products including the [4 + 2 + 2] cycloadducts, the oxoisochroman derivatives and other ring-rearranged products were derived from competitive pathways via 1,6-biradical recombination. The presence of oxygen in the reaction solution was found to be relevant to the distribution of different types of products. Mechanisms were proposed to rationalize the chemo- and regioselectivity in the photoreactions and the origin of the different types of products.

Abstract:The cycloaddition of ketenes and imines (Staudinger cycloaddition) is a general method for the synthesis of various beta-lactams. However, reactions of imines and ketenes with electron-withdrawing substituents produce a,beta-unsaturated alkenamides, ring-opening products of the intermediates generated from imines and the ketenes, even as sole products, besides the desired beta-lactams. The mechanism of the formation of a,beta-unsaturated alkenamides was investigated. The results indicate that the a,beta-unsaturated alkenamides are generated via a base-induced C?C bond isomerization followed by electrocyclic ring opening of the formed azacyclobutenes (=1,2-dihydroazetes; cf. Scheme 3).

Abstract:Stereochemical models and mechanistic insights are proposed for [2t + 2i + 2i] annulations of thioketenes and imines on the basis of experimental and computational investigations. In the [2t + 2i + 2i] annulations involving cyclic imines, the zwitterionic intermediates generated from monosubstituted thioketenes and the cyclic imines undergo a stepwise nucleophilic endo-addition/Si-face attack pathway with a second imines molecule, giving initially (2,4)-cis-(4,5)-cis-[2t + 2i + 2i] annuladducts, which completely epimerize into the corresponding (2,4)-cis-(4,5)-trans-annuladducts under basic reaction conditions. The annuloselectivity of thio-Staudinger cycloadditions is dependent on the substituents of both thioketenes and imines. The reactivities and annuloselectivities of Staudinger, thio-Staudinger, and sulfa-Staudinger intermediates are compared.
