Abstract:3-Alkoxy/aryloxy-beta-lactams are synthesized in satisfactory to good yields from the reaction of imines and alkyl/aryl diazoacetates under photoirradiation conditions. Typically, trans-beta-lactams are obtained as the major products from linear imines using the current method. By contrast, the corresponding thermal reaction of imines and alkoxy/aryloxyacetyl chlorides, or their equivalents, in the presence of triethylamine affords cis-beta-lactams as the major or exclusive products. The formation of trans-beta-lactams from linear imines is attributed to isomerization of the imines from their trans-isomers into syn-isomers under UV irradiation. The reported method represents a metal-free and neutral approach for the synthesis of 3-alkoxy/aryloxy-beta-lactams.

Abstract:alpha-Diazocarbonyl compounds are good precursors of ketenes in the Staudinger reaction. On the basis of the reactions of S-phenyl diazothioacetate with imines Linder the catalysis of Rh-2(OAc)(4) a method for the synthesis of 3-phenylthio beta-lactain derivatives has been developed previously. In this paper, a more convenient and improved procedure was achieved, which simplified the synthesis of S-phenyl diazothioacetate and made the reaction work well without the expensive Rh-2(OAc)(4) catalyst. The catalyst-free reactions of S-phenyl diazothioacetate with a variety of imines produced monocyclic and polycyclic beta-lactams stereospecifically in good to excellent yields.

Abstract:A photocatalytic methodology has been devised for the stereoselective construction of trisubstituted alkenes incorporating 3,3‐difluoro‐γ‐lactams in 17‐93% yields via a radical cascade process utilizing MBH acetates and N‐allylbromodifluoroacetamides as starting materials. The reaction mechanism involves single‐electron transfer, 5‐exo‐trig cyclization, radical addition, and elimination in a cascade fashion.

Abstract:Quaternary carbons are useful motifs in chemical synthesis but can be challenging to prepare using many chemical methods. Here, we report a stereoselective synthesis of β-quaternary lactams using flavin-dependent 'ene'-reductases via a 5-exo-trig radical cyclization. The products are formed in moderate to good levels of enantioselectivity using an 'ene'-reductase variant from Zymomonas mobilis. This method highlights the opportunity for biocatalysis to form quaternary centers using non-natural reactions.

Abstract:The Staudinger reaction is one of the most important methods in synthesis of beta-lactams. 3-(Thiophen-2-yl)-,beta-lactam is one class of important beta-lactam derivatives. A convenient and new method was developed to synthesize 2-diazo-1-(thiophen-2-yl)-ethanone from 4,4,4-trifluoro-1-(thiophen-2-yl)1,3-butanedione and tosyl azide. A series of 3-(thiophen-2-yl)-beta-lactams were synthesized via the Staudinger reaction of imines and thiophen-2-ylketene generated thermally from 2-diazo-1-(thiophen-2-yl)-ethanone. The stereoselectivity of the Staudinger reaction was also investigated. The results indicate that thiophen-2-ylketene is a more electron-rich Moore ketene than phenylketene and its electronic property locates between those of 4-methoxyphenylketene and 4-methylphenylketene.

Abstract:[reaction: see text] Ketene-imine cycloaddition reactions (the Staudinger reaction) of ethoxycarbonyl(phenylthio)ketene with various imines and subsequent desulfurization reactions were employed to synthesize 3-ethoxycarbonyl beta-lactam derivatives. The results indicate that the current approach provides a convenient, mild, and versatile method for synthesizing a variety of 3-alkoxycarbonyl trans-beta-lactam derivatives with good to excellent yields and diastereoselectivities.

Abstract:A general method for accessing 5-alkyl-5-aryl-gamma-lactams has been developed using readily available aryl bromides, nitroalkanes, and methyl acrylate as the starting materials. The palladium-catalyzed arylation of nitroalkanes gave the 1-aryl-substituted nitroalkanes, which underwent the DBU-mediated Michael addition with methyl acrylate at room temperature to afford the methyl 4-aryl-4-nitroalkanoates. The latter were then subjected to the nitro reduction using NaBH4-NiCl2 center dot 6H(2)O in MeOH at 0 degrees C to furnish, after treatment with aqueous K2CO3 at room temperature, the 5-alkyl-5-aryl-gamma-lactams in good to excellent overall yields. Abstract:Reactions of ketenes generated from alpha-diazoketones with a series of acyclic and cyclic imines were investigated under both microwave and photoirradiation conditions. The results indicate that the zwitterionic azabutadiene-type intermediates yielded from imines and ketenes undergo a conrotatory ring closure exclusively to produce beta-lactams. It is notable that no Woodward-Hoffmann product was found under the ultraviolet irradiation. The photoirradiation-induced Staudinger reaction shows a different stereo selectivity from the electrocyclic reaction of substituted 1,3-butadiene.

Abstract:A facile and convenient anion cascade strategy was developed for the synthesis of bridged-ring amides in moderate to excellent yields in one step in the presence of tBuOK in EtOH under mild conditions, starting from various cheap and commercially available 2-cyanoacetamides and precisely designed straight-chain and annular 1,4-dienones. This simple protocol was generally applicable to a wide range of substrates with high chemical conversion and diastereoselectivity.

Abstract:A rhodium-catalyzed reaction of N-hydroxyanilines with diazo compounds to produce α-imino esters was developed. Distinct from the commonly accepted 1,2-H transfer for normal X–H insertion reactions, density functional theory calculations indicate that this transformation proceeds via a novel rebound hydrolysis mechanism. Furthermore, a three-component reaction was explored to synthesize highly functionalized β-lactams in good yields and diastereoselectivities.

Abstract:A series of trans-3-chloro-beta-lactams was synthesized stereospecifically from imines and chloroacetyl chloride or a mixture of chloroacetyl chloride and nitroacetyl chloride, prepared from vinylidene chloride and a mixture of concentrated nitric acid and sulfuric acid, in the presence of triethylamine. The reaction of vinylidene chloride and the mixed acid was investigated. The formation mechanism of chloroacetyl chloride and nitroacetyl chloride and their reaction process with imines were proposed.

Abstract:We have developed a strategy for constructing nitrogen heterocycles by photoinduced PhSe group transfer radical cyclization. trans-alpha,beta-Disubstituted gamma-butyrolactams (2-pyrrolidinones) were prepared in good yields (38-73%) with high regioselectivity and stereoselectivity from N-alkenyl alpha-PhSe beta-keto amides. Reaction outcomes were modulated by the steric effect of the substituents on the nitrogen atom of the cyclization precursors and the stereoelectronic effect of the substrates. Diphenyldiselenide, as an additive, was found to promote ring closure, The advantage of this strategy in natural product synthesis is demonstrated by a formal synthesis of (+/-)-isocynometrine.
