Abstract:It is shown here for the first time that diethyl azodicarboxylate promotes dehydrogenation of tertiaryamines to afford enamines, which subsequently take place in tandem reactions with sulfonyl azides to give the N-sulfonyl amidine derivatives. A number of different substituted tertiaryamines and sulfonyl azides can successfully be coupled, and several functionalized groups are tolerated in this system. The reaction described here is mild, general, and efficient, thus providing an extremely preferable method for synthesis of a variety of N-sulfonyl amidine derivatives.

Abstract:AbstractIn contrast to the well‐established [3+2] cycloaddition reactions, the catalytic enantioselective [3+n] (n≥3) cycloaddition reaction of activated isocyanides for the preparation of six‐membered or larger ring systems has remained underdeveloped. Herein, we report the first example of highly diastereo‐ and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. By employing silver catalysis, a wide range of biologically important bicyclic 1,2,4‐triazines were obtained in high yields (up to 99 %) with good to excellent stereoselectivities (up to >20 : 1 dr, 99 % ee). In addition, the same catalytic system could be applied to both the late‐stage functionalization of complex bioactive molecules and the kinetic resolution of racemic azomethine imines, further highlighting its versatility and synthetic utility.

Abstract:Pd(OAc)(2)-catalyzed intramolecular dehydrogenative cross-coupling reaction between tertiary enamides, which were derived from the condensation of 2-arylethylamines and methyl o-acetylbenzoate, and arenes enabled synthesis of 7,8-dihydro-5H-benzo[4,5]azepino[2,1-a]-isoindol-5-one derivatives under mild conditions. The synthetic method was applied in the total synthesis of aporhoeadane alkaloids palmanine, lennoxamine, and chilenamine in only three or four steps.

Abstract:Novel annulated azaheterocycles of benzo[1,2,4]triazoloazepine and tetrahydronaphtho[1,2-e][1,2,4]triazine derivatives have been synthesized. Treatment of 2-diazenyl-1,2,3,4-tetrahydronaphthalen-2-yl acetates with BF3.Et2O generates 1-aza-2-azoniaallenium cation intermediates (or azocarbenium ions), which are intercepted by nitriles via cascade polar [3(+) + 2]-cycloaddition/rearrangement reactions to afford benzo[1,2,4]triazoloazepinium salts. These literature unprecedented fused tricycle compounds have been shown to exhibit antimicrobial activity against Gram-positive Staphylococcus aureus with in silico docking studies, suggesting that they may exhibit their antibiotic activity through inhibition of DNA gyrase. Additionally, when ethyl 2-(1-acetoxy-1,2,3,4-tetrahydronaphthalen-2-yl)diazene-1-carboxylate is employed, the reaction with BF3.Et2O produces 1,2-diaza-1,3-diene, which reacts with nitriles via a diaza-Diels-Alder reaction with inverse electron demand, leading to ethyl tetrahydronaphtho[1,2-e][1,2,4]triazine carboxylates. The DFT calculation has been performed to further prove the D-A reaction speculation.

Abstract:In this paper, a novel and sustainable synthesis of the hitherto unreported 5H-benzo[c]imidazo[1,2-a]azepine-6-carboxylic acids via the cascade reactions of 2-arylimidazoles (1) with methylene-oxetanones (2) is presented. Mechanistically, the formation of the title compounds is triggered by a Rh(iii)-catalyzed C(sp2)-H alkenylation of 1 with 2 followed by an intramolecular N-nucleophilic substitution. With this method, a series of hybrid compounds combining the biologically promising imidazole and benzoazepine moieties decorated with a synthetically versatile carboxyl group were prepared in moderate to good efficiency. In addition, the utility of the products thus obtained was remarkably showcased by their efficient transformations into some otherwise difficult-to-obtain pentacyclic compounds.

Abstract:A cascade [4 + 3] annulation of o‐alkenyl bromoarenes and o‐bromoaniline derivatives was described. Various dibenzo[b,f]azepines with substitutions on the 10/11 position were obtained in 14‐97% yields. The synthetic versatility of this protocol is highlighted by the preparation of a precursor of the drug molecule oxcarbazepine, a gram‐scale synthesis, and two product transformations. Unlike previous amination/Heck sequence, this cascade process is supposed to undergo a C(vinyl), C(aryl)‐palladacycle involved pathway.

Abstract:The bicyclic 1-aza-2-azoniaallenium salt intermediates, generated from the azoester species upon treatment with a Lewis acid, have been demonstrated to participate in Huisgen-type cycloaddition with nitriles to result in the formation of fused 6,7,8,9-tetrahydro-5 H-[1,2,4]triazolo[1,5- d][1,4]diazepinium salts. This transformation is interpreted as a regular [3++2] cycloaddition between intermediates as the reactive 1,3-monopole reactants and nitriles as the nucleophilic reagents followed by spontaneous [1,2]-cationic rearrangement. The azoester precursors were easily accessible via oxidation of the corresponding hydrazones using hypervalent iodine oxidant PhI(OAc)2 under mild conditions. The [1,2,4]triazolodiazepine compounds represent a class of N-containing biologically important heterocycles with a new type of scaffold.

Abstract:(1-Chloro-2-trifluoromethylcyclohexyl)azo compounds 3a-d, prepared by oxidation of the hydrazones 2a-d of 2-trifluoromethylcyclohexanone (1) with a slightly excess amount of tertbutyl hypochlorite, react with antimony pentachloride and a nitrile to afford the respective 2,3-disubstituted-9-trifluoromethyl-6,7,8,9-tetrahydro-5H-[1,2,4]triazolo[1,5-a]azepinium hexachloroantimonates 6a-j. The reaction mechanism comprises of the initial cycloaddition of the in situ generated 1-aza-2-azoniaallene salts 4 to the triple bond of nitrile, giving the spiro-substituted 5a-j, followed by rearrangement and concomitant ring enlargement. From 3a-c where R-1 = aryl, the salts 6a-h are isolated in high yields. However, if ethyl (1-chloro-2-trifluoromethyl) azocarboxy late (3d) is employed, the neutral N(3)-unsubstituted analogues 7a and 7b are obtained and after usual basic work-up, are converted to picrates 8a and 8b respectively. For one compound 6f, the X-ray diffraction analysis has been carried out in order to confirm the structural assignment. A practical modified procedure for preparation of the crucial starting ketone 1 is also described.

Abstract:In the presence of a catalytic amount of Zn(OTf)2 in DCE at ambient temperature, a number of tertiary enamides underwent highly efficient reaction with imines of salicylaldehydes to afford diverse functionalized 4-chromanamine derivatives in high yields. The reaction proceeds most probably through an enaminic addition of tertiary enamides to imine functionality followed by the intramolecular interception of the resulting iminium intermediate by the phenolic hydroxy group. The synthesis was also practically implemented by means of a three-component reaction starting from salicylaldehyde, para-nitroaniline, and a tertiary enamide.
