Abstract:Although copper-catalyzed amination of activated C(sp3)-H bonds through radical relay has been developed, amination of unactivated C(sp3)-H bonds is rare. Herein, copper-catalyzed intramolecular amination of remote unactivated C(sp3)-H bonds is reported. The reaction is conducted in a mild and effective manner with moderate to good yields, demonstrating broad tolerance toward various functional groups and exhibiting complete regio- and chemoselectivities. This innovation supplies novel synthetic pathways for the construction of saturated nitrogenated heterocycles.

Abstract:A palladium-/copper-catalyzed intermolecular C-H amination reaction of indoles has been developed. This reaction proceeds in good to excellent yields to produce a variety of 2-amino-substituted indoles and exhibits excellent regioselectivity at room temperature. Abstract:The Iodine-mediated oxidative intramolecular amination of anilines via cleavage of unactivated (sp3)C-H and N-H bonds for the production of indolines is described. This transition-metal-free approach provides a straightforward strategy for producing (sp3)C-N bonds for use in the preferential functionalization of unactivated (sp3)C-H bonds over (sp2)C-H bonds. The reaction could be performed on a gram scale for the synthesis of functionalized indolines.

Abstract:An efficient palladium-catalyzed [2 + 2 + 1] annulation of 3-iodochromones, bridged olefins, and iodomethane is described, affording a range of chromone-containing polycyclic compounds. Additionally, the corresponding deuterated products were smoothly obtained with iodomethane- d 3 instead of iodomethane. Moreover, the synthetic utility of this method is further substantiated by gram scale preparation and application to late-stage modification of estrone.

Abstract:An additive-assisted iridium-catalyzed directed C(sp3)-H amination with 2,2,2-trichloroethoxycarbonyl azide as an amino source is reported. Both carboxylate anions and the corresponding cations in the additives are crucial to achieve satisfactory efficiency. Sodium acetate or n-pentanoic acid can promote the amination of various primary C(sp3)-H bonds adjacent to secondary, tertiary, and quaternary carbons in ketoximes or N-aromatic heterocycles, respectively, providing a practical route to versatile β-amino ketoxime and N-heteroaryl ethanamine derivatives. The amination products can be treated as isocyanate analogues and can be converted to other useful amino functionalities. An iridacyclic compound was isolated and identified as a plausible intermediate.

Direct C4- and C2 C–H Amination of Heteroarenes Using I(III) Reagents via a Cross Azine Coupling

Abstract:Aminated nitrogen heterocycles are valuable motifs across numerous chemical industries, perhaps most notably small mole-cule pharmaceuticals. While numerous strategies exist to install nitrogen atoms onto azaarenes, most require pre-functionalization and methods for direct C–H amination are almost entirely limited to the C2-position. Herein, we report a method for the direct C2 and C4 C–H amination of fused azaarenes via in situ activation with a bispyridine-ligated I(III)-reagent, [(Py)2IPh]2OTf, or Py-HVI. Unlike commonly used N-oxide chemistry, the method requires no pre-oxidation of the azaarene and it provides unprecedented access to C4-amination products. The resulting N-heterocyclic pyridinium salts can be isolated via simple trituration, and the free amine can be liberated under mild Zincke aminolysis, or the amination and cleav-age can be telescoped to a one-pot process. The scope of the method is broad, the conditions are mild and operationally sim-ple, and the aminated products are produced in good to excellent yields. Computational studies provide insights into the mech-anism of activation, which involves an unusual direct nucleophilic functionalization of an I(III)-ligand, as well as a kinetic basis for the observed C2 and C4 amination products.

Abstract:An ammonium iodide/hydrogen peroxide-mediated intramolecular oxidative amination of 3-aminoalkyl-2-oxindoles was achieved, affording the corresponding 3,2'-pyrrolidinyl spirooxindoles and their 6- or 7-membered analogous in moderate to high yields. This metal-free procedure features very mild reaction conditions, non-toxicity and easily handled hydrogen peroxide as a clean oxidant.

Abstract:C-H amination of arenes directed by weakly coordinating Weinreb amides has been achieved with an iridium catalyst and 2,2,2-trichloroethoxycarbonyl (Troc) azide as an aminating agent, providing a robust method of producing synthetic useful ortho-TrocNH aryl Weinreb amides. Taking advantage of the reactivity of Weinreb amide and Troc groups in the amination products, selective hydrolysis was achieved as an attractive process for the synthesis of ortho-NH2 aryl Weinreb amides, which are the building blocks useful in the synthesis of bioactive compounds, and cascade aminocyclization with primary amines was successful and provided an efficient pathway for the construction of quinazolin-2,4-diones, which are present in various alkaloids and natural products.
