Abstract:We report a cobalt-catalyzed desymmetrizing isomerization of exo-cyclic alkenes to generate chiral 1-methylcyclohexene derivatives with good yields and enantioselectivities. A novel chiral thiazolinyl iminoquinoline ligand and its cobalt complex were designed and synthesized to control the establishment of tertiary or quaternary carbon centers at a remote position. This protocol is operationally simple, and a model for the stereochemical outcome has been proposed.

Abstract:An asymmetric three‐component reaction of α‐H diazoacetate with sulfonamide and imine has been developed under the co‐catalyzation of Rh2(OAc)4 and a chiral phosphoric acid (CPA), which proceed through Mannich‐type interception of transient ammonium ylide. The reaction offers a convenient access to α, β‐diamine acid derivatives with two adjacent chiral centers in 59%‐74% yields with 91%‐99% ee and 92:8 to > 95:5 dr. Moreover, the generated products could be easily converted to free chiral diamines and other useful chiral motifs.

Abstract:Chiral vicinal diamines represent an important class of organic building blocks that play essential roles in various fields in chemical and material sciences. Their syntheses have continued to attract attention to enhance their yield, selectivity, and scope, but all of the procedures reported so far in the literature are solvent-based reactions. We herein report the first example of mechanochemical diaza-Cope rearrangement to convert a wide variety of aldehyde substrates into chiral diimines, which upon hydrolysis lead to the formation of chiral vicinal diamines. This solvent-free protocol is highlighted with high yield, fast reaction, and simple synthetic operation with commercially available precursors and is easily scalable to gram quantity. Mechanistic investigations revealed a distinct reaction profile for mechanochemical-induced reactions compared to solution-based reactions. Reaction time course studies showed that the diaza-Cope rearrangement step, which requires a highly ordered six-membered chair-like TS, is rapid in mechanochemical-induced reactions. This enhanced rate stems from the formation of effective solid-state packing in mechanochemical reaction settings. The powder X-ray diffraction analysis of the milled sample confirmed the formation of a new crystalline phase during the mechanochemical reaction, which leads to continuation of the reaction even after ball milling is stopped. Furthermore, we developed a one-pot two-step procedure to synthesize chiral salen metal complexes, a “privileged” chiral catalyst, directly from arylaldehydes without the need for isolating the rearrangement products. From a practicality perspective, this work demonstrates a mechanochemical synthesis of chiral vicinal diamines that can be readily adapted in academic laboratories and, potentially, in industrial settings.

Abstract:The enantioselective construction of all-carbon quaternary stereocenters is one of the most challenging fields in asymmetric synthesis. An asymmetric desymmetrization strategy offers an indirect and efficient method for the formation of all-carbon stereocenters. An enantioselective formation of cyano-bearing all-carbon quaternary stereocenters in 1,2,3,4,-tetrahydroquinolines and 2,3,4,5-tetrahydro-1H-benzo[b]azepines by copper-catalyzed desymmetric N-arylation is demonstrated. The cyano group at the prochiral center plays a key role for the high enantioselectivity and works as an important functional group for further transformations. DFT studies provide a model which successfully accounts for the origin of enantioselectivity.

Abstract:A cascade vinylogous 1,6-Michael addition/1,4-proton shift/aza-Michael addition/hemiaminal formation sequence of 1,2-diaza-1,3-dienes and β-substituted 2-butenals has been developed under the influence of dienamine activation of a chiral secondary amine. This method exhibits high regio- and chemoselectivity and provides an efficient and straightforward approach to bicyclic l,8-diazabicyclo[3.3.0]octane skeletons with a tetrasubstituted stereogenic center with fair to excellent enantioselectivity.

Abstract:Pseudo-C2-symmetric dodecaheterocyclic structures, which possess two acyl/aroyl groups disposed on either a cis- or trans-relative configuration, were prepared from the naturally occurring (-)-(1R)-myrtenal. Addition of Grignard reagents (RMgX) to the diastereoisomeric mixture of these compounds unexpectedly showed that nucleophilic additions to the two prochiral carbonyl centers gave the same stereochemical result in both cis/trans diastereoisomers, making unnecessary the separation of this mixture. Noticeably, both carbonyl groups showed different reactivity because one of them is attached to an acetalic carbon and the other to a thioacetalic carbon. Furthermore, addition of RMgX to the carbonyl attached to the former carbon takes place through the re face, while addition to the second one proceeds through the si face, thus affording the corresponding carbinols in a highly diastereoselective process. This structural feature allowed the sequential hydrolysis of both carbinols, yielding separately (R)- and (S)-1,2-diols after reduction with NaBH4. The mechanism of the asymmetric Grignard addition was explained by density functional theory calculations. This approach contributes to the development of the divergent synthesis of structurally and/or configurationally different chiral molecules.

Abstract:Transition-metal catalyzed asymmetric allylic substitution with alkyl and heteroaryl carbon nucleophiles has been well-established. However, the asymmetric allylic arylation of acyclic internal alkenes with aryl nucleophiles remains challenging and underdeveloped. Herein we report a stereospecific 3-aza-Cope rearrangement interrupted asymmetric allylic substitution-isomerization (Int-AASI) that enables asymmetric allylic arylation. By means of this stepwise strategy, both enantioenriched allylic arylation products and axially chiral alkenes could be readily obtained in high enantioselectivities. Experimental studies support a mechanism involving a cascade of asymmetric allylic amination, stereospecific aryl 3-aza-Cope rearrangement and alkene isomerization. Density functional theory studies detailed the reasons of achieving the high chemoselectivity, regioselectivity, stereoselectivity and stereospecificity, respectively.

Abstract:We developed a novel asymmetric Friedel-Crafts alkylation/transamidation tandem reaction for the enantio- and diastereoselective synthesis of pyrazolo[3,4-b]pyridin-6-ones bearing a -CF3 unit via synergistic chiral phosphoric acid and MgSO4 catalysis. This [3 + 3] annulation protocol allows the formation of trifluoromethylated pyrazolo[3,4-b]pyridin-6-ones with two adjacent tertiary stereocenters in moderate to high yields (up to 90%), enantioselectivities (up to 97% ee), and diastereoselectivities (up to >20:1 dr).
