Abstract:Due to the uniqueness of fluorine atom and C—F bond,difluoromethylene has special properties.As a bioisostere of an oxygen or a carbonyl group,it plays an important role in medicines,pesticides and materials.Difluoromethyl heteroaryl sulfones,represented by 2-PySO2CF2H (Hu reagent),have been developed recently as difluoromethylation reagents,and widely recognized by synthetic chemists for their ease of preparation,good functional group tolerance and universal applicability to a wide range of carbonyl compounds.Through different types of reactions such as nucleophilic substitution,nucleophilic addition,Julia-Kocienski olefination reaction,and radical-mediated difunctionalization,they introduced difluoromethyl,difluoromethylene,gem-difluoroalkene and other fluorine-containing groups into aldehydes,ketones,and heterocyclic compounds.For the first time,the synthesis of fluorine-containing organic compounds involved in various difluoromethyl heteroaryl sulfones in the past decade is reviewed from the perspective of reaction types and their application studies.

Abstract:A novel intramolecular Smiles rearrangement of α-fluoro-β-keto-pyrimidylsulfones (usually used as a carbon nucleophile) was developed, providing a versatile avenue for synthesis of tri/tetra-substituted monofluorinated pyrimidyl enol ethers. Among these, diverse (Z)-monofluorovinylsulfones and sulfinates were efficiently assembled by adding extra electrophile and fine-tuning reaction conditions. The process is triggered by a keto-enol tautomerism from enol oxyanion to pyrimidine 2-carbon, completely different from the classical carbon nucleophilic addition reaction approach.

Abstract:A highly diastereoselective [3 + 2] cycloaddition of aziridines with difluorinated silyl enol ethers has been developed. This approach provides a facile methodology for highly functionalized gem-difluorinated pyrrolidines in good to excellent yields with good functional group tolerance. A one-pot, two-step approach for synthesis of structurally interesting fluorinated pyrroles has also been developed through a cycloaddition/aromatization/desulfonation sequence. Moreover, readily available substrates, mild reaction conditions, and easy scale-up synthesis show practical advantages.

Abstract:A cascade AgOTf-catalyzed chemoselective approach to 3,5/1,3-disubstitued pyrazoles from propargylic alcohols and para-tolylsulfonohydrazide has been developed. Good chemoselectivity is observed depending on the different substituents in the alkyne moiety of the propargylic alcohols, generating two different kinds of products through different aromatization mechanisms. The pyrazolo[5,1-a]isoquinoline skeleton can also be effectively constructed by this method through a cascade bicyclization process.

Abstract:Optically pure diarylmethanes are frequently presented in pharmaceuticals and bioactive molecules. However, minor efforts have been devoted to chiral polyfluoroarene-containing diarylmethanes, and their synthesis is still challenging. Herein, we describe an enantioselective Cu/sulfoxide phosphine (SOP) catalyzed nucleophilic substitution reaction by using polyfluoroarenes as the polyfluoroaryl reagent. Under mild conditions, this protocol enables the efficient synthesis of chiral polyfluoroaryl diarylmethanes with fluorinated quaternary stereogenic center in good yields (up to 93%), high regioselectivties, and excellent enantioselectivities (up to 99% ee). Moreover, gram-scale experiments, product derivations, and late-stage diversifications were performed to demonstrate the utility of this method.
