Abstract:A novel synthesis of multisubstituted pyrroles was developed where in trisubstituted olefins were reacted with p-toluenesulfonylmethyl isocyanide (TosMIC) derivatives. Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A new and efficient method for the synthesis of 4-methoxyl-2,3,5-trimethylpyiidine with an overall yield of 76% from cheap and easily available 2,3,5-trimethylpyridine was developed. The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:An alternative route has been developed for the construction of multisubstituted pyrrole derivatives from readily available, doubly activated cyclopropanes and anilines using an iron-mediated oxidation domino reaction (i.e., sequential ring-opening, cyclization, and dehydrogenation reactions). This reaction uses readily available reactants and is tolerant of a broad range of substrates, with the desired products being formed in good to excellent yields.

Abstract:A facile synthesis of polysubstituted pyrroles from tosylmethyl isocyanide (TOSMIC) and readily synthesized vinyl azides was developed. The reaction proceeded under mild conditions in the presence of base. 2-Tosyl-substituted pyrroles were obtained in moderate to good isolated yields. Additionally, a base-initiated one-pot pyrrole synthesis also was developed using carboxaldehydes, ethyl 2-azidoacetate, and TOSMIC.

Abstract:An efficient copper-catalyzed tandem oxidation/alpha-formylation reaction of 2,3-dihydro-1H-pyrroles in an oxygen atmosphere has been developed. The new reaction is conducted in dimethyl sulfoxide (DMSO), which serves as both the carbonyl carbon source and solvent, in the presence of trifluoroacetic acid to directly generate structurally diverse alpha-formylpyrrole derivatives in one pot in moderate to good yields. Sensitive aldehyde groups remain intact in the presence of an oxidant, trifluoroacetic acid and a transition metal salt. A possible mechanism is presented.

Abstract:An efficient and Brønsted acid free one-pot protocol to directly generate structurally sophisticated α-formylpyrrole derivatives in moderate to good yields has been demonstrated, involving an iron-mediated domino oxidation/formylation reaction of readily available 2,3-dihydro-1H-pyrroles in dimethyl sulfoxide and air atmosphere, in which dimethyl sulfoxide acts as the formyl donor. Abstract:A series of polysubstituted pyrroles were rapidly synthesized in moderate to good yields via catalyst-free one-pot three-component reaction of 1,3-dicarbonyl compounds with primary amines and nitrostyrenes by using liquid-assisted grinding. This protocol provides several advantages over the conventional chemical synthesis, such as simple work-up procedure, wide substrate scope, short reaction time, and environmental friendliness. Structures of polysubstituted pyrroles were confirmed by H-1 NMR, C-13 NMR, and ESI-MS studies.

Abstract:A heating‐induced desulfuration annulation of pyridinium 1,4‐zwitterionic thiolates and CF3‐substituted imidoyl sulfoxonium ylides has been achieved, allowing a route to biologically important 5 trifluoromethylpyrroles. The transformation proceeds through an unusual ((3+3)‐1) pathway under metal‐free conditions with the extrusion of 4‐methoxypyridine, sulfur and DMSO. The scale‐up reaction and further transformation of the obtained pyrrole products have been performed to demonstrate the synthetic utility of the developed method.

Abstract:Under bifunctional squaramide and thiourea organocatalysis, a series of vicinally bis(trifluoromethyl)-substituted 3,3′-pyrrolidinyl spirooxindoles were successfully synthesized via 1,3-dipolar cycloaddition reactions of 3-(trifluoroethylidene)oxindoles with diethyl 2-((2,2,2-trifluoroethyl)imino)malonate and N-(2,2,2-trifluoroethyl)isatin ketimines. The highlight of this protocol is the efficient construction of the vicinally bis(trifluoromethyl)-substituted pyrrolidine unit of spirooxindoles, including multiple contiguous stereogenic centres, in excellent yields and stereoselectivities.
