Abstract:A TfOH-promoted synthesis of fluorinated poly-fused heterocycles via the cascade cyclization of azadienes and difluoroenoxysilanes has been developed, leading to the facile construction of fluorinated benzofuro[3,2-b]pyridines, 5H-indeno-[1,2-b]pyridines, and 5,6-dihydrobenzo[h]quinolines. This one-pot formal [4 + 2] approach involves 1,4-difluoroalkylation, desulfo-nylation, cyclization, and dehydrated and dehydrofluorinated aromatization and represents the first application of difluoroenoxysilane in cascade transformations. Furthermore, this methodology is highlighted by the synthesis of three fluoro analogues of bioactive molecules with potent topoisomerase inhibitory activities.

Abstract:Difluoroenoxysilane, a commonly used difluoroallylating reagent, has attracted considerable attention in recent years. However, its application in the annulation reaction for the construction of fluorinated heterocyclic compounds remains relatively limited. Presented here is the Bronsted acid-catalyzed efficient formal [4 + 2] annulation of difluoroenoxysilanes with alpha-cyano chalcones. The developed protocol demonstrates tolerance to various substituents under mild reaction conditions, providing a reliable approach to construct gem-difluoro-3,4-dihydro-2H-pyrans in good to excellent yields with high diastereoselectivities.

Abstract:In the presence of KI and (diacetoxyiodo)benzene (PIDA), primary amines bearing an alpha-hydrogen can be efficiently converted to fluoroalkylated imidazoles by reacting with fluoroalkylated alkynes and sodium azide via a cascade Michael addition/azidation/cycloamination process. KI plays a crucial role in this reaction by generating iodine(I) species to promote the cyclization. Substrate scope was explored, and the application of this protocol was demonstrated. Additionally, a plausible reaction mechanism was proposed.

Abstract:A highly diastereoselective [3 + 2] cycloaddition of aziridines with difluorinated silyl enol ethers has been developed. This approach provides a facile methodology for highly functionalized gem-difluorinated pyrrolidines in good to excellent yields with good functional group tolerance. A one-pot, two-step approach for synthesis of structurally interesting fluorinated pyrroles has also been developed through a cycloaddition/aromatization/desulfonation sequence. Moreover, readily available substrates, mild reaction conditions, and easy scale-up synthesis show practical advantages.

Abstract:An effective [3+2] cycloaddition reaction of difluoromethyl or trifluoromethyl hydrazonoyl bromides with alkylidene pyrazolones was disclosed. The method provides an efficient approach to access a variety of highly functionalized fluoroalkyl spiropyrazolones in good yields. This protocol also features some advantages such as easy available and stable substrates, simple operation procedure, atom and step economy. The formation of (cis)- and (trans)-products was discussed.

Abstract:A novel intramolecular Smiles rearrangement of α-fluoro-β-keto-pyrimidylsulfones (usually used as a carbon nucleophile) was developed, providing a versatile avenue for synthesis of tri/tetra-substituted monofluorinated pyrimidyl enol ethers. Among these, diverse (Z)-monofluorovinylsulfones and sulfinates were efficiently assembled by adding extra electrophile and fine-tuning reaction conditions. The process is triggered by a keto-enol tautomerism from enol oxyanion to pyrimidine 2-carbon, completely different from the classical carbon nucleophilic addition reaction approach.

Abstract:A copper catalyzed annulation-aromatization of benzyl trifluoromethyl ketimines with 3-acryloyloxazolidin-2-ones for the synthesis of 3-fluoropyridines through double C-F bond cleavages has been developed. In this approach, the annulation occurred between the in situ formed dienes from trifluoromethyl ketimines via the first C-F bond cleavage and 3-acryloyloxazolidin-2-ones. Then the aromatization afforded 3-fluoropyridines in moderate yields through the second C-F bond cleavage. The 3-fluoropyridine products could be further hydrolyzed to multi-substituted 3-pyridinecarboxylic acids.

Abstract:A simple and novel method to the synthesis of 3-fluoro-4-aryl-2-pyridone from a Michael-adduct of fluoronitroacetate and α,β-unsaturated ketone is reported. With (NH 4 ) 2 CO 3 as the N-source and TsOH as the promoted acid, a series of fluorinated-pyridones was obtained with moderate to good yields.

Abstract:Presented herein is a selective synthesis of 3-(α-fluorovinyl)indoles and 3-acylindoles via the coupling of 1-phenylpyrazolidinones with α,α-difluoromethylene alkynes. Mechanistically, the formation of 3-(α-fluorovinyl)indoles is resulted from a cascade process including Rh(III)-catalyzed ortho-C–H bond cleavage, regioselective triple bond insertion, pyrazolidinone ring-opening, indole ring-formation and HF elimination. Interestingly, when this reaction was carried out in CH3OH/H2O instead of CH3CN, the in situ formed 3-(α-fluorovinyl)indoles readily undergo a hydration process to afford 3-acylindole derivatives. Of note, this protocol features with controllable selectivity, simultaneous formation of both the heterocyclic scaffold and the monofluoroalkenyl moiety, atom-economy, substrate diversity, good functional group tolerance and redox-neutral reaction conditions. Furthermore, anticancer screening of selected products against two human cancer cell lines demonstrated their potential as lead compounds for drug development.

Abstract:Under strongly acidic conditions, nucleophilic attack on tetrafluoropyridazine and on 3,5,6-trifluoro-4-methoxypyridazine by water or methanol leads to substitution of the fluorine atoms at C-3 and C-6 in preference to those at C-4 and C-5 which have previously been shown to be more reactive towards nucleophilic reagents under basic conditions. Treatment of tetrafluoropyridazine with water in sulphuric acid solution gives 3,4,5-trifluoro-1H-pyridazin-6-one, and similar treatment with methanol gives 3,4,5-trifluoro-6-methoxypyridazine (at 0°) or 4,5-difluoro-3,6-dimethoxypyridazine (at 20°). All four fluorine atoms of tetrafluoropyridazine are replaced by chlorine when it is treated with ethereal hydrogen chloride at 18°. Spectroscopic evidence suggests that the acid-catalysed reactions involve nucleophilic attack on protonated polyfluoropyridazinium cations, and two N-alkyl-tetrafluoro-pyridazinium tetrafluoroborates have been prepared and shown to give exclusively 1-alkyl-3,4,5-trifluoropyridazin-6-ones on treatment with water. Factors affecting the orientation of the products of nucleophilic attack on poly-fluoropyridazinium cations are discussed. Structures have been assigned to the two isomeric difluoromethoxy-pyridazinones resulting from the acid-catalysed hydroxylation of 3,5,6-trifluoro-4-methoxypyridazine, by relating them to 3,4-difluoro-5,6-dimethoxypyridazine and 3,5-difluoro-4,6-dimethoxypyridazine.

Abstract:Palladium-catalyzed CF bond formation via directed CH activation has recently emerged as a significant method to introduce fluorine atoms into organic molecules in an atom- and step-economical path. At the interface between fluorine chemistry and synthetic organic chemistry, the development of direct fluorinations of the CH bond remains sluggish, due to the major challenge of reductive elimination to afford the CF bond. One strategy is to oxidize palladium(II) species to more electron-deficient palladium(IV or III) fluorides to promote the CF bond forming reductive elimination, which opens up palladium-catalyzed CH fluorination as a powerful transformation. This focused review gives a brief summary of the mere handful of examples, including C(sp(2))F and C(sp(3))F bond formations, in this field so far.

Abstract:A copper-catalyzed, redox-neutral cascade difluoroalkylation/5-endo annulation/β-fluorine cleavage of ynones is developed, providing a direct and stereoselective method to access synthetically important α-monofluoroalkenyl cyclopentanones. Mechanistic studies suggest an unprecedented CuII-assisted β-fluorine fragmentation, which may be valuable for the challenging but important C-F bond activation. Moreover, the in situ generated difluorocarbene was found to serve as an effective reductant for the regeneration of copper(I) catalyst, thus avoiding the addition of external reductants.
