Abstract:Two enantiomeric chalcone epoxides 2a and 2b are synthesized under phase-transfer conditions using 1-benzylquinidinium chloride and 1-benzylquininium chloride as catalysts, respectively Stereoselective cyclization of 2a and 2b, followed by methylation and preparative high performance liquid chromatography gives pure (+)-aromadendrin trimethyl ether (4a) and its enantiomer 4b. Reduction of pure 4a and 4b with NaBH4. affords four pure flavan-3, 4-diol trimethyl ethers, 5a, 6a, 5b, and 6b.

Abstract:The (3S,5R) and (3R,5R) isomers of the title compound were synthesized from enantiopure epoxide chiral building blocks readily accessible from D-gluconolactone. The former (a syn-1,3-diol) gave H-1 and C-13 NMR spectroscopic data and optical rotation in good agreement with those reported for the natural product, whereas distinct differences were found with the data for the latter (an anti-1,3-diol). These results unambiguously showed that the absolute configuration of the natural product must be (3S,5R).

Abstract:A total synthesis of optically pure (+)-catechin pentaacetate has been established using the (-)-chalcon epoxide (100% ee) derived from 3,4,2',4',6'-pentakis(methoxymethoxy)chalcon as the starting material. The optical purity of the product is confirmed by H-1 nmr analysis in the presence of a shift reagent.

Abstract:The effect of substitutents on the formation of products in the reactions of 2"-methoxymethoxychalcone epoxides under acidic conditions has been examined. Regardless of the 6"-substitutent, the presence of a 4'-hydroxyl group results in the exclusive formation of flavanonols. The presence of a 4'-methoxyl group directs the reactions toward the competitive formation of flavanonols and glycol monomethyl ethers or toward the exclusive formation of glycol monomethyl ethers, depending on the bulk of the 6"-substituent.

Abstract:Alkylation under phase transfer conditions of the chiral glycine synthon 2 (prepared from the commercially available Oppolzer chiral sultam) with fluorinated analogues of 3,4-dimethoxybenzyl chloride proceeded with high diastereoselectivity. Hydrolysis of the Schiff base, removal of the chiral auxiliary and HBr demethylation produced 2-, 5-, 6-fluoro- and 2,6-difluoro-l-DOPA in e.e. of >99%.

Abstract:The structures of flavacitropones A ( 1 )-B ( 2 ), which are new homoacridone-flavanone dimers from Glycosmis parviflora (Sims) Kurz. (Rutaceae), were elucidated by spectroscopic studies. Flavacitropone A ( 1 ) was evaluated for its biological effects on cytotoxicity and release of histamine and TNF-α in 2,4-dinitrophenyl-human serum albumin-stimulated RBL-2H3 cells (rat basophilic cell line). The compound downregulated TNF-α release, but not histamine release.

Abstract:A new compound, 2-(3-(4-hydroxyphenyl)-3-methoxypropyl)-5-methoxycyclohexa-2,5-diene-1,4-dione (1), was isolated from the heartwood of Dalbergia cochinchinensis along with 15 known compounds, 2,4-dihydroxy-5-methoxybenzophenone (2), 3,5-dimethoxy-4′ -hydroxy-trans-stilbene (3), 7,8-dihydroxyflavanone (4), 2,3-dimethoxyxanthone (5), medicarpin (6), galangin (7), 5,7-dihydroxy-2′,3′,4′- trimethoxyisoflavanone (8), 6-hydroxy-2-(2-hydroxy-4-methoxyphenyl)-benzofuran (9), 3′-O-methylviolanone (10), 2′,5,7-trihydroxy-4′-methoxyisoflavone (11), 7-hydroxy-6-methoxyflavone (12), formononetin (13), pterocarpol (14), 2′-hydroxyformonetin (15), and naringenin (16). Their structures were elucidated by spectroscopic means and comparison with published data. Compounds 1–5, 7–9, and 11 were isolated from Dalbergia L. for the first time, and compounds 1–11, 13, 15, and 16 were isolated from D. cochinchinensis for the first time.

Abstract:Chalcone was prepared in a new route by reacting o-hydroxyacetophenone with 4-dimethylaminobenzaldehyde using piperidine as a catalyst. 3-Hydroxy-2-[4-(dimethylamino)phenyl] benzopyran-4-one were prepared by Algar-Flynn-Oyamada method by cyclization of chalcone using Hydrogen peroxide. A series of alkyl and ester derivatives of the flavonoid 3-hydroxy-2-[4-(dimethylamino)phenyl] benzopyran-4-one were prepared by reacting the above mentioned compound with different chemical reagents (Methyl iodide, Allyl bromide, Benzyl chloride, Bromoacetylcoumarin, Chloroacetamide, Chloroacetyl chloride, Phthalic anhydride, Maleic anhydride, Phthalimide, Cinnamoyl chloride) with potassium carbonate and acetone or DMF as a solvent. The physical and spectroscopic properties of the new compounds were studied by (FT-IR, 13C-NMR and 1H-NMR) spectral methods. The purity of the synthesized compounds were confirmed using TLC thin layer chromatography. The biological activity of some synthetic flavonoids (A2, A5, A7, A8, A9, A12) at two different concentrations (0.5 mg/ml, 0.25 mg/ml) were studied on three types of fungi: Aspergillus flavus, Acremonium strictum, Penicillium expansum. Some of this compounds showed high activity against the tested fungi.
