Abstract:The preparation of a series of heterocycles obtained by the condensation of 4,5-diazafluoren-9-one with various amines is described with particular emphasis on bridged systems formed by the condensation of 4,5-diazafluoren-9-one with diamine compounds such as hydrazine. Due to sp2 hybridization of the nitrogen atom in the bridge, the protons in the two pyridine units of each bipyridine group are non-equivalent. The projection of a hydrogen atom from one of the pyridine rings into the ring current of an adjacent phenyl ring shifts the proton resonance upfield. Proton NMR resonances have been uniquely assigned for all the compounds and electronic absorptions have been attributed to π → π∗ transitions located in the ultraviolet region of the spectrum. Reduction of the heterocycles occurs at the imine nitrogen bridge and is irreversible, except for the ligand resulting from the condensation of 4,5-diazafluoren-9-one with hydrazine.

Abstract:Pyrazole compounds are one kind of heterocyclic compounds with extensive bioactivity,and lots of pyrazole compounds have been commercialized both in the region of medicament and pesticide.In the process of finding out some novel pyrazole compounds,a~d four novel pyrazole intermediates were synthesized.Taking diethyl oxalate as starting substance,the novel compounds can be synthesized via condensation,cyclic condensation,alkylation,(bromination) and hydrolysis reactions.The yield of products was about 78%-90%.Their structures were confirmed by elementary analysis,1H-NMR,MS and IR.

Abstract:Aim and objective: According to the literature survey, pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the present work, pyrazol- 3-one 1 was reacted with cyanoacetic acid hydrazide and elemental sulfur to afford the corresponding thieno[3,2-c]pyrazol-6-carbohydrazide 3 derivatives. The latter compound reacted with some electrophilic reagents such as DMF-DMA, triethylorthoformate, arylidenemalononitriles and chalcones under neat conditions to give substituted oxadiazole and pyrazole, respectively. The treatment of compound 3 with active methylene reagents such as acetylacetone, diethylmalonate, ethyl acetoacetate and ethyl cyanoacetate under suitable conditions afforded pyrazole derivatives 10, 11, 13, and 15, respectively. Novel pyrazolothienopyrimidine 27 and 30 were prepared from precursor 26 with carbon disulfide and triethylorthoformate, respectively. The chemical structures of the newly synthesized compounds were established by elemental and spectral analyses including IR, and 1HNMR in addition to 13C-NMR and mass spectra. Materials and methods: A novel substituted pyrazole, pyrimidine and pyrazolothienopyrimidine were obtained via Gewald synthesis of thiophene and fused thiophene and Mannich reactions of 5-amino-3-phenyl-1Hthieno[ 3,2-c]pyrazole-6-carbohydrazide. Results and discussion: A series of some newly azoles and azines were prepared via reaction of thieno[3,2- c]pyrazol-6-carbohydrazide derivative 3 as starting material with some electrophilic and nucleophilic reagents. The structures of target compounds were established by elemental analyses and spectral data. Conclusion: Pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the current investigation, we have developed new and efficient methods for the synthesis of thieno[3,2-c]pyrazol-6-carbohydrazide derivatives. In addition, we have explored the preparative potential of these substances as intermediates for the synthesis of substituted pyrazoles and fused pyrazoles 10-30, respectively.

Abstract:Cyclocondensation reactions of 3-phenyl-1-(3-methyl-5-oxo-1-phenyl-2-pyrazolin-4-yl) prop-2-en-1-one (2) with active methylene reagents such as ethyl cyanoacetate, malononitrile, ethyl acetoacetate, and acetoacetanilide afforded pyrazolinyl pyridine derivatives (3 and 12), pyrazolinyl pyran derivative (9), and pyrazolinyl cyclohexene derivatives (10 and 11). Reaction of 11 with hydrazines and hydroxyl amine led to the formation of indazole derivatives (13 and 14), and benzisoxazole derivative 15. Also, reaction of 9 with different reagents gave pyrano[2,3-d][1,2,3]triazine (19) and pyrano[2,3-d]pyrimidine derivatives (20 and 21). The anti-inflammatory, analgesic, and antimicrobial activities of these compounds were determined. Compounds 13 and 14 showed good anti-inflammatory activity in comparison with the standard indomethacin, whereas compounds 4, 7, and 12 showed higher analgesic activity than the reference aspirin drug. In addition, compounds 7, 11, and 15 exhibited broad spectrum activities against all bacterial and fungal species tested.Graphical Abstract

Abstract:This paper uses basic-catalysed method to convert 1H-pyrazole-4-carboxamides into tri-fused heterocyclic systems containing two kinds of pyrazolotriazolopyrimidine framework. The synthesis of compounds 5-6 and reactions of them with some electrophiles have been investigated. Preliminary bioassay study showed that some compounds displayed high activity.
