Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.

Abstract:A series of novel 1,5-diaryl-1H-pyrazol-3-oxy derivatives containing oxyacetic acid or oxy(2-thioxothiazolidin-3-yl)ethanone moieties were prepared from methyl 3-arylacrylates via a serial of reactions included addition-cyclization, oxidation, substitution, hydrolysis, and condensation. Their structures were confirmed by 1H-NMR, 13C-NMR, IR, and elemental analysis. In addition, the crystal structure of the compound 2-(1,5-diphenyl-1H-pyrazol-3-yloxy)-1-(2-thioxothiazolidin-3-yl)ethanone was determined by single crystal X-ray diffraction analysis. Bioassay results indicated that the compound 2-(5-(4-chlorophenyl)-1-phenyl-1H-pyrazol-3-yloxy)-1-(2-thioxo-thiazolidin-3-yl)ethanone exhibited moderate inhibitory activity against Gibberella zeae at the dosage of 10 mu g mL-1.

Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

Abstract:Seven novel 1,5-diaryl-3-benzoyloxy-1H-pyrazoles (2a-2g) were prepared by the DCC condensation of 1,5-diaryl-1H-pyrazol-3-ols (1a-1g) with benzoic acid. Their structures were characterized by elemental analysis, IR, H-1 NMR and mass spectral data. The structure of 5(3,4-dimethoxyphenyl)-1-phenyl-3-benzoyloxy-1H-pyrazole (2d) was also determined by single crystal X-ray diffraction analysis. Bioassay results indicated that the compounds 5-(4-fluorophenyl)-1-phenyl-3-benzoyloxy-1H-pyrazole (2f) and 1,5-diphenyl-3-benzoyloxy-1H-pyrazole (2a) exhibited excellent-to-medium fungicidal activity against Sclerotinia sclerotiorum at a dosage of 10 mu g/mL. The structure-activity relationships (SAR) were also discussed.

Abstract:Five novel pyrazole-coupled glucosides, 1,5-diaryl-1H-pyrazol-3-yl 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranosides 5a5e, were synthesized by the phase-transfer catalytic reaction of 1,5-diaryl-1H-pyrazol-3-ols 4a4e with acetobromo-a-D-glucose in H2O/CHCl3 under alkaline conditions, using Bu4N+Br- as catalyst. Then, glucosides 5a5c were deacetylated in a solution of Na2CO3/MeOH to yield the 1,5-diaryl-3-(beta-D-glucopyranosyloxy)-1H-pyrazoles 6a6c. Their structures were characterized by 1H,1H-COSY, 1H-, 13C-, and 19F-NMR spectroscopy, as well as elemental analysis. The structures of 5d and 6c were also determined by single-crystal X-ray diffraction analysis. A preliminary in vitro bioassay indicated that compounds 4e and 5d exhibited excellent-to-medium fungicidal activity against Sclerotinia sclerotiorum at the dosage of 10 mu g/ml.

Abstract:Succinate dehydrogenase inhibitor is a low-toxic and high active fungicide. In order to develop novel and broad-spectrum succinate dehydrogenase inhibitor (SDHI) fungicides, thiophene was introduced into the structure of pyrazole carboxamide fungicides. Twenty four pyrazole-thiophene carboxamides were designed, synthesized and characterized by H NMR, C NMR and HRMS. The crystal structure of N-(4-methoxyphenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene2-carboxamide (7i) was determined by X-ray diffraction. The antifungal activity of all the synthesized compounds was determined against six plant pathogenic fungi, and preliminary bioassays suggested that some compounds exhibited good antifungal activity against Rhizoctonia solani, Fusarium graminearum and Botrytis cinerea. Among them, N-(4fluorophenethyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7c) exhibited the best antifungal activities against R. solani in vitro with EC50 value of 11.6 μmol/L, and N-(2-fluorophenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7j) against F. graminearum with EC50 value of 28.9 μmol/L. And N-(4-chlorophenyl)-4-(1-methyl-1H-pyrazol-4yl)thiophene-2-carboxamide (7h) showed similar inhibition abilities with thifluzamide against B. cinerea with EC50 value of 21.3 μmol/L. The molecular docking results showed that the high antifungal activitie compounds form stronger interactions with important amino acid residues of succinate dehydrogenase.

Abstract:In an attempt to search for potent fungicide, a series of novel N-aryl-1H-pyrazole-5-carboxylate derivatives was designed and synthesized. Their chemical structures were characterized by 1H NMR spectra and high resolution mass spectrometry(HRMS). The preliminary bioassay results indicated that some target compounds displayed better fungicidal activities against certain fungi at 50 µg/mL or favorable antitumor activities at 5 µg/mL compared with chlorothalonil and 5-fluorouracil, respectively. The structure-activity relationship demonstrated that the introduction of ester group and amide bond was favorable to the improvement of activities against Physalospora piricola and Phytophthora capsici.

Abstract:A series of pyrazolyl-heterocycles, including 1,3,4-thiodiazoles, 1,3,4-oxadiazoles and 1,3,4-triazores were synthesized from 4-nitro-pyrazole formhydrazide. The structures of air new compounds were confirmed by H-1 NMR and elementary analysis. Their fungicidal activity was evaluated against Rhizoctonia solani. Structure-activity relationships for the screened compounds were evaluated and discussed. It was found that same compounds showed a higher fungicidal activity and systematic activity.

Abstract:Fifteen new pyrazole-4-carboxylic oxime ester derivatives were synthesized from benzaldehyde oxime and 3-(trifluoromethyl)-1-methyl-1H-pyrazole-4-carbonyl chloride conveniently. These pyrazole-4-carboxylic oxime ester structures were confirmed by 1H NMR and HRMS. The compound 5f was further confirmed by X-ray diffraction. They crystallized in the monoclinic system, space group C2/c, Z = 8. The fungicidal activity results indicated that some of these compounds possessed good activity against S. sclerotiorum, B. cinerea, R. solani, P. oryae and P. piricola at 50 ppm. Furthermore, the structure-activity relationship was studied by molecular docking simulation.

Abstract:On the basis of the three-component synthetic methodology developed by us, a total of twenty-six pyrazole compounds bearing aryl OCF3 were designed and synthesized. Their chemical structures were characterized by 1H and 13C nuclear magnetic resonance and high-resolution mass spectrometry. These compounds were evaluated systematically for antifungal activities in vitro against six plant pathogenic fungi by the mycelium growth rate method. Most of the compounds showed some activity against each of the fungi at 100 μg/mL. Compounds 1t and 1v exhibited higher activity against all the tested fungi, and 1v displayed the highest activity against F. graminearum with an EC50 value of 0.0530 μM, which was comparable with commercial pyraclostrobin. Structure–activity relationship analysis showed that, with respect to the R1 substituent, the straight chain or cycloalkyl ring moiety was a key structural moiety for the activity, and the R2 substituent on the pyrazole ring could have significant effects on the activity. Simple and readily available pyrazoles with potent antifungal activity were obtained, which are ready for further elaboration to serve as a pharmacophore in new potential antifungal agents.

Abstract:In this study, 32 novel quinazolinone-scaffold-containing pyrazole carbamide derivatives were designed and synthesized in a search for a novel fungicide against Rhizoctonia solani. Single-crystal X-ray diffraction of 3-(difluoromethyl)-N-(2-((6,7-difluoro-4-oxoquinazolin-3(4H)-yl)methyl)phenyl)-1-methyl-1H-pyrazole-4-carboxamide (6a11) confirmed the structure of the target compounds. The in vitro antifungal activity of the target compounds against R. solani was evaluated at 100 µg/mL. The structure-activity relationship analysis results revealed that antifungal activity was highest when the substitution activity was at position 6. Moreover, the position and number of chlorine atoms directly affected the antifungal activity. Further in vitro bioassays revealed that 6a16 (EC50 = 9.06 mg/L) had excellent antifungal activity against R. solani that was higher than that of the commercial fungicide fluconazole (EC50 = 12.29 mg/L) but lower than that of bixafen (EC50 = 0.34 mg/L). Scanning electron microscopy), 7.33 (SEM) revealed that N-(2-((6,8-dichloro-4-oxoquinazolin-3(4H)-yl)methyl)phenyl)-3-(difluoromethyl)-1-methyl-1H-pyrazole-4-carboxamide (6a16) also affected the mycelial morphology. The findings revealed that molecular hybridization was an effective tool for designing antifungal candidates. Meanwhile, pyrazolecarbamide derivatives bearing a quinazolinone fragment exhibited potential antifungal activity against R. solani.

Abstract:A practical method of four-step synthesis towards novel (E)-5-(methoxyimino)-3,5-dihydrobenzo[e][1,2]oxazepin-4(1H)-one antifungals is presented, where a commercially available pesticide and pharmacology intermediate, (E)-methyl 2-(2-(bromomethyl)phenyl)-2-(methoxyimino)acetate (1), was used as starting material. These compounds were confirmed by 1H NMR, 13C NMR, high-resolution mass spectroscopy and X-ray crystal structure. Via in vitro fungicidal evaluation, the moderate to high activities of several compounds against eight phytopathogenic fungi were demonstrated. Especially, the fungicidal activities of compounds 5-03 and 5-09 were comparable to those of the controls azoxystrobin and trifloxystrobin in precise virulence measurements for four fungi. These results suggested that dihydrobenzo[e][1,2]oxazepin-4(1H)-one analogues could be considered as potential fungicidal candidates for crop protection.
Synthesis and Bioactivity of Novel Sulfonate Scaffold-Containing Pyrazolecarbamide Derivatives as Antifungal and Antiviral Agents

Abstract:Novel pyrazolecarbamide derivatives bearing a sulfonate fragment were synthesized to identify potential antifungal and antiviral agents. All the structures of the key intermediates and target compounds were confirmed by nuclear magnetic resonance (NMR) and high-resolution mass spectrometry (HRMS). The single-crystal X-ray diffraction of the compound T22 showed that pyrazole carbamide is a sulfonate. The in vitro antifungal activities of the target compounds against Colletotrichum camelliae, Pestalotiopsis theae, Gibberella zeae, and Rhizoctonia solani were evaluated at 50 μg/ml. Among the four pathogens, the target compounds exhibited the highest antifungal activity against Rhizoctonia solani. The compound T24 (EC50 = 0.45 mg/L) had higher antifungal activity than the commercial fungicide hymexazol (EC50 = 10.49 mg/L) against R. solani, almost similar to bixafen (EC50 = 0.25 mg/L). Additionally, the target compounds exhibited protective effects in vivo against TMV. Thus, this study reveals that pyrazolecarbamide derivatives bearing a sulfonate fragment exhibit potential antifungal and antiviral activities.

For this reason, a series of novel pyrazole‐4‐carboxamide derivatives with an N‐methoxy group were designed and synthesized. All the target compounds were characterized by 1H NMR, 13C NMR, and HRMS. The compound 3c was further confirmed by X‐ray diffraction, which crystallized in the triclinic system, space group P‐1, Z = 2. The fungicidal activity results indicated that most of these compounds possessed good activity against Sclerotinia sclerotiorum at 50 ppm. Especially, compound 3h exhibited the best activity with the EC50 is 7.80 μg/mL, which is comparable with the positive control bixafen (EC50 = 6.70 μg/mL). Furthermore, the physicochemical properties, molecular docking simulation, ADMET analyses, DFT of compounds 3h, 3k and two commercial SDHIs, isoflucypram and pydiflumetofen, were investigated in this study.

Abstract:The title compound N-((3,5-dimethylphenyl)carbamoyl)-1-methy1-3-(trifluoromethyl)-1H-pyrazole-4-carboxamide (C15H15F3N4O2) was synthesized, and its structure was confirmed by H-1 NMR, H RMS and X-ray diffraction. It crystallizes in the triclinic system, space group P (1) over bar with a = 11.7147(5), b = 11.7935(5), c = 13.6620(5) angstrom, alpha = 69.755(7)degrees, beta = 66.182(6)degrees, gamma = 72.100(7)degrees, D-c = 1.423 g/cm(3) , Z = 4, V = 1588.88(11) angstrom(3) , the final R = 0.0347 and wR = 0.1005 for 7171 observed reflections with I > 2 sigma(I). The preliminary biological test showed that the title compound has antifungal activities against Fusarium oxysporum, Pseudomonas syringae, Corynespora mazei and Botrytis cinerea at 100 mu g/mL as 5.19%, 53.50%, 88.55% and 70.62%, respectively. The docking results indicated the hydrogen bonds formed between the compound and SHD.

Abstract:A series of novel 1,2,4-triazole derivatives containing oxime ether and phenoxy pyridine moiety were designed and synthesized. The new compounds were identified by nuclear magnetic resonance (NMR) spectroscopy and high-resolution mass spectrometry (HRMS). Compound (Z)-1-(6-(4-nitrophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-methyl oxime (5a18) was further confirmed by X-ray single crystal diffraction. Their antifungal activities were evaluated against eight phytopathogens. The in vitro bioassays indicated that most of the title compounds displayed moderate to high fungicidal activities. Compound (Z)-1-(6-(4-bromo-2-chlorophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-methyl oxime (5a4) exhibited a broad-spectrum antifungal activities with the EC50 values of 1.59, 0.46, 0.27 and 11.39 mg/L against S. sclerotiorum, P. infestans, R. solani and B. cinerea, respectively. Compound (Z)-1-(6-(2-chlorophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-benzyl oxime (5b2) provided the lowest EC50 value of 0.12 mg/L against S. sclerotiorum, which were comparable to the commercialized difenoconazole. Moreover, homologous modeling and molecular docking disclosed possible binding modes of compounds 5a4 and 5b2 with CYP51. This work provided useful guidance for the discovery of new 1,2,4-triazole fungicides.

Abstract:Compounds containing N‐pyridylpyrazole motif have aroused interest and brought about research hotspots due to their highly‐efficient insecticidal activity. The fungicidal potential of N‐pyridylpyrazole derivatives has gradually been disclosed in recent years. To discover new agrochemicals with poly‐heterocyclic features, a series of novel triazole thione/thioether derivatives containing pyridylpyrazole motif (8‐11) was synthesized. The new compounds were identified by melting point, 1H NMR, 13C NMR, 19F NMR, HRMS, and elemental analysis. The bioassays showed that most of the pyridylpyrazole‐containing triazole thione Mannich bases possessed favorable in vitro fungicidal activity toward pathogenic fungi, such as Magnaporthe oryzae, Sclerotinia sclerotiorum, Botrytis cinerea and Fusarium verticillioides, and were comparable with those of the contrast compounds A and triadimefon. Some of them exhibited moderate to good in vivo fungicidal activity against S. sclerotiorum at 0.2 mg/mL (e.g. 8f control efficacy: 60.9±3.2%). The SEM observation displayed that 8f might cause disruption of cell membrane and wall of S. sclerotiorum. Compounds 8a, 8c, 8f‐8h, 8p and 9b can serve as promising new fungicidal agents to make further structural optimization. The findings in this article provide useful clue and guidance for the design and development of new poly‐heterocyclic agrochemicals.
