Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.

Abstract:A series of novel 1,5-diaryl-1H-pyrazol-3-oxy derivatives containing oxyacetic acid or oxy(2-thioxothiazolidin-3-yl)ethanone moieties were prepared from methyl 3-arylacrylates via a serial of reactions included addition-cyclization, oxidation, substitution, hydrolysis, and condensation. Their structures were confirmed by 1H-NMR, 13C-NMR, IR, and elemental analysis. In addition, the crystal structure of the compound 2-(1,5-diphenyl-1H-pyrazol-3-yloxy)-1-(2-thioxothiazolidin-3-yl)ethanone was determined by single crystal X-ray diffraction analysis. Bioassay results indicated that the compound 2-(5-(4-chlorophenyl)-1-phenyl-1H-pyrazol-3-yloxy)-1-(2-thioxo-thiazolidin-3-yl)ethanone exhibited moderate inhibitory activity against Gibberella zeae at the dosage of 10 mu g mL-1.

Abstract:Seven novel 1,5-diaryl-3-benzoyloxy-1H-pyrazoles (2a-2g) were prepared by the DCC condensation of 1,5-diaryl-1H-pyrazol-3-ols (1a-1g) with benzoic acid. Their structures were characterized by elemental analysis, IR, H-1 NMR and mass spectral data. The structure of 5(3,4-dimethoxyphenyl)-1-phenyl-3-benzoyloxy-1H-pyrazole (2d) was also determined by single crystal X-ray diffraction analysis. Bioassay results indicated that the compounds 5-(4-fluorophenyl)-1-phenyl-3-benzoyloxy-1H-pyrazole (2f) and 1,5-diphenyl-3-benzoyloxy-1H-pyrazole (2a) exhibited excellent-to-medium fungicidal activity against Sclerotinia sclerotiorum at a dosage of 10 mu g/mL. The structure-activity relationships (SAR) were also discussed.

Abstract:Five novel pyrazole-coupled glucosides, 1,5-diaryl-1H-pyrazol-3-yl 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranosides 5a5e, were synthesized by the phase-transfer catalytic reaction of 1,5-diaryl-1H-pyrazol-3-ols 4a4e with acetobromo-a-D-glucose in H2O/CHCl3 under alkaline conditions, using Bu4N+Br- as catalyst. Then, glucosides 5a5c were deacetylated in a solution of Na2CO3/MeOH to yield the 1,5-diaryl-3-(beta-D-glucopyranosyloxy)-1H-pyrazoles 6a6c. Their structures were characterized by 1H,1H-COSY, 1H-, 13C-, and 19F-NMR spectroscopy, as well as elemental analysis. The structures of 5d and 6c were also determined by single-crystal X-ray diffraction analysis. A preliminary in vitro bioassay indicated that compounds 4e and 5d exhibited excellent-to-medium fungicidal activity against Sclerotinia sclerotiorum at the dosage of 10 mu g/ml.

Abstract:Succinate dehydrogenase inhibitor is a low-toxic and high active fungicide. In order to develop novel and broad-spectrum succinate dehydrogenase inhibitor (SDHI) fungicides, thiophene was introduced into the structure of pyrazole carboxamide fungicides. Twenty four pyrazole-thiophene carboxamides were designed, synthesized and characterized by H NMR, C NMR and HRMS. The crystal structure of N-(4-methoxyphenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene2-carboxamide (7i) was determined by X-ray diffraction. The antifungal activity of all the synthesized compounds was determined against six plant pathogenic fungi, and preliminary bioassays suggested that some compounds exhibited good antifungal activity against Rhizoctonia solani, Fusarium graminearum and Botrytis cinerea. Among them, N-(4fluorophenethyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7c) exhibited the best antifungal activities against R. solani in vitro with EC50 value of 11.6 μmol/L, and N-(2-fluorophenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7j) against F. graminearum with EC50 value of 28.9 μmol/L. And N-(4-chlorophenyl)-4-(1-methyl-1H-pyrazol-4yl)thiophene-2-carboxamide (7h) showed similar inhibition abilities with thifluzamide against B. cinerea with EC50 value of 21.3 μmol/L. The molecular docking results showed that the high antifungal activitie compounds form stronger interactions with important amino acid residues of succinate dehydrogenase.

Abstract:A series of novel 1,2,4-triazole derivatives containing oxime ether and phenoxy pyridine moiety were designed and synthesized. The new compounds were identified by nuclear magnetic resonance (NMR) spectroscopy and high-resolution mass spectrometry (HRMS). Compound (Z)-1-(6-(4-nitrophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-methyl oxime (5a18) was further confirmed by X-ray single crystal diffraction. Their antifungal activities were evaluated against eight phytopathogens. The in vitro bioassays indicated that most of the title compounds displayed moderate to high fungicidal activities. Compound (Z)-1-(6-(4-bromo-2-chlorophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-methyl oxime (5a4) exhibited a broad-spectrum antifungal activities with the EC50 values of 1.59, 0.46, 0.27 and 11.39 mg/L against S. sclerotiorum, P. infestans, R. solani and B. cinerea, respectively. Compound (Z)-1-(6-(2-chlorophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-benzyl oxime (5b2) provided the lowest EC50 value of 0.12 mg/L against S. sclerotiorum, which were comparable to the commercialized difenoconazole. Moreover, homologous modeling and molecular docking disclosed possible binding modes of compounds 5a4 and 5b2 with CYP51. This work provided useful guidance for the discovery of new 1,2,4-triazole fungicides.

Abstract:A practical method of four-step synthesis towards novel (E)-5-(methoxyimino)-3,5-dihydrobenzo[e][1,2]oxazepin-4(1H)-one antifungals is presented, where a commercially available pesticide and pharmacology intermediate, (E)-methyl 2-(2-(bromomethyl)phenyl)-2-(methoxyimino)acetate (1), was used as starting material. These compounds were confirmed by 1H NMR, 13C NMR, high-resolution mass spectroscopy and X-ray crystal structure. Via in vitro fungicidal evaluation, the moderate to high activities of several compounds against eight phytopathogenic fungi were demonstrated. Especially, the fungicidal activities of compounds 5-03 and 5-09 were comparable to those of the controls azoxystrobin and trifloxystrobin in precise virulence measurements for four fungi. These results suggested that dihydrobenzo[e][1,2]oxazepin-4(1H)-one analogues could be considered as potential fungicidal candidates for crop protection.

Abstract:To find more effective agricultural antimicrobial agents, a family of new 4-oxyquinazoline derivatives bearing a benzimidazole moiety were synthesized and evaluated for their antimicrobial properties. All the newly-synthesized compounds were characterized by 1 H NMR, 13 C NMR and high-resolution mass spectrometry (HRMS) techniques, and the structure of compound D16 was further confirmed by single-crystal X-ray diffraction method. The obtained results indicated that compound C with a N -unsubstituted benzimidazole moiety had an EC 50 (effective concentration for 50% activity) value of 1.17 μg/mL against Gibberella zeae in vitro , significantly more effective than the commercialized fungicides Chlorothalonil and Azoxystrobin (EC 50 = 12.98 and 4.88 μg/mL, respectively). Further in vivo bioassays revealed that this compound was also capable of inhibiting the proliferation of G. zeae on the potted corn plants, having an obvious curative efficacy of 50.8% at 100 μg/mL, superior to control agent Azoxystrobin (39.2%). Finally, antifungal mechanisms of compound C were preliminarily explored by use of several biophysical and biochemical methods, including the determinations of cytoplasmic content leakage and cell membrane permeability, scanning electron microscopy (SEM) observations, and fluorescence imaging studies. Compound C had the potential for further development as a new fungicide for the control of G. zeae .

Abstract:Fifteen new pyrazole-4-carboxylic oxime ester derivatives were synthesized from benzaldehyde oxime and 3-(trifluoromethyl)-1-methyl-1H-pyrazole-4-carbonyl chloride conveniently. These pyrazole-4-carboxylic oxime ester structures were confirmed by 1H NMR and HRMS. The compound 5f was further confirmed by X-ray diffraction. They crystallized in the monoclinic system, space group C2/c, Z = 8. The fungicidal activity results indicated that some of these compounds possessed good activity against S. sclerotiorum, B. cinerea, R. solani, P. oryae and P. piricola at 50 ppm. Furthermore, the structure-activity relationship was studied by molecular docking simulation.

Abstract:(E)-3-(2-chlorophenyl)-1-(2,4-dichlorophenyl) prop-2-en-1-one was prepared from 2-chlorobenzaldehyde followed by cyclization with hydrazine monohydrate. Eight new 3-(2,4-Di-chlorophenyl)-5-(2-chlorophenyl)-4,5-dihydro-N-acylpyrazole derivatives were synthesized and characterized by elemental analysis, IR and 1H NMR spectroscopy. The experimental results show that the inhibition ratio of compounds 3f towards H. Oryzae and P. oryzae at 50 mg·L−1 is 55.2% and 57.1%, respectively. The inhibition ratio of compounds 3g towards H. Oryzae, P. oryza, S. Sclerotiorum at 50 mg·L−1 is 53.3%, 60.0%, 50.4% respectively.
Synthesis and Fungicidal Activities of Biaryl Methanone O-Benzyl Oximes

Abstract:To discover new strobilurin analogs with high pesticidal activity, 11 novel methoxy acrylates bearing biaryl methanone oxime ether moieties were synthesized from aroyl pyridines, and characterized by H-1 and C-13 NMR and HRMS. In vitro fungicidal evaluation indicated that all of the target compounds exhibited excellent activities against tested seven phytopathogenic fungi, and compounds 5d and 5g displayed comparable overall activities to azoxystrobin and trifloxystrobin. Some of the target compounds also exhibited excellent in vivo control effects at 400 mg/L, for example, compound 5h could 100% control wheat powdery mildew and cucumber anthracnose. Furthermore, at 25 mg/L, these compounds, including control agents, completely inhibited spores germination of rice blast, but no effect at all for cucumber gray mold. These results suggested that this class of compounds might deserve to be developed as new potential agricultural fungicides.

Abstract:A series of oxazolidinone-based strobilurin analogues were efficiently synthesized by the reaction of 3-(2-bromomethyl-phenyl)oxazolidin-2-one with 1-substituted phenyl-2H-pyrazolin-3-one. Their structures were confirmed and characterized by 1 H-NMR, 13 C-NMR, elemental analysis, and mass spectroscopy. In addition, the crystal structure of the target compound 3-(2-((1-phenyl-2H-pyrazol-3-yloxy)methyl)phenyl) oxazolidin-2-one was determined by single crystal X-ray diffraction. The bioassay results ofthese compounds indicated that some ofthe oxazolidin-2-one derivatives containing N-substituted phenyl 2H-pyrazol ring exhibited potential in vivo fungicidal activities against M grisea at the dosage of 1 g L -1 .

Abstract:A series of pyrazolyl-heterocycles, including 1,3,4-thiodiazoles, 1,3,4-oxadiazoles and 1,3,4-triazores were synthesized from 4-nitro-pyrazole formhydrazide. The structures of air new compounds were confirmed by H-1 NMR and elementary analysis. Their fungicidal activity was evaluated against Rhizoctonia solani. Structure-activity relationships for the screened compounds were evaluated and discussed. It was found that same compounds showed a higher fungicidal activity and systematic activity.

Abstract:A series of 1,4-benzoxazin-3-one derivatives containing an acylhydrazone moiety were designed, synthesized and evaluated for their in vitro antifungal activities against Gibberella zeae , Pellicularia sasakii , Phytophthora infestans , Capsicum wilt , and Phytophthora capsica . The structures of target compounds were characterized by 1 H NMR, 13 H NMR, 19 F NMR and HRMS. The preliminary antifungal evaluation of all target compounds showed that some target compounds possessed moderate to good activities against G. zeae , P. sasakii , P. infestans and C. wilt . Among them, compounds 5L and 5o exhibited noticeable inhibition effects against G. zeae with the EC 50 values (effective concentration for 50% activity) of 20.06 and 23.17 μ g/ml, respectively, which were even nearly double effective than that of hymexazol (40.51 μ g/ml). Meanwhile, compound 5q displayed a notable inhibitory effect toward P. sasakii , with the EC 50 value of 26.66 μg/ml, which was better than that of hymexazol (32.77 μ g/ml). In addition, compound 5r yielded the EC 50 value of 15.37 μ g/ml against P. infestans , which was less than those of hymexazol (18.35 μ g/ml) and carbendazim (34.41 μ g/ml). Eventually, compound 5p showed higher inhibitory effect against C. wilt , with EC 50 value of 26.76 μ g/ml, which was better than that of hymexazol (>50 μ g/ml).

Abstract:At present, phenolic acid derivatives and triazole derivatives have a good antifungal effect, which has attracted widespread attention. A series of novel phenolic acid triazole derivatives were synthesized, and their structures were characterized by IR, MS, NMR, and X-ray crystal diffraction. Compound methyl 4-(2-bromoethoxy)benzoate, methyl 4-(2-(1H-1,2,4-triazol-1-yl) ethoxy)benzoate, 4-(2-(1H-1,2,4-triazol-1-yl)ethoxy)benzoic acid and 4-(2-(1H-1,2,4-triazol-1-yl) ethoxy)-3-methoxybenzoic acid crystallize in the monoclinic system with space group P21/n, the monoclinic system with space group P21, the monoclinic system with space group P21 and the orthorhombic system with space group Pca21, respectively. At a concentration of 100 μg/mL and 200 μg/mL, the antifungal activity against seven plant pathogen fungi was determined. Compound methyl 4-(2-bromoethoxy)benzoate has the best inhibitory effect on Rhizoctonia solani AG1, and the inhibitory rate reached 88.6% at 200 μg/mL. The inhibitory rates of compound methyl 4-(2-(1H-1,2,4-triazol-1-yl) ethoxy)benzoate against Fusarium moniliforme and Sphaeropsis sapinea at a concentration of 200 μg/mL were 76.1% and 75.4%, respectively, which were better than that of carbendazim.

Abstract:Ethyl 2-oxo-2H-chromene-3-carboxylate derivatives were synthesized and characterized by FT-IR, NMR, MS and X-ray crystal diffraction. Compound 3d crystallizes in the triclinic system with space group P-1. Compound 3e crystallizes in the monoclinic system with space group P21/c. The potential anti-fungal activities have been studied against five kinds of common fungi at concentrations of 200 ppm and 500 ppm. The research showed that the target compounds exhibit certain anti-fungal activity against the tested fungal strains. The inhibition rate of compound 3b was the highest against Fusarium oxysporum, up to 60.29% at 500 ppm. Compound 3b is promising to become the lead compound of pesticide in the future, which is worthy of further study.

Abstract:Compounds containing N‐pyridylpyrazole motif have aroused interest and brought about research hotspots due to their highly‐efficient insecticidal activity. The fungicidal potential of N‐pyridylpyrazole derivatives has gradually been disclosed in recent years. To discover new agrochemicals with poly‐heterocyclic features, a series of novel triazole thione/thioether derivatives containing pyridylpyrazole motif (8‐11) was synthesized. The new compounds were identified by melting point, 1H NMR, 13C NMR, 19F NMR, HRMS, and elemental analysis. The bioassays showed that most of the pyridylpyrazole‐containing triazole thione Mannich bases possessed favorable in vitro fungicidal activity toward pathogenic fungi, such as Magnaporthe oryzae, Sclerotinia sclerotiorum, Botrytis cinerea and Fusarium verticillioides, and were comparable with those of the contrast compounds A and triadimefon. Some of them exhibited moderate to good in vivo fungicidal activity against S. sclerotiorum at 0.2 mg/mL (e.g. 8f control efficacy: 60.9±3.2%). The SEM observation displayed that 8f might cause disruption of cell membrane and wall of S. sclerotiorum. Compounds 8a, 8c, 8f‐8h, 8p and 9b can serve as promising new fungicidal agents to make further structural optimization. Synthesis And Antifungal Activity Of Novel 1,5-Diphenyl-1,4-Pentadien-3-One Oxime Esters

Abstract:Fifteen 1,5-diphenyl-1,4-pentadien-3-one oxime esters were synthesized by the reaction of 1,5-diphenyl-1,4-pentadien-3-one oxmie with acyl chloride. Their structures were identified by elemental analysis, IR, H-1 NMR and C-13 NMR spectra. The preliminary bioassay showed that compounds 3i and 3o had certain antifungal activities against Fusarium graminearum with rate of 51.1% and 53.45% at a concentration of 50 mg center dot L-1 respectively, similarly to the control (hymexozole).
