Abstract:Seven 6-alkyl-2,3,4,5-tetrahydropyridines (5a-5g) that mimic the natural piperideines that were recently identified in fire ant venom have been synthesized. Compounds 5c-5g with C-6 alkyl chain lengths from C14 to C18 showed varying degrees of antifungal activities, with 5e (6-hexadecyl-2,3,4,5-tetrahydropyridine) and 5f (6-heptadecyl-2,3,4,5-tetrahydropyridine) being the most active. Compound 5e exhibited minimum fungicidal concentrations of 3.8, 15.0, 7.5, and 7.5 μg/mL against Cryptococcus neoformans, Candida albicans, Candida glabrata, and Candida krusei, respectively. The antifungal activities of these compounds appear to be associated with the C-6 side chain length. This study represents the first effort to evaluate antifungal activities of synthetic analogues of the newly identified fire ant venom alkaloids.

Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.

Abstract:Twenty-three novel trifluoromethyl pyrimidine derivatives containing an amide moiety were designed and synthesized through four-step reactions and evaluated for their antifungal, insecticidal, and anticancer properties. Bioassay results indicated that some of the title compounds exhibited good in vitro antifungal activities against Botryosphaeria dothidea ( B. dothidea ), Phompsis sp., Botrytis cinereal ( B. cinerea ), Colletotrichum gloeosporioides ( C. gloeosporioides ), Pyricutaria oryzae ( P. oryzae ), and Sclerotinia sclerotiorum ( S. sclerotiorum ) at 50 μg/ml. Meanwhile, the synthesized compounds showed moderate insecticidal activities against Mythimna separata ( M. separata ) and Spdoptera frugiperda ( S. frugiperda ) at 500 μg/ml, which were lower than those of chlorantraniliprole. In addition, the synthesized compounds indicated certain anticancer activities against PC3, K562, Hela, and A549 at 5 μg/ml, which were lower than those of doxorubicin. Notably, this work is the first report on the antifungal, insecticidal, and anticancer activities of trifluoromethyl pyrimidine derivatives bearing an amide moiety.

Abstract:Two series of N-pyridylpyrazole derivatives containing 1,2,3-triazole and quinazolin-4(3H)-one substructures were designed and synthesized. In total, 18 novel compounds were prepared, and all compounds were characterized by H-1 NMR, C-13 NMR and elemental analysis (EA). Preliminary bioassay results revealed that a few of new compounds with quinazolin-4(3H)-one moiety exhibited good insecticidal activity against the oriental armyworm (Mythimna separata). In addition, the compounds Ia-h with 1, 2, 3-triazole moiety showed broad-spectrum antifungal activities against Fusarium oxysporum f.sp.cucumerinum, Cercospora arachidicola Hori, Botryosphaeria dothidea, Alternaria solani, Gibberella zeae and Phytophthora capsici at 50 [mu g/mL concentration. The EC50 values of Ib and If against Botryosphaeria dothidea were 13.9 and 11.0 mu g/mL, respectively, which were comparable to Chlorothalonil. These results indicated the potential application of N-pyridylpyrazole derivatives as fungicide in further study.

Abstract:For this work, two series of new piperazine derivatives (3a–o) and triazolo-pyrazine derivatives (3p–t) were synthesized in a single-step reaction. All twenty adducts were obtained in good to high yields and fully characterized by 1H NMR, 13C NMR, IR, and mass spectrometry techniques. To further confirm the chemical identity of the adducts, a crystal of N-{[(4-chlorophenyl)-3-(trifluoromethyl)]-5,6-dihydro-[1,2,4]triazolo[4,3-a]}pyrazine-7(8H)-carboxamide (3t) was prepared and analyzed using X-ray crystallography. In vitro screening of the antimicrobial activity of all compounds (3a–t) was evaluated against five bacterial and two fungal strains. This study disclosed that N-{[(3-chlorophenyl)]-4-(dibenzo[b,f][1,4]thiazepin-11-yl)}piperazine-1-carboxamide (3o) was the superior antimicrobial with good growth inhibition against A. baumannii. Furthermore, the results from the performed molecular docking studies were promising, since the observed data could be used to develop more potent antimicrobials.

Abstract:Succinate dehydrogenase inhibitor is a low-toxic and high active fungicide. In order to develop novel and broad-spectrum succinate dehydrogenase inhibitor (SDHI) fungicides, thiophene was introduced into the structure of pyrazole carboxamide fungicides. Twenty four pyrazole-thiophene carboxamides were designed, synthesized and characterized by H NMR, C NMR and HRMS. The crystal structure of N-(4-methoxyphenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene2-carboxamide (7i) was determined by X-ray diffraction. The antifungal activity of all the synthesized compounds was determined against six plant pathogenic fungi, and preliminary bioassays suggested that some compounds exhibited good antifungal activity against Rhizoctonia solani, Fusarium graminearum and Botrytis cinerea. Among them, N-(4fluorophenethyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7c) exhibited the best antifungal activities against R. solani in vitro with EC50 value of 11.6 μmol/L, and N-(2-fluorophenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7j) against F. graminearum with EC50 value of 28.9 μmol/L. And N-(4-chlorophenyl)-4-(1-methyl-1H-pyrazol-4yl)thiophene-2-carboxamide (7h) showed similar inhibition abilities with thifluzamide against B. cinerea with EC50 value of 21.3 μmol/L. The molecular docking results showed that the high antifungal activitie compounds form stronger interactions with important amino acid residues of succinate dehydrogenase.

Abstract:According to the principle of combination of active groups, a series of piperazine derivatives containing pyridinemethyl/thiazolemethyl were designed and synthesized from 2-chloro-5-(chloromethyl) pyridine or 2-chloro-5-(chloromethyl)thiazole via N-alkylation and addition reactions. Some compounds showed weak herbicidal activity and insecticidal activity against Aphis fabae but good insecticidal activity against Culex pipiens, among which compound 4f exhibited 100% mortality against Culex pipiens at 50 mg/L.

Abstract:In order to find high activity and broad-spectrum insecticide,ten novel pyrimidone derivatives were designed via the replacement of pyridine ring of pymetrozine with the different charge density five-or six-membered rings.A facile and efficient synthetic method for the preparation of pyrimidone derivatives was described,in which the triazone ring and dihydroquinazolin-2(1H)-one was conveniently constructed.The structures of all the newly synthesized compounds were characterized by HRMS,1H NMR and IR spectroscopy.The insecticidal activity tests showed that these compounds didn’t exhibit significant insecticidal activity against Aphid scraccivora,which implied that the pyridine ring moiety of pyrimidone is essential for its insecticidal activity.

Abstract:To further explore the structure-activity relationship(SAR) of amide bridge moeity of anthranilic diamides derivatives, a series of N -pyridylpyrazole derivatives was designed, synthesized and their biological activities were evaluated. The chemical structures of novel target compounds were confirmed by 1 H nuclear magnetic resonance (NMR), 13 C NMR and elemental analyses(EA). Bioassay results of insecticidal activity demonstrated that the target compound 6 h displayed 70% lethality rate against oriental armyworms at 200 mg/L. Moreover, most compounds displayed moderate to excellent antifungicidal activities against Fusarium oxysporum f. sp . cucumerinum , Cercospora arachidicola Hori , Botryosphaeria dothidea , Alternaria solani , Gibberella zeae and Phytophthora capsici at 50 mg/L. In particular, compound 6 e showed 61.5% and 92.3% inhibition rate against Cercospora arachidicola Hori and Botryosphaeria dothidea , which was superior to the commercial positive control Chlorothalonil . These results will provide a potential clue for exploring novel high-effective agrochemicals.

Abstract:In this research, with an aim to develop novel pyrazole oxime ether derivatives possessing potential biological activity, thirty-two pyrazole oxime ethers, including a substituted pyridine ring, have been synthesized and structurally identified through 1H NMR, 13C NMR, and HRMS. Bioassay data indicated that most of these compounds owned strong insecticidal properties against Mythimna separata, Tetranychus cinnabarinus, Plutella xylostella, and Aphis medicaginis at a dosage of 500 μg/mL, and some title compounds were active towards Nilaparvata lugens at 500 μg/mL. Furthermore, some of the designed compounds had potent insecticidal effects against M. separata, T. cinnabarinus, or A. medicaginis at 100 μg/mL, with the mortalities of compounds 8a, 8c, 8d, 8e, 8f, 8g, 8o, 8s, 8v, 8x, and 8z against A. medicaginis, in particular, all reaching 100%. Even when the dosage was lowered to 20 μg/mL, compound 8s also expressed 50% insecticidal activity against M. separata, and compounds 8a, 8e, 8f, 8o, 8v, and 8x displayed more than 60% inhibition rates against A. medicaginis. The current results provided a significant basis for the rational design of biologically active pyrazole oxime ethers in future.

Abstract:A series of novel 1,3,4-thiadiazole derivatives bearing 1,2,4-triazolo[1,5-a]pyrimidine moiety were synthesized by the method of splicing active substructures. Among these derivatives, compounds 12, 13, 15–22 and 24–31 were firstly reported. All the compounds were assayed for antimicrobial activities against five fungi strains and four bacteria strains. The preliminary results indicated that compounds 25 and 28–31 showed good antifungal activities against Physaclospora piricola and Rhizoctonia solani. Compound 26 exhibited good antifungal activities against Cercospora beticola and R. solani. Most of the compounds showed better antibacterial activities against Gram-negative bacteria strains than Gram-positive bacteria strains. Compounds 25 and 28 showed the best activities against Pseudomonas fluorescence while compounds 30–31 showed good activities against Escherichia coli.

Abstract:A series of new piperidine-4-carbohydrazide derivatives bearing a quinazolinyl moiety were prepared and evaluated for their fungicidal activities against agriculturally important fungi. Among these derivatives, the chemical structure of compound A45 was clearly verified by X-ray crystallographic analysis. The antifungal bioassays revealed that many compounds in this series possessed good to excellent inhibition effects toward the tested fungi. For example, compounds A13 and A41 had EC50 values of 0.83 and 0.88 μg/mL against Rhizoctonia solani in vitro, respectively, superior to those of positive controls Chlorothalonil and Boscalid (1.64 and 0.96 μg/mL, respectively). Additionally, the above two compounds also exhibited notable inhibitory activities against Verticillium dahliae (with EC50 values of 1.12 and 3.20 μg/mL, respectively), far better than the positive controls Carbendazim and Chlorothalonil (19.3 and 11.0 μg/mL, respectively). More importantly, compound A13 could potently inhibit the proliferation of R. solani in the potted rice plants, showing good in vivo curative and protective efficiencies of 76.9% and 76.6% at 200 μg/mL, respectively. Furthermore, compound A13 demonstrated an effective inhibition of succinate dehydrogenase (SDH) activity in vitro with an IC50 value of 6.07 μM. Finally, the molecular docking study revealed that this compound could be well embedded into the active pocket of SDH via multiple noncovalent interactions, involving residues like SER39, ARG43, and GLY46.

Abstract:In this study, a series of novel difluoromethyl pyrazole derivatives were synthesized and their structures were confirmed by 1 H NMR, 13 C NMR, and HRMS techniques. Subsequently, their antifungal activities in vitro against Mucor fragilis ( M. fragilis ), Trichoderma atroviride ( T. atroviride ), and Mucor bainieri ( M. bainieri ) at concentrations of 50 and 100 μg/mL were evaluated using the mycelium growth rate assay. Bioactivity results showed that the target compounds revealed lower antifungal activities against M. fragilis , T. atroviride , and M. bainieri than those of prochloraz and carbendazim. Additionally, the antibacterial activities against Xanthomonas axonopodis pv. citri ( Xac ) and Xanthomonas oryzae pv. oryzicola ( Xoo ) were determined using the turbidimeter test method. The bioassay results demonstrated that some of the target compounds exhibited moderate to good antibacterial activities. Notably, 2-methoxyphenyl 3-(difluoromethyl)-1-methyl-1 H -pyrazole-4-carboxylate displayed superior antibacterial effects against Xoo and Xac with inhibition rates of 62, 86 and 53, 80%, respectively, compared to those of thiodiazole copper.

Abstract:Cyclocondensation reactions of 3-phenyl-1-(3-methyl-5-oxo-1-phenyl-2-pyrazolin-4-yl) prop-2-en-1-one (2) with active methylene reagents such as ethyl cyanoacetate, malononitrile, ethyl acetoacetate, and acetoacetanilide afforded pyrazolinyl pyridine derivatives (3 and 12), pyrazolinyl pyran derivative (9), and pyrazolinyl cyclohexene derivatives (10 and 11). Reaction of 11 with hydrazines and hydroxyl amine led to the formation of indazole derivatives (13 and 14), and benzisoxazole derivative 15. Also, reaction of 9 with different reagents gave pyrano[2,3-d][1,2,3]triazine (19) and pyrano[2,3-d]pyrimidine derivatives (20 and 21). The anti-inflammatory, analgesic, and antimicrobial activities of these compounds were determined. Compounds 13 and 14 showed good anti-inflammatory activity in comparison with the standard indomethacin, whereas compounds 4, 7, and 12 showed higher analgesic activity than the reference aspirin drug. In addition, compounds 7, 11, and 15 exhibited broad spectrum activities against all bacterial and fungal species tested.Graphical Abstract

Abstract:Nine novel pyrazole-acyl fluoro-substituted aromatic thiourea derivatives were synthesized from phenylhydrazine and ethyl acetoacetate.The intermediate 4 was prepared via a series of reactions involving cyclization, formylation,oxidation and acylation,and was subsequently reacted with amonium thiocyanate to generate the corresponding intermediate 5 with good yield in the presence of phase transfer catalyst PEG-600.The obtained intermidiate 5 was then reacted with fluoro-substituted anilines to form the desired pyrazole-acyl fluoro-substituted aromatic thiourea derivatives,and their structures were identified by IR,1H NMR and elemental analysis.Preliminary bioassays indicated that compounds 6a,6b,6d,6e,6f,6g, 6h and 6i showed inhibitory activities(100%) against Rhizoctonia sasakii,and compound 6e showed inhibitory activities(98.2%) against Gibberella zeae at a concentration of 50 mg/L.

Abstract:In an attempt to search for potent fungicide, a series of novel N-aryl-1H-pyrazole-5-carboxylate derivatives was designed and synthesized. Their chemical structures were characterized by 1H NMR spectra and high resolution mass spectrometry(HRMS). The preliminary bioassay results indicated that some target compounds displayed better fungicidal activities against certain fungi at 50 µg/mL or favorable antitumor activities at 5 µg/mL compared with chlorothalonil and 5-fluorouracil, respectively. The structure-activity relationship demonstrated that the introduction of ester group and amide bond was favorable to the improvement of activities against Physalospora piricola and Phytophthora capsici.

Abstract:Novel pyrazolecarbamide derivatives bearing a sulfonate fragment were synthesized to identify potential antifungal and antiviral agents. All the structures of the key intermediates and target compounds were confirmed by nuclear magnetic resonance (NMR) and high-resolution mass spectrometry (HRMS). The single-crystal X-ray diffraction of the compound T22 showed that pyrazole carbamide is a sulfonate. The in vitro antifungal activities of the target compounds against Colletotrichum camelliae, Pestalotiopsis theae, Gibberella zeae, and Rhizoctonia solani were evaluated at 50 μg/ml. Among the four pathogens, the target compounds exhibited the highest antifungal activity against Rhizoctonia solani. The compound T24 (EC50 = 0.45 mg/L) had higher antifungal activity than the commercial fungicide hymexazol (EC50 = 10.49 mg/L) against R. solani, almost similar to bixafen (EC50 = 0.25 mg/L). Additionally, the target compounds exhibited protective effects in vivo against TMV. Thus, this study reveals that pyrazolecarbamide derivatives bearing a sulfonate fragment exhibit potential antifungal and antiviral activities.
