Abstract:A gentle and effective method for the photocatalytic dual functionalization of allenes with high regio- and stereoselectivity using a nonmetallic catalyst is described. Inexpensive and easily available sulfinates and TMSN3 were employed as sulfone and azido sources, respectively. The method is characterized by satisfactory substrate compatibility and tolerance toward functional groups. The straightforward initial mechanistic experiments suggested that the reaction could follow a radical pathway. The synthesis of vinylsulfone azide derivatives presented here offers a promising scaffold for the future development of vinyl sulfone-based drugs and functional bioorthogonal reagents.

Abstract:A visible-light-induced highly efficient C(sp3)-H amination of ethers with amides and azoles has been presented under mild conditions via a nitrogen- and carbon-centered radical coupling process. This protocol successfully utilizes 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) and tert-butyl nitrite (TBN) as cocatalysts to deliver the aminated products of ethers under aerobic conditions. Notably, the developed reaction features the corresponding products in good yields (up to 93%) with a wide substrate scope. The mechanistic study indicates that C-N bond formation proceeds via a direct radical cross-coupling process. Preliminary biological activity analysis indicates that the resulting products have good and selective inhibitory activity on osteosarcoma (OS) cell lines and are promising for use as hits for drug discovery.

Abstract:Enamides are versatile precursors for synthesizing bioactive compounds. As their alkylations often require perstoichiometric amounts of oxidants, transition metals, or photocatalysts, we herein report a simple alternative for their alkylations by just using visible light to irradiate the mixture of the readily available N-hydroxyphthalimide esters and enamides without an additive. Abstract:A visible-light-promoted olefinic C-H trifluoromethylation of enamides was developed by employing cheap and stable Langlois' reagent as the CF3 source. A series of β-CF3 enamides were obtained in moderate to good yields with high E-isomer selectivity under mild conditions. Preliminary mechanistic studies suggest that molecular oxygen acts as the terminal oxidant for this net oxidative process, and the E isomer selectivity could be well explained by a base-assisted deprotonation of the cation intermediate.

Abstract:The enantioselective reduction of enamides is an efficient and atom-economical way to construct chiral amine motifs. In contrast to transition-metal complex based asymmetric hydrogenation, an alternative biocatalytic way is more sustainable but remains unexploited. The challenge lies in the redox potential mismatch between the ground-state enzymes and abiotic enamide substrates. Herein, we report a photoenzymatic strategy, expanding the reaction repertoires of biocatalysts to a previously elusive enantioselective reduction of enamides for the synthesis of enantioenriched amines (with up to a 99:1 enantiomeric ratio). Mechanistic wet experiments and computational studies revealed that direct visible-light excitation of the hydroquinone state flavin cofactor enhances the reducing capacity of ene-reductases, thereby triggering a biocatalytic single-electron reduction process. Mild and green reaction conditions as well as broad functional group tolerance make this unnatural photoenzymatic protocol a practical complement to traditional chemocatalytic approaches.

Abstract:A direct oxidative C-H amidation of heteroarenes with sulfonamides via nitrogen-centered radicals has been achieved. Nitrogen-centered radicals are directly generated from oxidative cleavage of N-H bonds under visible-light photoredox catalysis. Sulfonamides, which are easily accessed, are used as tunable nitrogen sources and bleach (aqueous NaClO solution) is used as the oxidant. A variety of heteroarenes, including indoles, pyrroles and benzofurans, can undergo this amidation with high yields (up to 92 %). These reactions are highly regioselective, and all the products are isolated as single regioisomer.

Abstract:Utilization of carbon dioxide (CO2) via visible‐light photocatalysis is an attractive but challenging research area. Herein, we report a novel strategy of arylcarboxylation of enamides with CO2 and aryl iodides to generate a range of α,α‐disubstituted α‐amino acids via visible‐light photoredox catalysis under mild conditions. This three‐component coupling reaction rapidly delivers valuable α‐amino acids from readily available enamides and aryl iodides, providing a distinct tool for synthetic chemists. To harness light for converting CO2 into value‐added chemicals is an attractive synthetic method. A novel strategy of arylcarboxylation of enamides with CO2 and aryl iodides under mild conditions is reported, leading to a range of α,α‐disubstituted α‐amino acids that are important building blocks. This visible‐light‐induced three‐component coupling reaction provides a distinct tool to form valuable α‐amino acids rapidly.

Abstract:In this work, we report a regioselective sulfonamidation of N-(2-hydroxyaryl)amides with iminoiodinanes and iodine in visible light at room temperature. The method does not require a strong oxidant, metal or photocatalyst and enables direct functionalization of C-H bond to C-N bond. Mechanistic investigations suggest an in-situ generation of N-centered radical from N,N-diiodo-sulfonamide by homolytic N–I bond cleavage followed by its site-specific addition to N-(2-hydroxyaryl)amides to furnish para-sulfonamide derivatives.
