Abstract:A Pd‐catalyzed C‐glycosylation reaction was developed by allyl‐allyl coupling process using Achmatowicz rearrangement products as donors and methylcoumarins as acceptors under mild conditions. This method featured regio‐ and diastereoselectivities, stereodivergent synthesis of C‐glycosides. The glycosyl donors with controlled stereodiversity and glycosyl acceptors with fluorescent performace further highlighted this methodology.

Abstract:Synthesis of 3,5-cis-3-amino glycals with a cis-fused cyclic sulfamidate group has been achieved by selective reduction of sulfamidate ketimine groups. The efficient access to the structurally unique glycals allowed the subsequent divergent synthesis of various naturally occurring 3-amino-2,3,6-trideoxysugars. In addition, Lewis acid-promoted glycosylation of the glycals provided a simple solution for the stereoselective installation of O and C-linked aglycons on the amino sugar scaffolds.

Abstract:A one-pot propargylation-amination-cycloisomerization protocol for the synthesis of highly substituted pyrroles from simple starting materials (1,3-dicarbonyls, primary amines and propargylic alcohols) is reported. Extensive screening of reaction conditions led to the development of InCl3 as a multifunctional catalyst for this process. The substrate scope was extensively studied and the results show moderate to very good yields of products. However, the reaction fails when diethyl malonate primary or secondary aliphatic propargylic alcohols or ammonium acetate or chloride are used as substrates. Instead, ammonium carbamate gave the desired result in the synthesis of N-H pyrroles.

Abstract:Precise and selective modification of carbohydrates is a critical strategy in producing diverse carbohydrate derivatives for exploiting their functions. We disclosed a simple, efficient, and highly regioselective and stereoselective protocol to controllable amination of 2-nitroglycals under mild conditions in 5 mins. A range of 3-amino-carbohydrates including 3-arylamino-2-nitro-glycals and 1,3-di-amino-carbohydrate derivatives were obtained in good to excellent yield with excellent stereoselectivity. The produced 3-amino-2-nitro-glycals can be used as a precursor for further transformation.

Abstract:C‐alkyl glycosides play a crucial role in various bioactive compounds. However, the synthesis of C‐alkyl glycosides poses significant challenges, particularly through C(sp3)‐H glycosylation. Here, we report a set of reactions for constructing C‐alkyl glycosides through directing‐group‐mediated functionalization of unactivated γ‐C(sp3)‐H bonds under mild conditions. Those reactions not only achieve high regioselectivity and stereoselectivity in glycosylation, but also exhibit a wide substrate scope. They are compatible with both arene and alkane substrates, as well as natural and unnatural amino acid substrates. Mechanistic studies have shown that the directing‐group 8‐aminoquinoline (AQ) and picolinamide (PA) may affect the chirality of the β‐carbon of L‐valine through a sterically favorable trans‐palladacycle intermediate, resulting in (R) or (S) configuration of glycosylated amino acid, respectively. These reactions are promising to provide a convenient and powerful tool for constructing C‐alkyl glycosides and carbohydrate‐based drugs in the future.
