Abstract:C‐alkyl glycosides play a crucial role in various bioactive compounds. However, the synthesis of C‐alkyl glycosides poses significant challenges, particularly through C(sp3)‐H glycosylation. Here, we report a set of reactions for constructing C‐alkyl glycosides through directing‐group‐mediated functionalization of unactivated γ‐C(sp3)‐H bonds under mild conditions. Those reactions not only achieve high regioselectivity and stereoselectivity in glycosylation, but also exhibit a wide substrate scope. They are compatible with both arene and alkane substrates, as well as natural and unnatural amino acid substrates. Mechanistic studies have shown that the directing‐group 8‐aminoquinoline (AQ) and picolinamide (PA) may affect the chirality of the β‐carbon of L‐valine through a sterically favorable trans‐palladacycle intermediate, resulting in (R) or (S) configuration of glycosylated amino acid, respectively. These reactions are promising to provide a convenient and powerful tool for constructing C‐alkyl glycosides and carbohydrate‐based drugs in the future.

Abstract:An efficient Pd-catalyzed C(sp3)-H selective iodination of 8-methylquinolines is reported herein for the first time. Because of the versatility of organic iodides, the method offers a facile access to various C8-substituted quinolines. By slightly switching the reaction conditions, an efficient C(sp3)-H acetoxylation of 8-methylquinolines has also been enabled. Both approaches feature mild reaction conditions, good tolerance of functional groups, and a broad substrate scope.

Abstract:A new protocol for the synthesis of 2‐deoxy‐C‐aryl‐glycosides and 2‐deoxy‐C‐alkyl‐glycosides via iridium‐catalyzed directed C—H glycosylation with glycals is reported. Abstract:This report describes the first example of palladium-catalyzed ortho-C-H glycosylation/ipso-alkenylation of aryl iodides, and the easily accessible glycosyl chlorides are used as a glycosylation reagent. The reaction is compatible with the functional groups of the substrates, and a series of C-aryl glycosides have been synthesized in good to excellent yield and with excellent diastereoselectivity. It is found that a cheap 5-norbornene-2-carbonitrile as a transient mediator can effectively promote this reaction. In addition, ipso-arylation and cyanation were also realized by the strategy.

Abstract:C-Aryl glycosides are of high value as drug candidates. Here a novel and cost-effective nickel catalyzed ortho-C-Ar-H glycosylation reaction with high regioselectivity and excellent alpha-selectivity is described. Abstract:C-oligosaccharides are pharmacologically relevant because they are more hydrolysis-resistant than O-oligosaccharides. Despite indisputable advances, C-oligosaccharides continue to be underdeveloped, likely due to a lack of efficient and selective strategies for the assembly of the interglycosidic C-C linkages. In contrast, we, herein, report a versatile and robust strategy for the synthesis of structurally complex C-oligosaccharides via catalyzed C(sp3 )-H activations. Thus, a wealth of complex interglycosidic (2→1)- and (1→1)-C-oligosaccharides becomes readily available by palladium-catalyzed C(sp3 )-H glycoside glycosylation. The isolation of key palladacycle intermediates and experiments with isotopically-labeled compounds identified a trans-stereoselectivity for the C(sp3 )-H glycosylation. The glycoside C(sp3 )-H activation manifold was likewise exploited for the diversification of furanoses, pyranoses and disaccharides.

Abstract:C-aryl glycosides are widely found in nature and play important roles in drug design. Despite the significant progress made over the past few decades, efficient and stereoselective synthesis of complex C-aryl glycosides remains challenging, lagging far behind the state of the art of the synthesis of O- or N-glycosides. Here, we report a simple and powerful bioinspired strategy for the stereoselective synthesis of C-aryl glycosides via palladium-catalysed ortho-directed C(sp2)−H functionalization of arenes and heteroarenes with easily accessible glycosyl chloride donors. The catalytic palladacycle intermediate generated via C−H palladation provides a soft aryl nucleophile that can react with glycosyl oxocarbenium ion partners with high efficiency and excellent stereocontrol. The method can be applied to a wide range of arene and heteroarene substrates, glycosyl chloride donors and auxiliary groups. It can simplify the synthesis of a variety of complex C-aryl glycosides and offers a tool for late-stage modification of drug molecules.

Abstract:C-Aryl glycoside is a class of structural motifs with important medicinal value. Herein, a remote C-H glycosylation strategy enabled by Ruthenium(II)-catalyzed with mostly alpha-stereoselectivity is described. This approach exhibits great functional group compatibility with various glycosides and arylamides. A series of experiments provided strong evidence for the free radical mechanism and reversible C-H activation process, and a possible mechanism was proposed.

Abstract:The construction of 2-deoxy-C-glycosides has gradually become a hotspot of carbohydrate chemistry in recent years. In this work, we present an efficient, regioselective, stereoselective and widely applicable strategy for the synthesis of 2-indolyl-C-deoxyglycosides via Ir(I)-catalyzed, pyridine-group-directed C-H functionalization. This method exhibits a high tolerance for the functional groups of indoles and the protecting groups of carbohydrates. stereoselectivity and mainly produces β-configuration products. Gram-scale synthesis and several practical transformations were conducted for further applications. Moreover, this protocol has good stereoselectivity and mainly produces β-configuration products. Abstract:Indole and quinoline structures are present in numerous biologically active molecules, making the synthesis of their glycosylation products a subject of extensive research and interest in drug development. Here, we report a photoredox strategy for the synthesis of C-indolyl and C-quinolyl glycosides using 2-styrylisocyanides and glycosyl bromides as building blocks. This approach offers mild reaction conditions, high α-selectivity, and scalability for large-scale reactions. The radical cyclization mode switching from 5-exo-trig to 6-endo-trig is achieved by selecting the substituents on the 2-vinyl group. This strategy enriches the toolbox of heterocyclic glycosylation methods and benefits advances in research on heteroaryl-based pharmaceuticals.

Abstract:α-Quaternary amino and glycolic acids have found application in many biologically relevant compounds and pharmaceuticals. For example, α-quaternary amino acids can act as modifiers of peptide conformation, compared with natural amino acids. Although there are numerous enantioselective methods for the synthesis of α-quaternary amino and glycolic acids through α-alkylation, α-arylation routes are challenging. Here we report two protocols for the enantioselective synthesis of chiral α-aryl quaternary amino acids and glycolic acids derivatives, respectively, using palladium catalysis with two unique sulfinamide phosphine (Sadphos) ligands. The methods employ two common heterocycles, azlactones and 5 H -oxazol-4-ones, as amino acid and glycolic acid precursors and show a broad substrate scope, with high yields and excellent enantioselectivity. Density functional theory calculations of the transition-state structures, studying non-covalent interactions and using natural bond orbital analysis, reveal that C–H···O interactions between the ligand and the substrate play a critical role in the efficient stereocontrol.

Abstract:An efficient and step-economical ligand-free palladium-catalyzed decarboxylative functionalization of quinolinone-3-carboxylic acids has been well developed. Note that the reaction could proceed smoothly under an argon atmosphere at relatively low temperature by using Pd(OAc) 2 as the catalyst and Ag 2 CO 3 as the oxidant. In this protocol, various functionalities were compatible under the reaction condition, and a series of biologically important azaisoflavone derivatives were obtained in moderate to high yields. Abstract:We report a method for the monoselective alkylation of ortho-C-H bonds of N-quinolyl benzamides with both primary and secondary alkyl halides under palladium catalysis. With promotion by NaHCO3 and (BnO)(2)PO2H or (PhO)(2)PO2H, symmetric benzamide substrates can be selectively ortho-alkylated to give either mono- or dialkylated products by simply adjusting the amount of NaHCO3 applied. The use of phosphate notably improves the alkylation yield, although it may not be directly involved in C-H palladation or the subsequent functionalization step. Kinetic isotope effect studies indicate that C-H palladation is not the rate-limiting step. Examination of the reactions of an isolated palladacycle intermediate with both cis- and trans-4-methylcyclohexyl iodides revealed surprising stereoretentive couplings of these alkyl iodides. This evidence strongly suggests that the functionalization of the palladacycle with secondary alkyl iodides proceeds via a rarely precedented concerted oxidative addition pathway.

Abstract:In recent years, the synthesis of C-aryl glycosides hrough C-H functionalization has attracted extensive attention of organic synthesis chemists due to its steps and atomic economy. In this concept, we systematically summarizes the synthesis of C-aryl glycosides with diverse regioselectivity and diastereoselectivity from the perspective of C-H arylation of glycosides and C-H glycosylation of arenes. It can be found that a series of recently developed C-H glycosylation reactions have higher site-selectivity and diastereomeric selectivity than Friedel-Crafts glycosylation reaction. The reaction conditions are milder, which can be compatible with acid-sensitive protective groups, such as acetals or ketals, and the deprotection is more convenient. It can be seen that there are few reports on remote C-H glycosylation of aromatic hydrocarbons, which is a new field and needs further research. In addition, C-H glycosylation has a lot of shortcomings, which need to be further explored: a) the precise regulation of stereoselectivity in the reaction process also needs further optimization; b) the research on the reaction mechanism is almost limited to DFT calculation, and there is no exact experimental evidence. For key parts, such as the specific reaction mechanism between cyclo-metal intermediates and glycosyl donors in ortho-C-Ar-H glycosylation is still unclear; c) due to the fact that aryl glycoside compounds contain bare hydroxyl groups in practical applications, it is an urgent problem to realize the compatibility of glycoside substrates containing naked hydroxyl groups or to remove the protective groups on hydroxyl groups by a mild and efficient method after the reaction; d) In this rapidly developing field, we need to study a greener, more economical and more practical C-H glycosylation of arenes in the future, which will be conducive to the synthesis of C-aryl glycosides with more biological application significance.
