Abstract:Catalytic or stoichiometric amounts of Lewis acids were found to be very effective α-directing additives in the stereoselective glycosylations of diverse 2,3-O-carbonate-protected glucose and galactose thioglycoside donors by preactivation protocol. The poor stereoselectivities of 4,6-di-O-acetyl-2,3-O-carbonate protected thioglycoside donors in glycosyl coupling reactions were greatly improved, and excellent α-stereoselectivities were achieved by the addition of 0.2 equiv of BF(3)·OEt(2). On the other hand, the β-selectivities of 4,6-di-O-benzyl-2,3-O-carbonate-protected thioglucoside donor toward glycosylations were reversed completely to the α-selectivities by the use of 1 equiv of SnCl(4), making the stereoselectivity controllable. Furthermore, the poor stereoselectivities of 4,6-di-O-benzyl-2,3-O-carbonate-protected thiogalactoside donor in glycosylations were also improved by using SnCl(4) as additive.

Abstract:Based on a pre-activation protocol, the stereoselectivity of oxazolidinone-protected amino sugar thioglycoside donors towards glycosylations can be controlled by additives. Either alpha-or beta-selectivity could be obtained by changing additives. 2,4,6-Tri-tert-butylpyrimidine (TTBP) was the best beta-directing additive, while thiophene worked as the best alpha-directing additive. The bifunctional additives such as tetrabutyl ammonium iodide (TBAI) afforded either alpha-or beta-selectivity depending on the amount added. Poor alpha-selectivity of some glycosylations without any additives was greatly improved by adding TBAI or thiophene.

Abstract:A series of novel glycosyl allenoates were prepared by condensions of hemiacetals with 3,3-dimethyl-4,5-allenoic acid. Their glycosylations with various alcohols under the promotion of bromodimethylsulfonium bromide/silver triflate smoothly proceeded to produce disaccharides. Studies reveal that allenic group is crucial for the efficient glylcosylation of glycosyl allenoate.

Abstract:A BF3 center dot CH3CN-promoted synthesis of aryl alpha-O-glycosides from readily available glycosyl donors was developed. This reaction could proceed well on a gram-scale under mild condition with alpha/beta ratio up to 95: 5. Moreover, a variety of the glycosyl donors including acetylated/benzoylated pyranoses, di- and trisaccharide derivatives as well as glycosyl acceptors from substituted phenols, sterically hindered aliphatic alcohols and carboxylic acid, were well tolerant. Importantly, the potential of this strategy was highlighted with a successful gram-scale synthesis of oligosaccharide EPS-G7, which is a commercial detection reagent for quantitative determination of alpha-amylase.
Pre-activation Based Stereoselective Glycosylations: Stereochemical Control by Additives and Solvent

Abstract:Stereochemical control is an important issue in carbohydrate synthesis.Glycosyl donors with participating acyl protective groups on 2-O have been shown to give 1,2-trans glycosides reliably under the pre-activation based reaction condition.In this work,the effects of additives and reaction solvents on stereoselectivity were examined using donors without participating protective groups on 2-O.While several triflate salt additives did not have major effects,the amount of AgOTf was found to significantly impact the reaction outcome.Excess AgOTf led to lower stereochemical control presumably due to its coordination with the glycosyl triflate intermediate and a more SN1 like reaction pathway.In contrast,the stereoselectivity could be directed by reaction solvents,with diethyl ether favoring the formation of glycosides and dichloromethane leading to β isomers.The trend of stereochemical dependence on reaction solvent was applicable to a variety of building blocks including the selective formation of β-mannosides.

Abstract:The generation of thioesters through thioesterification of carboxylic acid has significant synthetic value given the easy availability of various carboxylic acids and the extensive application of thioesters. An economical and rapid thioester synthesis approach is still needed. Herein, we achieved thioesterification of carboxylic acids with valuable glycosyl thiol, aromatic and aliphatic thiols mediated by sulfonyl fluoride (SO2F2). The compatibility of numerous carboxylic acid substrates was tested, including (hetero)aromatic and aliphatic carboxylic acids and the reaction proceeded smoothly under mild conditions at good yields. Late‐stage modification of drug molecules rac‐Naproxen and Loxoprofen proved feasible. Besides, the gram‐scale reaction demonstrated the utility of this protocol.

Abstract:Reported herein is a new reaction for glycosylation with thioglycosides in the presence of iron(III) chloride. Previously, FeCl3 was used for the activation of thioglycosides as a Lewis acid co-promoter paired with NIS. In the reported process, although 5.0 equiv of FeCl3 are needed to activate thioglycosides most efficiently, no additives were used, and the reactions with reactive glycosyl donors smoothly proceeded to completion in 1 h at 0 °C. This work showcases a new direction in developing glycosylation methods using greener and earth-abundant activators.

Abstract:Abstract Based on SnCl 2 ‐catalyzed acetalation and selective benzoylation, a one‐pot strategy to efficiently synthesize orthogonally protected glycosyl acceptors with 2‐OH/3‐OH was developed. Consequently, 2‐OBz or 3‐OBz 4,6‐ O ‐benzylidene galactosides and glucosides were efficiently prepared in moderate to high yields starting from free galactosides and glucosides, and were used as valuable glycosyl acceptors for the synthesis of blood group antigens O and B analogues in this study.

Abstract:Whereas aminosugars are an important component in a variety of bioactive molecules, their stereoselective formation is made challenging by the Lewis basic nature of amino substituents. Additionally, the use of N-acyl protecting groups is often problematic due to competing formation of oxazolines during the glycosylation of 2-aminosugar derivatives. Herein, we report a boron-catalyzed strategy utilizing silyl ether glycosyl acceptors and 2-aminosugar donors that utilize the 2,2,2-trichloroethoxycarbonyl (Troc) protecting group for the C2 amino functionality in glycosyl fluorides. This modification allows for operationally simple, room-temperature glycosylations and features a rapid reaction profile that addresses some of the limitations in the synthesis of 2-amino-2-deoxy sugar containing glycosides. Tailoring the order of reactivity of the silyl acceptors enables one-pot iterative glycosylations, thus streamlining the synthesis of complex oligosaccharides while allowing fewer intermediates and purification steps.

Abstract:Glycosylation reaction involving the coupling of a glycosyl donor with a glycosyl acceptor is one of the cornerstones of chemical preparation of pure glycans and glycoconjugates of biological relevance. Catalytic glycosylation of glycosyl ester donors and thioglycosides is an attractive but underexplored topic in carbohydrate chemistry. Herein, triflic imide (Tf2NH)‐catalyzed glycosylation of various O‐, S‐, and C‐nucleophiles has been achieved using disarmed glycosyl ortho‐isopropenylphenylacetates (GIPPAs) and ortho‐isopropenylbenzyl thioglycosides as glycosylating agents. The reactions proceed under mild conditions to give the desired glycosides in good‐to‐excellent yields. Of particular note, the comparable reactivity for the α‐isomers and the β‐ones of GIPPAs is observed. The mechanistic investigation demonstrates that the isopropenyl group is essential for the reaction and its preferential protonation triggers the reaction. This work provides another member to the arsenals of glycosyl ester and thioglycoside donors suitable for acid‐catalyzed glycosylation to create various glycosidic bonds. An efficient protocol for Tf2NH‐catalyzed glycosylation of disarmed glycosyl ortho‐isopropenylphenylacetates and ortho‐isopropenylbenzyl thioglycosides has been established. Mechanistically, the reaction involves the preferential protonation of the isopropenyl group, the generation of reactive glycosylating species by the cation‐triggering intramolecular cyclization, and the ensuing glycosylation event.

Abstract:3,5-Di(trifluoromethyl)phenyl(cyano)iodonium triflate is described as an accessible, stable, and powerful thiophile that can activate batches of p-tolyl thioglycoside donors at room temperature. Various alcoholic acceptors were efficiently glycosylated, providing the desired glycosides. The novel activation protocol features mild conditions as well as high compatibility with some classic strategies for the stereoselective construction of some biologically relevant glycosidic linkages, as exemplified by α-idosides, α-galactoamines, β-mannosides, and β-rhamnosides.

Abstract:Here, we present 3,4‐dicyano‐2,5,6‐trifluorophenyl glycosides as bench‐stable, and reactive electron‐deficient fluoroarene based glycosyl donors that can be prepared in one‐step and be activated with catalytic amounts of TMSOTf. The glycosylation reaction conditions show tolerance towards acid/base sensitive protecting groups and have a different substrate scope with 68‐89% yields. The electron‐deficient fluoroarene‐containing glycosyl donor enables the construction of O‐ and C‐glycosides using both free alcohols and silylated alcohols, as well as C‐ nucleophiles as acceptors.

Abstract:AbstractWe have developed a highly efficient and practical approach for palladium‐catalyzed trifluoroacetate‐promoted N‐quinolylcarboxamide‐directed glycosylation of inert β‐C(sp3)−H bonds of N‐phthaloyl α‐amino acids with glycals under mild conditions. For the first time, C(sp3)−H activation for glycosylation was achieved to build C‐alkyl glycosides. This method facilitates the synthesis of various β‐substituted C‐alkyl glycoamino acids and offers a tool for glycopeptide synthesis.
