Abstract:A new eremophilane acid was isolated in the phytochemical investigation of the roots of Ligularia. sagitta. Its structure and stereochemistry were elucidated by ID NMR and 2D NMR techniques including H-1-H-1 COSY, C-13-H-1 COSY, C-13-H-1 COLOC and NOESY.

Abstract:One new triterpene and seven new eremophilane-type sesquiterpenes were isolated from the roots of Ligularia sagitta, among them la, an O-acetylated oleanane type triterpene, 2a and 3a, two 11-nor eremophilane-type sesquiterpenes, 4a, 5, and 6, three standard eremophilane-type sesquiterpenes, and 7 and 8, two tri-nor eremophilane-type sesquiterpenes. Their structures were established by extensive spectral analyses (1D, 2D-NMR, IR, and MS). Compound la showed moderate cytotoxicity against two tumor cell lines.

Abstract:OBJECTIVE:To study the chemical constituents of radix and rhizome of Ligularia sagitta.METHOD:Isolation and purification were carried out on the column chromatography of silica gel and sephadex LH -20. The structures were identified by spectral analysis.RESULT:Seven terpenoid compounds were isolated from the radix and rhizome of L. sagitta. Five of them are sesquiterpenoids, the structures were determined as 7alpha-hydroxy-9(10) -ene-1, 8-dioxo-6, 7-dihydrofuranoeremophilane (1), 1p, 10P3-epoxy-6beta, 8 beta-dihydroxy-eremophil7 (11) -en-12, 8alpha-olide (2) , 1-oxo-9-desoxycacalol (3), benzofuranoeremophil-l-ene (4) and bakkenolide (5); other two compounds belong to triterpenoid. 3p, 16P3-dihydroxy-12-oleanen-28-al (6) and lupeol (7).CONCLUSION:Compoundsl-3 and 6 were: isolated from L. sagitta for the first time, and compounds 3 and 6 were obtained from Ligularia for the first time.

Abstract:A new sesquiterpene, 7α-H-11-hydroxyeremophil-1(10)-en-2-one (1), together with 14 known sesquiterpenoids (2–15), were isolated from the extract of the whole plant of Ligularia fischeri. Among them, compounds 2–6 and 15 were isolated from Ligularia fischeri for the first time. The new compound was characterized by means of spectroscopic methods including 1D and 2D NMR and HR-ESI-MS, and the known ones were established by comparing their NMR data with those of the corresponding compounds in the literature.

Abstract:Phytochemical investigation of the roots of Ligularia dentata has resulted in the isolation of three new bisabolane sesquiterpenes: 1alpha,3beta,10,11 -tetrahydroxy-2alpha,5alpha,8-triangeloyl-oxybisabola-(7)14-en-4-one, 1alpha,3beta,10-trihydroxy-11 -methoxy-2alpha,5alpha,8-triangeloyloxybisabola-7(14)-en-4-one, and lalpha, 3beta,10-trihydroxy-2alpha,5alpha,8-triangeloyloxybisabola-7(14),11(12)-dien-4-one. Their structures were elucidated with spectro-scopic methods and 2D-NMR techniques as well as chemical transformations. In addition, three known phenolic norsesquiterpenes, liguhodgsonal, ligujapone, and 2-hydroxy-platyphyllide have also been obtained.

Abstract:A new eremophilane norsesquiterpenoid (1), together with a known eremophilane sesquiterpenoid (2), was isolated from the leaves of Ligularia virgaurea. The structure of 1 was elucidated by a combination of spectroscopic analysis (IR, 1D NMR, 2D NMR, and HR-ESI-MS), and its absolute configuration was determined by a single-crystal X-ray diffraction experiment (with copper radiation). The known compound 2 was identified by comparison of its physical and spectral data with those reported in the literature. Compound 1 was assayed for its cytotoxic activities against human cervical carcinoma cell (HeLa) and human small cell lung cancer cell (NCI-446) lines.

Abstract:Chemical investigation of the roots of Ligularia sagitta has resulted in the characterization of six eremophilenolides 60,80-dimethoxy-10beta-hydroxyeremophil-7(11)-en-12,8alpha-olide (1), 6beta-angeloyloxy-10beta-hydroxy-8beta-methoxyeremophil-7(11)-en-12,8alpha- olide (2), 6beta-(2'-methylbutanoyloxy)-10beta-hydroxy-8beta-methoxyeremophil-7(11)-en-12,8alpha-olide (3), 6beta-angeloyloxy-10beta-hydroxy-8alpha-methoxyeremophil-7(11)-en-12,8alpha-olide (4), 6beta-(2'-methylbutanoyloxy)-10beta-hydroxy-8alpha-methoxyeremophil-7(11)-en- 12,8beta-olide (5) and 8beta,10beta-dihydroxy-6beta-methoxyeremophil-7(11)-en-12,8alpha-olide (6), together with one monoterpene (3R,4R,6S)-3,6-dihydroxy-1-menthene (7), two triterpenes lupeol (8) and ursolic acid (9), and beta-sitosterol (10). The structures of five new constituents (1-5) were elucidated by spectroscopic methods including 2D-NMR experiments. The compounds 1, 5 and 7 showed antibacterial activity by being assayed against Staphylococcus aureus, Bacillus subtilis and Escherichia coli.

Their structures have been elucidated on the basis of their spectroscopic studies (UV, IR, MS, H-1, C-13 NMR and DEFT) including 2D NMR experiments (HMQC and HMBC).

Abstract:Three new eremophilane-type sesquitererpenes (1, 2 and 3) were isolated from the roots of Ligularia macrophylla. On the basis of spectroscopic methods including 2D NMR and X-ray diffraction experiment, their structures were elucidated as 6 alpha, 15 beta-epoxy-8 beta-hydroxy-7(11)-8 alpha, 12-olide (1), 7-acetyl-6(7)-en-noreremophil- 13 beta-carboxylic acid (2) and eremophil-7(11)-en-6 alpha, 15 beta-olide (3). In addition, antibacterial activity and cytotoxicity activity of compound 1 were tested.
