Abstract:To improve repeatability and efficiency and to simplify the operation procedure of capillary electrophoresis (CE), a pressurized CE system (p-CE) with injection valve sampling was developed. It consisted of one high-pressure pump, a six-port injection valve, a PEEK cross, a separation and back pressure capillary, an ultraviolet-visible detector and a high voltage power supply. The pressure-driven flow ranging from 4.5 nL min(-1) to 0.81 μL min(-1) in the separation capillary was produced by splitting to the flow from the high-pressure pumps (0.005-0.4 mL min(-1)). Nano-volume sample injection (<10 nL) was conducted by a micro-volume rotary injector (0.5-5 μL) with flow splitting. In the p-CE system, the new commercial capillary could be directly used without any wash, and the capillary-flush process between runs was also eliminated. In this case, the analytes were driven toward the outlet of the separation capillary by the pressurized flow, the electric field force and minute electroosmotic flow, and they were separated owing to the electrophoretic mobility. The p-CE system allows for the independent variation of the pressurized flow rate and electrical field and electrophoretic separation of good repeatability (below 3%) under high electrical fields (500-1000 V cm(-1)) and flow rate gradient modes. The feasibility of the p-CE system in real analysis was demonstrated by iodate quantification in iodized table salts. The separation of iodide and iodate was realized within 0.3 min, proving its high analytical speed.

Abstract:Micromachined capillary electrophoresis systems with integrated sample injector have been fabricated on glass chips using standard photolithography and chemical wet etching techniques. Experiments have been done to find proper ways of obtaining regular and smooth channels, and the performance of the chip was evaluated. Voltages of 4 kV could be applied over an 20 mm long capillary channel with sufficient dissipation of the Joule heat generated (1.12 W/cm). Electroosmotic flow was used to "load" the sample into the microchip and to "pump" the mobile phase during the experiments. Using laser-induced fluorescence detection and CCD imaging, the mixture of FITC-OH and FITC-labeled amino acid were injected and separated. Two methods of sample introduction ("floating" and "pinched") were evaluated. The results showed that applications of potentials to the intersecting channels could cut the sample plug clearly during the process of separation which made the tailing unserious. Frequent flushing of channels with alkaline solution and water was demonstrated to be important to keep the reproducibility of sample loading and separation.

Abstract:Microfabrication techniques used for the production of MEMS (micro electro-mechanical systems) have been successfully used to produce highly efficient microfluidic capillary electrophoresis chip systems. A limitation of this approach are the difficulties associated with the creation of the micrometer-sized structures in glass or other substrates, which currently involve specialized and expensive lithographic and etching processes. A further limitation is that hitherto most microfluidic chips are not designed for continuous introduction of a series of different samples, which limits the overall throughput of such systems. This article reviews the development of a microfluidic system for rapid CE separations, produced at a low cost of less than a dollar each, using equipment and materials readily available in the ordinary laboratory. Applications of the system, after coupling to flow-injection and/or sequential-injection sample introduction, for the determination of FITC-labeled amino acids by laser-induced fluorescence, trace metals by chemiluminescence, carbohydrates by amperometry, and inorganic and organic anions by indirect UV absorbance are exemplified to illustrate the performance and versatility of the microfluidic system.
An Automated Electrokinetic Continuous Sample Introduction System for Microfluidic Chip-Based Capillary Electrophoresis

Abstract:An automated and continuous sample introduction system for microfluidic chip-based capillary electrophoresis (CE) was developed in this work. An efficient world-to-chip interface for chip-based CE separation was produced by horizontally connecting a Z-shaped fused silica capillary sampling probe to the sample loading channel of a crossed-channel chip. The sample presentation system was composed of an array of bottom-slotted sample vials filled alternately with samples and working electrolyte, horizontally positioned on a programmable linearly moving platform. On moving the array from one vial to the next, and scanning the probe, which was fixed with a platinum electrode on its tip, through the slots of the vials, a series of samples, each followed by a flow of working electrolyte was continuously introduced electrokinetically from the off-chip vials into the sample loading channel of the chip. The performance of the system was demonstrated in the separation and determination of FITC-labeled arginine and phenylalanine with LIF detection, by continuously introducing a train of different samples. Employing 4.5 kV sampling voltage (1000 V cm(-1) field strength) for 30 s and 1.8 kV separation voltage (400 V cm(-1) field strength) for 70 s, throughputs of 36 h(-1) were achieved with <1.0% carryover and 4.6, 3.2 and 4.0% RSD for arginine, FITC and phenylalanine, respectively (n = 11). Net sample consumption was only 240 nL for each sample.
A Compact and Low-Cost Miniaturized Analysis System Composed of Microchip Electrophoresis and Chemiluminescence Detection Manipulated by a Simple Subatmospheric Pressure Fluid-Driven Device

Abstract:A portable and low-cost miniaturized analysis system was proposed, in which microchip electrophoresis (MCE) and chemiluminescence (CL) were used as the separation and detection units, respectively. A porous monolithic plug was created in the separation channel of the microchip as a select valve, which prevented the pressure-driven CL reagents flowing back into the separation channel but allowed electrophoretic migration along the separation channel. The sensitivity was greatly enhanced by improving the mixing efficiency using a spiral detection channel with an increased length facing the photomultiplier tube (PMT). Peak width could be significantly reduced by increasing the flow rate of CL reagents. Putting grounding electrode before the detection channel significantly improved the reproducibility. A simple and compact subatmospheric pressure fluid-driven device was developed for manipulating the whole analytical process, including variable-volume sample loading, electrophoretic separation and CL reagents transportation. All miniaturized components for constructing a portable MCE-CL system are commercially available. This approach considerably simplified the operation and equipments for constructing an efficient MCE-CL system. Nine metal cations, such as Cr(III), Co(II), Cu(II), Ni(II), Au(III), Mn(II), Zn(II), Pt(II), Pb(II) were successfully separated within 200 s. Migration time precisions ranging from 0.39% for Cr(III) to 2.1% for Cu(II) were obtained for ten consecutive determinations with peak height precisions from 1.67% for Co(II) to 5.73% for Pb(II). Detection limits ranging from 7.5 x 10(-11) mol L(-1) for Co(II) to 8.3 x 10(-9) mol L(-1) for Pb(II) were achieved, which were about three orders lower than the present MCE-CL system. It has been applied for the determination of metals in tea. The results with the recoveries from 97.0% to 102.3% proved that the proposed MCE-CL system offers a number of benefits including miniaturization, high sensitivity and better resolution.

Abstract:A simple method for injecting well-defined non-biased sample plugs into the separation channel of a microfluidic chip-based capillary electrophoresis system was developed by a combination of flows generated by negative pressure, electrokinetic and hydrostatic forces. This was achieved by using only a single syringe pump and a single voltage supply at constant voltage. In the loading step, a partial vacuum in the headspace of a sealed sample waste reservoir was produced using a syringe pump equipped with a 3-way valve. Almost instantaneously, sample was drawn from the sample reservoir across the injection intersection to the sample waste reservoir by negative pressure. Simultaneously, buffer flow from the remaining two buffer reservoirs pinched the sample flow to form a well-defined sample plug at the channel intersection. In the subsequent separation stage, the vacuum in headspace of the sample waste reservoir was released to terminate all flows generated by negative pressure, and the sample plug at the channel intersection was electrokinetically injected into the separation channel under the potential applied along the separation channel. The liquid levels of the four reservoirs were optimized to prevent sample leakage during the separation stage. The approach considerably simplified the operations and equipment for pinched injection in chip-based CE, and improved the throughput. Migration time precisions of 3.3 and 1.5% RSD for rhodamine123 (Rh123) and fluorescein sodium (Flu) in the separation of a mixture of Flu and Rh123 were obtained for 56 consecutive determinations with peak height precisions of 6.2% and 4.4% RSD for Rh123 and Flu, respectively.

Abstract:A novel microfluidic picoliter-scale sample introduction approach was developed by combining the spontaneous injection approach with a capillary electrophoresis (CE) system based on a short capillary and slotted-vial array. A droplet splitting phenomenon at the capillary inlet end during the spontaneous sample introduction process was observed for the first time. On the basis of this phenomenon, a translational spontaneous injection approach was established to reduce sample injection volumes to the sub-100 pL range. A versatile high-speed capillary electrophoresis (HSCE) system was built on the basis of this sample injection approach with separation performance comparable to or even better than those reported in microfluidic chip-based CE systems. The HSCE system was composed of a short fused-silica capillary and an automated sample introduction system with slotted sample and buffer reservoirs and a computer-programmed translational stage. The translational spontaneous sample injection was performed by linearly moving the stage, allowing the capillary inlet first to enter the sample solution and then removing it. A droplet was left at the tip end and spontaneously drawn into the capillary by surface tension effect to achieve sample injection. The stage was continuously moved to allow the capillary inlet to be immersed into the buffer solution, and CE separation was performed by applying a high voltage between the buffer and waste reservoirs. With the use of the novel system, high-speed and efficient capillary zone electrophoresis (CZE) separation of a mixture of five fluorescein isothiocyanate (FITC) labeled amino acids was achieved within 5.4 s in a short capillary with a separation length of 15 mm, reaching separation efficiencies up to 0.40 microm plate height. Outstanding peak height precisions ranging from 1.2% to 3.7% RSD were achieved in 51 consecutive separations. By extension of the separation length to 50 mm, both high-speed and high-resolution CZE separation of eight FITC-labeled amino acids could be obtained in less than 21 s with theoretical plates ranging from 163,000 to 251,000 (corresponding to 0.31-0.20 microm plate heights). The present HSCE system also allowed fast chiral separations of FITC-labeled amino acids under micellar electrokinetic chromatography (MEKC) mode within 6.5 s.

Abstract:A suction-free interfacing method was developed for microchip electrophoresis hyphenated with inductively coupled plasma mass spectrometry (MCE-ICP-MS). The hyphenated system was composed of a microchip, a demountable capillary microflow nebulizer (d-CMN) combined with a heated single pass spray chamber, a negative pressure sampling device, a high voltage power supply, a syringe pump and an ICP-MS. To eliminate the nebulizer suction generated by the pneumatic nebulizer and to ensure that the makeup solution flowed into the nebulizer, two porous polymer plugs were fabricated in the microchip. As a result, reasonably true electropherograms were obtained when compared to the CE separation performed in the traditional MCE-ICP-MS mode without porous polymer plugs. Electrophoretic separation of I I- and IO3- was achieved within 25 s in a microchip with an effective separation length of only 15 mm at an electric field of 857 V cm(-1) using 10 mmol L-1 borate (pH 9.2) as the running buffer. A resolution of 1.3 was obtained and the absolute detection limits for I- and IO3- were 0.12 and 0.13 fg, respectively. The precisions (RSD, n = 10) of the migration time and peak height for I- and IO3- were in the range of 1.1-1.6% and 2.5-2.8%, respectively. Two table salt samples were analyzed by an external calibration method. The iodate contents were in accordance with their labeled values. Abstract:A sequential injection micro-sample introduction system was coupled to a microfluidic-chip based capillary electrophoresis system through a split–flow sampling interface integrated on the micro-chip. The microfluidic system measured 20×70×3mm in dimension, and was produced using a non-lithographic approach with components readily available in the analytical laboratory. In the H-configuration channel design the horizontal separation channel was a 75μm I.D.×60mm quartz capillary, with two vertical side arms produced from plastic tubing. The conduits were embedded in silicon elastomer with a planar glass base. Sequential introduction of a series of samples with about 2.5% carryover was achieved at 48h−1 throughput with samples containing a mixture of fluorescein isothiocyanate (FITC)-labeled amino acids using SI sample volumes of 3.3μl and carrier flow-rate of 2.0mlmin−1. Baseline separation was achieved for FITC-labeled arginine, phenylalanine, glycine and FITC (laser induced fluorescence detection) in sodium tetraborate buffer (pH 9.2) within 8–80s, at separation lengths of 25–35mm and electrical field strengths of 250–1500Vcm−1, with plate heights in the 0.7–3μm range.

Abstract:A miniaturized CE system has been developed for fast DNA separations with sensitive fluorimetric detection using a rectangle type light-emitting diode (LED). High sensitivity was achieved by combining liquid-core waveguide (LCW) and lock-in amplification techniques. A Teflon AF-coated silica capillary on a compact 6x3 cm baseplate served as both the separation channel for CE separation and as an LCW for light transmission of fluorescence emission to the detector. An electronically modulated LED illuminated transversely through a 0.2 mm aperture, the detection point on the LCW capillary without focusing, and fluorescence light was transmitted to the capillary outlet. To simplify the optics and enhance collection of light from the capillary outlet, an outlet reservoir was designed, with a light transmission window, positioned directly in front of a photomultiplier tube (PMT), separated only by a high pass filter. Automated sample introduction was achieved using a sequential injection system through a split-flow interface that allowed effective release of gas bubbles. In the separation of a phiX174 HaeIII DNA digest sample, using ethidium bromide as labeling dye, all 11 fragments of the sample were effectively resolved in 400 s, with an S/N ratio comparable to that of a CE system with more sophisticated LIF.

Abstract:A newly designed capillary electrophoresis (CE) microchip with a simple and efficient sample introduction interface is described. The sample introduction is carried out directly on the separation channel through a sharp inlet tip placed in the sample vial, without an injection cross, complex microchannel layouts or hardware modification. Alternate placement of the inlet tip in vials containing the sample and buffer solutions permits a volume defined electrokinetic sample introduction. Such fast and simple sample introduction leads to highly reproducible signals with no observable carry over between different analyte concentrations. The performance of the system was demonstrated in flow-injection and CE measurements of nitroaromatic explosives and for on-chip enzymatic assays of glucose in the presence of ascorbic acid. Employing an 8 cm long separation channel and a separation voltage of 4000 V it offers high-throughput flow-injection assays of 100 samples h(-1) with a relative standard deviation of 3.7% for TNT ( n = 100). Factors influencing the analytical performance of the new microchip have been characterized and optimized. Such ability to continuously introduce discrete samples into micrometer channels indicates great promise for high-speed microchip analysis.

Abstract:A portable heterogeneous immunoassay system is presented in this paper. It consists of a poly(dimethylsiloxane) (PDMS) based microfluidic chip as the immunoreactor, a miniaturized programmable high voltage sequencer as the power supply and the flow controller, and a laser-optical fiber fluorescence detection module as the signal reader. The operation of this immunoassay system is automatic. The sequential reagent dispensing and washing processes are controlled by the programmable sequencer. The reagent consumption was only 12 microL and the assay time was only 26 min. The detection limit for Escherichia coli O157:H7 bacterial antigen with this miniaturized system was 0.3 ng/muL, lower than that obtained using fluorescence microscope in previous studies.

Abstract:A high-speed DNA fragment separation system was developed based on a short capillary and a slotted-vial array automated sample introduction system. The injection process of DNA sample in a short capillary was investigated systematically with three injection techniques including constant-field-strength, low-field-strength and translational spontaneous injections. Under the optimized conditions, picoliter-scale sample plugs (corresponding to ca. 20-mu m plug length) were obtained, which ensure the high-speed and high-efficiency separation for DNA fragments with a short effective separation length. Other separation conditions including the sieving matrix concentration, separation field strength and effective separation length were also optimized. The present system was applied in the separation of Phi X174-Hae III digest DNA marker. With an effective separation length of 2.5 cm, the separation could be achieved in <100 s with plate heights ranging from 0.21 to 0.74 mu m (corresponding to plate numbers from 4.86 x 10(6) to 1.36 x 10(6)/m). The repeatabilities for the migration time of the eleven fragments were between 0.4 and 1.1% RSD (n = 8). By using the automated continuous injection method, the separation for four different DNA samples could be achieved within 250 s. The present system was further applied in the fast sizing of real DNA samples of PCR products.

Abstract:The development of efficient sample introduction and pretreatment systems for microfluidic chip-based analytical systems is important for their application to real-life samples. In this work, world-to-chip interfacing was achieved by a novel flow-through sampling reservoir featuring a guided overflow design. The flow-through reservoir was fabricated on a 30 x 60 x 3 mm planar glass chip of crossed-channel design used for capillary electrophoresis separations. The 20-microL sample reservoir was produced from a section of plastic pipet tip and fixed at one end of the sampling channel. Sample change was performed by pumping 80-microL samples sandwiched between air segments at approximately 0.48 mL/min flow rate through the flow-through reservoir, introduced from an access hole on the bottom side of the chip. A filter paper collar wrapped tightly around the reservoir guided the overflowing sample solution into a plastic trough surrounding the reservoir and then to waste. The performance of the system was demonstrated in the separation and determination of FITC-labeled arginine, glycine, phenylalanine, and glutamic acid with LIF detection, by continuously introducing a train of different samples through the system without electrical interruption. Employing a separation channel of 4 cm (2-cm effective separation length) and 1.4-kV separation voltage, maximum throughputs of 80/h were achieved with <4.1% carryover and precisions ranging from 1.5% for arginine to 2.6% RSD (n = 11) for glycine. The sampling system was tested in the continuous monitoring of the derivatizing process of amino acids by FITC over a period of 4 h, involving 166 analytical cycles. An outstanding overall precision of 4.8% RSD (n = 166) was achieved for the fluorescein internal standard.

Abstract:Microfluidic chip electrophoresis has been widely employed for separation of various biochemical species owing to its advantages of low sample consumption, low cost, fast analysis, high throughput, and integration capability. In this article, we reviewed the development of four different modes of microfluidics-based electrophoresis technologies including capillary electrophoresis, gel electrophoresis, dielectrophoresis, and field (electric) flow fractionation. Coupling detection schemes on microfluidic electrophoresis platform were also reviewed such as optical, electrochemical, and mass spectrometry method. We further discussed the innovative applications of microfluidic electrophoresis for biomacromolecules (nucleic acids and proteins), biochemical small molecules (amino acids, metabolites, ions, etc.), and bioparticles (cells and pathogens) analysis. The future direction of microfluidic chip electrophoresis was predicted.
