Abstract:A critical review is given for the development of modern experimental testing of engineering structures.Main achievements in this area are introduced along with comments on recent topics.The evolution of structural experimentation is closely related to scientific breakthroughs and major technological development.The vital importance of structural experimentation is recognized within the framework of physics for defining the nonlinear characteristics of structures under various complicated loading and environmental conditions.Progresses,problems and future challenges related to structural experimentation are discussed.The authors emphasize that methodology of structural experimentation should follow the approach of first-principle.

Abstract:Some problems from the principal stress axis rotation experiment with hollow cylinder apparatus are analyzed in detail herein;which consists of three aspects,such as the preparation of the hollow cylinder sample and the acquisition of the isotropic one;the estimation and elimination of the size effect and the end effect in the experiment;and the membrane penetration effect on both the drained and the undrained tests results,etc.The relevant countermeasures are put forward based on the systematic study concerned,and then some beneficial recommendations for obtaining and modifying the reasonable test results are presented as well.
An Investigation of Two Finite Element Modeling Solutions for Biomechanical Simulation Using a Case Study of a Mandibular Bone.

Abstract:The method used in biomechanical modeling for finite element method (FEM) analysis needs to deliver accurate results. There are currently two solutions used in FEM modeling for biomedical model of human bone from computerized tomography (CT) images: one is based on a triangular mesh and the other is based on the parametric surface model and is more popular in practice. The outline and modeling procedures for the two solutions are compared and analyzed. Using a mandibular bone as an example, several key modeling steps are then discussed in detail, and the FEM calculation was conducted. Numerical calculation results based on the models derived from the two methods, including stress, strain, and displacement, are compared and evaluated in relation to accuracy and validity. Moreover, a comprehensive comparison of the two solutions is listed. The parametric surface based method is more helpful when using powerful design tools in computer-aided design (CAD) software, but the triangular mesh based method is more robust and efficient.

Abstract:In this review, a critical look at the research progress of experimental solid mechanics in China for the past years is presented. Issues are discussed of the discovery and development of new fundamental methods and techniques versus performance benchmarking for many of their applications. Included herein are photoelasticity and various forms of modern photomechanics, acoustical techniques, image processing and videometrics, radial and spectrum techniques, and experimental mechanics on micro/nano scale. It is also noticed that both the ever developed instrumentation and specialized synthetical techniques have played important roles in advancing experimental mechanics in scientific researches and industrial applications. Finally, an attempt is made to look into the future of experimental solid mechanics with personal opinions offered on what the future trends will be for the researches in the field.

Abstract:Since the cell was discovered by humans, it has been an important research subject for researchers. The mechanical response of cells to external stimuli and the biomechanical response inside cells are of great significance for maintaining the life activities of cells. These biomechanical behaviors have wide applications in the fields of disease research and micromanipulation. In order to study the mechanical behavior of single cells, various cell mechanics models have been proposed. In addition, the measurement technologies of single cells have been greatly developed. These models, combined with experimental techniques, can effectively explain the biomechanical behavior and reaction mechanism of cells. In this review, we first introduce the basic concept and biomechanical background of cells, then summarize the research progress of internal force models and experimental techniques in the field of cell mechanics and discuss the latest mechanical models and experimental methods. We summarize the application directions of cell mechanics and put forward the future perspectives of a cell mechanics model.
[Biomechanical Research on Upper Femur with Vertical and Bending Stress].

Abstract:OBJECTIVE:To study the bone state with ultimate stress by examining biomechanical distribution of upper femur in Chinese, in order to accumulate more experiences for clinical work.METHODS:Totally 60 Chinese femurs from fresh cadavers were randomly divided into two groups. All the femurs were cleaned, and the body age ranged from 36 to 72 years old, averaged 56.4 years, including 41 from males, and 19 from females. These two groups underwent mechanical stress and bending stress tests. Special mechanical laboratory and machines were used to get the information. Results about the loading value at each testing point under stress were collected.RESULTS:The four faces of the upper femur suffered different stress under external forces. The bone on upper femur can tolerate more mechanical stress than bending stress. Medial and lateral region of the femur neck and the rear side of the small tuberosity section were themain position enduring the vertical stress. The rear position of the base femur neck and the small tuberosity section were the main regions enduring the bending stress. Those main positions had strong cancellous bones. The intertrochanteric fracture fixation and artificial femoral stems were designed depending on this biomechanical basis.CONCLUSION:According to our experiment result, doctors need to chose more effective fixations for upper femur fracture, and femoral stems for the patients. More information should be collected by further researches.

Abstract:Over the past decades, increasing evidence has indicated that mechanical loads can regulate the morphogenesis, proliferation, migration, and apoptosis of living cells. Investigations of how cells sense mechanical stimuli or the mechanotransduction mechanism is an active field of biomaterials and biophysics. Gaining a further understanding of mechanical regulation and depicting the mechanotransduction network inside cells require advanced experimental techniques and new theories. In this review, the fundamental principles of various experimental approaches that have been developed to characterize various types and magnitudes of forces experienced at the cellular and subcellular levels are summarized. The broad applications of these techniques are introduced with an emphasis on the difficulties in implementing these techniques in special biological systems. The advantages and disadvantages of each technique are discussed, which can guide readers to choose the most suitable technique for their questions. A perspective on future directions in this field is also provided. It is anticipated that technical advancement can be a driving force for the development of mechanobiology.

Abstract:Studying the effects of mechanical loads on living cells provides valuable information about their activity, vitality and intercellular communication. Mechanical stimuli have often a significant impact on cell migration, tissue remodeling and healing and other phenomena. This effects in micro scale affect also tissue structure, its mechanical characteristics and functionality. Special methods are needed to study deformation and biological responses of such small objects as cells. Moreover, in order to keep the cells alive during such tests, it is necessary to ensure appropriate environmental conditions. The methods of loading and examination cells can be categorized into several groups depending on the physical effects used in experiment. This paper provides a systematic review of the methods used in such studies and examples of mathematical modeling the mechanical response of cells to mechanical loads. Such models enable computational simulation of changes in cells and tissues excited by mechanical stress.

Abstract:As an intriguing interdisciplinary research field, cell and molecular biomechanics is at the cutting edge of mechanics in general and biomechanics in particular. It has the potential to provide a quantitative understanding of how forces and deformation at tissue, cellular and molecular levels affect human health and disease. In this article, we review the recent advances in cell and molecular biomechanics, examine the available computational and experimental tools, and discuss important issues including protein deformation in mechanotransduction, cell deformation and constitutive behavior, cell adhesion and migration, and the associated models and theories. The opportunities and challenges in cell and molecular biomechanics are also discussed. We hope to provide readers a clear picture of the current status of this field, and to stimulate a broader interest in the applied mechanics community.

Abstract:Interactions between cells and their microenvironment trigger a series of complex mechanotransduction signals that regulate cell behavior. Mechanical characterization of cell interactions with extracellular matrix (ECM)/substrate is a prerequisite for quantitative investigations of the mechanisms of mechanotransduction. Here, we present recent progress in the quantification of traction forces generated during cell-ECM/substrate interaction, with an emphasis on force/stress reconstructions on functionalized ECM/substrates fabricated by microfluidics techniques such as elastomeric micropillar arrays, patterned and continuous substrates, and three-dimensional hydrogel substrates. We also present an overview of how to measure displacement fields induced by cell activity, and reliably and accurately capture cellular traction force/stress on different substrates. Lastly, we describe recent applications in mechano-biology.

Abstract:Cells tend to form hierarchy structures in native tissues. Formation of cell aggregates in vitro such as cancer spheroids and embryonic bodies provides a unique means to study the mechanical properties and biological behaviors/functions of their counterparts in vivo. In this paper, we review state-of-the-art experimental approaches to assess the mechanical properties and mechanically-induced responses of cell aggregates in vitro. These approaches are classified into five categories according to loading modality, including micropipette aspiration, centrifugation, compression loading, substrate distention, and fluid shear loading. We discussed the advantages and disadvantages of each approach, and the potential biomedical applications. Understanding of the mechanical behavior of cell aggregates provides insights to physical interactions between cells and integrity of biological functions, which may enable mechanical intervention for diseases such as atheromatosis and cancer.

Abstract:Mechanics, as a key physical factor which affects cell function and tissue regeneration, is attracting the attention of researchers in the fields of biomaterials, biomechanics, and tissue engineering. The macroscopic mechanical properties of tissue engineering scaffolds have been studied and optimized based on different applications. However, the mechanical properties of the overall scaffold materials are not enough to reveal the mechanical mechanism of the cell-matrix interaction. Hence, the mechanical detection of cell mechanics and cellular-scale microenvironments has become crucial for unraveling the mechanisms which underly cell activities and which are affected by physical factors. This review mainly focuses on the advanced technologies and applications of cell-scale mechanical detection. It summarizes the techniques used in micromechanical performance analysis, including atomic force microscope (AFM), optical tweezer (OT), magnetic tweezer (MT), and traction force microscope (TFM), and analyzes their testing mechanisms. In addition, the application of mechanical testing techniques to cell mechanics and tissue engineering scaffolds, such as hydrogels and porous scaffolds, is summarized and discussed. Finally, it highlights the challenges and prospects of this field. This review is believed to provide valuable insights into micromechanics in tissue engineering.

Abstract:In this article,we review recent advances in solid experimental techniques for the mechanical characterization of materials and structures at micro and nano scales with emphasis in the optical and probe techniques.Advantages and disadvantages of various approaches are discussed to highlight the need for clamping,loading,force and displacement measuring,as well as carefully designed experiments and rigorous and coupling analysis of experimentally obtained data to yield unambiguous results.By introducing typical studies we demonstrate that the development of robust and innovative experimentation and system is crucial for the tests in micro/nano experiments.

Abstract:The cells and biological tissues need to adapt the complex physiological and mechanical environment in human body. They must withstand the mechanical loads from external environment, and equally important, they often actively produce forces to change their architecture and shape during physiological processes such as tissue growth and repair. The mechanical properties of cells are mainly determined by cytoskeleton, and the stiffness of biological tissues is greatly affected by extracellular matrix. Microscopically, cytoskeleton and extracellular matrix are intricate, heterogeneous 3D networks of crosslinked biopolymers. Early studies mainly focused on explaining the universal features such as the nonlinear response and strain stiffening of these biopolymer networks by constructing various network models. In recent years, with the simultaneous progress in experimental methods, theoretical models and computational techniques, more intriguing mechanical behaviors and underlying mechanisms of these living matters have been revealed. In this review, we online some of the major advances in modeling and analyzing cytoskeleton and extracellular matrix, including the dynamic crosslinking, active materials originated from mechanochemical coupling of biopolymers, plasticity/fracture of crosslinked networks, and self-adaption triggered by mechanical training. These modeling and analyses may help to quantify the complex behaviors of cells and tissues, deepen our understanding of the underlying mechanobiological mechanisms, and provide guidance for synthetic biological materials and tissue engineering.
Biomechanical Study and Clinical Evaluation of Four Techniques in the Treatment of Intertrochanteric Fracture of the Femul

Abstract:Objective: To find an optimal method for the treatment of intertrochanteric fracture from multiple and complex treatment methods.Method:20 embalmed cadaver femoral specimens were collected and randomly allocated into four groups,the intertrochanteric fracture were made and each group was fixed with one of four techniques;After vertical load and deflective axis load test,the anti-rotation strength and anti-bending strength were tested.The clinical effect and incidence of complication of 271 cases of intertrochanteric fracture were analyses.Result: Dynamic hip screw(DHS) is the most stable technique of all,external fixators to be the second,and Steinmann pins was the poorest.The clinical effect was coincident with the result of the biomachanical test.Conclusion:To the patients,who can tolerate an operation,the authors prefer to using of DHS,and to the patients,who can not or will not receive an operation,we prefer to use unilateral external fixator.

Abstract:Objective: To compare the different methods for the fixation of femoral neck fractures and determine the best one according to the biomechanical principal. Methods: Four different methods were compared by biomechanical experiment to observe the subside, open and rotation migration. Results: The external fixator, which was devised by ourselves, had the minimal open, subside and rotation migration. Conclusion: The external fixator was a good, economical, effective and simple method in the treatment of femoral neck fracture, which is valuable to disseminated. The placement of the pins should be in accordance with the compressive trabeculae and clingy to the femoral calcar so as to fit to the principle of biomechanics.

Abstract:The objective of this study is to use the finite element (FE) method to predict the mechanical signals (interstitial fluid velocity, strain, pore pressure, and pore fluid velocity) produced by osteocyte during physiological activities. The model predicts that the amplitude and distribution of the mechanical signals are mainly affected by the loading rate. The magnitude of mechanical signals in the lacunar-canalicular system increases as the amplitude, frequency and amount of direction of load increase. Collagen hillocks can effectively amplify strain signals at the process. The established model can be used for studying the mechanism of bone mechanotransduction at the micro-level.
The effect of different mechanical stimuli on numerical simulation of bone remodeling

Abstract:Objective To study the effect of three different kinds of mechanical stimuli( i. e. strain energy density,equivalent stress and equivalent strain) on numerical simulation of bone remodeling. Methods A two-dimensional finite element model of the proximal femur was constructed. Based on the mechanostat theory and finite element method,the inner structure of the proximal femur and its density distributions under the three different stimuli were predicted. Then the simulation results were compared quantitatively with calculation results obtained from CT images. Results The predicted density distributions on the proximal femur under different stimuli were all well matched with the real structure of the proximal femur. By comparing the values and shapes of the calculated bone density curves,the predictions from the model using equivalent stress as mechanical stimuli were mostly consistent with the CT images. Conclusions The equivalent stress might play a leading role in mechano-regulation algorithms of bone remodeling. The accurate prediction of bone remodeling process will provide a theoretical basis for clinical practices such as orthopedic surgery,treatment of bone diseases and personalized design and optimization of prosthesis.

Abstract:Cell mechanics plays a key role in determining physical performances and physiological functions of cells, as well as the early detection of diseases and development of biomedical engineering. In this study, we utilized a combination of atomic force microscopy (AFM) and finite element method (FEM) to compare the cellular elasticity (Young's modulus) and viscoelasticity (stress-relaxation time) of living and fixed endothelial cells (ECs) across varying loading rates. The results showed that both mechanical properties of normal ECs are more sensitive to loading speed compared with fixed ECs. The Young's modulus of normal endothelial cells (ECs) exhibits an increasing trend with the growing loading rate, whereas the Young's modulus of fixed ECs is almost not affected by the loading rate. Among various viscoelastic properties of cells under varying loading rates, the long-term relaxation time, especially at a loading rate of 5μm/s, showed the most significant difference between living and fixed cells. This work comprehensively evaluated the effectiveness of using different mechanical properties to distinguish cells with different physiological characteristic. This research would improve our knowledge of single-cell mechanical behaviors and provide new ideas for distinguishing various types of cells by AFM-based cellular elastic and viscoelastic properties with varying loading rates.

Abstract:Osteoarthritis caused by knee sports injuries and cumulative fatigue damage is very common.The research result of biomechanical behavior of the human knee is not only important to know the etiology and pathogenesis,improve the treatment of joint diseases,but also helpful to design the knee rehabilitation and orthopedic brace.The experimental method and simulation method are mainly used in the biomechanical research of the knee joints.The finite element simulation method has been widely used in recent years,and its further development has to depend on the knowledge of macroscopic mechanical properties and microscopic structure of joint bone and tissue.
