Abstract:One-pot synthesis of 6-bromo-4,4-dimethylthiochroman from bromobenzene is reported. The crucial connection between the three steps is that the byproducts in the first two steps are used as the catalyst in the third step. Abstract:A domino one‐pot synthesis of 2‐(trifluoromethyl)benzothiazole via copper‐mediated three‐component cascade reaction starting from the easily accessible starting materials such as o‐iodoanilines, methyl trifluoropyruvate, and elemental sulfur is reported. The present strategy displayed a comprehensive substrate scope and good functional group tolerance and enabled access to a variety of substituted 2‐(trifluoromethyl)benzothiazoles. A 2‐(trifluoromethyl)benzoselenazole has also been synthesized utilizing this reaction methodology.

Abstract:2-Trifluoromethyl and 2-difluoromethyl substituted benzimidazole, benzoxazole and benzothiazole derivatives were efficiently prepared through a one-pot reaction of trifluoroacetic acid and difluoroacetic acid, respectively, with commercially available o-phenylenediamines, 2-aminophenols, and 2-aminobenzenethiols in good to excellent yields. Subsequential bromination of 2-difluoromethyl groups by photolysis with NBS led to the formation of bromodifluoromethyl benzo-1,3-diazoles which may be utilized to prepare the new generation of gem-difluoromethylene linked identical or non-identical twin molecules for drug synthesis.

Abstract:A novel and efficient copper-catalyzed one-pot procedure for the synthesis of 3-N-substituted 1,4,2-benzodithiazine 1,1-dioxide derivatives from 2-halobenzenesulfonamides, amines and CS2 is described. The reaction proceeds through Ullmann-type S-arylation, intramolecular addition of NH2 with CS and dehydrosulfide, which provides a new and useful strategy for construction of cyclic aromatic sulfonamides.

Abstract:We have developed an efficient method for the synthesis of benzimidazoles via cascade reactions of o-haloacetoanilide derivatives with amidine hydrochlorides. The protocol uses 10 mol % CuBr as the catalyst, Cs2CO3 as the base, and DMSO as the solvent, and no ligand is required. The procedure proceeds via the sequential coupling of o-haloacetoanilide derivatives with amidines, hydrolysis of the intermediates (amides), and intramolecular cyclization with the loss of NH3 to give 2-substituted 1H-benzimidazoles.

Abstract:The analogous three‐component synthesis strategy for substituted 1,2,4‐oxadiazole and quinazoline derivatives from readily available benzaldehyde, benzylamine and hydroxylamine or aniline has been developed. Both the cascade reaction sequences involves nucleophilic addition of C‐N bond, introduction a halogen donor, nucleophilic substitution and Cu(II)‐catalyzed aerobic oxidation. This synthesis methodology demonstrated good yields, broad substrate scope and oxygen as a green oxidant. Thus, this synthesis protocol provides strategies for the construction of substituted 1,2,4‐oxadiazole and quinazolines from readily and simple starting materials.

Abstract:The one-pot synthesis of 2,3-dihydro-1,5-benzodiazepins-2-one bearing phosphanylidene (ylide) or phosphono-succinate substituent is described. In this four-component reaction, benzodiazepine derived from condensation of o-phenylenediamine and diketene is trapped with the trialkyl phosphite–dialkyl acetylenedicarboxylate zwitterion. In the presence of H2O, the ylide functional group is hydrolyzed to the corresponding phosphonate. The configuration of the products is selective and only one of the two possible rotamers or diastereomers is formed exclusively in high yield.
