Abstract:A novel way of synthesizing quinoxalines has been developed that involves condensation and C-N bond formation in a copper-catalyzed, one-pot, three-component reaction. The reaction was optimized when 2-iodoanilines (1.0 equiv), arylacetaldehydes (2.0 equiv), sodium azide (1.2 equiv), CuI (10mol%), DMEDA (10mol%), and K2CO3 (1.0 equiv) were reacted in DMSO at 80 degrees C for 20hours. This approach was used to directly synthesize a variety of quinoxalines in moderate to good yields.

Abstract:The analogous three‐component synthesis strategy for substituted 1,2,4‐oxadiazole and quinazoline derivatives from readily available benzaldehyde, benzylamine and hydroxylamine or aniline has been developed. Both the cascade reaction sequences involves nucleophilic addition of C‐N bond, introduction a halogen donor, nucleophilic substitution and Cu(II)‐catalyzed aerobic oxidation. This synthesis methodology demonstrated good yields, broad substrate scope and oxygen as a green oxidant. Thus, this synthesis protocol provides strategies for the construction of substituted 1,2,4‐oxadiazole and quinazolines from readily and simple starting materials.

Abstract:A Cu-catalyzed tandem synthesis of azole-fused pyrimido[1,2-c]quinazolines and imidazo[1,2-c]quinazolines has been developed. The reaction is based on a C-N cross-coupling/C-H functionalization reaction of 2-(2-bromophenyl)-1,4,5,6-tetrahydropyrimidines with azoles. Abstract:We have developed a versatile and efficient method for copper-catalyzed synthesis of both 2-amino-4(3H)-quinazolinone and 2-aminoquinazoline derivatives. The protocol uses readily available substituted 2-halobenzoic acids, 2-bromobenzaldehyde, 2-bromophenyl ketones and guanidines as the starting materials, inexpensive copper(I) iodide as the catalyst, and the method has important application values for construction of N-heterocycles in organic chemistry and medicinal chemistry.

Abstract:Via an imine-protection strategy, we herein present an unprecedented copper-catalyzed oxidative multicomponent annulation reaction for direct synthesis of quinazolinones. The construction of various products is achieved via formation of three C-N and one C-C bonds in conjunction with the benzylic functionalization. The merits of easily available feedstocks, naturally abundant catalyst, good functional group and substrate compatibility, and release of H2O as the byproduct make the developed chemistry a practical way to access quinazolinones.

Abstract:An easy and efficient method has been developed for the synthesis of pyrazolo[1,5-c]quinazolines via one-pot two-step reactions of readily available substituted 1-(2-halophenyl)-3-akylprop-2-yn-1-ones, hydrazine hydrochloride and amidine hydrochlorides under mild conditions, and the corresponding pyrazolo[1,5-c]quinazolines were obtained in good to excellent yields. The novel method affords a new strategy for the construction of diverse and useful poly N-heterocyclic compounds.

Abstract:The synthesis of functionalized (trichloromethyl)quinazoline‐4(1H)‐one through a novel intramolecular CH activation reaction from trichloroacetonitrile, benzoyl chlorides, and various primary amines is a remarkable achievement in organic chemistry. Furthermore, the product with phenylacetylene and sodium azide in the presence of a copper catalyst in water solvent at room temperature demonstrates a new (1,2,3‐triazole)‐quinazoline‐4(1H)‐one derivative. The synthesis of functionalized (trichloromethyl)quinazoline‐4(1H)‐one with high yields through a novel three‐component intramolecular CH activation reaction from trichloroacetonitrile, benzoyl chlorides, and various primary amines is a remarkable achievement in organic chemistry. This strategy offers a direct and efficient route to access complex molecular structures from readily available starting materials. The use of copper (I) as a catalyst and L‐proline as a ligand in tetrahydrofuran at room temperature highlights the importance of transition metal catalysis in enabling selective CH activation processes. Furthermore, the subsequent transformation of the obtained product with phenylacetylene and sodium azide in the presence of a copper catalyst in water solvent at room temperature demonstrates the versatility of this synthetic approach in accessing new (1,2,3‐triazole)‐quinazoline‐4(1H)‐one derivative. The combination of available starting materials, catalytic systems, mild reaction conditions, and ease of purification procedures contributes to the attractiveness of this method for the synthesis of diverse (trichloromethyl)quinazoline‐4(1H)‐one and (1,2,3‐triazole)‐quinazoline‐4(1H)‐one derivative.

Abstract:The 1-arylpyrazolo[5,1-b]quinazolin-9(1H)-one derivatives were obtained via intramolecular alkyne hydroamination reaction of 2-amino-N′-arylbenzohydrazide and alkynal diethyl acetal in the presence of CuBr and Cs2CO3.
