Abstract:A new practical synthesis of 1H-indazole is presented. A previous mechanism for the cyclization step is proved to be nonfeasible and a hydrogen bond propelled mechanism is proposed. The new mechanism is suitable for similar cyclization, and a new reaction is predicted.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:A convenient synthesis of 1-oxo-1,2,3,4-tetra-hydrocarbazoles has been developed by reaction of 2-aminocyclohexanone hydrochlorides with various phenylhydrazine hydrochlorides via Fischer indole synthesis under mild conditions. The method is more satisfactory in terms of the easy availability of starting materials and the simple one-pot operation.

Abstract:An efficient intermolecular annulation of indazole aldehydes and propargylic amines has been developed for the synthesis of pyrazinoindazoles under catalyst- and additive-free conditions. This straightforward methodology features a wide substrate scope, high atom economy and environmental advantages. The bioactivity results of these new pyrazino[1,2-b]indazoles showed that some of them exhibited significant antifungal activity.

Abstract:AbstractCyclization of 2‐methoxy‐6‐benzyloxy acetophenone hydrazone gave 3‐methyl‐4‐methoxy indazole and 3‐methyl‐4‐methoxy‐7‐benzyl indazole in the presence of polyphosphoric acid (PPA). The hydrazone was probably converted to 2‐hydroxy‐6‐methoxy acetophenone hydrazone and 2‐hydroxy‐3‐benzyl‐6‐methoxy acetophenone hydrazone followed by cyclization to the corresponding indazoles in acidic conditions. Cyclization of 2‐methoxy‐6‐(halo or alky] or aryl benzyloxy) acetophenone hydrazones gave similar products. Cyclization of 2‐methoxy‐6‐(p‐nitrobenzyloxy) acetophenone hydrazone gave 2‐(p‐nitrophenyl)‐3‐methyl‐4‐methoxy benzofuran and 3‐methy]‐4‐methoxy indazole while 2‐methoxy‐6‐(m‐nitrobenzyloxy) acetophenone hydrazone gave 3‐methyl‐4‐methoxy indazole, 3‐methyl‐4‐methoxy‐7‐(m‐nitrophenyl) indazole and 3‐methyl‐4‐(m‐nitrobenzyloxy) indazole.

Abstract:An efficient cascade intramolecular cyclization/intermolecular nucleophilic addition reaction of allenyl benzoxazinone with isatin or isatin-derived ketimine has been established by using Pd0-π-Lewis base catalysis. A series of 3-hydroxy-2-oxindoles and 3-amino-2-oxindoles with quaternary carbon atoms at the C3 position were synthesized in good yields under mild conditions through this protocol.

Abstract:A palladium-catalyzed synthesis of polysubstituted indoles and isoquinolines through the coupling of aryl bromides with 2-alkynyl arylazides or 2-alkynyl benzylazides has been developed. This method provides straightforward access to indoles and isoquinolines with high efficiency and excellent functional group compatibility. In this transformation, the iminophosphorane in situ generated from azides is served as the nucleophile that attacks the alkyne moiety in the cyclization process.

Abstract:An appropriate method for the synthesis of 5-aryl-9-hydroxypyrano[3,2-f]indole-2(8H)-one was described. The targeted compounds were obtained starting from vanillin via nine steps. Interestingly, in the final cyclization step, the intermediate 4-(2-halogeno phenyl)-7-methoxy-1H-indole-6-yl propiolate could convert directly into the final product in one step reaction using PtCl4 or Pd(PPh3)4/trifluoroacetic acid as catalysts. The possible catalytic mechanism for PtCl4 and Pd(PPh3)4/trifluoroacetic acid was discussed.

Abstract:A copper-catalyzed [3 + 2] annulation of O-acyl oximes with 2-electron-withdrawing group substituted p-hydroquinones for the efficient synthesis of polysubstituted 5-hydroxyindoles is developed. Further intramolecular cyclization leads to the concise and rapid construction of several kinds of 3,4- and 4,5-fused polycyclic indoles.

Abstract:A facile and general one-step method is presented for the synthesis of aromatic indolizine compounds 3a–n in moderate to high yields (53–99%) by reaction of the pyridinium N-ylides 1a–e, the quinolinium N-ylide 1f and the isoquinolinium N-ylide 1g with various olefinic dipolarophiles, such as acrylonitrile 2a, methyl acrylate 2b, acrylamide 2c, diethyl maleate 2d and methyl crotonate 2e, respectively, in the presence of a new oxidant TPCD [Copy4(HCrO4)2, tetrapyridinecobalt(II) dichromate] at 90 °C for 2 h in DMF.

Abstract:A novel cyclization reaction of pyridinium 1,4-zwitterionic thiolates and propiolic acid derivatives mediated by triethylamine is described, which allows the facile synthesis of indolizines under mild reaction conditions. The net transformation involves an acetylide-driven formal [5 + 1] annulation reaction followed by a spontaneous ring-contraction/sulfur extrusion process of transient pyridothiazine intermediates.

Abstract:An efficient and practical one-pot domino synthesis of 1-phenyl-1H-indol-2-amine and 5-amino-indolo[1,2-a]quinazoline derivatives from readily available 2-(2-bromophenyl)acetonitriles was developed. The overall protocol involves a Buchwald-Hartwig type coupling and a base-promoted intramolecular nucleophilic reaction. The reaction scope, advantages and limitations are discussed.

Abstract:The efficient synthesis of 4-hydroxy-3-arylindolin-2-ones via phosphoric acid-mediated annulation of various β-nitrostyrenes and 1,3-cyclohexanedione is described. This annulation reaction gives a practical method for affording a diverse set of oxindoles, having simple experimentation, readily available starting materials, and very good yields. Additionally, substituted 1,3-cyclohexanediones under the same conditions afforded tetrahydrobenzofuran oxime compounds.

Abstract:A facile and efficient method for the synthesis of 1,2-dihydropyrrolo[3,4-b]indol-3(4H)-one derivatives via a two-step reaction has been developed. The starting materials of 1H-indole-2-carboxylic acid, aldehyde, and amine are mixed and refluxed in ethanol to generate Mannich products, which are dehydrated in the presence of 2-(7-aza-1H-benzotriazole-1-yl)-1,1,3,3-tetramethyluroniumto form the desired products, 1,2-dihydropyrrolo[3,4-b] indol-3(4H)-ones.
