Abstract:3-Benzoylindolizine-5-carbaldehydes (4a-f), which could be used as derivatization reagents for amino compounds in HPCE were synthesized based on the 1,3-dipolar cycloaddition of 1-phenacyl-2-(1,3-dioxolan-2-yl)pyridinium ylide with alkenes in the presence of TPCD.

Abstract:A facile and general one-step method is presented for the synthesis of aromatic indolizine compounds 3a–n in moderate to high yields (53–99%) by reaction of the pyridinium N-ylides 1a–e, the quinolinium N-ylide 1f and the isoquinolinium N-ylide 1g with various olefinic dipolarophiles, such as acrylonitrile 2a, methyl acrylate 2b, acrylamide 2c, diethyl maleate 2d and methyl crotonate 2e, respectively, in the presence of a new oxidant TPCD [Copy4(HCrO4)2, tetrapyridinecobalt(II) dichromate] at 90 °C for 2 h in DMF.

Abstract:3-Acylindolizines (5a-5f) and their benzo-analogues 1-acylpyrrolo[1,2-a]quinolines (6a-6f) and 1-acylpyrrolo[2,1-a]isoquinolines (7a-7i) are regioselectively synthesized by a convenient one pot reaction of the corresponding pyridinium (quinolinium, isoquinolinium) ylide with maleic anhydride (MA) in the presence of the mild oxidant tetrakispyridinecobalt(II) dichromate (TPCD). These reactions proceed via a tandem reaction sequence of 1,3-dipolar cycloaddition of azomethine ylide with MA, anhydride hydrolysis and oxidative bisdecarboxylation of the primary cycloadducts followed by dehydrogenative aromatization of the dihydroindolizines. TPCD serves as both decarboxylation and dehydrogenation reagent in the reactions. These results show that TPCD is a promising new reagent for bisdecarboxylation of aliphatic carboxylates.

Abstract:A convenient synthesis of 1-oxo-1,2,3,4-tetra-hydrocarbazoles has been developed by reaction of 2-aminocyclohexanone hydrochlorides with various phenylhydrazine hydrochlorides via Fischer indole synthesis under mild conditions. The method is more satisfactory in terms of the easy availability of starting materials and the simple one-pot operation.

Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:Two selective procedures for the synthesis of potentially important agrochemicals, indolizine-3-carboxamides (6) and indolizine-3-carbonitriles (7), were developed. In the presence of tetrakispyridinecobalt(II) dichromate (TPCD), compounds 6 are readily synthesized by a one-pot reaction sequence which consists of a 1,3-dipolar cycloaddition of pyridinium N-ylides to alkenes, followed by an aromatization and a regiospecific hydration reaction. When MnO2 instead of TPCD was used in the 1,3-dipolar addition reaction, the nitrile group of the indolizine product was not hydrated and compounds 7 were obtained as final products. Both procedures utilize convenient conditions and inexpensive reagents, and give products in good to high yields.

Abstract:2-Aryl-1-haloindolizines were synthesized from pyridinium salts and 2-aryl-1,1-dihaloalk-1-enes using a DBU/THF system. 2-Aryl-1,1-dihaloalk-1-enes containing electron-withdrawing or -donating groups were efficiently converted into the corresponding 2-aryl-1-haloindolizines in moderate to excellent yields.

Abstract:A novel cyclization reaction of pyridinium 1,4-zwitterionic thiolates and propiolic acid derivatives mediated by triethylamine is described, which allows the facile synthesis of indolizines under mild reaction conditions. The net transformation involves an acetylide-driven formal [5 + 1] annulation reaction followed by a spontaneous ring-contraction/sulfur extrusion process of transient pyridothiazine intermediates.

Abstract:A facile and efficient method for the synthesis of 1,2-dihydropyrrolo[3,4-b]indol-3(4H)-one derivatives via a two-step reaction has been developed. The starting materials of 1H-indole-2-carboxylic acid, aldehyde, and amine are mixed and refluxed in ethanol to generate Mannich products, which are dehydrated in the presence of 2-(7-aza-1H-benzotriazole-1-yl)-1,1,3,3-tetramethyluroniumto form the desired products, 1,2-dihydropyrrolo[3,4-b] indol-3(4H)-ones.

Abstract:An appropriate method for the synthesis of 5-aryl-9-hydroxypyrano[3,2-f]indole-2(8H)-one was described. The targeted compounds were obtained starting from vanillin via nine steps. Interestingly, in the final cyclization step, the intermediate 4-(2-halogeno phenyl)-7-methoxy-1H-indole-6-yl propiolate could convert directly into the final product in one step reaction using PtCl4 or Pd(PPh3)4/trifluoroacetic acid as catalysts. The possible catalytic mechanism for PtCl4 and Pd(PPh3)4/trifluoroacetic acid was discussed.

Abstract:A novel [3+3] annulation reaction of 2‐alkenylindoles with hydrazonyl chlorides in the presence of base for the formation of pyridazino[4,5‐b]indoles was developed. This approach provided a convenient and efficient way to synthesize a series of functionalized pyridazino[4,5‐b]indoles derivatives. The method exhibited a broad substrate scope and provided the corresponding pyridazino[4,5‐b]indoles products with excellent yields.

Abstract:An approach to hydrocarbazoles bearing an all‐carbon quaternary center at C4a position was developed via a Brønsted acid‐initiated Diels‐Alder cycloaddition/retro‐aza‐Michael addition cascade process from azepino[4,5‐b]indoles and commercially available dienophiles. The method provided a range of hydrocarbazoles in 63–99% yields. The practicality of this transformation was demonstrated by a scale‐up experiment and various transformations to several hydrocarbazole derivatives and the tetracyclic indoline scaffold. Moreover, hetero‐Diels‐Alder cycloadditions of azepino[4,5‐b]indoles were explored with 4‐phenyl‐3H‐1,2,4‐triazole‐3,5(4H)‐dione (PTAD) and 82–99% yields were obtained.

Abstract:Dearomatization reactions provide a rapid approach to construct complicated molecules that are difficult to synthesize by traditional methods from simple aromatic compounds. Herein, we report an efficient dearomative [3+2] cycloaddition reaction of 2-alkynyl pyridines with diarylcyclopropenones, leading to the synthesis of densely functionalized indolizinones in moderate to good yields under metal-free conditions. In addition, this strategy can also be employed in dearomative cyclization of isoquinolines to access a variety of benzo-fused indolizinones. Density functional theory (DFT) calculations revealed that an appropriate substituent at the 2-position of pyridine is crucial to the dearomatization process.
