Abstract:A new practical synthesis of 1H-indazole is presented. A previous mechanism for the cyclization step is proved to be nonfeasible and a hydrogen bond propelled mechanism is proposed. The new mechanism is suitable for similar cyclization, and a new reaction is predicted.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:Ethyl and phenyl oxanilates were readily prepared from N-(2-bromobenzyl)anilines and oxalyl chloride monoethyl and monophenyl esters, respectively. It was found that the ethyl oxanilate survived in the presence of K2CO3 in DMA at 120 degrees C and underwent an intramolecular direct arylation using Pd(OAc)(2)-dppf, furnishing the 5,6-dihydrophenanthridine derivative. In contrast, the corresponding phenyl oxanilates decomposed upon exposure to K2CO3 in DMA at 120 degrees C and were transformed into N-arylisoindolin-1-ones via Pd(OAc)(2)-dppf-catalyzed intramolecular decarbonylative coupling. Except for the 4-methoxy-substituted oxanilic acid phenyl ester, other phenyl oxanilates possessing electron-withdrawing (NO2, Cl) and weak electron-donating (Me) substituents provided the N-arylisoindolin-1-ones in 43-80% yields.

Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:A novel [3+3] annulation reaction of 2‐alkenylindoles with hydrazonyl chlorides in the presence of base for the formation of pyridazino[4,5‐b]indoles was developed. This approach provided a convenient and efficient way to synthesize a series of functionalized pyridazino[4,5‐b]indoles derivatives. The method exhibited a broad substrate scope and provided the corresponding pyridazino[4,5‐b]indoles products with excellent yields.

Abstract:A novel domino intramolecular [3+2] cycloaddtion and ring-opening aromatization process has been successfully developed for the efficient direct synthesis of pyrrolyl or indolyl substituted 9,10-dihydrophenanthren-9-ol analogues. And 1-(phenanthren-9-yl)-1H-pyrroles can be easily obtained via dehydration of 10-(1H-pyrrol-1-yl)-9,10-dihydrophenanthren-9-ols. Furthermore, the MU assay indicated that four compounds with indolyl substitutions showed obvious inhibitory activities against HepG2 cells, and compound anti-4jb displayed a lowest IC50 value of 9.99 mu M. Abstract:An efficient cascade intramolecular cyclization/intermolecular nucleophilic addition reaction of allenyl benzoxazinone with isatin or isatin-derived ketimine has been established by using Pd0-π-Lewis base catalysis. A series of 3-hydroxy-2-oxindoles and 3-amino-2-oxindoles with quaternary carbon atoms at the C3 position were synthesized in good yields under mild conditions through this protocol.

Abstract:A facile and efficient method for the synthesis of 1,2-dihydropyrrolo[3,4-b]indol-3(4H)-one derivatives via a two-step reaction has been developed. The starting materials of 1H-indole-2-carboxylic acid, aldehyde, and amine are mixed and refluxed in ethanol to generate Mannich products, which are dehydrated in the presence of 2-(7-aza-1H-benzotriazole-1-yl)-1,1,3,3-tetramethyluroniumto form the desired products, 1,2-dihydropyrrolo[3,4-b] indol-3(4H)-ones.

Abstract:The synthesis of novel spirobenzazepinoindole derivatives has been achieved through a highly efficient and synthetic route. The approach involves a two‐step reaction, utilizing indole derivatives, 2‐amino benzyl alcohol, and ninhydrin as key starting materials under mild reaction conditions. The reaction proceeds via a sequential cascade process involving cyclization, condensation and spiro‐annulation, leading to the formation of the spirobenzazepinoindole core structure in good to excellent yields. The method offers broad substrate scope, high atom economy, and operational simplicity. The synthesized spirobenzazepinoindoles were fully characterized by spectroscopic techniques, including NMR (1H & 13C), IR and mass spectrometry. The methodology provides a valuable tool for the rapid generation of structurally complex spirobenzazepinoindoles, which could serve as scaffolds for the development of new therapeutic agents.

Abstract:An efficient and novel route for the synthesis of 1H-indol-2-yl-(4-aryl)-quinolin-2(1H)-one 1 via palladium-catalyzed site-selective cross-coupling reaction and cyclization process was described. Reaction of 3-bromo-4-trifloxy-quinolin-2(1H)-one 3 with arylboronic acid catalyzed by PdCl2(PPh3)2 afforded 3-bromo-4-aryl-quinolin-2(1H)-one 4, which then reacted with 2-ethynylaniline 5 via Pd-catalyzed Sonogashira coupling and CuI-mediated cyclization leading to the desired 1H-indol-2-yl-(4-aryl)-quinolin-2(1H)-one 1 in good yields.

Abstract:A facile and general one-step method is presented for the synthesis of aromatic indolizine compounds 3a–n in moderate to high yields (53–99%) by reaction of the pyridinium N-ylides 1a–e, the quinolinium N-ylide 1f and the isoquinolinium N-ylide 1g with various olefinic dipolarophiles, such as acrylonitrile 2a, methyl acrylate 2b, acrylamide 2c, diethyl maleate 2d and methyl crotonate 2e, respectively, in the presence of a new oxidant TPCD [Copy4(HCrO4)2, tetrapyridinecobalt(II) dichromate] at 90 °C for 2 h in DMF.

Abstract:An efficient and practical one-pot domino synthesis of 1-phenyl-1H-indol-2-amine and 5-amino-indolo[1,2-a]quinazoline derivatives from readily available 2-(2-bromophenyl)acetonitriles was developed. The overall protocol involves a Buchwald-Hartwig type coupling and a base-promoted intramolecular nucleophilic reaction. The reaction scope, advantages and limitations are discussed.

Abstract:A simple and mild approach to access polycyclic 8H-indolo[3,2,1-de]phenanthridin-8-ones through a visible-light induced free radical cyclization of (2-bromophenyl)(9H-carbazol-9-yl)methanones using benzothiadiazole-containing photocatalyst is reported. This transition metal free method can be utilized for the synthesis of 7H-thieno[2′,3′:4,5]pyrido[3,2,1-jk]carbazol-7-one, 14H-isoquinolino[4,3,2-kl]phenoxazin-14-one, 14H-isoquinolino[4,3,2-kl]phenothiazin-14-one, and 10,11,12,13-tetrahydro-8H-indolo[3,2,1-de]phenanthridin-8-one. Moreover, some products were found to emit strong blue or green fluorescence.

Abstract:A novel cyclization reaction of pyridinium 1,4-zwitterionic thiolates and propiolic acid derivatives mediated by triethylamine is described, which allows the facile synthesis of indolizines under mild reaction conditions. The net transformation involves an acetylide-driven formal [5 + 1] annulation reaction followed by a spontaneous ring-contraction/sulfur extrusion process of transient pyridothiazine intermediates.
