Abstract:A convenient synthesis of 1-oxo-1,2,3,4-tetra-hydrocarbazoles has been developed by reaction of 2-aminocyclohexanone hydrochlorides with various phenylhydrazine hydrochlorides via Fischer indole synthesis under mild conditions. The method is more satisfactory in terms of the easy availability of starting materials and the simple one-pot operation.

Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:An efficient [bis-(trifluoroacetoxy)iodo]benzene (PIFA)-mediated oxidative C-N bond formation is developed for the synthesis of 1H-indazoles from readily available arylhydrazones. The reaction tolerates a wide range of functional groups and has broad scope of substrates. Moreover, this method is a relative green and reliable method for rapid preparation of substituted 1H-indazoles under mild conditions.

Abstract:An oxa-6 pi-electrocyclization of difluoroenoxysilanes with diaryl 2-indolylmethanols has been developed. In addition, a rarely reported C3-nucleophilic [3+2] cycloaddition of difluoroenoxysilanes with dialkyl 2-indolylmethanols has been disclosed. This divergent cycloaddition approach affording readily available difluoroenoxysilanes as three-atom and C2 synthons provides rapid access to fluoro 2H-pyrano[3,4-b]indoles and gem-difluoro cyclopenta[b]indoles in good to excellent yields with good functional group tolerance. The metal-free and mild conditions using only HFIP as the solvent without any external acid catalyst illuminate practical and environmentally benign advantages.

Abstract:Polysubstituted carbazoles were efficiently synthesized through direct benzannulation reaction between 1,4‐dicarbonyl compounds and indoles with catalytic amount of inexpensive zirconium(IV) chloride. This transformation proved to be regioselective and furnishes 1,4‐disubstituted and 1,2,4‐trisubstituted carbazoles with yields ranging from 26% to 91% and broad substrate scope. Moreover, this protocol benefits from using readily accessible starting materials without the need of their pre‐functionalization. The synthetic utility of the products was exemplified by functionalization of an iodocarbazole by means of Suzuki‐Miyaura reactions and by the synthesis of a 3‐deaza‐derivative of the natural product canthin‐6‐one.

Abstract:The indole unit is one of the most abundant and relevant heterocycles in natural products and pharmaceuticals. The synthesis of indoles has been a topic of research for over 100 years, and a variety of well-established classical methods are now available. The venerable Fischer indole synthesis, discovered in 1883, remains one of the most powerful and versatile routes to the indole heterocycle, although this method suffers from several drawbacks. A variety of synthetic modifications have been introduced to increase the practicality and lessen the environmental impact of the Fischer cyclization such as using mild acids, reusable catalysts, and ionic liquids. However, it is important to note that hydrazines are carcinogenic and that certain hydrazines are difficult to prepare, are unstable, or display only moderate functional-group tolerance under acidic conditions (Scheme 1a).

Abstract:The Fischer indole synthesis is perhaps the most powerful method for indole preparation, but it often suffers from low regioselectivities with unsymmetric aliphatic ketone substrates and strong acidic conditions and is not suitable for α,β-unsaturated ketones. In this article, we disclose an efficient synthesis of N-protected indoles from N-arylhydroxamic acids/N-aryl-N-hydroxycarbamates and a variety of alkynes via a cooperative gold and zinc catalysis. The zinc catalysis is similar to the related zinc ion catalysis in metalloenzymes such as human carbonic anhydrase II and substantially enhances the O-nucleophilicity of N-acylated hydroxamines by forming the corresponding Zn chelates. The Zn chelates can attack gold-activated alkynes to form O-alkenyl-N-arylhydroxamates, which can undergo facile 3,3-sigmatropic rearrangements and subsequent cyclodehydrations to yield N-protected indole products. This new chemistry offers several important improvements over the Fischer indole synthesis: a) the reaction conditions are mildly acidic and can tolerate sensitive groups such as Boc; b) broader substrate scopes including substrates with pendant carbonyl groups (reactive in the Fischer chemistry) and alkyl chlorides (e.g., 3f); c) better regioselectivities for the formation of 2-substituted indoles under much milder conditions; d) 2-alkenylindoles can be prepared readily in good to excellent yields, but the Fischer chemistry could not; e) with internal alkynes both steric and electronic controls are available for achieving good regioselectivities, while the Fischer chemistry is in general problematic.

Abstract:A rhodium-catalyzed efficient method for the synthesis of 2,3-dihydro-1H-indazoles is described. The reaction of arylhydrazines with olefins results in the corresponding 2,3-dihydro 1H-indazoles with exclusive regioselectivity via C-H bond activation. The utility of the methodology is illustrated by a rapid synthesis of 1H-indazoles under mild reaction conditions in half an hour.

Abstract:A dehydrogenative annulation of 2-arylindazoles with maleimides for the switchable synthesis of indazolo[2,3-a]pyrrolo[3,4-c]quinolinones or spiroindolo[1,2-b]indazole-11,3'-pyrrolidinones is presented. Mechanistically, the formation of the title compounds involves a Rh(III)-catalyzed C-H metalation of 2-arylindazole, followed by maleimide insertion and intramolecular cyclization. Interestingly, the selectivity to form the fused or spiro compounds could be switched by resorting to different additives. The notable features of this protocol include simple substrates and excellent atom economy and regioselectivity.

Abstract:Three-component reactions of 2-alkynylbenzaldehydes, amines, and indoles catalyzed by AgOTf under mild conditions afford the 1-(1H-indol-3-yl)-1,2-dihydroisoquinolines in good yields. This silver-catalyzed tandem reaction is found to be workable with various indoles, anilines or alkyl amines, and 2-alkynylbenzaldehydes with electron-withdrawing groups attached on the aromatic backbone.

Abstract:An efficient intermolecular annulation of indazole aldehydes and propargylic amines has been developed for the synthesis of pyrazinoindazoles under catalyst- and additive-free conditions. This straightforward methodology features a wide substrate scope, high atom economy and environmental advantages. The bioactivity results of these new pyrazino[1,2-b]indazoles showed that some of them exhibited significant antifungal activity.

Abstract:An unexpected isomerization of azomethine ylides generated in situ from isatin with indoline-2-carboxylic acid has been disclosed, providing direct access to N -functionalized indole scaffolds. This protocol has good functional group tolerance and provides various 3-(1 H -indol-1-yl)indolin-2-one derivatives in moderate to high yields simply by using alcohol as the solvent, with no additional additive being required.
