Abstract:A heterogeneous catalyst of Ir(I)-9-amino epi-cinchonine complex immobilized on the surface of mesoporous SBA-15 was firstly synthesized and used in the asymmetric transfer hydrogenation of aromatic ketones. Enhanced enantioselectivity compared with homogeneous catalyst was obtained at room temperature. Furthermore, the heterogeneous system with very low leaching of iridium can be easily regenerated.

Abstract:A series of novel and easily accessed ferrocene-based amino-phosphine-sulfonamide (f-Amphamide) ligands have been developed and applied in Ir-catalyzed asymmetric hydrogenation of aryl ketones, affording the corresponding chiral secondary alcohols with excellent results (up to >99% conversion, >99% ee and TON up to 200 000). Abstract:A highly enantioselective iridium-catalyzed hydrogenation of α-keto amides to form α-hydroxy amides has been achieved with excellent results (up to >99% conversion and up to >99% ee, TON up to 100 000). As an example, this protocol was applied to the synthesis of (S)-4-(2-amino-1-hydroxyethyl)benzene-1,2-diol, the enantiomer of norepinephrine, which is widely used as an injectable drug for the treatment of critically low blood pressure. Density functional theory (DFT) calculations were also carried out to reveal the reaction mechanism.

Abstract:Although the transition metal-catalyzed asymmetric hydrogenation of aromatic ketones has been extensively explored, the enantioselective hydrogenation of aliphatic ketones remains a challenge because chiral catalysts cannot readily discriminate between the re and si faces of these ketones. Herein, we report a carboxyl-directing strategy for the asymmetric hydrogenation of aliphatic γ-ketoacids. With catalysis by iridium complexes bearing chiral spiro phosphino-oxazoline ligands, hydrogenation of aliphatic γ-ketoacids afforded chiral γ-hydroxylacids with high enantioselectivity (up to 99% ee). Mechanistic studies revealed that the carboxyl group of the substrate directs hydrogen transfer and ensures high enantioselectivity. Abstract:Research on the chemoselective metal‐catalyzed hydrogenation of conjugated p‐systems has mostly been focussed on enones. Herein, we communicate the understudied asymmetric hydrogenation of enimines catalyzed by N,P‐iridium complexes and chemoselective toward the alkene. A number of enoxime ethers underwent hydrogenation smoothly to yield the desired products in high yield and stereopurity (up to 99% yield, up to 99% ee). No hydrogenation of the C=N p‐bond was observed under the applied reaction conditions (20 bar H2, rt, DCM). It was demonstrated that the chiral oxime ether could be hydrolyzed into the ketone with complete preservation of the installed stereogenity at the α‐carbon. At last, a binding mode of the substrate to the active iridium catalyst and the consequence for the stereoselective outcome was proposed.

Abstract:With the assistance of hydrogen bonds, the first asymmetric hydrogenation of beta-yanocinnamic esters is developed, affording chiral beta-cyano esters with excellent enantioselectivities (up to 99% ee). This novel methodology provides an efficient and concise synthetic route to chiral GABA-derivatives such as (S)-Pregabalin, (R)-Phenibut, (R)-Baclofen. Interestingly, in this system, the catalyst with a single H-bond donor performs better than that with double H-bond donors, which is a novel discovery in the metalorganocatalysis area.

Abstract:Catalytic enantioselective functionalization of unactivated 1,1-disubstituted alkenes is challenging due to the difficulty to discriminate the enantiotopic faces. Enantioselective hydrofunctionalization of unactivated 1,1-disubstituted alkenes with tunable Markovnikov and anti -Markovnikov selectivity remains elusive. We report here an amide directed, regiodivergent and enantioselective hydroalkynylation of unactivated alkenes. The regioselectivity can be readily tuned by the choice of a proper ligand. Catalytic alkynylations occurred with tunable Markovnikov and anti -Markovnikov selectivity to afford products containing acyclic tertiary or quaternary stereocenters β to an amide. Combining a sequence of alkene isomerization and regioselective hydroalkynylation, we further realized an iridium catalyzed formal asymmetric conjugated alkynylation of β,β-disubstituted α,β-unsaturated amides. Computational studies suggest that the regioselectivity is dictated the ligand structures.

Abstract:A general and highly efficient method for asymmetric sequential hydrogenation of alpha,beta-unsaturated ketones has been developed by using an iridium/f-Ampha complex as the catalyst, furnishing corresponding chiral alcohols with two contiguous stereocenters in high yields with excellent diastereo- and enantioselectivities (up to 99% yield, >20 : 1 dr and >99% ee). Control experiments indicated that the C=C and C=O bonds of the enones were hydrogenated sequentially, and the final stereoselectivities were determined by the dynamic kinetic resolution of ketones. Moreover, DFT calculations revealed that an outer sphere pathway was involved in both reduction of C=C and C=O bonds of enones. The synthetic utility of this method was demonstrated by a gram-scale reaction with very low catalyst loading (S/C = 20 000) and a concise synthetic route to key chiral intermediates of the antiasthmatic drug CP-199,330.

Abstract:The stereoselective synthesis of 2,5-disubstituted piperidines, which is a common structural motif in bioactive compounds and organocatalysts, remains a challenging task and is relatively rare. Herein, we report a novel iridium-catalyzed asymmetric hydrogenation of 5-hydroxypicolinate salts for the first time. The asymmetric dearomatization strategy exhibits a broad substrate scope, enabling the synthesis of valuable cis-configurated hydroxypiperidine esters with distinct functionalities (ester, amino and hydroxy groups) in excellent yield, enantioselectivity, and diastereoselectivity (up to 96% yield, 97% ee and >20:1 dr). The synthetic potential of this reaction is explored through gram-scale reactions and subsequent diverse transformations, leading to the generation of chiral amine scaffolds with substantial synthetic significance (such as crucial building blocks in the manufacturing of diazabicyclooctane β-lactamase inhibitors). The successful implementation of this approach in a continuous flow manner has additionally expanded its practical applicability. Consequently, this method expands the realm of catalytic asymmetric hydrogenation for N-heteroarenes.

Abstract:We report a highly enantioselective radical-based hydroamination of enol esters with sulfonamides jointly catalyzed by an Ir photocatalyst, Brønsted base, and tetrapeptide thiol. This method is demonstrated for the formation of 23 protected β-amino-alcohol products, achieving selectivities up to 97:3 er. The stereochemistry of the product is set through selective hydrogen atom transfer from the chiral thiol catalyst to a prochiral C-centered radical. Structure-selectivity relationships derived from structural variation of both the peptide catalyst and olefin substrate provide key insights into the development of an optimal catalyst. Experimental and computational mechanistic studies indicate that hydrogen-bonding, π-π stacking, and London dispersion interactions are contributing factors for substrate recognition and enantioinduction. These findings further the development of radical-based asymmetric catalysis and contribute to the understanding of the noncovalent interactions relevant to such transformations.
