Abstract:A novel asymmetric hydrogenation of vinylthioethers was developed using a ruthenium(II) NHC complex. This method provides an efficient approach to optically active 1,5-benzothiazepines featuring stereocenters with C-S bonds. Excellent enantioselectivities (up to 95%ee) and high yields (up to 99%) were obtained for a variety of substrates bearing a range of useful functional groups. Moreover, this methodology could be directly applied to the synthesis of the antidepressant drug R-(-)-thiazesim.

Abstract:A novel class of chiral glycosyl-triazole-based P,N ligands were synthesized by 'click chemistry' and were applied to the Rh-catalyzed asymmetric hydrosilylation of a range of substituted acetophenones. Enantioselective hydrosilylation of acetophenone gave (S)-1-phenylethanol in moderate enantioselectivity (72% ee) and in good conversion (93%).

Abstract:A series of chiral spiroketal bisphosphine ligands containing 1,1'-spirobi(3 H,3' H)isobenzofuran backbones was accessed through asymmetric synthesis and subsequently tested in enantioselective Rh-catalyzed hydrogenation of α-dehydroamino acid esters. The ligand providing the highest enantioselectivity (up to 99.5%) was obtained in seven steps in an overall 38% yield. The synthesis could be performed on a gram scale, and no kinetic resolution of enantiomers is required. Overall, the developed ligand provides an easily accessible alternative to SDP ligands as well as other chiral bisphosphine ligands.

Abstract:Chiral iminopyridine oxazoline (IPO) ligands were designed, synthesized and utilized for the first cobalt-catalyzed highly regio- and enantioselective anti-Markovnikov hydroboration of 1,1-disubstituted aryl alkenes. Abstract:The highly regio- and enantioselective iron-catalyzed anti-Markovnikov hydroboration of 1,1-disubstituted aryl alkenes is reported by using a novel chiral iminopyridine oxazoline (IPO) ligand, in which the iminopyridine group is proposed to stabilize the iron and chiral oxazoline group to control enantioselectivity. This distinct class of reactive IPO ligands will likely be of high value for a large variety of asymmetric transformations using first-row transition metals.

Abstract:A series of novel and easily accessed ferrocene-based amino-phosphine-sulfonamide (f-Amphamide) ligands have been developed and applied in Ir-catalyzed asymmetric hydrogenation of aryl ketones, affording the corresponding chiral secondary alcohols with excellent results (up to >99% conversion, >99% ee and TON up to 200 000). Abstract:Air-stable P-chiral dihydrobenzooxaphosphole oxazoline ligands were designed and synthesized. When they were used in the iridium-catalyzed asymmetric hydrogenation of unfunctionalized 1-aryl-3,4-dihydronaphthalenes under one atmosphere pressure of H2 , up to 99:1 e.r. was obtained. High enantioselectivities were also observed in the reduction of the exocyclic imine derivatives of 1-tetralones.

Abstract:A series of bis‐cyclometalated chiral‐at‐metal iridium(III) complexes containing a coordinated secondary phosphine oxide (SPO) have been synthesized and evaluated as catalysts in the asymmetric transfer hydrogenation (ATH) of acetophenone. The catalytic results show that SPO ligands have a non‐innocent role in activating the catalytic process. Additionally, it has been observed that for the same chiral descriptor (D‐at‐Ir or L‐at‐Ir), the major enantiomer formed depends on the nature of the cyclometalating ligand. These enantiodivergent results contravene the general assumption that the chiral‐at‐metal core's chirality dictates the sense of the chiral induction. A combined analysis of the main structural features of the catalysts deduced from XRD structures and in situ NMR spectroscopy allowed us to propose a simplified catalytic cycle and a working hypothesis to explain the observed enantioselectivities

Abstract:Since its early stages, ligands incorporating P-stereogenic phosphine/aminophosphine donor groups have shown significant potential in metal-catalyzed asymmetric hydrogenation. Despite the initial promise, their synthesis presented challenges that led to a period of reduced attention from the scientific community. However, recent advancements in the development of more straightforward methodologies for introducing chirality to the phosphine moiety have given rise to new P-stereogenic phosphine/aminophosphine-containing ligands for this process. This chapter summarizes the progress made in this field from the end of 2010 to the present.

Abstract:Research on the chemoselective metal‐catalyzed hydrogenation of conjugated p‐systems has mostly been focussed on enones. Herein, we communicate the understudied asymmetric hydrogenation of enimines catalyzed by N,P‐iridium complexes and chemoselective toward the alkene. A number of enoxime ethers underwent hydrogenation smoothly to yield the desired products in high yield and stereopurity (up to 99% yield, up to 99% ee). No hydrogenation of the C=N p‐bond was observed under the applied reaction conditions (20 bar H2, rt, DCM). It was demonstrated that the chiral oxime ether could be hydrolyzed into the ketone with complete preservation of the installed stereogenity at the α‐carbon. At last, a binding mode of the substrate to the active iridium catalyst and the consequence for the stereoselective outcome was proposed.

Abstract:Iridium-based asymmetric reduction of ketones to chiral enantiomerically enriched alcohols has recently attracted attention by a number of research groups and interest in this area is growing. This review presents the different catalytic systems based on iridium complexes that have been used in asymmetric hydrosilylation, in asymmetric transfer hydrogenation (ATH) with alcohols or formic acid derivatives as reducing agents, and in asymmetric hydrogenation (H2 as reducing agent). A large variety of chiral ligands of various denticities and bearing various combination of coordinating atoms (N, P, S, O, C, …) have been used and will be presented. The last part critically reviews the mechanistic understanding of all the above transformations with specific reference to iridium catalysts.
