Abstract:A heterogeneous catalyst of Ir(I)-9-amino epi-cinchonine complex immobilized on the surface of mesoporous SBA-15 was firstly synthesized and used in the asymmetric transfer hydrogenation of aromatic ketones. Enhanced enantioselectivity compared with homogeneous catalyst was obtained at room temperature. Furthermore, the heterogeneous system with very low leaching of iridium can be easily regenerated.

Abstract:The stereoselective synthesis of 2,5-disubstituted piperidines, which is a common structural motif in bioactive compounds and organocatalysts, remains a challenging task and is relatively rare. Herein, we report a novel iridium-catalyzed asymmetric hydrogenation of 5-hydroxypicolinate salts for the first time. The asymmetric dearomatization strategy exhibits a broad substrate scope, enabling the synthesis of valuable cis-configurated hydroxypiperidine esters with distinct functionalities (ester, amino and hydroxy groups) in excellent yield, enantioselectivity, and diastereoselectivity (up to 96% yield, 97% ee and >20:1 dr). The synthetic potential of this reaction is explored through gram-scale reactions and subsequent diverse transformations, leading to the generation of chiral amine scaffolds with substantial synthetic significance (such as crucial building blocks in the manufacturing of diazabicyclooctane β-lactamase inhibitors). The successful implementation of this approach in a continuous flow manner has additionally expanded its practical applicability. Consequently, this method expands the realm of catalytic asymmetric hydrogenation for N-heteroarenes.

Abstract:With the assistance of hydrogen bonds, the first asymmetric hydrogenation of beta-yanocinnamic esters is developed, affording chiral beta-cyano esters with excellent enantioselectivities (up to 99% ee). This novel methodology provides an efficient and concise synthetic route to chiral GABA-derivatives such as (S)-Pregabalin, (R)-Phenibut, (R)-Baclofen. Interestingly, in this system, the catalyst with a single H-bond donor performs better than that with double H-bond donors, which is a novel discovery in the metalorganocatalysis area.

Abstract:A stable iridium catalyst bearing a chiral 2-pyridyl imidazoline ligand is described for the asymmetric transfer hydrogenation (ATH) of quinolines and N-heteroaryl compounds. This ATH system employs formic acid as the hydrogen source and water as the solvent. Various quinolines and N-heteroaryl compounds can be efficiently converted into desired products with moderate to good enantioselectiviy at as low as 0.001 mol% of catalyst loading under mild conditions.

Abstract:Research on the chemoselective metal‐catalyzed hydrogenation of conjugated p‐systems has mostly been focussed on enones. Herein, we communicate the understudied asymmetric hydrogenation of enimines catalyzed by N,P‐iridium complexes and chemoselective toward the alkene. A number of enoxime ethers underwent hydrogenation smoothly to yield the desired products in high yield and stereopurity (up to 99% yield, up to 99% ee). No hydrogenation of the C=N p‐bond was observed under the applied reaction conditions (20 bar H2, rt, DCM). It was demonstrated that the chiral oxime ether could be hydrolyzed into the ketone with complete preservation of the installed stereogenity at the α‐carbon. At last, a binding mode of the substrate to the active iridium catalyst and the consequence for the stereoselective outcome was proposed.

Abstract:A highly regio- and enantioselective cobalt-catalyzed hydroboration/hydrogenation of internal alkynes with HBpin and a hydrogen balloon in one pot was developed. A new type of chiral imidazoline iminopyridine (IIP) ligand was introduced for the first time in this novel and efficient strategy. This protocol used relatively simple and available starting materials with good functional group tolerance to construct more valuable chiral secondary organoboronates. The primary mechanistic studies illustrated that the cobalt-catalyzed regioselective hydroboration of alkynes did initially occur followed by HBpin-promoted and cobalt-catalyzed enantioselective hydrogenation of alkenylboronates.

Abstract:New chiral phosphine-oxazoline ligands (7, SIPHOX) with a rigid and bulky spirobiindane scaffold were synthesized, starting with optically pure 7-diphenylphosphino-7'-trifluoromethanesulfonyloxyl-1,1'-spirobiindane, in four steps in 40-64% overall yield. Iridium complexes of 7, the chiral analogues of the Crabtree catalyst, were generated by coordination of ligands 7 and [Ir(COD)Cl](2) in the presence of sodium tetrakis-3,5-bis(trifluoromethyl)phenylborate. The complexes were characterized by NMR, ESI-MS, and X-ray diffraction analysis. The Ir-SIPHOX complexes can catalyze the hydrogenation of acyclic N-aryl ketimines under ambient pressure with excellent enantioselectivities (up to 97% ee) and full conversions. This result represents the highest enantioselectivity and the first example of the hydrogenation of imines catalyzed by chiral analogues of the Crabtree catalyst at ambient pressure. Studies on the stability of the catalysts revealed that the catalysts Ir-SIPHOX are very stable and resistant to the formation of inactive trimers under hydrogenation conditions. On the basis of the X-ray diffraction analysis of the structures of catalysts and amine products, a rational explanation for the enantiocontrol of the chiral catalysts in the hydrogenation of imines is proposed.

Abstract:AbstractIonic hydrogenation has not been extensively explored, but is advantageous for challenging substrates such as unsaturated intermediates. Reported here is an iridium‐catalyzed hydrogenation of oxocarbenium ions to afford chiral isochromans with high enantioselectivities. A variety of functionalities are compatible with this catalytic system. In the presence of a catalytic amount of the Brønsted acid HCl, an α‐chloroether is generated in situ and subsequentially reduced. Kinetic studies suggest first‐order kinetics in the substrate and half‐order kinetics in the catalyst. A positive nonlinear effect, together with the half kinetic order, revealed a dimerization of the catalyst. Possible reaction pathways based on the monomeric iridium catalyst were proposed and DFT computational studies revealed an ionic hydrogenation pathway. Chloride abstraction and the cleavage of dihydrogen occur in the same step.

Abstract:The asymmetric transfer hydrogenation of various dibenzo‐fused azepines including 5H‐dibenzo[b,e][1,4]diazepines, dibenzo[b,f][1,4]thiazepines and 11H‐dibenzo[b,e]azepines using chiral iridium diamine catalysts and HCO2H/NEt3 as the hydrogen source has been accomplished. A rang of chiral 10,11‐dihydro‐5H‐dibenzo[b,e][1,4]diazepines, 10,11‐dihydrodibenzo[b,f][1,4]thiazepine and 6,11‐dihydro‐5H‐dibenzo[b,e]azepines have been prepared in 82‐94% yields with 82‐99% ee. Diversely substituted substrates are suitable for this transformation, and a number of functional groups are tolerated. The enantiocontrol is achieved via judicious choice of catalyst, additive and hydrogen source. The synthetic potential of this reaction is explored through gram‐scale reactions without loss of reactivity and optical purity and further transformations on products.

Abstract:A series of novel and easily accessed ferrocene-based amino-phosphine-sulfonamide (f-Amphamide) ligands have been developed and applied in Ir-catalyzed asymmetric hydrogenation of aryl ketones, affording the corresponding chiral secondary alcohols with excellent results (up to >99% conversion, >99% ee and TON up to 200 000). Abstract:Highly efficient asymmetric hydrogenation of dibenzo[b,f][1,4]oxazepine, dibenzo[b,f][1,4]thiazepine and dibenzo[b,e]azepine derivatives with chiral cationic ruthenium diamine catalysts has been developed, giving diverse chiral seven-membered N-heterocycles with excellent results (up to 99% yield and 99% ee).
