The reaction proceeds through alkene isomerization followed by regioselective hydroalkynylation. Abstract:Catalytic enantioselective functionalization of unactivated 1,1-disubstituted alkenes is challenging due to the difficulty to discriminate the enantiotopic faces. Enantioselective hydrofunctionalization of unactivated 1,1-disubstituted alkenes with tunable Markovnikov and anti -Markovnikov selectivity remains elusive. We report here an amide directed, regiodivergent and enantioselective hydroalkynylation of unactivated alkenes. The regioselectivity can be readily tuned by the choice of a proper ligand. Catalytic alkynylations occurred with tunable Markovnikov and anti -Markovnikov selectivity to afford products containing acyclic tertiary or quaternary stereocenters β to an amide. Combining a sequence of alkene isomerization and regioselective hydroalkynylation, we further realized an iridium catalyzed formal asymmetric conjugated alkynylation of β,β-disubstituted α,β-unsaturated amides. Computational studies suggest that the regioselectivity is dictated the ligand structures.

Abstract:We report iridium-catalyzed C-C formation between benzamides and terminal alkynes. With the choice of a suitable ligand, a C-H alkynylation or alkenylation product could be obtained selectively. The directed C-H alkynylation proceeded without the need for an external oxidant, while the directed C-H alkenylation likely involves an unusual vinylidene mechanism. This divergent reactivity provides access to both alkynylation and alkenylation products from the same set of starting materials.

Abstract:Transition-metal-catalyzed asymmetric hydroalkenylation of alkenes provides an atom-economical method to build molecular complexity from easily available materials. Herein we report an iridium-catalyzed asymmetric hydroalkenylation of unconjugated alkenes with acrylamides and acrylates. The catalytic hydroalkenylation of norbornene derivatives occurred to form products with allylic stereocenters with high chemo-, regio-, and stereoselectivities. DFT calculations revealed that the migratory insertion is irreversible and the enantiodetermination step.

Abstract:Iodine-mediated intermolecular C-N and C-I bond formation of alkenes is described. A series of alkenes could be selectively converted into the corresponding aminoiodination products, which are versatile building blocks in organic synthesis and medicinal chemistry. Abstract:We report here an iridium-catalyzed hydroalkynylation of allylic alcohols protected by an oxime group. Catalytic alkynylation occurs exclusively at the distal position of the alkene. This method generates γ-alkynyl alcohol oximes directly from internal alkenes and terminal alkynes. The oxime group can be readily removed to afford a free alcohol, thus providing an indirect route for the catalytic hydroalkynylation of allylic alcohols.

Abstract:Herein we report an iridium(I)-catalyzed atroposelective alkenylation of isoquinoline-derived heterobiaryls with terminal alkynes. In the presence of a cationic iridium(I) catalyst with (R)-SEGPHOS as the chiral ligand, this atom-economical alkenylation protocol allows the rapid construction of a series of axially chiral alkenylated heterobiaryls in moderate to good yields with good to high enantioselectivities.

Abstract:A novel iridium-catalyzed hydroxyl-enabled cycloaddition of azides and alkynes has been developed. In the presence of catalytic amount (2 mol%) of [Ir(cod)Cl](2), the 2-alkynyl phenols would engage in a regioselective cycloaddition with various azides for rapid access to diverse triazoles, and the hydroxyl group acts as directing group to enable this reaction. The process is featured mild and biocompatible reaction condition with the achievement of brevity and diversity. Furthermore, the hydroxyl group offers ample opportunity for late-stage divergent transformations of the products.

Abstract:C-H amination of arenes directed by weakly coordinating Weinreb amides has been achieved with an iridium catalyst and 2,2,2-trichloroethoxycarbonyl (Troc) azide as an aminating agent, providing a robust method of producing synthetic useful ortho-TrocNH aryl Weinreb amides. Taking advantage of the reactivity of Weinreb amide and Troc groups in the amination products, selective hydrolysis was achieved as an attractive process for the synthesis of ortho-NH2 aryl Weinreb amides, which are the building blocks useful in the synthesis of bioactive compounds, and cascade aminocyclization with primary amines was successful and provided an efficient pathway for the construction of quinazolin-2,4-diones, which are present in various alkaloids and natural products.

Abstract:An iridium-catalyzed enantioselective hydroalkynylation of α,β-unsaturated amides was described. The selectivity of this reaction is distinct from that observed in many catalytic hydroalkynylations of α,β-unsaturated carbonyl compounds. It occurs selectively at the γ instead of the β position. Preliminary mechanistic studies suggest that the reaction proceeds through alkene isomerization followed by hydroalkynylation. This method provides a straightforward route for the synthesis of amides with a remote stereocenter at the γ position.

Abstract:An electrochemical approach in promoting directed C-H alkynylation with terminal alkyne via iridium catalysis is reported. This work employed anodic oxidation of Ir(III) intermediate (characterized by X-ray crystallography) to promote reductive elimination, giving the desired coupling products in good yields (up to 95%) without the addition of any other external oxidants. This transformation is suitable for various directing groups with H2 as the only by-product, which warrants a high atom economy and practical oxidative C-C bond formation under mild conditions.

Abstract:The general protocol for site-specific alkylation of anilines and phenols would provide an ideal strategy for affording densely functionalized aniline and phenol derivatives. Herein, we disclose a new iridium-catalyzed and acid-promoted process for selective para -alkylation of anilines and phenols using aryloxy or alkoxy aryl alkynes as the alkylated sources, delivering a wide range of para - C -alkylated phenols and anilines in good to excellent yields. The low catalyst loading (up to 0.05 mol%), scale-up of synthesis, operation under air conditions, as well as recycling of catalyst (5 times without deactivation) evidenced the promising application in organic synthesis.

Abstract:An iridium(i)-catalyzed hydration/esterification of 2-alkynylphenols and carboxylic acids is described. Various 2-alkynylphenols and carboxylic acids could be used in this process to furnish aromatic ortho-acyloxyketones via a regio- and stereo-selective addition reaction followed by intramolecular rearrangement.
