Abstract:The convenient and precise fabrication of drug-hydrogel formulations with satisfactory degradability and a well-controlled drug release profile are crucial factors for injectable hydrogel formulations in clinical applications. Here a new injectable thermosensitive hydrogel formed from poly(ε-caprolactone) (PCL)-poly(ethylene glycol)-poly(ε-caprolactone) amphiphilicco-polymers with 1,4,8-trioxa[4.6]spiro-9-undecanone (TOSUO) moieties incorporated in the poly(ε-caprolactone) (PCL)block (PECT) was constructed to provide a route to tailor the degradation and drug release behavior. The effect of hydrophilic cyclic ether moieties on the degradation of and drug release by PECT hydrogels were evaluated in vitro and in vivo. The results indicated that a freeze-dried powder of paclitaxel-loaded PECT nanoparticles rapidly dissolved in water at ambient temperature with slightly shaking and formed a stable injectable in situ drug-hydrogel formulation at body temperature, which is convenient for clinical operations because it avoids the need for pre-quenching or long-term incubation. The paclitaxel distribution was also more quantitative and homogeneous on entrapping paclitaxel in PECT nanoparticles. Further, the small number of pendant cyclic ether groups in PCL could decrease the cystallinity and hydrophobicity and, as a result, the in vitro and in vivo retention time of PECT hydrogels and the release of entrapped paclitaxel could be tuned from a few weeks to months by varying the amount of PTOSUO in the hydrophobic block. Significantly, paclitaxel-loaded PECT nanoparticles and free paclitaxel could be simultaneously released during the in vitro paclitaxel release from PECT hydrogels. A histopathological evaluation indicated that in vivo injected PECT hydrogels produced only a modest inflammatory response. Thus pendant cyclic ether modification of PCL could be an effective way to achieve the desired degradation and drug release profiles of amphiphilicco-polymer thermosensitive hydrogels and PECT hydrogels may be suitable for local drug delivery.

Abstract:Poly(vinylcaprolactam) (PVCL)-based biodegradable microgels were prepared for the biomedical application as drug delivery system via precipitation polymerization, where N,N-bis(acryloyl) cystamine (BAC) served as cross-linker, methacrylic acid (MAA) and polyethylene glycol (PEG) methyl ether methacrylate acted as comonomers. The microgels with excellent stability had distinct temperature sensitivity as largely observed in the case of PVCL-based particles and their volume phase transition temperature (VPTT) shifted to higher temperature with increasing MAA content and ambient pH. In the presence of reducing agent glutathione (GSH) or dithiothreitol (DTT), the microgels could be degraded into individual linear polymer chains by the cleavage of the disulfide linkages coming from the cross-linker BAC. The microgels could effectively encapsulate Doxorubicin (DOX) inside and presented stimuli-triggered drug release in acidic or reducing environment. The results of the cytotoxicity assays further demonstrated that the blank microgels were nontoxic to normal cells while DOX-loaded microgels presented efficient antitumor activity to HeLa cells.

Abstract:To improve the biocompatibility and biodegradability of nanocarriers, well-defined poly(vinylcaprolactam)-based acid degradable nanogels were fabricated for drug delivery via precipitation polymerization in water, where synthetic ketal-based 2,2-dimethacroyloxy-1-ethoxypropane (DMAEP) acted as a cross-linker, and N-(2-hydroxypropyl)methacrylamide (HPMA) served as a co-monomer. Expectedly, we observed that the temperature and pH of the environment play important roles in the performance of the nanogels. The nanogels were reduced in size upon increasing the temperature and showed higher volume phase transition temperature (VPTT) with higher concentration of HPMA. With the incorporation of ketal linkages, the nanogels showed accelerated degradation profiles by lowering the pH and increasing temperature of the incubation medium. When used as nanocarriers of anticancer drug doxorubicin (DOX), compared to non-degradable nanogels with similar components, the acid-degradable nanogels displayed more effective drug controlled release behaviour, low drug leakage of DOX at neutral pH while rapid and sufficient release from the nanogels under acidic conditions. The results of the cytotoxicity and hemolysis assays further highlighted that the acid-degradable nanogel produced no hemolysin but showed excellent viability to normal cells, and the DOX-loaded nanogel exhibited higher proliferation inhibition against tumor cells.

Abstract:This paper investigates drug release from a novel series of mPEG-functionalised PLLA polymers whose individual components (PEG and PLLA) have regulatory FDA approval. Two processing methods were explored to understand their effect on the morphology and drug release profiles of the polymers, with and without mPEG functionalisation. In the first method the polymer and Propranolol.HCl drug powders were mixed together before injection moulding. In the second method, supercritical CO2 was used to mix the polymer and drug before injection moulding. When non-functionalised PLLA was processed through injection moulding alone, there were no signs of polymer-drug interaction, and the drug was confined to crystals on the surface. This resulted in up to 85 wt% burst release of propranolol.HCl after one day of incubation. By contrast, injection moulding of mPEG-functionalised polymers resulted in the partial dissolution of drug in the polymer matrix and a smaller burst (50 wt% drug) followed by sustained release. This initial burst release was completely eliminated from the profile of mPEG-functionalised polymers processed via supercritical CO2. The addition of mPEG facilitated the distribution of the drug into the bulk matrix of the polymer. Paired with supercritical CO2 processing, the drug release profile showed a slow, sustained release throughout the 4 months of the study.
GSH-responsive polymeric micelles based on the thio–ene reaction for controlled drug release

Abstract:We report a glutathione (GSH) responsive drug carrier that is based on the Michael addition reaction between an olefinic bond and GSH. Amphiphilic copolymers with multi-linear-cyclized and branched structures that contain vinyl groups were successfully synthesized by deactivation enhanced atom transfer radical polymerization (DE-ATRP). The polymers self-assembled into GSH responsive micelles with sizes of 68 to 97 nm and spherical morphology. The core cross-linked micelles were confirmed to greatly decrease doxorubicin (DOX) leakage due to the concentration gradient. Drug release experiments indicated that the drug carrier containing no disulfide has the same responsive ability as a conventional GSH reduction responsive carrier with a similar polymer structure. MTT assays showed that the blank micelles are totally non-cytotoxic, while the DOX-loaded micelles exhibit GSH-dependent behaviour.
Biodegradable Strain-Promoted Click Hydrogels for Encapsulation of Drug-Loaded Nanoparticles and Sustained Release of Therapeutics

Abstract:Biodegradable polycarbonate-based ABA triblock copolymers were synthesized via organocatalyzed ring-opening polymerization and successfully formulated into chemically cross-linked hydrogels by strain-promoted alkyne-azide cycloaddition (SPAAC). The synthesis and cross-linking of these polymers are copper-free, thereby eliminating the concern over metallic contaminants for biomedical applications. Gelation occurs rapidly within a span of 60 s by simple mixing of the azide- and cyclooctyne-functionalized polymer solutions. The resultant hydrogels exhibited pronounced shear-thinning behavior and could be easily dispensed through a 22G hypodermic needle. To demonstrate the usefulness of these gels as a drug delivery matrix, doxorubicin (DOX)-loaded micelles prepared using catechol-functionalized polycarbonate copolymers were incorporated into the polymer solutions to eventually form micelle/hydrogel composites. Notably, the drug release rate from the hydrogels was significantly more gradual compared to the solution formulation. DOX release from the micelle/hydrogel composites could be sustained for 1 week, while the release from the micelle solution was completed rapidly within 6 h of incubation. Cellular uptake of the released DOX from the micelle/hydrogel composites was observed at 3 h of incubation of human breast cancer MDA-MB-231 cells. A blank hydrogel containing PEG-(Cat)12 micelles showed almost negligible toxicity on MDA-MB-231cells where cell viability remained high at >80% after treatment. When the cells were treated with the DOX-loaded micelle/hydrogel composites, there was a drastic reduction in cell viability with only 25% of cells surviving the treatment. In all, this study introduces a simple method of formulating hydrogel materials with incorporated micelles for drug delivery applications.

Abstract:Recently, nano- and micro-particulate systems have been widely utilized to deliver pharmaceutical compounds to achieve enhanced therapeutic effects and reduced side effects. Poly (DL-lactide-co-glycolide) (PLGA), as one of the biodegradable polyesters, has been widely used to fabricate particulate systems because of advantages including controlled and sustained release, biodegradability, and biocompatibility. However, PLGA is known for low encapsulation efficiency (%) and insufficient controlled release of water-soluble drugs. It would result in fluctuation in the plasma levels and unexpected side effects of drugs. Therefore, the purpose of this work was to develop microcapsules loaded with alginate-coated chitosan that can increase the encapsulation efficiency of the hydrophilic drug while exhibiting a controlled and sustained release profile with reduced initial burst release. The encapsulation of nanoparticles in PLGA microcapsules was done by the emulsion solvent evaporation method. The encapsulation of nanoparticles in PLGA microcapsules was confirmed by scanning electron microscopy and confocal microscopy. The release profile of hydrophilic drugs can further be altered by the chitosan coating. The chitosan coating onto alginate exhibited a less initial burst release and sustained release of the hydrophilic drug. In addition, the encapsulation of alginate nanoparticles and alginate nanoparticles coated with chitosan in PLGA microcapsules was shown to enhance the encapsulation efficiency of a hydrophilic drug. Based on the results, this delivery system could be a promising platform for the high encapsulation efficiency and sustained release with reduced initial burst release of the hydrophilic drug.

Abstract:Stimuli responsive drug delivery systems have attracted considerable attention due to their ability to deliver drugs on demand to special tissues and cells (e.g. cancer cells). In this paper, we fabricated a new kind of redox/pH dual stimuli-responsive biodegradable poly(methacrylic acid) (MAA) nanohydrogels crosslinked by N,N-bis(acryloyl)cystamine (BACy) through a facile reflux-precipitation polymerization. Subsequently polyethylene glycol (PEG) was grafted onto the surface of nanohydrogels to improve their long circulation time. This kind of PEGylated nanohydrogels could be degraded into short linear polymer chains in the solution of 10 mM glutathione (GSH) due to the sensitive disulfide bonds in the cross-linked networks. Interestingly, we found that the hydrophilic PEGylated nanohydrogels could be simultaneously loaded with hydrophilic drug doxorubicin (DOX) and hydrophobic drug paclitaxel (PTX) with high drug loading efficiency because of its regional hydrophobic nature in the nanohydrogels. The cumulative release profile showed that either lower pH (5.0) or reducing environment (GSH) or a combination could significantly accelerate the drug release from the drug-loaded nanohydrogels. In addition, the experimental results on living cells indicated that the multi-drug-loaded system had synergistic effect on cancer therapy. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:Novel water-soluble polyphosphazene containing carboxylatophenamino groups (PCPAP) was synthesized by the substitution reaction of ethyl p-amino benzonate with poly(dichlorophosphazene), followed by alkali hydrolysis. Characterizations by IR, H-1-NMR, differential scanning calorimetry, and elemental analysis indicated that the reaction brought about an almost complete introduction of carboxylatophenamino to the polymer side chain. Calcium-crosslinked PCPAP hydrogel beads were accomplished with an extremely mild method. The erosion experiments were conducted in vitro in various pH environments. The erosion duration of the beads at pH 7.4 and 37degreesC was effectively extended by an increase in the concentration of the PCPAP or CaCl2 solution during the preparation process. Moreover, the bead erosion was sensitive to the pH. The sample dissolved 39.4% in a pH 8.0 buffer within 34 days but only 5.36% in a pH 5.0 buffer. Furthermore, PCPAP underwent degradation into macromolecular segments through the breaking of the backbone, and this could prevent accumulation in the body. Abstract:Zwitterionic polymers play an attractive role in the application of stealthy nanocarriers for their excellent antifouling property. Herein, a zwitterionic nanogel with temperature sensitivity and redox-responsive degradability prepared by copolymerization of N-vinylcaprolactam (VCL) and 2-(methacryloyloxy) ethyldimethyl-(3-sulfopropyl) ammonium hydroxide (DMAPS) via aqueous precipitation polymerization. The prepared nanogels own ultra-high colloidal stability and non-specific protein adsorption resistance as a result of the incorporation of zwitterionic groups. Meanwhile, they exhibit sensitive temperature-induced swelling/collapse transition in aqueous solution and excellent redox-degradability ascribed to the presence of disulfide bonds. The nanogels loaded with anticancer drug doxorubicin (DOX) exhibit low leakage of DOX under physiological conditions (merely 23.8 % within 24 h), whereas striking release amount of DOX under reducing conditions combined with elevated temperature (93.4 % within 24 h). The measurement of cell viability showed that the cytotoxicity of blank nanogels to tumor cells (HeLa cells) was negligible, while the nanogels loaded with DOX had a prominent inhibitory impact on tumor cells.
Controlled Release of Lysozyme-Loaded Microspheres in Hydrogel Implants

Abstract:Drug-loaded poly(lactic-co-glycolic) acid(PLGA) microspheres were dispersed in methylcellulose reverse thermo-sensitive hydrogel for long-term sustained release of biomacromolecular drugs.The in vitro release properties of the model drug lysozyme in the mixture formulation were examined.The influence of the hydrogel on the degradation of the PLGA was investigated using gel permeation chromatography(GPC) and a scanning electron microscope(SEM).The mechanisms for drug release from the microspheres in the hydrogel were deduced by Peppas' formula.The results show that the mixture formulation exhibits nearly zero-order release of lysozyme for over 30 days without any burst effect.The drug release is controlled by both drug diffusion and material erosion.

Abstract:The injectable gelling drug delivery system incorporated with biodegradable microspheres has great potential in implanted drug delivery for long-term controlled drug release.We investigated drug release in vitro in a model system which was composed by naltrexone-loaded PLGA microspheres and thermosensitive hydrogel consisted of methylcellulose,polyethylene glycol,sodium citrate and sodium alginate.In the microspheres-hydrogel composite system,drug release was dominated by drug diffusion in microspheres.The hydrogel affected the porosity,erosion and degradation of the microspheres,which resulted in the smaller drug diffusion coefficient,lower erosion rate and prolonged period to reach the maximal erosion rate.Therefore,it reduced and stabilized the drug release rate.These impacts of hydrogel were presented in the drug release model based on drug release from biodegradable polymeric microspheres,and the developed model was well consistent with the experiment.The microspheres-hydrogel composite system realized the over 60-day steady release of naltrexone by optimizing the molecular weight and hyhydrophilicity of PLGA.
Bioresponsive Controlled Drug Release Based on Mesoporous Silica Nanoparticles Coated with Reductively Sheddable Polymer Shell

Abstract:The design of bioresponsive controlled drug delivery systems is a promising approach in cancer therapy, but it still is a major challenge capable of optimum therapeutic efficacy, i.e. no premature drug leakage in blood circulation while having a rapid and complete release in tumor tissues. In this work, a kind of PEGylated core/shell structured composite nanoparticle was developed via precipitation polymerization, where a disulfide-cross-linked poly(N-vinylcaprolactam-co-methacrylic acid) (P(VCL-s-s-MAA)) polymer shell was created to act as sheddable thermo/pH-sensitive gatekeepers, and a carboxylic acid modified mesoporous silica nanoparticles (MSN-COOH) core was applicable as an accessible reservoir to encapsulate high drug doses. At physiological conditions, the P(VCL-s-sMAA)-PEG shell underwent a distinct transition from a swollen state in pH 74 to a collapsed state in pH 5.0. Though sufficiently stable in water, composite nanoparticles were prone to fast dissociation and rupture when subjecting to 10 mM glutathione (GSH), due to the shedding of polymer walls through reductive cleavage of intermediate disulfide bonds, so that the polymer shell was active in moderating the diffusion of embedded drugs in-and-out of MSN channels. The cumulative in vitro release of DOX-loaded composite nanoparticles allowed a low trace of DOX diffusion below volume phase transition temperature (VPTT) and a significant release rate above its VPTT, while the most rapid and perfect release was achieved under a reductive environment (pH 6.5 and 10 mM GSH), mimicking that of intracellular cytosol compartments. The in vitro cell assay of blank carriers to normal cells indicated that the composite nanoparticles were suitable as drug carriers, but DOX-loaded carriers had a similar intensive toxicity to cancer cells compared with free DOX. Therefore, these stimuli responsive composite nanoparticles with a reductively sheddable and thermo/pH-responsive polymer shell gate could, in principle, be applied for in vivo cancer therapy, and synergistic drug delivery can be accomplished "just in time in a precise event over the location.

Abstract:Folate-conjugated poly(N-(2-hydroxypropyl)methacrylamide-co-methacrylic acid) nanohydrogels were facilely prepared via distillation–precipitation polymerization and subsequent folate modification. Due to the nanohydrogels being crosslinked with disulphide bonds, they could be easily degraded into short polymer chains in the presence of glutathione, which will be beneficial for easily discharging the nanohydrogels from the body. Doxorubicin (DOX), a clinical anti-cancer drug, was efficiently loaded in the nanohydrogels (up to 39.3 wt%) by the electrostatic interactions between the amine group in doxorubicin (DOX) and the carboxyl groups in the nanohydrogels at neutral conditions. The cumulative release profile of the DOX-loaded nanohydrogels showed a relatively low level of drug release (23 wt% in 48 h) at pH 7.4 and a quick release (over 95 wt% in 2 h) at pH 5.0 with reducing environment, exhibiting pH/redox dual-stimuli-responsive drug release. The dose-dependent cytotoxicity of the drug-loaded nanohydrogels was studied by the CCK-8 assay. The nanohydrogels possess many favourable merits of drug carriers, such as excellent biocompatibility, high drug loading capacity, minimal drug release under an extracellular condition, rapid drug release in response to the intracellular level of pH and reducing environment, and folate-mediated endocytosis, which endow them as great candidates for targeted delivery of anti-cancer drugs.

Abstract:The microphase inversion of water-insoluble poly(L-lactide), "poly[(L-lactide)-co-glycolide] and polylactide-block-poly(ethylene oxide)-block-polylactide from THF to water can result in narrowly distributed stable particles. Gliclazide, a commercial drug, can be encapsulated inside during the process. The formation and degradation of such particles was studied by laser light scattering. In comparison with the corrosion of a bulk material, the degradation of each particle is so fast that we only detect the decrease of the particle number, not the corrosion of individual particles. Therefore, the degradation is a "one-by-one" random process, just like the chemical reaction of molecules in solution. The disappearing rate of the particle number is nearly independent of time, ideal for the controlled release of drugs encapsulated inside. The amount of encapsulated gliclazide depends on the copolymer's hydrophobicity (composition), while the releasing rate mainly is, directly related to the disappearing rate of the particles. The correlation between the fluorescence intensity and the degradation was used to study the kinetics of gliclazide releasing. The releasing pattern is controllable with a proper choice of the drug loading, copolymer composition, pH and temperature.
Biodegradation and Toxicity of Protease/Redox/Ph Stimuli-Responsive Peglated Pmaa Nanohydrogels for Targeting Drug Delivery

Abstract:The application of nanomaterials in intelligent drug delivery is developing rapidly for treatment of cancers. In this paper, we fabricated a new kind of protease/redox/pH stimuli-responsive biodegradable nanohydrogels with methacrylic acid (MAA) as the monomer and N,N-bis(acryloyl)cystamine (BACy) as the cross-linker through a facile reflux-precipitation polymerization. After that, the polyethylene glycol (PEG) and folic acid (FA) were covalently grafted onto the surface of the nanohydrogels for enhancement of their long in vivo circulation lifetime and active targeting ability to the tumor cells and tissues. This kind of nanohydrogels could be disassembled into short polymer chains (Mn<1140; PDI<1.35) both in response to glutathione (GSH) through reduction of the sensitive disulfide bonds and protease by breakage of the amido bonds in the cross-linked networks. The nanohydrogels were utilized to simultaneously load both hydrophilic drug doxorubicin (DOX) and hydrophobic drug paclitaxel (PTX) with high drug loading efficiency. The cumulative release profile showed that the drug release from the drug-loaded nanohydrogels was significantly expedited by weak acidic (pH 5.0) and reducing environment (GSH), which exhibited an distinct redox/pH dual stimuli-responsive drug release to reduce the leakage of drugs before they reach tumor site. In addition, the in vitro experiment results indicated that the multidrug-loaded system had synergistic effect on cancer therapy. Meanwhile, the acute toxicity and intravital fluorescence imaging studies were adopted to evaluate the biocompatibility and biotoxicity of the nanohydrogels, the experimental results showed that the PEG modification could greatly enhance the long in vivo circulation lifetime and reduce the acute toxicity (LD50: from 138.4 mg/kg to 499.7 mg/kg) of the nanohydrogels.

Abstract:In the presence of surfactant, water-insoluble poly(D,L-lactide) (PLA) was dispersed into narrowly distributed nanoparticles stable in water via microphase inversion. The structure and degradation of such formed nanoparticles were investigated by a combination of static and dynamic laser light scattering. Our results revealed that the degradation rate increased with the temperature and pH so that the degradation could be regulated from minutes to days. Using anionic sodium dodecyl sulfate (SDS) as stabilizer resulted in a slower degradation than using cationic hexadecyltrimethylammonium bromide (HTAB). The phthalocyanine chromophores (PC) could be encapsulated inside these PLA nanoparticles. The degradation of individual PLA nanoparticles led to a controllable releasing of PC. The absorption and fluorescence studies revealed a correlation between the degradation and the releasing of PC. Our results showed that a higher PC/PLA ratio could lead to a faster degradation.
