Abstract:Nanoparticles are appealing drug delivery systems since they promise prolonged circulation time and predictable release behaviors. The current work reports a novel hyperbranched amphiphilic block copolymer synthesized using deactivation-enhanced atom transfer radical polymerization (DE-ATRP) for smart drug delivery. PEG2000-Br was applied as a macroinitiator to initiate acid-cleavable divinyl (ACD) monomers linked by acetal groups, formulating the hydrophilic (PEG) and hydrophobic (ACD) segments. The polymer appeared to self-assemble into micelles with diameters in the range of 70-100 nm, and was examined for the controlled release of doxorubicin (DOX). Results showed that DOX-loaded micelles (similar to 90 nm) could achieve drug loading as high as 8.2 wt%, with interesting pH-dependent release behaviors. Studies with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) showed that DOX-loaded micelles exhibited a high cellular uptake performance by HeLa cells, which indicated promising antitumor efficacy for such a drug delivery system. Additionally, such DOX-loaded micelles exhibited remarkable cytotoxicity against HeLa cells in a dose-and time-dependent manner due to the enhanced cell uptake behavior of micelles. These results indicated that the polymeric micelles might be used as a promising candidate for a pH-responsive drug delivery for cancer therapy.

Abstract:In this paper, a novel kind of photo-cross-linked biodegradable hydrogels based on n-arm-poly(ethylene glycol) (n=2, 3, and 4) and poly(-caprolactone) was prepared by ultraviolet-initiated free radical polymerization. The resulting n-arm-poly(ethylene glycol)-poly(-caprolactone) and n-arm-poly(ethylene glycol)-poly(-caprolactone) acrylate (n-arm-PEG-PCL-AC, macromer) were characterized by proton nuclear magnetic resonance and fourier transform infrared spectra. The influences of arm numbers and concentration of macromer on the properties of hydrogel were researched systematically, and the results showed that the gelation time, equilibrium swelling ratio, invitro degradation, and drug release rate decreased with the increase of arm numbers and concentration of macromer. The degradation and drug release rate could be controlled by varying the cross-linking density of hydrogel, indicating a potential application as controlled drug delivery system. Cytotoxicity test of hydrogel extracts was conducted using L929 mouse fibroblasts, and the relative growth rate exceeded 75% (cytotoxicity type: class 1) after incubation for 24h, showing excellent cytocompatibility. In addition, the paper presented a pH-sensitive hydrogel (G4(pH)) based on 4-arm-PEG-PCL-AC and acrylic acid, and the influences of pH value on swelling behaviors and invitro drug release of the pH-sensitive hydrogel were examined. The hydrogels shrank under acidic condition and would swell in neutral or basic medium. The pH-dependent drug release behaviors indicated a promising application of the materials as oral drug delivery vehicles.

Abstract:Hydrogels with pH-sensifive poly(acrylic acid) (PAAc) chains and biodegradable acryloyl-poly(E-caprolactone)-2-hydroxylethyl methacrylate (AC-PCL-HEMA) chains were designed and synthesized. The morphology of hydrogel was observed by scanning electron microscopy. The degradation of the hydrogel in the presence of Pseudomonas lipase was studied. The in vitro release of bovine serum albumin from the hydrogel was investigated. Cytotoxicity study shows that the AC-PCL-HEMA/AAc copolymer exhibits good biocompatibility. Cell adhesion and migration into the hydrogel networks were evaluated by using different cell lines. The hydrogel with a lower cross-linking density and a larger pore size exhibited a better performance for cells migration.

Abstract:Cellulose-based biocompatible, tunable and injectable hydrogels embedded with pH-responsive diblock copolymer micelles were constructed to achieve localized drug delivery with prolonged, stimuli-driven and slow-release function. First, we prepared two types of modified carboxymethyl cellulose (CMC) including hydrazide-modified carboxymethyl cellulose (CMC-NH2) and oxidized carboxymethyl cellulose (CMC-CHO) with varying degrees of oxidation. Then, pH-responsive poly (ethylene oxide)-block-poly (2-(diisopropylamino) ethyl methacrylate) (PEO-b-PDPA) copolymers as micelle cores to carry hydrophobic substances were also synthesized through atom transfer radical polymerization (ATRP). An injectable hydrogel composite system was finally obtained by mixing CMC-NH2 and CMC-CHO polymer suspensions containing PEO-b-PDPA copolymer micelles through a Schiff base reaction. This newly-synthesized, tunable, cellulose-based double barrier system exhibits a pH-triggered, prolonged, and slow-release profile based on the release test using both Nile Red dye and doxorubicin. The hydrogel system also exhibited comparable storage moduli and tunable degradation properties.

Abstract:In this paper, modeling and simulation of a new type of controlled drug delivery micro-device based on biodegradable polymers is reported. The micro-device consists of micro-chambers arrays for drug storage to achieve linear release. The micro-chambers are fabricated with polyanhydrides (CPP-SA) using the UV-LIGA technology and the controlled release process are the combined results of the design of the micro-chambers and the biodegradable characteristics of the polymer. This type of drug delivery system has some unique advantages in controlled long-term drug delivery, such as larger loading volume than the matrices release systems, easier control for the release rate, etc. It is necessary to optimize the structure for the long-term and zero-order drug release. Based on the Monte Carlo erosion model, the drug release model is founded for the drug delivery system and using the new model, the drug release profiles from the delivery systems with different structures are simulated. The simulated results indicate that the effect of the drug delivery is dependent on the micro-structure of the delivery system and the simulated drug profiles of coaxial rings micro-cavity shape equal to zero-order released model approximatively. The simulated results are very important to the application research of the new biodegradable polymer micro-device.

Abstract:In this work, a novel biodegradable pH-sensitive hydrogel based on poly(ɛ-caprolactone) (PCL), methoxpoly(ethylene glycol) (MPEG) and methacrylic acid (MAA) was prepared by UV-initiated free radical polymerization. The resulting macromonomers and hydrogels were characterized by FTIR and/or 1H NMR. Swelling behaviour and pH sensitivity of the hydrogels were studied in detail. With increase in pH of aqueous medium from 1.2 to 7.2, swelling ratio of the hydrogels increased accordingly. The hydrolytic degradation behaviour was also investigated. The prepared biodegradable pH-sensitive hydrogel based on PCL, MPEG, and MAA might have great potential application in smart drug delivery system.

Abstract:Abstract A new double-layer, pH-sensitive, composite hydrogel sustained-release system based on polysaccharides and synthetic polymers with combined functions of different inner/outer hydrogels was prepared. The polysaccharides inner core based on sodium alginate (SA) and carboxymethyl cellulose (CMC), was formed by physical crosslinking with pH-sensitive property. The synthetic polymer out-layer with enhanced stability was introduced by chemical crosslinking to eliminate the expansion of inner core and the diffusion of inner content. The physicochemical structure of the double-layer hydrogels was characterized. The drug-release results demonstrated that the sustained-release effect of the hydrogels for different model drugs could be regulated by changing the composition or thickness of the hydrogel layer. The significant sustained-release effect for BSA and indomethacin indicated that the bilayer hydrogel can be developed into a novel sustained delivery system for bioactive substance or drugs with potential applications in drugs and functional foods.

Abstract:In this paper, a new kind of pH-sensitive hydrogel based on poly(lactic acid) (PLA), methoxyl poly(ethylene glycol) (MPEG) and itaconic acid (IA) was prepared by heat-initiated free radical polymerization method without any organic solvent. The obtained macromonomers and hydrogels were characterized by H-1 NMR and FTIR. Detailed swelling behavior of hydrogels under variational pH-conditions revealed that the dynamic sensitivity of P(MPEG-PLA-co-IA-MEGMA) hydrogels [P(LE-IA-MEG) hydrogels] as well as the diffusional mechanisms might be causing the network expansion and collapse. In vitro drug release behavior was investigated using VB12 and paclitaxel (PTX) as model drugs, revealing that the P(LE-IA-MEG) hydrogel could control the diffusion of the drug. The prepared new kind of pH-sensitive hydrogel with great pH-responsiveness and good properties for drug delivery might have great potential application in smart drug delivery systems.

Abstract:Drug-loaded poly(lactic-co-glycolic) acid(PLGA) microspheres were dispersed in methylcellulose reverse thermo-sensitive hydrogel for long-term sustained release of biomacromolecular drugs.The in vitro release properties of the model drug lysozyme in the mixture formulation were examined.The influence of the hydrogel on the degradation of the PLGA was investigated using gel permeation chromatography(GPC) and a scanning electron microscope(SEM).The mechanisms for drug release from the microspheres in the hydrogel were deduced by Peppas' formula.The results show that the mixture formulation exhibits nearly zero-order release of lysozyme for over 30 days without any burst effect.The drug release is controlled by both drug diffusion and material erosion.

Abstract:The P(CL-PDO)-PEG-(P(CL-PDO)(PECP) copolymer hydrogel is an attractive thermogelling material for practical applications due to the fast dissolution of the copolymer in water and good stability of the resultant sol solution that is beneficial for guest matter mixing. In this paper, the degradation properties and drug release behavior of the hydrogel loaded with various types of drugs were evaluated. The integrity of the PECP hydrogel could preserve for 2 weeks and became viscous liquid after degradation time of 21 weeks. With the degradation time, both the molecular weight and weight loss of the hydrogel decreased gradually. NMR analysis of the degraded products indicated that the chain breaking of the copolymer mainly occurred within the sequence structures of PDO-PDO or PDO-PCL, implying the acceleration effect of PDO unit to the degradation of hydrogel. Three distinct types of drugs including small molecular hydrophobic, small molecular hydrophilic and hydrophilic macromolecular drugs were loaded into the PECP hydrogel to evaluate their release profiles. The result showed that the releasing periods for macromolecular protein or hydrophobic drug were extended to more than one month. Since these two types of drugs are loaded within different regions of the hydrogel due to their different hydrophobic/hydrophilic nature, the PECP hydrogel is expected to develop injectable system loaded with a versatility of drugs or guest matter for synergetic effect.

Abstract:The poor solubility of many newly discovered drugs has resulted in numerous challenges for the time-controlled release of therapeutics. In this study, an advanced drug delivery platform to encapsulate and deliver hydrophobic drugs, consisting of poly (lactic-co-glycolic acid) (PLGA) nanoparticles incorporated within poly (ethylene glycol) (PEG) microgels, was developed. PLGA nanoparticles were used as the hydrophobic drug carrier, while the PEG matrix functioned to slow down the drug release. Encapsulation of the hydrophobic agents was characterized by fluorescence detection of the hydrophobic dye Nile Red within the microgels. In addition, the microcomposites prepared via the droplet-based microfluidic technology showed size tunability and a monodisperse size distribution, along with improved release kinetics of the loaded cargo compared with bare PLGA nanoparticles. This composite system has potential as a universal delivery platform for a variety of hydrophobic molecules.

Abstract:Injectable hydrogels with pH and temperature triggered drug release capability were synthesized based on biocompatible glycol chitosan and benzaldehyde-capped poly(ethylene glycol)-block-poly(propylene glycol)-block-poly(ethylene glycol) (PEO-PPO-PEO). Aqueous solutions of the above polymers formed hydrogel under physiological conditions, allowing a desirable injectability, through the formation covalent benzoic-imine bond with pH and temperature changes. Rheological characterization demonstrated that the gelation rate and the moduli of the hydrogels were able to be tuned with chemical composition as well as pH and temperature of the polymer solution. Both hydrophobic and hydrophilic drugs could be incorporated inside the hydrogel through the in situ gel forming process and undergo a controlled release by altering pH or temperature. In vivo tests proved the formation and biocompatibility of the hydrogel in rat model.

Abstract:With the rapid development of biomedicine applications, more flexible and efficient fabrication strategies are required for drug delivery vehicles. Naturally resourced polymers have gained considerable attention for construction of drug carriers. Here, biocompatible hydrogel reservoir system (named PHG-PDA) based on pullulan and polydopamine (PDA) are designed and fabricated by one-pot incorporation of PDA fibers into pullulan hydrogel (PHG). The structure and performance of the PHG-PDAs can be nicely adjusted by regulating the content of PDA fibers in precursor hydrogel solutions. Crystal violet is used as a drug prototype and introduced into hydrogels via swelling-diffusion approach. We found that the drug loading process followed the pseudo-second-order model and Langmuir isotherm model, while the drug releasing process was pH-responsive, and the cumulative release reached 60.3% and 87% at pH 7.4 and 5.0, respectively. In vitro cytotoxicity studies on L929 cells demonstrated that our hydrogels were nontoxic even in a high concentration (3.4 mg/mL). In sum, these biocompatible PHG-PDAs with tunable physicochemical properties are promising systems for drug delivery.

Abstract:A novel pH and oxidation dual-responsive and injectable supramolecular hydrogel was developed, which was formed from multi-block copolymer poly(ether urethane) (PEU) and α-cyclodextrin (α-CD) inclusion complexes (ICs). The PEU copolymer was synthesized through a simple one-pot condensation polymerization of poly(ethylene glycol), di(1-hydroxyethylene) diselenide, dimethylolpropionic acid and 3-isocyanatomethyl-3,5,5-trimethylcyclohexyl isocyanate. In aqueous solution, the amphiphilic PEU copolymers could self-assemble into nanoparticles with dual pH and oxidation sensitivities, which can efficiently load and controllably release a hydrophobic drug indomethacin (IND). Then a dual-drug loaded supramolecular hydrogel was obtained by addition of α-CD and hydrophilic model drug (rhodamine B, RB) into the resulting IND-loaded PEU nanoparticle solution. The rheology studies showed that the supramolecular hydrogels with good injectability underwent a pH-induced reversible sol-gel transition and an oxidation-triggered degradation behavior. The in vitro drug release results demonstrated that the hydrogels showed dual drug release behavior and the release rates could be significantly accelerated by addition of an oxidizing agent (H2O2) or increasing the environmental pH. Therefore, this injectable and dual stimuli-responsive supramolecular hydrogel based codelivery systems could potentially be a promising candidate for controlled drug delivery systems.

Abstract:A novel pulsed-release system based on bilayer coated tablets containing an osmotically active agent is presented. Hydroxypropylmethylcellulose (HPMC) and the mixture of Eudragit RS and RL were applied as the swelling layer and semipermeable outer coat, respectively. To examine the mechanism of drug release from this pulsed-release system, drug release behaviors were investigated under conditions of various osmotic pressures. Both lag time and release rate were dependent on the coating level and the osmotic pressure of the dissolution medium. The swelling of tablets and the dynamics of water uptake during the dissolution were investigated to further elucidate the mechanism of drug release. The osmotic active agent induces a continuous water influx resulting in a rapid expansion of the membrane. The subsequent formation of fractures leads to a fast drug release after an initial lag time. All the results obtained in the present study indicated that both diffusion and osmotic pumping effect were involved in drug release from the device, but the latter was more dominant.
