Near‐Infrared Light‐Responsive Nanogels with Diselenide‐Cross‐Linkers for On‐Demand Degradation and Triggered Drug Release

Abstract:On the above-mentioned bases, we expect to realize the NIRtriggered disassembly of diselenide-cross-linked nanogels by using ICG to generate ROS. Here, a novel diselenide cross-linker is synthesized through a one-step reaction, and the diselenide-cross-linked poly(methacrylic acid) (PMAA)-based nanogels are prepared through distillation-precipitation polymerization ( Figure 1 ). The polymerization technique used here is a simple one-pot technique to prepare monodisperse nanospheres free of any added surfactant or stabilizer. [ 33,34 ] ICG is introduced into nanogels to produce ROS under NIR irradiation, thus cleaving the diselenide bond and triggering the disassembly of nanogels. We further demonstrate that the control of illumination times allows for on-demand nanogel degradation and controlled drug release. We anticipate that the convenient method of obtaining NIR-response based on diselenide group presented here can provide a favorable platform to develop light-sensitive nanocarriers for remote controllable drug delivery. Prior to nanogel fabrication, a novel diselenide cross-linker, N,N′ -bis(methacryloyl) selenocystamine (BMASC), was synthesized in a facile way, where a commercial selenocystamine dihydrochloride was selected as the original reactant to introduce diselenide moiety, and the whole synthesis route was hence shortened to a one-step reaction (Details of the synthesis and characterization, Figure S1, Supporting Information). The diselenide cross-linker can be used in the cross-linking of hydrophilic polymer chains, thereby making nanohydrogel. Furthermore, it opens a pathway to prepare selenium-containing polymers through radical polymerization. Next, diselenidecross-linked nanogels were prepared by distillation-precipitation copolymerization of methacrylic acid (MAA) and BMASC dissolved in acetonitrile using 2,2-azobisisobutyronitrile (AIBN) as initiator, and denoted as P(MAASe-Se -MAA).
