Abstract:Zwitterionic polymers play an attractive role in the application of stealthy nanocarriers for their excellent antifouling property. Herein, a zwitterionic nanogel with temperature sensitivity and redox-responsive degradability prepared by copolymerization of N-vinylcaprolactam (VCL) and 2-(methacryloyloxy) ethyldimethyl-(3-sulfopropyl) ammonium hydroxide (DMAPS) via aqueous precipitation polymerization. The prepared nanogels own ultra-high colloidal stability and non-specific protein adsorption resistance as a result of the incorporation of zwitterionic groups. Meanwhile, they exhibit sensitive temperature-induced swelling/collapse transition in aqueous solution and excellent redox-degradability ascribed to the presence of disulfide bonds. The nanogels loaded with anticancer drug doxorubicin (DOX) exhibit low leakage of DOX under physiological conditions (merely 23.8 % within 24 h), whereas striking release amount of DOX under reducing conditions combined with elevated temperature (93.4 % within 24 h). The measurement of cell viability showed that the cytotoxicity of blank nanogels to tumor cells (HeLa cells) was negligible, while the nanogels loaded with DOX had a prominent inhibitory impact on tumor cells.

Abstract:A novel temperature-response hydrogel was developed for drug-delivery applications. The hydrogel matrix (PES) was synthesized by melt polycondensation of poly(ether–ester) diacid based on PEG with low molecular weight and sebacic acid. The sol–gel–sol phase transitions of PES nanoparticle (NP) hydrogel were investigated. In vitro erosion of hydrogel was characterized by Fourier transform infrared spectroscopy, environmental scanning electron microscopy and dynamic light scattering. In vitro release behaviors of hydrophilic and hydrophobic drugs and in vivo histopathological evaluation were studied in detail. The study results revealed that an aqueous dispersion of PES nanoparticle freeze-dried powder exhibited reversible sol–gel transition behavior with increasing temperature. The hydrogel could maintain steadily at least a month during in vitro erosion process. There were sustained release behaviors of hydrophilic and hydrophobic drugs from PES NP hydrogel and histopathological studies confirmed that the PES NP hydrogel only provoked an acceptable modest inflammatory response. Thus, PES NP hydrogel is biodegradable, biocompatible and promising in controlling the incorporated drugs for sustained release.

Abstract:The strategy to efficiently deliver antitumor drugs via nanocarriers to targeted tumor sites and achieve controllable drug release is attracting great research interest in cancer therapy. In this study, a novel type of disulfide‐bonded poly(vinylcaprolactam) (PVCL)‐based nanogels with tunable volume phase transition temperature and excellent redox‐labile property are prepared. The nanogels are hydrophilic and swell at 37 °C, whereas under hyperthermia (e.g., 41 °C), the nanogels undergo sharp hydrophilic/hydrophobic transition and volume collapse, which enhances the cellular uptake and drug release. The incorporation of disulfide bond linkers endows the nanogels with an excellent disassembly property in reducing environments, which greatly facilitates drug release in tumor cells. Nanogels loaded with doxorubicin (DOX) (DOX‐NGs) (DOX‐NGs) are stable in physiological conditions with low drug leakage (15% in 48 h), while burst release of DOX (92% in 12 h) can be achieved in the presence of 10 × 10−3 m glutathione and under hyperthermia. The DOX‐NGs possess improved cell killing efficiency under hyperthermia (IC50 decreased from 1.58 μg mL−1 under normothermia to 0.5 μg mL−1). Further, the DOX‐NGs show a pronounced tumor inhibition rate of 46.6% compared with free DOX, demonstrating that this new dual‐responsive nanogels have great potential as drug delivery carriers for cancer therapy in vivo.

Abstract:Stimuli-responsive, readily functionalizable, and degradable nanogels have great potential in targeted drug delivery applications. While crosslinked polymeric nanoparticles such as nanogels modified with cell-targeting moieties can effectively deliver various hydrophobic drugs to cancer cells, they should be engineered to undergo degradation upon exposure to endogenous stimuli and release the payload once inside the cell. Herein, we report the preparation of a nanogel system crosslinked through the thiol–maleimide Michael addition reaction, which can be degraded in a reducing environment through a thiol–disulfide exchange reaction. The maleimide groups on the nanogels provided handles for conjugating cell-targeting motifs. To this end, a new monomer was designed to synthesize poly(ethylene glycol) methacrylate-based copolymers containing pendant maleimide groups linked to the polymer backbone through disulfide linkages. These copolymers were utilized to yield nanogels by crosslinking the nanoaggregates formed upon heating the copolymers in aqueous media. Nanogels were loaded with a clinically administered anticancer drug, docetaxel, and the drug release was investigated in the reductive cellular environment. Nanogels conjugated with cell-targeting peptides demonstrated preferential cellular internalization in breast cancer cells and also exhibited enhanced cytotoxicity in cells rich in glutathione. One can envision that such nanogels, employed to deliver various drugs and target motifs depending on the type of cancer, will be an attractive platform for such applications.

Abstract:Degradable and thermosensitive microgels were successfully prepared via simultaneous quaternization and siloxane condensation during surfactant-free emulsion polymerization, with N-vinylcaprolactam as the main monomer and 1-vinylimidazole (VIM) as the comonomer, in the presence of (3-bromopropyl)trimethoxysilane (BPTMOS). The formation mechanism of cross-linking network was attributed to the hydrolysis and condensation of the methoxysilyl groups of BPTMOS and the quaternization of imidazole moiety of VIM by the bromine group of BPTMOS, leading to the microgels. The microgels were spherical in shape with a narrow size distribution, stable in an acidic buffer solution, but degradable in neutral and alkaline solutions. The presence of quaternized imidazolium in the same chain segment of Si-O-Si cross-linking points promoted the decomposition of Si-O-Si bonds and hence the degradation of the microgels. The obtained microgels could load and release the model drug, doxorubicin. The size, thermosensitivity, stability, degradation rate, and drug release behavior of the resultant microgels could be tuned by controlling the cross-linking degree, chemical composition, and degradation medium.
Acid Degradable Poly(vinylcaprolactam)-Based Nanogels with Ketal Linkages for Drug Delivery.

Abstract:To improve the biocompatibility and biodegradability of nanocarriers, well-defined poly(vinylcaprolactam)-based acid degradable nanogels were fabricated for drug delivery via precipitation polymerization in water, where synthetic ketal-based 2,2-dimethacroyloxy-1-ethoxypropane (DMAEP) acted as a cross-linker, and N-(2-hydroxypropyl)methacrylamide (HPMA) served as a co-monomer. Expectedly, we observed that the temperature and pH of the environment play important roles in the performance of the nanogels. The nanogels were reduced in size upon increasing the temperature and showed higher volume phase transition temperature (VPTT) with higher concentration of HPMA. With the incorporation of ketal linkages, the nanogels showed accelerated degradation profiles by lowering the pH and increasing temperature of the incubation medium. When used as nanocarriers of anticancer drug doxorubicin (DOX), compared to non-degradable nanogels with similar components, the acid-degradable nanogels displayed more effective drug controlled release behaviour, low drug leakage of DOX at neutral pH while rapid and sufficient release from the nanogels under acidic conditions. The results of the cytotoxicity and hemolysis assays further highlighted that the acid-degradable nanogel produced no hemolysin but showed excellent viability to normal cells, and the DOX-loaded nanogel exhibited higher proliferation inhibition against tumor cells.

Abstract:Degradable thermosensitive ionic microgels were synthesized via surfactant-free emulsion polymerization.(SEEP), N-isopropylacrylamide (NIPAm) and 1-vinylimidazole :(VIM) at 70 degrees C with degradable 1,4-phenylene bis(4-bromobutanoate) or 1,6-hexanediol bis(2-bromopropionate) as quaternized cross-linkers. VIM could be,quaternized by 1,4-phenylene bis(4-bromobutanoate) or 1,6-hexanediol bis(2-bromopropionate), leading to the formation of degradable cross-linking network and ionic microgels. Combined techniques of transmission electron microscopy (TEM), dynamic light scattering (DLS), electrophoretic light scattering (ELS), UV-vis spectroscopy, FT-IR spectra, and gel permeation chromatography (GPC) were employed to systematically investigate the sizes, morphologies, and properties of the obtained microgels before and after degradation as well as the degradation mechanism. The obtained microgels were spherical in shape with narrow size distribution and exhibited thermosensitive behavior and controllable degradation. The disintegration of the microgels was confirmed to be resulted from the hydrolysis of ester bonds of the cross-linkers. The degradation rate of the obtained microgels could be regulated by tuning the pH value of niicrogel suspensions. The PNI-Ph series of microgels fabricated with 1,4-phenylene bis(4-bromobutanoate) as the cross-linking agent could gradually degrade even in neutral solution with lifetime S' of 44-53 h, depending On the quaternizaiton ratio. The degradation of PNI-Ph series of microgels experienced two reaction processes, that is, the hydrolysis of ester bonds of the cross-linkers and the oxidation of generated hydroquinone to form benzoquinone. It was also demonstrated that different silica nanostructures could be fabricated by Using such degradable thermosensitive ionic microgels as the template at various temperatures.

Abstract:Of the multitude of stimuli-responsive microgels, it is still a challenge to achieve multiple responsivenesses to one single stimulus, which can even revert to the corresponding original state autonomously after stimulus. In this work, we reported a series of anthraquinone functionalized microgels (PNI-xVAQ) with thermosensitivity and redox-actuated self-regulating color, size, and fluorescent properties, which were easily synthesized via surfactant-free emulsion copolymerization (SFEP) with N-isopropylacrylamide (NIPAm) as the monomer, 2-vinylanthraquinone (VAQ) as the comonomer, and N,N'-methylenebis(acrylamide) (BIS) as the cross-linker in an aqueous solution at 70 °C. The hydrophobic interactions of comonomer VAQ also led to the formation of internal phase-separated hydrophobic nanodomains in the obtained PNI-xVAQ microgels. The self-regulating color, size, and fluorescence changes of the PNI-xVAQ microgels were reliant on the nonequilibrium redox process of anthraquinone moieties by the addition of sodium dithionite as the chemical fuel to activate the positive feedback that was the reduction of anthraquinone to transient anthraquinone radical anions, following the slow oxidation of anthraquinone radical anions by autonomous "breathing" oxygen in air as the delayed negative feedback. These autonomous self-regulating properties of the PNI-xVAQ microgel were recyclable to a certain extent by repeated feeding of sodium dithionite.

Abstract:A family of multi-responsive nanogels with different compositions and crosslinking degrees have been prepared by the miniemulsion copolymerization of monomethyl oligo(ethylene glycol) acrylate (OEGA) and an ortho ester-containing acrylic monomer, 2-(5,5-dimethyl-1,3-dioxan-2-yloxy) ethyl acrylate (DMDEA), with bis(2-acryloyloxyethyl) disulfide (BADS) as a crosslinker. These nanogels are thermoresponsive and labile in the weakly acidic or reductive environments. The thermoresponsive behaviors, acid-triggered hydrolysis, and reduction-induced degradation of these nanogels were studied by means of dynamic light scattering (DLS), transmission electron microscopy (TEM) and atomic force microscopy (AFM). The results indicate that the volume phase transition temperature (VPTT), thermally induced deswelling ratio, and acid-triggered swelling ratio of the nanogels are closely relevant to their compositions and crosslinking degrees. Although these nanogels could be reductively disrupted by dithiothreitol (DTT), single polymer chains with sizes smaller than 20nm were not detected by DLS. This is probably due to the existence of some unbreakable linkages formed by chain transfer to the disulfide bond during the radical polymerization. These nanogels are capable of encapsulating hydrophobic compounds. The loading capability of the nanogels for Nile Red (NR), paclitaxel (PTX), and doxorubicin (DOX), and the release behaviors of the drug-loaded nanogels were investigated by UV–vis spectrometry and HPLC. As expected, drug release can be greatly accelerated by a cooperative effect of both acid-triggered hydrolysis and DTT-induced degradation. Finally, the PTX-loaded nanogels exhibit a concentration-dependent toxicity to MCF-7 cells while the intact unloaded nanogels are non-toxic, thereby they may be used as potential carriers for hydrophobic anticancer drugs.

Abstract:In this study, we designed and synthesized hybrid microgels with a positively charged cross-linking network and embedded gold nanoparticles. The hybrid microgels presented reversible thermosensitive behavior and the volume transition temperature of similar to 37 degrees C, which was close to body temperature. These microgels were successfully applied as vectors to simultaneously deliver hydrophobic anticancer drug, doxorubicin, and anionic anti-inflammatory drug, diclofenac sodium. In the tumor microenvironment of low pH, high temperature, reduction condition, and the presence of ions, both drugs could controllably release from the microgels. Such microgels might be a promising platform for further development as an effective strategy for combination therapy.

Abstract:Stimuli responsive drug delivery systems have attracted considerable attention due to their ability to deliver drugs on demand to special tissues and cells (e.g. cancer cells). In this paper, we fabricated a new kind of redox/pH dual stimuli-responsive biodegradable poly(methacrylic acid) (MAA) nanohydrogels crosslinked by N,N-bis(acryloyl)cystamine (BACy) through a facile reflux-precipitation polymerization. Subsequently polyethylene glycol (PEG) was grafted onto the surface of nanohydrogels to improve their long circulation time. This kind of PEGylated nanohydrogels could be degraded into short linear polymer chains in the solution of 10 mM glutathione (GSH) due to the sensitive disulfide bonds in the cross-linked networks. Interestingly, we found that the hydrophilic PEGylated nanohydrogels could be simultaneously loaded with hydrophilic drug doxorubicin (DOX) and hydrophobic drug paclitaxel (PTX) with high drug loading efficiency because of its regional hydrophobic nature in the nanohydrogels. The cumulative release profile showed that either lower pH (5.0) or reducing environment (GSH) or a combination could significantly accelerate the drug release from the drug-loaded nanohydrogels. In addition, the experimental results on living cells indicated that the multi-drug-loaded system had synergistic effect on cancer therapy. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:Thermoresponsive phospholipid-poly(N-isopropylacrylamide) (PL-PNIPAM) conjugates were synthesized via reversible addition fragmentation chain transfer polymerization mediated by a phospholipid-modified trithiocarbonate. Temperature triggered the micellization of the PL-PNIPAM conjugate to form phosphate group-decorated micelles in the aqueous solution. Driven by the chelation of phospholipids and Ca2+, the PL-PNIPAM conjugate and Ca2+ ions formed size-tunable nanoclusters at a temperature beyond the lower critical solution temperature. To fabricate cross-linked nanogels, NIPAM was copolymerized with N-succinimidyl acrylate (NSA) to obtain the PL-P(NIPAM-co-NSA) conjugate bearing pendent cross-linkable functionalities. Subsequently, the size-controllable nanogels containing disulfide linkages were generated at 37 °C by cross-linking the PL-P(NIPAM-co-NSA)/Ca2+ nanoclusters with cystamine through modulation of Ca2+ concentrations. These negatively charged nanogels demonstrate temperature/pH/reduction triple responsiveness. The nanogels can be efficiently loaded with doxorubicin (DOX) and proteins with various isoelectric points. The DOX-loaded nanogels exhibited a temperature/pH/reduction triple-responsive release profile. The immobilized RNase A, BSA, and GOx retained the protein bioactivity. The release of RNase A-loaded nanogels possesses a temperature-responsive profile. The immobilization of Lys and cytochrome C in nanogels inhibited protein bioactivity. However, the addition of NaCl triggered the recovery of bioactivity. These multistimuli-responsive nanogels can provide a versatile platform applicable in biotechnology and drug/protein delivery.

Abstract:The convenient and precise fabrication of drug-hydrogel formulations with satisfactory degradability and a well-controlled drug release profile are crucial factors for injectable hydrogel formulations in clinical applications. Here a new injectable thermosensitive hydrogel formed from poly(ε-caprolactone) (PCL)-poly(ethylene glycol)-poly(ε-caprolactone) amphiphilicco-polymers with 1,4,8-trioxa[4.6]spiro-9-undecanone (TOSUO) moieties incorporated in the poly(ε-caprolactone) (PCL)block (PECT) was constructed to provide a route to tailor the degradation and drug release behavior. The effect of hydrophilic cyclic ether moieties on the degradation of and drug release by PECT hydrogels were evaluated in vitro and in vivo. The results indicated that a freeze-dried powder of paclitaxel-loaded PECT nanoparticles rapidly dissolved in water at ambient temperature with slightly shaking and formed a stable injectable in situ drug-hydrogel formulation at body temperature, which is convenient for clinical operations because it avoids the need for pre-quenching or long-term incubation. The paclitaxel distribution was also more quantitative and homogeneous on entrapping paclitaxel in PECT nanoparticles. Further, the small number of pendant cyclic ether groups in PCL could decrease the cystallinity and hydrophobicity and, as a result, the in vitro and in vivo retention time of PECT hydrogels and the release of entrapped paclitaxel could be tuned from a few weeks to months by varying the amount of PTOSUO in the hydrophobic block. Significantly, paclitaxel-loaded PECT nanoparticles and free paclitaxel could be simultaneously released during the in vitro paclitaxel release from PECT hydrogels. A histopathological evaluation indicated that in vivo injected PECT hydrogels produced only a modest inflammatory response. Thus pendant cyclic ether modification of PCL could be an effective way to achieve the desired degradation and drug release profiles of amphiphilicco-polymer thermosensitive hydrogels and PECT hydrogels may be suitable for local drug delivery.

Abstract:During the past decade, a series of thermo-responsive hydrogels based on poly(N-isopropylacrylamide) (PNIPAM) were prepared for drug delivery. However, the release performance of those hydrogels was limited because the LCST (lower critical solution temperature) of PNIPAM (about 32 °C) is slightly lower than human body temperature, and the drug loading is complicated. To develop a new drug delivery matrix with the suitable responsive interval, a novel, temperature-sensitive copolymer with two monomers, N-isopropylacrylamide and methylacrylic polyethylene glycol monomethyl ether ester (MPEGMA) [poly(NIPAM-co-MPEGMA)], was synthesized by free radical polymerization. Then, the supramolecular hydrogels with tunable response temperature, whose structures were confirmed by X-ray diffraction (XRD), were fabricated via inclusion complexation with α-cyclodextrin (α-CD) and the copolymers in aqueous solution. The rheology results indicated that response interval could be modulated by controlling the contents of PNIPAM in the copolymers. Compared to the normal PNIPAM ge1, the succinct method of gel preparation streamlined the process of loading and made the drug loading capacity controllable. The release time was proved to be prolonged by the release kinetics of 5-fluoroucrail (5-Fu), which showed feasibility as a drug delivery.

Abstract:To explore the potential biomedical application of thermoresponsive nanosystems, it is important to enhance their thermosensitivity to improve the controllability in delivery of therapeutic agents. The present work develops multifunctional nanogels with enhanced thermosensitivity through copolymerization of N-isopropylacrylamide (NIPAM) and acrylic acid (AA) in the presence of β-cyclodextrin (β-CD), using N,N'-bis(acryloyl)cystamine (BAC) as a biodegradable crosslinker. The resulting nanogels display significantly improved sensitivity in deswelling (swelling) behavior upon temperature increase (decrease) around body temperature. The nanogels can effectively encapsulate doxorubicin (DOX), which can be released in an accelerated way under microenvironments that mimic intracellular reductive conditions and acidic tumor tissues. Release can also be remotely manipulated by increasing temperature. In vitro study indicates that the nanogels are quickly taken up by KB cells (a human epithelial carcinoma cell line), exerting improved anticancer cytotoxicity, showing their potential for delivery of therapeutic agents beyond anticancer drugs.

Microgels with Linear Thermosensitivity in A Wide Temperature Range

Abstract:A newly developed poly(2-methoxyethyl acrylate-co-oligo(ethylene glycol) methyl ether acrylate) (P(MEA-co-OEGA) microgel, synthesized by free radical precipitation copolymerization, displays a linear volume phase transition in water upon heating in a wide range of temperature. Varying cross-linking density in the microgels does not interrupt the linear thermosensitivity. By changing the feeding molar ratio of MEA and OEGA, the linear thermosensitive temperature range can be adjusted, where higher OEGA content results in higher plateau starting temperature. Furthermore, temperature- and pH-sensitive microgels could be achieved by adding another comonomer, acrylic acid (AA). The resulting P(MEA-co-OEGA-co-AA) microgels are ideal candidates to prepare hybrid microgels by in situ formation of silver nanoparticles (AgNPs).
