Abstract:Higher mode and bimodal atomic force microscopy (AFM) are two recently developed imaging modes of dynamic AFM for improving resolution. In higher mode, the higher flexural mode of the cantilever instead of the traditional fundamental eigenmode is excited. In bimodal mode, two flexural modes of the cantilever are simultaneously excited for obtaining more information about the properties of the material. The first three flexural modes for higher mode and superposition of two excitation signals for bimodal mode are explored and compared by imaging a polymer blend of polystyrene (PS) and low density polyethylene (LDPE). The effects of different operating conditions of the two imaging modes are researched to improve image contrast and material discrimination. Dissipated power and virial are employed to explain the origin of contrast for the complex and highly nonlinear dynamical tip-sample interfacial system. Amplitude and phase contrasts of each single mode and bimodal mode are calculated by Ashman's D statistical equation. It is found that higher single modes with small free amplitudes show enhanced phase contrast. The bimodal of the first and the third modes gains a clear advantage over the bimodal of the first and second modes for phase and amplitude image contrasts. In addition, the best contrast of bimodal imaging occurs when it is a combination of a large free amplitude for the first mode and a small free amplitude for the third mode.
Enhancement of Fluorescence Emission Difference Microscopy Using Conjugated Vortex Phase Modulation

Abstract:In this paper, we propose and demonstrate an improved fluorescence emission difference microscopy (FED) method, exploiting programmable phase modulation for image enhancement. The main novelty of the proposed approach lies in the matched size and intensity of two excitation spots. The proposed method improves the FED performance on image quality via artefact elimination. We demonstrate the feasibility of this method through theoretical studies and experimental tests. The experimental results of nanobeads and cells validate the practical performance of this method, which can enable reliable observations at superresolution in biomedical studies. LAY DESCRIPTION: In this paper, we propose a method to improve the imaging quality of regular fluorescence emission difference (FED) microscopy. In regular FED imaging, a solid and a doughnut excitation beam are successively used to acquire two images which are then subtracted with each other to improve the resolution of confocal microscopy. The doughnut beam can be generated by modulating the excitation beam with a vortex phase mask. Note that both of the excitation beam and the vortex phase mask must have the same handed direction in regular FED microscopy. However, some negative values may be produced and some information may be lost due to the subtraction process in regular FED imaging, which is mainly caused by the mismatched size and intensity of these two excitation spots. To address this issue, we propose conjugated FED (cFED) microscopy which additionally uses a conjugated vortex phase mask to modulate the solid beam to extend its focal spot size to be matched with the doughnut spot, which means the handed direction of the solid beam and the vortex phase mask is different. Besides, in order not to damage the resolution, the doughnut beam needs to be saturated to some degree. The experiment results show that, at the same resolution level, the negative values and the information loss in cFED image are all less than that of regular FED image.
Quantitative Phase and Amplitude Imaging with an Efficient Support Constraint.

Abstract:High-speed quantitative phase and amplitude imaging methods have led to numerous biological discoveries. For general samples, phase retrieval from a single-diffraction pattern has been an algorithmic and experimental challenge. Here we present a quantitative phase and amplitude imaging method applying an efficient support constraint to yield a rapid algorithmic convergence due to the removal of the twin image and spatial shift ambiguities. Compared to previous complex-valued imaging, our method is lenslet-free and relies neither on assumption based on sample sparsity nor interferometric measurements. Our method provides a robust method for imaging in materials and biological science, while its rapid imaging capability will benefit the investigation of dynamical processes.
The Phase Behaviour of Single Polyethylene Chains with and Without Fixing One End

Abstract:The phase behaviour of a single polyethylene chain is studied by using molecular dynamics simulations. A free chain and a chain with fixing one end are considered here, since the atomic force microscope (AFM) tip can play a significant role in polymer crystallization in experiment. For a free chain, it is confirmed in our calculation that the polymer chain exhibits an extended coil state at high temperatures, collapses into a condensed state at low temperatures, i.e. the coil-to-globule transition that is determined by a high temperature shoulder of the heat capacity curve, and an additional liquid-to-solid transition that is described by a low temperature peak of the same heat curve. These results accord with previous studies of square-well chains and Lennard-Jones homopolymers. However, when one of the end monomers of the same chain is fixed the results become very different, and the chain cannot reach an extended coil-like state as a free chain does at high temperatures, i.e. there exists no coil-to-globule-like transition. These results may provide some insights into the influence of AFM tip when it is used to study the phase behaviour of polymer chains. If the interaction force between AFM tip and polymer monomers is strong, some monomers or one of them can be seen as being fixed by the tip, which is similar to our simulation model, and it is also found that AFM tip could induce polymer crystallization.

Abstract:Multi-parameter fitting algorithms, which are currently used for the characterization of coated-bubbles, inevitably introduce uncertainty into the results. Therefore, a better technique that can accurately determine the microbubbles' mechanical properties is urgently needed. A comprehensive technology combining atomic force microscopy, optical, and acoustic measurements with simulations of coated bubble dynamics was developed. Using this technique, the mechanical parameters (size distribution, shell thickness, elasticity, and viscosity) of hybrid (ultrasound/magnetic-resonance-imaging) contrast microbubbles and their structure-property relationship were determined. The measurements indicate that when more superparamagnetic iron oxide nanoparticles are embedded in the microbubbles' shells, their mean diameter and effective viscosity increase, and their elastic modulus decreases. This reduces the microbubbles' resonance frequency and thus enhances acoustic scattering and attenuation effects. (C) 2015 Elsevier Ltd. All rights reserved.
Imaging contrast and tip-sample interaction of non-contact amplitude modulation atomic force microscopy with Q-control

Abstract:Active quality factor (Q) exhibits many promising properties in dynamic atomic force microscopy. Energy dissipation and image contrasts are investigated in the non-contact amplitude modulation atomic force microscopy (AM-AFM) with an active Q-control circuit in the ambient air environment. Dissipated power and virial were calculated to compare the highly nonlinear interaction of tip-sample and image contrasts with different Q gain values. Greater free amplitudes and lower effective Q values show better contrasts for the same setpoint ratio. Active quality factor also can be employed to change tip-sample interaction force in non-contact regime. It is meaningful that non-destructive and better contrast images can be realized in non-contact AM-AFM by applying an active Q-control to the dynamic system.
Different Directional Energy Dissipation of Heterogeneous Polymers in Bimodal Atomic Force Microscopy.

Abstract:Dynamic force microscopy (DFM) has become a multifunctional and powerful technique for the study of the micro-nanoscale imaging and force detection, especially in the compositional and nanomechanical properties of polymers. The energy dissipation between the tip and sample is a hot topic in current materials science research. The out-of-plane interaction can be measured by the most commonly used tapping mode DFM, which exploits the flexural eigenmodes of the cantilever and a sharp tip vibrating perpendicular to the sample surface. However, the in-plane interaction cannot be detected by the tapping mode. Here a bimodal approach, where the first order flexural and torsional eigenmodes of the cantilever are simultaneously excited, was developed to detect the out-of-plane and in-plane dissipation between the tip and the polymer blend of polystyrene (PS) and low-density polyethylene (LDPE). The vibration amplitudes and phases have been recorded to obtain the contrast, energy dissipation and virial versus the setpoint ratio of the first order vibration amplitude. The pull-in phenomenon caused by a strong attractive force can occur near the transitional setpoint ratio value, the amplitude setpoint at which the mean force changes from overall attractive to overall repulsive. The in-plane dissipation is much lower than out-of-plane dissipation, but the torsional amplitude image contrast is higher when the tip vibrates near the sample surface. The average tip-sample distance can be controlled by the setpoint ratio to study the in-plane dissipation. Both flexural and torsional phase contrasts and torsional amplitude contrast can also be significantly enhanced in the intermediate setpoint ratio range, in which compliant heterogeneous materials can be distinguished. The experiment results are of great importance to optimize the operating parameters of image contrast and reveal the mechanism of friction dissipation from the perspective of in- and out-of-plane energy dissipation at different height levels, which adds valuable ideas for the future applications, such as compliant materials detection, energy dissipation and the lateral micro-friction measurement and so on.
Nanomechanics of Polyphenylacetylene Derivatives by AFM-based SMFS

Abstract:We quantitatively investigated the effect of iodine and pH on the stability of the helical structure formed by polyphenylacetylene derivatives using atomic force microscopy (AFM) based single-molecule force spectroscopy (SMFS) and circular dichroism (CD). The AFM measurement results showed that the addition of iodine into the system caused the decrease of the plateau heights in the stretching curves recorded during the SMFS experiments. In addition, the pH value increase can weaken the intra. polymer hydrogen bond and interrupt the integrity of the helical structure. However, the CD result did not show any detectable changes upon the addition of iodine and the change of pH value, which indicate that SMFS is a very sensitive and powerful method for subtle structure study.

Abstract:In bimodal FM-AFM, two flexural modes are excited simultaneously. The total vertical oscillation deflection range of the tip is the sum of the peak-to-peak amplitudes of both flexural modes (sum amplitude). We show atomically resolved images of KBr(100) in ambient conditions in bimodal AFM that display a strong correlation between image quality and sum amplitude. When the sum amplitude becomes larger than about 200 pm, the signal-to-noise ratio (SNR) is drastically decreased. We propose this is caused by the temporary presence of one or more water layers in the tip-sample gap. These water layers screen the short range interaction and must be displaced with each oscillation cycle. Further decreasing the sum amplitude, however, causes a decrease in SNR. Therefore, the highest SNR in ambient conditions is achieved when the sum amplitude is slightly less than the thickness of the primary hydration layer.
Amplitude Contrast Imaging in High-Resolution Transmission Electron Microscopy of Ferroelectric Superlattice Film

Abstract:To date, high-resolution electron microscopy has largely relied on using the phase of the exit wave function at the exit surface to form a high-resolution electron microscopic image. We have for the first time used chromatic aberration correction to implement a new imaging mode to achieve amplitude contrast imaging in high-resolution electron microscopy, allowing us to obtain directly interpretable high-resolution electron microscopic images with discrimination between light and heavy atomic columns. Using this imaging approach, we have successfully visualized the atomic structure in a BaTiO3/CaTiO3 superlattice with high spatial accuracy and discrimination between Ba and Ca columns, providing direct visualization of the Ca and Ba associated oxygen octahedral tilt that controls ferroelectric behavior in these superlattice structures. Furthermore, this approach offers new opportunities to unravel the structure in a wide range of materials, especially complex oxides with exotic behaviors based on specific structural arrangements.

Simulations of Subnanometer Scale Image Contrast in Atomic Force Microscopy of Self-Assembled Monolayers in Water

Abstract:Achieving high-resolution images using dynamic atomic force microscopy (AFM) requires understanding how chemical and structural features of the surface affect image contrast. This understanding is particularly challenging when imaging samples in water. An initial step is to determine how well-characterized surface features interact with the AFM tip in wet environments. Here, we use molecular dynamics simulations of a model AFM tip apex oscillating in water above self-assembled monolayers (SAMs) with different chain lengths and functional groups. The amplitude response of the tip is characterized across a range of vertical distances and amplitude set points. Then relative image contrast is quantified as the difference of the amplitude response of the tip when it is positioned directly above a SAM functional group vs positioned between two functional groups. Differences in contrast between SAMs with different lengths and functional groups are explained in terms of the vertical deflection of the SAMs due to interactions with the tip and water during dynamic imaging. The knowledge gained from simulations of these simple model systems may ultimately be used to guide selection of imaging parameters for more complex surfaces.
Direct Imaging of Interfacial Fluctuations in Confined Block Copolymer with in Situ Slow-Scan-Disabled Atomic Force Microscopy

Abstract:Using environmentally controlled, high-speed atomic force microscopy (AFM), we examine dynamic fluctuations of topographically confined poly(styrene-block-methyl methacrylate) (PS-b-PMMA) cylinders. During thermal annealing, fluctuations drive perturbations of the block copolymer (BCP) interface between polymer domains, leading to pattern roughness. Whereas previous investigations have examined roughness in room-temperature and kinetically quenched samples, we directly visualize the dynamics of PS/PMMA interfaces in real space and time at in situ temperatures above the glass transition temperature, Tg. Imaging under these experimentally challenging thermal annealing conditions is critical to understanding the inherent connection between thermal fluctuations and BCP pattern assembly. Through the use of slow-scan-disabled AFM, we dramatically improve the imaging time resolution for tracking polymer dynamics. Fluctuations increase in intensity with temperature and, at high temperatures, become spatially coherent across their confining potential. Additionally, we observe that topographic confinement suppresses fluctuations and correlations in the proximity of the guiding field. In situ imaging at annealing temperatures represents a significant step in capturing the dynamics of chain mobility at BCP interfaces.

Abstract:Single molecule force spectroscopy (SMFS) is a powerful means in studying the intramolecular and intermolecular interactions in polymer and supramolecular systems, thus opening new horizons for nanomechanic chemistry. This feature article concerns mainly how to employ AFM-based SMFS to obtain some of the important information which is not available by conventional characterizing methods, including single chain mechanics, the interfacial conformation and adhesive energy of polymers, interaction between macromolecules and small molecules, and the direct measurement of intermolecular forces. Quantitative annular dark-field imaging of single-layer graphene—II: atomic-resolution image contrast

Abstract:We have investigated how accurately atomic-resolution annular dark-field (ADF) images match between experiments and simulations to conduct more reliable crystal structure analyses. Quantitative ADF imaging, in which the ADF intensity at each pixel represents the fraction of the incident probe current, allows us to perform direct comparisons with simulations without the use of fitting parameters. Although the conventional comparison suffers from experimental uncertainties such as an amorphous surface layer and specimen thickness, in this study we eliminated such uncertainties by using a single-layer graphene as a specimen. Furthermore, to reduce image distortion and shot noises in experimental images, multiple acquisitions with drift correction were performed, and the atomic ADF contrast was quantitatively acquired. To reproduce the experimental ADF contrast, we used three distribution functions as the effective source distribution in simulations. The optimum distribution function and its full-width at half-maximum were evaluated by measuring the residuals between the experimental and simulated images. It was found that the experimental images could be explained well by a linear combination of a Gaussian function and a Lorentzian function with a longer tail than the Gaussian function.

Abstract:<p>Recent developments in phase contrast microscopy have enabled the label‐free visualization of certain organelles due to their distinct morphological features, making this method an attractive alternative in the study of cellular dynamics. However tubular structures such as ER networks and complex dynamics such as the fusion and fission of mitochondria, due to their low phase contrast, still need fluorescent labeling to be adequately imaged. In this paper, we report a quantitative phase microscope with ultra‐oblique illumination that enables us to see those structures and their dynamics with high contrast for the first time without labeling. The imaging capability was validated through comparison to the fluorescence images with the same field‐of‐view. The high image resolution ( ~270nm) was validated using both beads and cellular structures. Further, we were able to record the vibration of ER networks at a frame rate of 250Hz. We additionally show complex cellular processes such as remodelling of the mitochondria networks through fusion and fission and vesicle transportation along the ER without labels. Our high spatial and temporal resolution allowed us to observe mitochondria "spinning", which has not been reported before, further demonstrating the advantages of the proposed method. Photo-induced Phase-Matched Second-Harmonic Generation in Azodye-Doped Polymer Films by Nonresonant Dual-Frequency Coherent Excitation

Abstract:All-optical control of the noncentrosymmetry of azodye-doped polymer films was demonstrated by nonresonant dual-frequency coherent excitation. Three kinds of noncentrosymmetries of the polymer films were optically tailored using appropriate combinations of the writing beam polarizations. The characteristic kinetics of the photoinduced molecular polar orientation and the tensor analysis were performed by monitoring SHG of the films. The SHG conversion efficiency of a 105)mum thick film was estimated to be about 2%. It was observed that when polarizations of the writing beams were changed from the linear polarization to the circular polarization the saturated SHG signals were enhanced even at the same readout condition. A relaxation retardation effect of the photoinduced (chi) (2) was also observed in the thick films. The quadratic dependence of SHG on film thickness showed that a (chi) (2)grating satisfying the phase-matching condition for SHG was optically induced in the polymer films, indicating this nonresonant all-optical poling can be used to tailor the symmetry of bulk samples.
Retrospecting the relationship between interlamellar amorphous structure and intrinsic modulus of semicrystalline polymers via in-situ positron annihilation lifetime spectroscopy and in-situ X-ray scattering

Abstract:The structure-property relationship of the interlamellar amorphous phase in semicrystalline polymers has been a fundamental yet controversial topic in polymer physics for several decades. The intricate hierarchical structure and limited availability of experimental techniques pose significant challenges in quantitatively characterizing the evolution of the amorphous structure and establishing its connection to the mechanical behavior. In this work, the unannealed and annealed isotactic polypropylene hard-elastic films composed of idealized series-arranged lamellar stacks are studied to simplify the mechanical coupling between the two phases. By utilizing the synchrotron-based in-situ small and wide-angle X-ray scattering (SAXS/WAXS) techniques, the apparent modulus and overall volumetric strain of the amorphous phase are estimated. Moreover, with our significant progress in the counting rate of positron annihilation lifetime spectroscopy (PALS), the minute-scale structure evolution of amorphous phase (free-volume pores) during the tensile process is now feasible to be captured. Combing in-situ X-ray scattering and in-situ PALS results, in the elastic regime, it is found that triaxial stress in the amorphous phase leads to the expansion of free-volume pores (about 23% volume increase for the annealed sample, 35% volumetric strain for the unannealed one) and density decrement within occupied volume by overcoming Van Der Waals force. Our quantitative analysis also reveals that the annealing process not only enhances the crystallinity and aspect ratio of lamellae but also weakens the intrinsic modulus and chain packing density of amorphous phase.

Abstract:The characterization of inverted structures (crystallographic, ferroelectric, or magnetic domains) is crucial in the development and application of novel multi-state devices. However, determining these inverted structures needs a sensitive probe capable of revealing their phase correlation. Here a contrast-enhanced phase-resolved second harmonic generation (SHG) microscopy is presented, which utilizes a phase-tunable Soleil–Babinet compensator and the interference between the SHG fields from the inverted structures and a homogeneous reference. By this means, such inverted structures are correlated through the π -phase difference of SHG, and the phase difference is ultimately converted into the intensity contrast. As a demonstration, we have applied this microscopy in two scenarios to determine the inverted crystallographic domains in two-dimensional van der Waals material MoS 2 . Our method is particularly suitable for applying in vacuum and cryogenic environments while providing optical diffraction-limited resolution and arbitrarily adjustable contrast. Without loss of generality, this contrast-enhanced phase-resolved SHG microscopy can also be used to resolve other non-centrosymmetric inverted structures, e.g. ferroelectric, magnetic, or multiferroic phases.
