Determination of the Content of Colchicine in Compound Colchicine Capsules by HPLC

Abstract:To establish a method for determination of the content of colchicine in compound colchicine capsules,a HPLC method was utilized.The sample was separated on a Kromasil 100～5 C 18 column (5 μm,4.6 mm×150 mm)and detected at 350 nm,using mixture of 0.05 mol/L potassium dihydrogen phosphate buffer-methanol (50∶50) as the mobile phase,the flow rate was 1.0 mL/min,the column temperature was kept at 20℃.The contents were calculated with external standard.The results showed that the calibration curve was linear within the range from 3.05 to 9.15 μg/mL,y=47.143 0 x-1.382 7,r=0.999 4(n=5),the average recovery was 100.19%,RSD=0.86%(n=9).It concludes this method is simple,accurate and reliable,and can be used to control the quality of compound colchicine capsules.
