Amperometric Glucose Biosensor with Toluidine Blue O Mediator

Abstract:An amperometric glucose biosensor has been fabricated by immobilizing glucose oxidase(GOD) along with the mediator toluidine blue O (TBO) to a functional sol-gel thin layer on the surface of a platinum electrode via the cross-linking agent glutaraldehyde. The functional sol-gel thin layer was prepared through the hydrolysis and polycondensation of γ-aminopropylmethyldimethoxysilane and tetramethoxysilane in a dilute hydrochloric acid medium. The IR spectra showed that GOD and TBO were covalently linked to the carrier sol-gel material through the interaction between the aldehyde groups in glutaraldehyde and the amino groups in GOD, TBO and their carrier. Covalent immobilization prevents possible shedding of the sensing reagent and mediator from the carrier. Cyclic voltammetry was employed to demonstrate that TBO is capable to transfer electrons effectively between the active sites of the immobilized GOD and the surface of a platinum electrode. The calibration curve of glucose is linear in the range of 1~8 mmol/L, with a detection limit of 0.05 mmol/L. The lifetime of the biosensor exceeds one month.
