Abstract:OBJECTIVE:To investigate the cytotoxicity of calcium hydroxide (CH) and composite resin on fibroblasts derived from human embryo body fibroblasts-H9 (EBf-H9), human dental pulp cells (hDPCs) and immortalized fibroblasts L929; and to evaluate the use of EBf-H9 as a cellular model for cytotoxicity screening of dental materials. METHODS:The EBf-H9 cells were derived from human embryonic stem cells (H9) via outgrowth of embryonic body (EB); hDPCs were isolated from healthy dental pulp, and identified by immunochemical staining. Cell Counting Kit-8 (CCK-8) assay was applied to analyze the cytotoxicity of CH and composite resin with serial concentrations on the 3 kinds of cells. RESULTS:Following 24 h and 48 h (or 72 h) post-treatment of CH and composite resin, the viability of L929 cells was significantly lower than that of EBf-H9 and hDPCs (P<0.05), and there was no significant difference between the last two groups (P>0.05). CONCLUSION:Immortalized fibroblasts L929 cells exhibited different response to CH and composite resin compared with EBf-H9 and hDPCs, and the last two cell types were similar to each other. This study indicated that fibroblasts derived from human embryonic stem cells were a potential cellular model instead of traditional immortalized murine cell line for cytotoxicity screening assay.

Abstract:Objectives: To compare the permeability of human dentin and a porous ceramic with two different compositions of perfusion fluid, in vitro. Methods: Dentin discs were cut from extracted third molar at a level close to the pulp chamber and ceramic discs were cut from a porous ceramic block. The convective permeability of the discs was measured in a fluid transport model using de-ionised water or 1:3 diluted bovine serum and the hydraulic conductance was determined. Results: In the negative controls, no flow was detected. The hydraulic conductance values for the dentin discs and the ceramic discs when water was used as the perfusion fluid were 0.069±0.023and 0.047±0.019ml min-1 cm-2 cm H2O-1, respectively. When the discs were perfused with the diluted bovine serum the mean (standard error of the mean) hydraulic conductance values for the dentin and the ceramic discs were 0.036±0.016 and 0.012±0.0055 ml min-1 cm-2 cm H2O-1, respectively. Conclusion: Basing on the modern technique it is possible to fabricate discs of hydroxylapatite ceramic to act as substitutes for dentin barriers in cytotoxicity test.
The characteristic and osteogenic effect of a nanoporous coating of zirconia implant

Abstract:Zirconia ceramics have become increasingly used as dental implant material which is mainly due to their "tooth-like" color, superior biocompatibility, and excellent mechanical properties. However, the bio-inertness of the zirconia surface limits its use for a wider range of clinical applications. To enhance the surface bioactivity, a combination of magnetron sputtering, and anodization has in the present study been used to deposit a homogeneous nano-porous coating (ZrNP) on the zirconia surface (forming ZrO2/ZrNP with a ZrNP pore diameter of 20-30 nm, and a ZrNP layer thickness of similar to 2 mu m). EDS and XRD analysis showed that the distribution of the surface chemical components, as well as the crystalline structure of the zirconia (ZrO2), remained unchanged. On the other hand, the hydrophilicity of the ZrO2/ZrNP surface became significantly improved (as compared with a control group). In addition, the bond strength of the coating was decreased as a result of the anodization treatment. Moreover, both the Zr and ZrNP nanolayers exhibited improved proliferation and osteogenic properties in the in vitro cell experiment. Also, the ZrNP nanolayer was found to promote the biocompatibility and osteogenic ability to the largest extent. The ZrNP nanolayer did also provide numerous adhesion sites for MC3T3-E1 whereby the cell morphology could be enhanced. Additionally, the nano porous ZrNP layer improved the new bone formation around the zirconia implant and did robustly enhance the push out bond strength, thereby indicating the best osteointegration ability. In conclusion, a nano porous zirconium layer has been found to be very promising for the improvement of the bioactivity of the zirconia implant material.
[Dentin Barrier Cytotoxicity Test with Three-Dimensional Cell Cultures].

Abstract:OBJECTIVE:To evaluate the cytotoxicity of four dentin filling materials and two dentin adhesives with a dentin barrier test and to compare the results with those in a conventional filter diffusion test in order to investigate the advantages of the dentin barrier test.METHODS:Eugenol cement, zinc phosphate cement, adhesive glass ionomer cement, composite resin and two self-etching adhesives (REMI BOND and Adper Easy One) were tested. In the dentin barrier test, L929 mouse fibroblasts were three-dimensional cultured in polystyrene meshes. The dentin disks were cut from the human third molars, near the pulp and in parallel with the occlusal surface, and their permeability within the measurement area was evaluated by a hydraulic permeability device. A mesh with the cells was placed in the "pulp cavity" of the chamber and one dentin disk was put on the cell mesh and its "pulp side" was in contact with the mesh. The test materials and controls were in contact with the "occlusal side" of the dentine disks for 24 h. The cell viability was obtained with MTT assay and the results were expressed as a percentage of control tissues. The Mann-Whitney U test was used to make the statistical analyses. In the filter diffusion test, after a 24 h contact between the test materials and the filters with monolayer cells, the filters were dyed and the grades of cytotoxicity were decided.RESULTS:A mean permeability of the dentin disks near the pulp was 0.293 μL/(min×cm(2)×cmH(2)O)(1 cmH(2)O=0.098 kPa); In the dentin barrier test, Eugenol cement, REMI BOND and Adper Easy One respectively reduced the cell survival rates to 82%, 63% and 54%. Other materials showed no or very low toxic reactions; In the filter diffusion test, the light-curing composite resin was moderately cytotoxic, the dental adhesive glass ionomer cement was mildly cytotoxic and the others were severely cytotoxic; All the six materials in the dentin barrier test had lower cytotoxicity than in the filter diffusion test.CONCLUSION:The cytotoxicity of the test materials using the dentin barrier test with three-dimensional cell cultures is lower than that in the filter diffusion test, which has good correlation with the clinical situation.

Abstract:Introduction: This study examined the effect of a new bioactive dentin substitute material (Biodentine) on the viability of human gingival fibroblasts. Methods: Biodentine, White Pro Root mineral trioxide aggregate (MTA), and glass ionomer cement were evaluated. Human gingival fibroblasts were incubated for 1, 3, and 7 days both in the extracts from immersion of set materials in culture medium and directly on the surface of the set materials immersed in culture medium. Fibroblasts cultured in Dulbecco modified Eagle medium were used as a control group. Cytotoxicity was evaluated by flow cytometry, and the adhesion of human gingival fibroblasts to the surface of the set materials was assessed by using scanning electron microscopy. The data of cell cytotoxicity were analyzed statistically by using a one-way analysis of variance test at a significance level of P < .05. Results: Cells exposed to extracts from Biodentine and MTA showed the highest viabilities at all extract concentrations, whereas cells exposed to glass ionomer cement extracts displayed the lowest viabilities (P < .05). There was no significant difference in cell viabilities between Biodentine and MTA during the entire experimental period (P > .05). Human gingival fibroblasts in contact with Biodentine and MTA attached to and spread over the material surface after an overnight culture and increased in numbers after 3 and 7 days of culture. Conclusions: Biodentine caused gingival fibroblast reaction similar to that by MTA. Both materials were less cytotoxic than glass ionomer cement.

Abstract:Zirconia ceramics with porous structure have been prepared by solid-state reaction using yttria-stabilized zirconia and stearic acid powders. Analysis of its microstructure and phase composition revealed that a pure zirconia phase can be obtained. Our results indicated that its porosity and pore size as well as the mechanical characteristics can be tuned by changing the content of stearic acid powder. The optimal porosity and pore size of zirconia ceramic samples can be effective for the increase of surface roughness, which results in higher cell proliferation values without destroying the mechanical properties.
In Vitro Cytotoxicity of Self-Adhesive Dual-Cured Resin Cement Polymerized Beneath Three Different Cusp Inclinations of Zirconia

Abstract:The aim of the present study was to evaluate the in vitro cytotoxicity of self-adhesive dual-cured resin cement (SADRC) polymerized beneath three different cusp inclinations of zirconia with different light curing time. A commercial SADRC (Multilink Speed) was polymerized beneath zirconia (ZrO 2 ) with three different cusp inclinations (0°, 20°, and 30°) for 20 s or 40 s. After being stored in light-proof box for 24 h, the ZrO 2 -SADRC specimens were immersed in DMEM for 72 h and then we got the extract solution, cultured the human gingival fibroblasts (HGF, 8 × 10 3 per well) with 100% or 50% concentrations of the extract solution for 24 h, 72 h, and 120 h, respectively, and evaluated cytotoxicity of the polymerized SADRC with CCK-8 assay in optical density (OD) values, relative growth rates (RGR), and cytotoxicity grades. Statistical analysis was conducted using a two-way ANOVA followed by post hoc Student–Newman–Keuls test. The OD values varied from 0.8930 to 3.2920, the RGR varied from 33.93% to 98.68%, and the cytotoxicity grades varied from 0 to 2. There was significant difference in the OD values among the different cusp inclinations of zirconia (P < 0.001), and there was significant difference in the OD values between the different light curing times in some situations (P < 0.05). The cusp inclination of zirconia affects the in vitro cytotoxicity of SADRC. Prolonging the light curing time from 20 s to 40 s can reduce the in vitro cytotoxicity of SADRC when the cusp inclination of zirconia is smaller than 20°.

Abstract:Objective To identify the cytotoxicity of a new type of diatomite-based machinable ceramic and preliminarily evaluate its biocompatibility.Methods In the present study,we used MTT to assess the cytotoxicity of the new ceramic,comparing with other four prosthodontics materials.The cell lines(L929)were treated with extract dilutions of the prosthodontics materials for 1d,3d and 5d.In order to observe the changes in cell morphology,we observed absorbance(OD value)to calculate the relative cell proliferation rate(RGR),and determine the cytotoxicity grade.Results MTT method found no significant difference in the cell viability among control,new ceramic,pressable ceramic and Co-Cr alloy-porcelain(P>0.05).However,other groups were much higher than control(P<0.05).The cytotoxicity of all the materials was grade 1.Conclusion The new diatomite-based machinable ceramic had no apparent cytotoxicity to L929 cell.
Surface Modification of Zirconia with Polydopamine to Enhance Fibroblast Response and Decrease Bacterial Activity in Vitro: A Potential Technique for Soft Tissue Engineering Applications

Abstract:The quality of soft-tissue integration plays an important role in the short- and long-term success of dental implants. The aim of the present study was to provide a surface modification approach for zirconia implant abutment materials and to evaluate its influence on fibroblast behavior and oral bacteria adhesion, which are the two main factors influencing the quality of pen-implant soft-tissue seal. In this study, polydopamine (PDA)-coated zirconia was prepared and the surface characteristics were evaluated using scanning electron microscopy, atomic force microscopy, a contact-angle-measuring device, X-ray photoelectron spectroscopy, and Raman spectroscopy. The responses of human gingival fibroblasts (HGFs) to PDA-coated zirconia; i.e., adhesion, proliferation, morphology, protein synthesis, and gene expression, were analyzed. Additionally, the adhesion of Streptococcus gordonii and Streptococcus mutans to zirconia after PDA coating was assessed by scanning electron microscopy and live/dead staining. The material surface analyses suggested the successful coating of FDA onto the zirconia surface. The PDA coating significantly increased cell adhesion and proliferation compared with pristine zirconia. HGFs exhibited a high degree of spreading and secreted a high level of collagen type Ion FDA-modified disks. Upregulation of integrin alpha(5), beta(1), beta(3) and fibronectin was noted in HGFs cultured on PDA-coated zirconia. The number of adherent bacteria decreased significantly on zirconia after PDA coating. In summary, our result suggest that PDA is able to modify the surface of zirconia, influence HGFs' behavior and reduce bacterial adhesion. Therefore, this surface modification approach holds great potential for improving soft-tissue integration around zirconia abutments in clinical application. (C) 2015 Elsevier B.V. In vitro Evaluation of the Cytotoxicity of Chairside CAD/CAM Block Materials for Fabricating Dental Restorations

Abstract:Background: The increasing utilization of chairside computer-aided design/computer-aided manufacturing (CAD/CAM) block materials in dentistry necessitates a comprehensive evaluation of their cytotoxicity to ensure patient safety. This study aimed to assess the cytotoxic effects of commonly used chairside CAD/CAM block materials for fabricating dental restorations. Materials and methods: Several chairside CAD/CAM block materials including zirconia, lithium disilicate, and resin-based composites were evaluated for cytotoxicity using an in vitro model. Human gingival fibroblasts were cultured and exposed to extracts from each material. Cytotoxicity was assessed using MTT assay after 24, 48, and 72 hours of exposure. Additionally, cell morphology and viability were examined using microscopy. Results: The MTT assay revealed varying degrees of cytotoxicity among the tested materials. Zirconia demonstrated the least cytotoxicity with cell viability of 85% ± 5%, 82% ± 4%, and 79% ± 6% at 24, 48, and 72 hours, respectively. Lithium disilicate exhibited moderate cytotoxicity with cell viability of 70% ± 6%, 65% ± 7%, and 60% ± 5% at the corresponding time points. Resin-based composites displayed the highest cytotoxicity with cell viability of 55% ± 8%, 45% ± 6%, and 40% ± 4% at 24, 48, and 72 hours, respectively. Microscopic examination revealed altered cell morphology and reduced cell viability in the presence of resin-based composites. Conclusion: Chairside CAD/CAM block materials exhibit varying degrees of cytotoxicity, with zirconia demonstrating the least and resin-based composites showing the highest cytotoxic effects. These findings underscore the importance of considering cytotoxicity profiles when selecting CAD/CAM materials for dental restorations to ensure patient safety and biocompatibility.

Abstract:Background: To synthesize mesoporous titanium dioxide composite hydroxyapatite (TiO2-HAP) and to evaluate its effectiveness in sealing of occluding dentine tubules. Methods: TiO2-HAP was synthesized by chemical precipitation method and characterized using infrared absorption spectrometer, X-ray diffraction, scanning electron microscope, and specific surface area detector. Forty completely extracted molars were prepared and randomly assigned into Control group, Gluma group, HAP group and TiO2-HAP group according to different treatments. The characteristics of HAP and TiO2-HAP and the sealing effectiveness of dentine tubules in these four groups, including infrared spectrum, surface contact angle, pore size distribution, and re-mineralized enamel surface profiles, were analyzed by suitable characterized techniques. The cytotoxicity of the synthesized TiO2-HAP was tested and compared using 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide (MTT) colorimetry. Results: Our results showed TiO2-HAP group had significantly lower contact angle, higher specific surface area, and wider range of pore size distribution than other groups. The majority of dentinal tubules in the TiO2-HAP group were blocked by white matter in a uniformed manner, and the crystals arranged in order grew along the axial direction. In addition, no significant difference in optical density (OD) value was found between control group and TiO2-HAP group (P > 0.05), and cell growth was good in TiO2-HAP group, indicating no cytotoxicity of TiO2-HAP. Conclusions: The MTT assay identified that TiO2-HAP had little effect on the L929 cell line. We showed TiO2-HAP might be used as a remineralization agent in enamel caries-like lesions.
Evaluation of the Bonding Shear Strength between Enamel and Dentin Feldspathic Porcelain and Two Different Monolithic Zirconia with Low and High Translucency

Abstract:Introduction: The utilization of ceramics in the field of dentistry has seen a significant rise owing to their esthetic appeal and excellent functional properties. The use of ceramics in the field of dentistry has witnessed a notable surge, driven by their appealing esthetics and exceptional functional attributes. Zirconia, distinguished by its exceptional mechanical strength, plays a pivotal role in the fabrication of posterior crowns and bridges. Among zirconia variants, monolithic zirconia stands out, where the entire restoration is crafted from zirconia material. In parallel, feldspathic porcelain, chosen for its remarkable resemblance to natural tooth enamel, represents another significant ceramic type. This study aims to evaluate the shear bond strength (SBS) between two types of monolithic zirconia with two types of feldspathic porcelain. Methods and materials: Forty-four monolithic zirconia veneered discs with feldspathic porcelain were subjected to SBS testing. The dimensions of the discs were 7 mm in diameter and 5 mm in height (3 mm of zirconia and 2 mm of porcelain). Subsequently, the specimens were subjected to a universal testing machine at a speed of 0.5 mm/min until failure occurred. The type of failure was examined using scanning electron microscopy. One-way analysis of variance (ANOVA), two-way ANOVA, Fisher's test, and multiple Tukey comparisons were used as statistical analyses. Results: The highest SBS was achieved by the high-translucency monolithic zirconia with enamel porcelain group (18.81 ± 3.18 MPa) and the high-translucency monolithic zirconia with dentin porcelain group (17.89 ± 2.75 MPa), followed by the low-translucency monolithic zirconia with dentin porcelain group (15.04 ± 2.24 MPa) and the low-translucency monolithic zirconia with enamel porcelain group (14.33 ± 2.00 MPa), respectively. Additionally, the most common type of failure pattern observed was mixed, followed by adhesive failure. Conclusion: The translucency of the porcelain did not significantly affect SBS, while the type of monolithic zirconia used had a significant impact. Furthermore, there was no discernible relationship between the four groups in terms of the distribution of failure patterns.
Promoted Abutment-Soft Tissue Integration Around Self-Glazed Zirconia Surfaces with Nanotopography Fabricated by Additive 3D Gel Deposition

Abstract:Introduction: Improving the biological sealing around dental abutments could promote the long-term success of implants. Although titanium abutments have a wide range of clinical applications, they incur esthetic risks due to their color, especially in the esthetic zone. Currently, zirconia has been applied as an esthetic alternative material for implant abutments; however, zirconia is purported to be an inert biomaterial. How to improve the biological activities of zirconia has thus become a popular research topic. In this study, we presented a novel self-glazed zirconia (SZ) surface with nanotopography fabricated by additive 3D gel deposition and investigated its soft tissue integration capability compared to that of clinically used titanium and polished conventional zirconia surfaces. Materials and methods: Three groups of disc samples were prepared for in vitro study and the three groups of abutment samples were prepared for in vivo study. The surface topography, roughness, wettability and chemical composition of the samples were examined. Moreover, we analyzed the effect of the three groups of samples on protein adsorption and on the biological behavior of human gingival keratinocytes (HGKs) and human gingival fibroblasts (HGFs). Furthermore, we conducted an in vivo study in which the bilateral mandibular anterior teeth of rabbits were extracted and replaced with implants and corresponding abutments. Results: The surface of SZ showed a unique nanotopography with nm range roughness and a greater ability to absorb protein. The promoted expression of adhesion molecules in both HGKs and HGFs was observed on the SZ surface compared to the surfaces of Ti and PCZ, while the cell viability and proliferation of HGKs and the number of HGFs adhesion were not significant among all groups. In vivo results showed that the SZ abutment formed strong biological sealing at the abutment-soft tissue interface and exhibited markedly more hemidesmosomes when observed with a transmission electron microscope. Conclusion: These results demonstrated that the novel SZ surface with nanotopography promoted soft tissue integration, suggesting its promising application as a zirconia surface for the dental abutment.

In this study, pre-sintered zirconia was placed in a suspension of titania (TiO2) and zirconium oxychloride (ZrOCl2) and heated in a water bath for dense sintering. A titania coating was prepared on the zirconia surface and subjected to UV irradiation. The surface morphology, elemental composition and chemical state of each group of samples were analyzed by scanning electron microscope (SEM), X-ray energy spectrometer (EDS), X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD). The responses of human gingival fibroblasts (HGFs) and common oral pathogens Streptococcus mutans (S. mutans) and Porphyromonas gingivalis (P. gingivalis) to modified zirconia were systematically assessed. Our findings demonstrated that the surface of titania-coated zirconia after UV irradiation produced a large number of hydroxyl groups, and its hydrophilicity was significantly improved. Meanwhile, the UV irradiation also greatly removed the hydrocarbon contaminants on the surface of the titania-coated zirconia. The UV-treated titania coating significantly promoted the proliferation, spreading, and up-regulation of adhesion-related genes and proteins of HGFs. Furthermore, the titania coating irradiated with UV could reduce the adhesion, colonization and metabolic activity of S. mutans and P. gingivalis. Therefore, UV irradiation of titania-coated zirconia can promote the biological behavior of HGFs and exert a significant antibacterial effect, which has broad clinical application prospects for improving soft tissue integration around zirconia abutments. &#xD.
The Effect of UV Treatment on Surface Contact Angle, Fibroblast Cytotoxicity, and Proliferation with Two Types of Zirconia-Based Ceramics

Abstract:UV photofunctionalization of Zirconia-based materials for abutment fabrication is a promising approach that might influence the formation of a sound peri-implant seal, thus promoting long-term soft and hard tissue implant integration. This study aimed to evaluate the effect of UV treatment of test specimens made by two different ZnO2-based ceramic materials on the hydrophilicity, cell cytotoxicity, and proliferation of human gingival fibroblasts (HGFs). Two Zirconia-based materials, high-translucent and ultra-translucent multi-layered Zirconia (Katana, Kuraray Noritake, Japan), were used to prepare a total of 40 specimens distributed in two equally sized groups based on the material (n = 20). The same surface finishing protocol was applied for all specimens, as suggested by the manufacturer. Half the specimens from each group were treated with UV-C light for 48 h. Water contact angle (WCA), fibroblast cytotoxicity, and proliferation were investigated. The WCA values for the high-translucent Zirconia ranged from 69.9° ± 6.4° to 73.7° ± 13.9° for the treated/non-treated specimens and from 79.5° ± 12.8° to 83.4° ± 11.4° for the ultra-translucent multi-layered Zirconia, respectively. However, the difference was insignificant (F(16) = 3.50, p = 0.292). No significant difference was observed for the fibroblast cytotoxicity test. The results for proliferation revealed a significant difference, which was material-dependent (F(8) = 9.58, p = 0.005). We found that UV surface photofunctionalization of ZrO2-based materials alters the human gingival fibroblast cell viability, which might produce favourable results for cell proliferation.

Abstract:The objective of this study was to investigate the effects of a silicate-based composite material on proliferation and mineralization of human dental pulp cells (hDPCs), which was compared with those of calcium hydroxide (Ca(OH)2, CH) and tricalcium silicate (Ca3SiO5, C3S). HDPCs were cultured with CH, C3S and tricalcium silicate/dicalcium silicate (Ca3SiO5/Ca2SiO4, C3S/C2S) composites extract. The CCK-8 assay showed that the composite material stimulated the proliferation of hDPCs. The odontogenic marker genes and DSPP protein expression were more significantly up-regulated by the C3S/C2S composite material compared with pure CH and C3S. HDPCs cultured with composite material extract exert stronger ALP activity and alizarin red S staining. C3S/C2S composite material was advantageous over pure C3S by showing enhanced ability to stimulate the proliferation and odontogenic differentiation of hDPCs, suggesting that the C3S/C2S composite materials possess desirable biocompatibility and bioactivity, and might be a new type of pulp-capping agent and dentin alternative materials.
In Vitro Direct and Indirect Cytotoxicity Comparative Analysis of One Pre-Hydrated versus One Dried Acellular Porcine Dermal Matrix

Abstract:Aim: The aim of the present study was to compare the direct and indirect cytotoxicity of a porcine dried acellular dermal matrix (PDADM) versus a porcine hydrated acellular dermal matrix (PHADM) in vitro. Both are used for periodontal and peri-implant soft tissue regeneration. Materials and methods: Two standard direct cytotoxicity tests—namely, the Trypan exclusion method (TEM) and the reagent WST-1 test (4-3-[4-iodophenyl]-2-[4-nitrophenyl]-2H-[5-tetrazolio]-1,3-benzol-desulphonated)—were performed using human primary mesenchymal stem cells (HPMSCs) seeded directly onto a PDADM and PHADM after seven days. Two standard indirect cytotoxicity tests—namely, lactate dehydrogenase (LTT) and MTT (3-[4,5-dimethyl-2-thiazolyl]-2,5-diphenyl-2H-tetrazoliumbromide)—were performed using HPMSCs cultivated in eluates from the matrices incubated for 0.16 h (10 min), 1 h, and 24 h in a serum-free cell culture medium. Results: The WST and the TEM tests revealed significantly lower direct cytotoxicity values of HPMSCs on the PHADM compared with the PDADM. The indirect cytotoxicity levels were low for both the PHADM and PDADM, peaking in short-term eluates and decreasing with longer incubation times. However, they were lower for the PHADM with a statistically significant difference (p < 0.005). Conclusions: The results of the current study demonstrated a different biologic behavior between the PHADM and the PDADM, with the hydrated form showing a lower direct and indirect cytotoxicity.

Abstract:Background: This in vitro study set out to find out how well oral cavity-dwelling bacteria can form biofilms and adhere on the surfaces of zirconium oxide samples created by 3D printing and milling technologies. Methods: 5 strains of microorganisms were used for the study, and 40 zirconium oxide samples were prepared, which were divided into two groups (n = 20)—20 samples produced using removal technology comprised the control group, while 20 samples produced by 3D printing technology comprised the test group. The prepared samples were placed in culture media of bacteria and fungi that naturally occur in the oral cavity. Then, the intensity of biofilm build-up on the samples was determined using qualitative and quantitative methods. The results for both materials were compared with each other. Results: No variations in the degree of biofilm deposition on zirconium oxide samples were found for the microorganisms Streptococcus mutans, Pseudomonas aeruginosa, Enterococcus faecalis, and Staphylococcus aureus. For Candida albicans fungi, more intense biofilm deposition was observed on samples made using 3D printing technology, but these differences were not statistically significant. Conclusion: The biofilm accumulation intensity of ceramics produced by additive technology is comparable to that of milled zirconium oxide, which supports the material's broader use in clinical practice from a microbiological perspective. This ceramic has demonstrated its ability to compete with zirconium oxide produced by milling techniques in in vitro experiments, but sadly, no in vivo tests have yet been found to determine how this material will function in a patient's oral cavity.
