Abstract:Microstructure of hydroxyapatite is known to influence cellular behavior, can be performed as a substrate for osteoblast growth, and exploited as a drug release platform. However, easy delamination and self-decomposition of hydroxyapatite caused by poor adhesion with substrates are the main problems currently. In this paper, we successfully fabricated TiO2/SrHA composite scaffolds by self-generated strontium substituted hydroxyapatite microspheres in TiO2 nanotubes. Moreover, the active compound 1-(5-bromo-2-hydroxy-methoxyphenyl)-ethanone (BHM) from Seahorse (Hippocampus kuda Bleeler) was loaded in this scaffold, and the controlled release kinetics of BHM was studied. It was found that in the first 5 hours, the release concentration and time of BHM had a good linear relationship, and the correlation coefficient reached 0.98. TiO2/SrHA/BHM composites exhibited favorable cytocompatibility at a given concentration of BHM (20 μmol/L). Compared to pure SrHA, TiO2 nanotubes and traditional TiO2/SrHA composites, the superior cytocompatibility (cell adhesion and proliferation) of MC3T3-E1 was obtained on TiO2/SrHA/BHM composites. The expression levels of osteogenic marker genes such as ALP, OPN and RunX2 are also up-regulated to varying degrees. This TiO2/SrHA composite scaffold mediated phenolic compounds BHM could be applied in bone tissue repair.

Abstract:For dental/orthopedic implants to achieve better bone apposition and bone-implant bonding, various approaches to improve titanium surfaces have been developed. Recently, a fluoridated hydroxyapatite (FHA) coating on titanium (Ti) implants was made by sol–gel method and shown to be a possible applicative bone implant. The purpose of the current study was to evaluate biological responses and biomechanical bonding strength of FHA coated Ti implants as compared with that of the conventional Ti alloys and hydroxyapatite (HA) coated Ti implants. In vitro assays were made using human osteoblast-like cell (MG63) culture on different implants with cell attachment, morphology and differentiation evaluations. The implant plates were also implanted into the proximal metaphysis of New Zealand White rabbit tibiae. After 8 and 16 weeks implantation, mechanical and histological assessments were performed to evaluate biomechanical and biological behavior in vivo. The results showed that the cell adhesion and cell growth rate on the FHA and HA surface was higher than that on cp Ti surface (p<0.01), and insignificant difference was observed between two coated groups. Mechanical test demonstrated that the FHA implants had a higher interface shear strength than the both controls at 8 and 16 wks, with no significant difference with HA-Ti. Histologically, the coated implants revealed a significantly greater percentage of bone-implant contact when compared with the uncoated implants. Results demonstrated that the new FHA surface improved cell adhesion and proliferation. The coating exhibited a bioactive mechanical and histological behavior at bone-implant interface, suggesting that a useful approach by combined coating processes could optimize implant surfaces for bone deposition and early implant fixation.
The Impact of Titanium Hydroxyapatite Doping on the Mechanical and Biological Properties of Photocured Resin

Abstract:Photocured resin materials are widely used in various fields, such as 3D printing, medical applications, and dentistry. However, the strength, wear resistance, and antibacterial properties of photocured resin are relatively limited, rendering it susceptible to potential failures. In this recent study, photocured composite resins incorporating titanium-doped hydroxyapatite (Ti-HAp) were fabricated to investigate their mechanical and biological properties. It was found that the hardness and wear resistance increased with the addition of an appropriate amount of hydroxyapatite (HAp). Specifically, the 6wt%HAp resin demonstrated superior hardness. Compared with the 6wt%HAp resin, the acid resistance and wear resistance improved when an appropriate amount of Ti-HAp was added. Notably, the resin containing 0.56%Ti-HAp demonstrated superior wear resistance. Additionally, the antibacterial performance improved with higher titanium (Ti) content, showcasing a 71.9% improvement in the resin containing 1.37%Ti-HAp compared with the 6wt%HAp resin, alongside commendable remineralization capabilities. In summary, the Ti-HAp composite resin showed enhanced mechanical and biological properties, meeting clinical standards in terms of mechanical and antibacterial properties.

Abstract:Dentine hypersensitivity (DH) is a common issue among humans caused by the erosion of tooth enamel as a result of chewing and brushing incorrectly. The most effective treatment method is occluding the exposed dentine tubules. Because calcium phosphate is an important mineral and the most common mineral for dentine and orthopaedic application, this study focuses on the synthesis of bioactive gels containing calcium phosphate nanoparticles. The synthesis is performed using water-in-oil emulsion and characterized by the use of TGA, Rheological, TEM, and SEM techniques. This method used oleic fatty acids as an external phase and Tween 20 as surfactant to form water-in-oil nanodroplets. Gelatine was used to form the substance into a gel and increase its stability. The resulting nanoparticles had the same spherical shape and a similar size. Therefore, the varying amounts of gelatin do not affect the particle size. However, viscoelastic properties of gels were found to be changed. The ability of the gel for dentine tubule occlusion was investigated using SEM techniques. After 1 day, it was found that the gels can occlude dentine tubules.
Disaggregated Nano-Hydroxyapatite (DnHAP) with Inhibitory Effects on Biofilms and Demineralization

Abstract:Nano-hydroxyapatite (nHAP) is considered a biocompatible agent that promotes the remineralization of dental hard tissue; however, its antibacterial efficacy is under scientific discussion. Therefore, this investigation aimed to specify the inhibitory effects of disaggregated nano-hydroxyapatite (DnHAP) on regrown biofilms and demineralization. Regrown biofilm models of single-species (Streptococcus mutans), dual-species (S. mutans and Candida albicans), and saliva-derived microcosm biofilms were established in vitro. Repeat treatment with DnHAP was applied to biofilms. The viability, lactic acid, biofilm structure, biomass, the inhibitory effect of demineralization, and virulence factors' expression were determined. In addition, the biofilm microbial community was analyzed by 16S ribosomal RNA gene sequencing. DnHAP inhibited metabolism, lactic acid production, biomass, and water-insoluble polysaccharide production (P < 0.05) of regrown single/dual-species biofilms. Concerning the saliva-derived biofilms, samples treated with DnHAP showed lower biofilm metabolic activity without significant differences from samples treated with sterile deionized water (P > 0.05); in addition, saliva-derived biofilms treated with DnHAP exhibited lower lactic acid production (P < 0.05). The demineralization of bovine enamel was the lowest in the DnHAP group, as detected by transverse microradiography, and the lesion depth and volume decreased significantly (P < 0.05). The application of DnHAP did not change the diversity of regrown saliva-derived microcosm biofilms. In conclusion, this investigation showed that DnHAP could be a promising solution for the management of regrown biofilms to combat dental caries.
Characterization of a Novel TiF 4 Inclusion Complex and in vitro Evaluation of Its Effect on Inhibiting Enamel Demineralization

Abstract:Introduction: Titanium tetrafluoride (TiF 4 ) is an anticariogenic agent with high remineralizing potential. However, the acidic pH of TiF 4 solution can limit its clinical application. The present study aimed to prepare and characterize a new TiF 4 -dendrimer inclusion complex and evaluate its ability to inhibit enamel demineralization under pH cycling conditions. Methods: PEG-citrate dendrimer and TiF 4 -dendrimer inclusion complex were synthesized and their molecular structures were evaluated using Fourier-transform Infrared Spectroscopy (FTIR), Hydrogen Nuclear Magnetic Resonance (HNMR), and Liquid Chromatography-Mass Spectrometry (LC-MS) tests. Forty-eight enamel samples were prepared and randomly divided into four groups: distilled water (negative control), TiF 4 solution (T), dendrimer solution (D), and TiF 4 -dendrimer solution (TD). The microhardness of the samples was measured initially. Next, the samples underwent pH cycling, were exposed to the solutions, the microhardness was measured again, and microhardness loss was calculated. EDX analysis was performed on the surface and cross-sectional segments of the samples. Results: The microhardness loss was significantly higher in control (−65.1 ± 6.0) compared to other groups. No significant difference was observed between T (−47.9 ± 5.6) and D (−41.7 ± 12.0) and also D and TD (−40.5 ± 9.4) in this regard. Microhardness loss was significantly higher in T compared to TD group. The TD samples showed similar fluoride and titanium content in both surface and subsurface regions, while the T group had higher concentrations in the surface region. Moreover, the TD solution had a higher pH of 3.4 compared to the T solution’s pH of 1.1. Conclusion: No significant difference was observed between the efficacy of TiF 4 -dendrimer and TiF 4 solution in inhibiting demineralization while TiF 4 -dendrimer solution had the added advantage of having a higher pH.

Abstract:Background: Maintaining a long-term biological effect of dental materials on dentinal tubule occlusion is one of the great technical challenges in dental clinics. In addition to physical treatment, chemical treatment to produce insoluble precipitates to seal dentinal tubules has been used. As dentin is mostly composed of calcium and phosphate complexes, in this study, we have developed a novel tubule-occluding material [Ca2+/PO43-@mesoporous silica nanoparticles (MSNs)] by separately conjugating either Ca2+ or PO43- with MSNs. Methods: The shape and structure of the MSNs were examined using transmission electron microscopy (TEM) and scanning electron microscopy (SEM). The surface morphology and chemical compositions of Ca2+@MSNs/PO43-@MSNs and Ca2+/PO43-@MSNs were examined using SEM and X-ray fluorescence (XRF). The element distribution of Ca2+/PO43@MSNs was detected using energy dispersive spectrometer (EDS). The sustained release ability of Ca2+@MSNs/PO43-@MSNs was detected using inductively coupled plasma atomic emission spectrometry (ICP-AES). The efficacy of Ca2+/PO43-@MSNs on dentinal tubule sealing was evaluated using SEM, and the results were analyzed by Image-Pro software to determine the best waterpowder ratio. We also compared the sealing efficacy between Ca2+/PO43-@MSNs and NovaMin, which is currently used in clinics, under the simulated conditions of oral acidic corrosion and mechanical friction. Results: Ca2+/PO43-@MSNs are a new type of tubule-occluding material with sustained release properties. The ratio of Ca2+@MSNs: PO43-@MSNs: H2O =0.015 g: 0.015 g: 150 mu L exhibited an excellent sealing effect on dentinal tubules as well as resistance to oral acid corrosion and daily oral friction. Conclusions: The novel dental material Ca2+/PO43-@MSNs demonstrates potential long-term effectiveness in sealing dentinal tubules and reducing dentin sensitivity, which is one of the most important problems in dental clinics.
Comparison of Nanofluoridated Hydroxyapatite of Varying Fluoride Content for Dentin Tubule Occlusion

Abstract:Purpose: To evaluate the viability of a series of nano-fluoridated hydroxyapatite (nano-FHA) formulations of varying fluoride content for the occlusion of exposed dentin tubules, in comparison to nano-hydroxyapatite (nano-HA). Methods: Nano-FHA powders with varying levels of fluoride ion were synthesized to substitute hydroxyl ions (-OH) present in hydroxyapatite (HA). Nano-FHA were defined as 2nFHA,4nFHA,6nFHA, 8nFHA and 10nFHA, with the molar ratio of F- to Ca-2+ of 0.002,0.004,0.006,0.008 and 0.01, respectively. The powders were synthesized using a wet chemistry route, and characterized by transmission electron microscopy, X-ray diffraction, and Fourier transform infrared spectroscopy. The methylthiazol tetrazolium (MTT) assay was used to assess cell viability toward nano-FHA. According to the content of F-, the nano-nFHA and nano-HA powders were divided into six groups, with the artificial saliva as control. They were spread over the dentin surface for 1 minute and repeated three times per day for 7 consecutive days. After washing and brushing with distilled water for an additional 7 days, scanning electron microscopy was used to evaluate the in vitro plugging rate of the tubules and penetrating depth. Results: The 2nFHA, 4nFHA, 6nFHA, 8nFHA and 10nFHA powders were prepared and characterized. The 8nFHA had less crystallinity compared to 2nFHA,4nFHA, and 6nFHA.The MTT assay (from 12 to 120 hours) showed that the cell viability of the L-929 fibroblasts in the 2nFHA, 4nFHA, 6nFHA, 8nFHA, 10nFHA, and nano-HA groups ranged from 80.54 +/- 3.35% to 112.9 +/- 4.8%. Most of the nanoFHA powders successfully occluded dentin tubules. The plugging rate of 8nFHA was significantly higher than that of the 2nFHA, 4nFHA, 6nFHA and nano-HA groups. The penetrating depth of the 8nFHA group into the tubules was also significantly higher than that of the other nano-FHA and nano-HA group.
Synthesis and characterization of different nano-hydroxyapatites and their impact on dental enamel following topical application for dental bleaching

Abstract:Objective: This study aims to synthesize, characterize, and assess the penetration of hydrogen peroxide (HP), color change (CC), and surface morphology changes after the application of two distinct nano-hydroxyapatite (nano-HAp). Methods: Two nano-HAp were previously synthesized by co-precipitation: one with rod-shaped particles (RS) and the other with spherical-shaped particles (SS). The surface charge of the nano-HAp particles was determined at varying pH levels and characterized by X-ray diffraction patterns and Fourier transform infrared spectroscopy. The morphology of the samples was assessed using scanning electron microscopy (SEM). The nano-HAp particles were applied before the dental bleaching procedure. Forty teeth were divided into four groups (n = 10) according to the bleaching treatment: no treatment, bleaching with 35% HP only, RS application and bleaching with 35% HP, and SS application and bleaching with 35% HP. HP concentration (µg mL-1) was measured using UV-Vis, while CC was evaluated with a digital spectrophotometer (ΔEab, ΔE00 and WID). Additionally, four teeth from each group were selected for SEM analysis. Statistical analysis encompassed one-way ANOVA, Tukey's, and Dunnet's tests. Results: RS and SS were successfully synthesized by coprecipitation, primarily differing in pH during synthesis. Both variations of nano-HAp morphology significantly reduced HP diffusion into the pulp chamber (p < 0.001). Regarding enamel morphology, groups analyzed post dental bleaching exhibited greater HAp deposition on the enamel surface. Notably, this deposition did not impede CC. Significance: The utilization of different nano-HAp morphologies prior to dental bleaching appears to be a promising strategy for mitigating adverse effects associated with dental bleaching procedures.
Effect of polyamidoamine dendrimer on the occlusion of dentinal tubule

Abstract:Objective To evaluate the occluding ability of polyamidoamine dendrimers (PAMAM) on human dentinal tubules,and to provide experimental basis for dentin remineralization and the treatment of dentin hypersensitivity.Methods Twenty-eight dentin disks from scvcn extracted sound human third molars were divided into seven groups (Group A,B,C,D,E,F and G) randomly.Group G was served as the control.After demineralized with citric acid and NaOCl aqueous solution,the dentin disks of Group A to E were treated with G2.0 PAMAM (glutaraldehyde crosslinked for 24 h) and subsequently immersed into remineralizing solution for a period of time (0,6,12,18 and 24 h) respectively and Group F was reminerabzed for 24 h without being treated by PAMAM.The dentin discs were characterized by field emission scanning electron microscope (FE-SEM),X-ray diffraction (XRD),and energy dispersive X-ray spectrometer(EDXS).Results SEM showed that the results were different in different groups.The minerals on the surfaces of the dentin disks were formed gradually with the time.The dentinal tubules were occluded effectively.The occluding depth of dentinal tubules was approximately 10 μm.The minerals were determined to be hydroxyapatite according to XRD and EDXS tests.Conclusions The experimental results showed that polyamidoamine dendrimers had the occluding ability by inducing remineralization on the demineralized collagen matrix.Consequently,this strategy may provide an experimental basis for dentin remineralization and is valuable for developing a new method to treat dentin hypersensitivity.
Electron Microscopy Evaluation of Mineralization on Peritubular Dentin with Amorphous Calcium Magnesium Phosphate Microspheres

Abstract:Dentin hypersensitivity can be reduced by the use of a remineralization agent to hinder movement of fluids within the tubules. Penetration of particles into the tubules and a continuous release of Ca2+ and phosphate ions can induce the mineralization of a material mimicking the mineral component of dentin, sealing the tubules. In this work, we have used complementary electron microscopy techniques to investigate the ultrastructure of dentin and crystallization and occlusion effects when using amorphous calcium magnesium phosphate (ACMP) microspheres on extracted human molars. Application of the particles in a gel intended for at-home use resulted in intra-tubular mineralization of a carbonate substituted hydroxyapatite (HA). Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) showed that crystallization was initiated on the peritubular dentin (PTD) with undirected crystal growth leading to the formation of a porous material. We additionally investigated the effects from using a fluoride toothpaste to potentially improve the remineralization and anti-cariogenic properties of the ACMP microspheres. Energy dispersive x-ray spectroscopy (EDX) using TEM in scanning mode (STEM) showed that fluoride incorporation resulted in an increase in aspect ratio of the crystals, crystal growth directed towards the center of the tubule lumen and densification of the mineralized material. Thus, ACMP microspheres are promising alternatives as occluding agents and the efficacy of the particles could be further improved with the use of a fluoride toothpaste.
Remineralizing Efficacy of Fluorohydroxyapatite Gel on Artificial Dentinal Caries Lesion

Abstract:The aim was to evaluate the remineralizing efficacy of fluorohydroxyapatite (FHA) gel on artificial dentinal caries lesion in vitro. Artificial carious lesions were made on occlusal cavities of teeth by exposing the dentin surface to a demineralizing solution. Each cavity was capped with a 3 mm thick FHA gel for 4 weeks. After the FHA gel was removed, the surface morphology and structure of the dentin were characterized by scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FT-IR). The dentin mineral density (DMD) was measured by micro-computed tomography (Micro-CT). A layer of dense and orderly hexagonal crystal structure, with average diameter of 1 μm and thickness of 4~5 μm, could be observed on dentin surface. These crystals exhibited elemental peaks for calcium, phosphorus, carbon, and oxygen and characteristic peaks of hydroxyapatite (HA) and fluorapatite (FA) via XRD and FT-IR. The DMD of dentin surface layer significantly increased after it was capped with FHA gel (P<0.05). In the present study, the FHA gel could rapidly construct apatite on the artificial dentin caries surface and significantly increase the mineral density, which suggests that FHA gel might be a proper IPT material with remineralizing function.

Abstract:The occlusion of dentinal tubules is considered to be an effective strategy to treat dentin hypersensitivity. This in vitro study introduced mesoporous silica nanoparticles (MSNs) for tubular occlusion to achieve deeper sealing. Further, MSNs with independently encapsulated calcium and phosphates (as calcium and phosphate sources) (Ca2+/PO43-@MSNs) were introduced to achieve improved efficacy of tubular occlusion and remineralization. MSNs or Ca2+/PO43-@MSNs were proportionally mixed with distilled water to make their respective desensitizing slurries, which were used to treat dentin surfaces. The efficacy of tubular occlusion was evaluated using scanning electron microscopy (SEM) images and an image analyzer, and compared with that achieved with Green-Or (TM)-a commonly used densitizer. The results demonstrated that both MSNs and Ca2+/PO43-@MSNs almost completely occluded dentinal tubules and formed a deeper seal which penetrated about 105 mu m deep into the dentinal tubules. Significant differences in tubular occlusion were observed between Green-Or (TM) densitizer and MSNs or Ca2+/PO43-@MSNs. The novel MSNs-based nanomaterials showed great potential as a treatment option for dentin hypersensitivity.
Use of poly (amidoamine) dendrimer for dentinal tubule occlusion: a preliminary study.

Abstract:The occlusion of dentinal tubules is an effective method to alleviate the symptoms caused by dentin hypersensitivity, a significant health problem in dentistry and daily life. The in situ mineralization within dentinal tubules is a promising treatment for dentin hypersensitivity as it induces the formation of mineral on the sensitive regions and occludes the dentinal tubules. This study was carried out to evaluate the in vitro effect of a whole generation poly(amidoamine) (PAMAM) dendrimer (G3.0) on dentinal tubule occlusion by inducing mineralization within dentinal tubules. Dentin discs were treated with PAMAM dendrimers using two methods, followed by the in vitro characterization using Attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR), X-ray diffraction (XRD), Field emission scanning electron microscopy (FE-SEM) and Energy-Dispersive X-ray Spectroscopy (EDS). These results showed that G3.0 PAMAM dendrimers coated on dentin surface and infiltrated in dentinal tubules could induce hydroxyapatite formation and resulted in effective dentinal tubule occlusion. Moreover, crosslinked PAMAM dendrimers could induce the remineralization of demineralized dentin and thus had the potential in dentinal tubule occlusion. In this in vitro study, dentinal tubules occlusion could be achieved by using PAMAM dendrimers. This could lead to the development of a new therapeutic technique for the treatment of dentin hypersensitivity.

Abstract:Calcium phosphate crystals, as the main component of dentin and enamel, have been widely used for the occlusion of dentinal tubules. However, the low bond strength and poor sealing effect limit their clinical practicality. In this study, a collagen/calcium dual-affinitive peptide E8DS (EEEEEEEEDSpESpSpEEDR) and nano-hydroxyapatite (nano-HAp) flocculi were developed to seal dentin tubules for reducing dentin hypersensitivity, whereas the E8DS peptides were pre-applied to improve the adhesion of occlusive hydroxyapatite coating on dentin collagen matrix for the long-lasting sealing effect and relief from hypersensitivity. Our study showed that E8DS peptides had a strong affinity with dentin collageneous matrix that almost 43.7% of initial E8DS peptides immobilized on exposed dentin samples remained detained after continuous washing by distilled water for four weeks at a rate of 1mL/min. Nano-HAp flocculi were obtained by re-neutralization of HAp-HCl solution and then brushed onto the surfaces of pre-treated human dentin disks with E8DS peptides, which showed a perfect occlusion of exposed dentinal tubules, as compared with Nano-HAp only and a commercial desensitizer, Green Or. With only around 10-min E8DS peptide pre-treatment, the occlusive mineral layers remained intact against consecutively stirred washing in phosphate-buffered saline or coke for 15min, and 6min of tooth-brushing, which implied that our E8DS peptide could comparatively improve the durability of sealant-dentin interface bonds for long-lasting dentine desensitization.

Abstract:In this study, a hydroxyapatite (HA) was coated on a pure titanium surface by means of a complex oxidation and hydrothermal treatment. First an anodic oxidation was done on the titanium plates, followed by micro-arc oxidation. The HA-coated specimens and pure titanium specimens were immersed in SLB for 1, 5, and 10 days, respectively, to study their electrochemical behavior. The corrosion currents of HA-coated specimens were less than pure titanium specimens. This indicated that HA coating prevented surface metal ions of the implant from dissolving, thereby, reducing the tissue toxicity. The cytotoxic effect on fibroblasts L929 cells was measured by cell counting after being seeded for 2, 4, 8, 12, and 24 h. The number of surface cell attachments on the HA-coated specimens was much greater than on pure titanium specimens. The morphology of the cells on the HA coating had normal shapes and spread well with some cells climbing onto surface pores while cells on the pure titanium were oval shaped. The results confirm that the cell compatibility on HA-coated ion titanium surfaces is much better than pure titanium.

Abstract:A simple method of synthesis of TiO2 nanotubes (TiNT) loaded with hydroxyapatite (HAP) is described. Such nanotubes find wide applications in various fields, including biomedicine, solar cells, and drug delivery, due to their bioactivity and potential for osseointegration. The Cp-Ti substrate was anodized at a constant voltage of 40 V, with the subsequent heat treatment at 450 °C. The resulting TiNT had a diameter of 100.3 ± 2.8 nm and a length of 3.5 ± 0.04 μm. The best result of the growth rate of HAP in Hanks’ balanced salt solution (Hanks’ BSS) was obtained in calcium glycerophosphate (CG = 0.1 g/L) when precipitates formed on the bottom and walls of the nanotubes. Structural properties, surface wettability, corrosion resistance, and growth rate of HAP as an indicator of the bioactivity of the coating have been studied. X-ray diffraction (XRD), scanning electron microscope (SEM), energy dispersive spectroscopy (EDS), potentiodynamic polarization test (PPC), electrochemical impedance spectroscopy (EIS), and contact angle (CA) measurements were used to characterize HAP-loaded nanotubes (HAP-TiNT). The CA, also serving as an indirect indicator of bioactivity, was 30.4 ± 1.1° for the TiNT not containing HAP. The contact angle value for HAP-TiNT produced in 0.1 g/L CG was 18.2 ± 1.2°, and for HAP-TiNT exposed to Hanks’ BSS for 7 days, the CA was 7.2 ± 0.5°. The corrosion studies and measurement of HAP growth rates after a 7-day exposure to Hanks’ BSS confirmed the result that TiNT processed in 0.1 g/L of CG exhibited the most significant capacity for HAP formation compared to the other tested samples.

Abstract:Dentin hypersensitivity is attributable to the exposed dentin and its patent tubules. We proposed the therapeutic management of demineralized dentin surfaces using a mineralizing adhesive to seal and remineralize dentin, dentinal tubules, and odontoblast processes. An experimental self-etch adhesive and a mineralizing adhesive consisting of the self-etch adhesive and 20 wt % poly-aspartic acid-stabilized amorphous calcium phosphate (PAsp-ACP) nanoparticles were prepared and characterized by X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM), and scanning electron microscopy. After 60 acid-etched midcoronal dentin disks were treated with distilled water (control), a desensitizing agent (Gluma), the experimental self-etch adhesive, and the mineralizing adhesive, dentin permeability was measured and mineralization was evaluated by Raman, FTIR, XRD, TEM, and selected-area electron diffraction, irrespective of abrasive and acidic challenges. In vitro cytotoxicity of the adhesive and the mineralizing adhesive was assessed by Cell Counting Kit-8. The mineralizing adhesive possessed excellent biocompatibility. We proposed a hybrid mineralization layer composed of the light-cured mineralizing adhesive and the mineralized dentin surfaces, as well as interiorly mineralized resin tags and odontoblast processes inside of the dentinal tubules. This hybrid mineralization not only reduced dentin permeability but also resisted abrasive and acidic attacks.
Development of TiF4-Dendrimer complex gel as an anti-demineralization agent for dentin: An in vitro study

Abstract:Objective The anti-caries effects of titanium tetrafluoride (TiF4) are well-documented, but its low pH challenges clinical application. This study evaluated PEG-citrate dendrimer as a carrier to enhance TiF4 stability and efficacy. Methods PEG-citrate dendrimer and TiF4-dendrimer gel were synthesized, and their structures confirmed using Fourier Transform Infrared Spectroscopy (FTIR), Hydrogen Nuclear Magnetic Resonance (1H NMR), and Liquid Chromatography–Mass Spectrometry (LC-MS). Thirty-six intact human teeth were prepared, randomly divided into three groups (n = 12) and subjected to pH cycling with the following treatments: titanium tetrafluoride (T), dendrimer (D), and dendrimer with TiF4 (TD). Vickers microhardness and Raman spectroscopy evaluated dentin demineralization. EDS analysis measured titanium and fluoride penetration into dentin in T and TD groups and mineral content (calcium and phosphorus) in all groups. Results The T group showed the highest microhardness loss (p < 0.001), followed by D and TD groups. EDS analysis revealed no significant difference in titanium and fluoride content between the surface and subsurface in TD (p = 0.344), while T had more titanium on the surface (p < 0.001). TD had higher subsurface calcium content compared to T (p = 0.008). Raman spectroscopy revealed significant changes in phosphate-to-amide and carbonate-to-amide ratios before and after pH cycling in all groups (p < 0.001), with no statistical differences among the groups. Conclusion Using dendrimer as a carrier for TiF4 increased pH and enhanced TiF44 ability to limit dentin demineralization and microhardness loss. Clinical Relevance The application of the newly-developed TiF4-dendrimer gel might be an effective approach to prevent/ limit dentin demineralization and dentin caries.
