Abstract:A simple, efficient and green protocol for chemoselective reduction of carbon-carbon double bond in conjugated alkenes with Hantzsch esters is described. Without any additional catalysts, a series of conjugated alkenes with strong electron-withdrawing groups were reduced in water with excellent yield. Functional groups such as nitrile, ester, nitro, fluoro, chloro, bromo, furanyl and benzyl are all tolerated by the reaction conditions employed.

Abstract:The Pd/C-catalyzed transfer hydrogenation with Hantzsch ester 1,4-dihydropyridine (HEH) provides an efficient protocol for the selective reduction of aromatic azides to the corresponding amines. The method is advantageous in terms of high yield and high selectivity, The reduction of aromatic nitro compounds can also be achieved with the method. In addition, the reagent combination of HEH-Pd/C is also effective for the reduction of conjugated olefins.

Abstract:A metal-free chemoselective reduction of α, β-unsaturated carbonyl compounds using water as hydrogen donor is described. Various functional groups attached to substrates were well tolerated to afford the corresponding products in moderate to good yields under simple reaction conditions. Abstract:Hantzsch esters are used extensively in photoredox catalysis as terminal reductants, donating an electron and a hydrogen atom. They generally exert their reducing potential by means of single-electron transfer to a photoredox catalyst, which acts as an electron shuttle. Recently, they were found to effect single-electron transfer upon visible-light irradiation in the absence of a photocatalyst. We found that this ability could be used for debromination of vicinal dibromo compounds to afford alkenes, as well as for hydrodifluoroacetamidation reactions. We also found that substituted Hantzsch esters could be used as reductive alkylation reagents to build highly congested ketones.

Abstract:A thiourea-catalyzed biomimetic reduction of conjugated nitroalkenes has been developed. Various aromatic and aliphatic conjugated nitroalkenes can be reduced to give the respective nitroalkanes with good yields under mild conditions. This protocol is not only practical, but may also provide insight into the mechanisms of redox transformations in biological systems.

Abstract:A cobalt-catalyzed asymmetric reduction of beta,beta-dialkyl alpha,beta-unsaturated esters with polymethylhydrosiloxane (PMHS) was reported to deliver the corresponding esters containing a chiral trialkyl carbon center at beta-position with up to 97 % yield and 98 % ee. The chiral tridentate ligand oxazoline iminopyridine (OIP) could perform well for the asymmetric reduction instead of chiral bidentate ligands. This operationally simple protocol shows a broad scope of substrates using one equivalent of readily available PMHS as a cheap and easy-to-handle reductive reagent.

Abstract:The chemical activation and functionalization of water are considered an ideal method for converting earth-abundant sources into valuable chemicals. Here, we show that a non-activated free water molecule can be applied directly as a hydrogen donor to achieve the carbanion-mediated alkene reduction with 9-HTXTF serving as an organophotocatalyst. Notably, direct syntheses of high-value-added drugs and bioactive molecules are readily achieved by utilizing plentiful energy and an earth-abundant resource, showcasing the usefulness of the protocol in chemical synthesis.

Abstract:Iron-catalyzed direct reduction of allylic halides with benzylic alcohol was achieved, providing a new, simple, and efficient method for conducting highly regioselective hydrodehalogenation. This method not only features a readily available reductant, an inexpensive catalyst, simple manipulation, and good tolerance of functional groups including nitriles, nitro, esters, and methoxyl groups, it also has mild reaction conditions and shows complete regioselectivity in that only halides sited at the allylic position are reduced. Alternatively, this method can be applied in the selective transformation of benzylic alcohols to aromatic aldehydes without overoxidation to carboxylic acids.

Abstract:Several bidentate dihydroimidazolines were prepared and investigated as catalysts for hydrogen transfer reduction of C═N bond with Hantzsch ester. Highly efficient reactions were observed for quinolines and imines with low catalyst loading of 2 mol %. The presence of halogen bonding was elucidated using NMR studies and isothermal calorimeric titrations. Binding constants of the XB donors were also measured using isothermal calorimeric titrations (ITC).

Abstract:Herein, we disclose a highly chemoselective room-temperature deoxygenation method applicable to various functionalized N-heterocyclic N-oxides via visible light-mediated metallaphotoredox catalysis using Hantzsch esters as the sole stoichiometric reductant. Despite the feasibility of catalyst-free conditions, most of these deoxygenations can be completed within a few minutes using only a tiny amount of a catalyst. This technology also allows for multigram-scale reactions even with an extremely low catalyst loading of 0.01 mol %. The scope of this scalable and operationally convenient protocol encompasses a wide range of functional groups, such as amides, carbamates, esters, ketones, nitrile groups, nitro groups, and halogens, which provide access to the corresponding deoxygenated N-heterocycles in good to excellent yields (an average of an 86.8% yield for a total of 45 examples).

Abstract:In aqueous media,Hantzsch reaction of hexamethylenetetramine and ethyl acetoacetate with ammonium carbonate occured to diethyl 1,4-dihydro-2,6-dimethylpyridine-3,5-dicarboxylate,in yield of 90% at 70 ℃.In 55-60 ℃ aldehyde and ethyl allylacetoacetate reacted with ammonium carbonate to give symmetrical Hantzsch ester in yield of 88%-99% in 1-4 h.Instead of ammonium carbonate with ammonium acetate,in green organic solvent(water or ethanol),catalyst(proline or guanidine hydrochloride) catalyzed Hantzsch reaction in medium to quantitative yield.Instead of another ethyl allylacetoacetate with 5,5-dimethyl-cyclohexane-1,3-dione,asymmetric Hantzsch reactions occured.Even without solvent and catalyst at ambient temperature,Hantzsch esters of aromatization were produced.With the green chemistry oriented,the basic research results of Hantzsch reaction were designed into were designed and comprehensive synthesis experiments to achieve Hantzsch esters,and they are widely used in chemistry,biology and medicine.Not only do they develop the innovative ability of students and cultivate their green environmental protection consciousness,but also they could link theory with practice,to use what you've learned.

Abstract:A cascade Knoevenagel condensation-reduction approach, which was carried out in water, has been reported. Using Hantzsch esters as reducing agent, under the promotion of base, a variety of reduced Knoevenagel adducts could be easily prepared by direct alkylation of malononitrile, ethyl 2-cyanoacetate, and 2-(4-nitrophenyl)acetonitrile, respectively. Meanwhile, a gram-scale synthesis of the protocol was also realized with excellent isolated yield.
