Abstract:The oxidation of N-heterocycles is key to their functionalization since it enhances their reactivity in the ortho position. How-ever, an efficient post-reduction is often required to access the targeted compound. Here, a rhenium-based photocatalyst enables the versatile and fast deoxygenation of N-heterocyclic and alkyl N-oxides, avoiding sacrificial oxophilic reagents. The strong influence of the reaction conditions (solvent, H2O as additive, irradiation set-up) is demonstrated. The system shows a high functional group tolerance towards other reducible functions (free alcohol, carboxylic acid, ester, nitrile, halo-gen,…) and light-sensitive N-heterocycles.

Abstract:We present a novel deoxygenation method of heterocyclic N-oxides which uses iodide as a catalyst. In this methodology, iodide acts as a reducing catalyst that is regenerated by formic acid, which also acts as a Brønsted activator and solvent. This method demonstrates high efficiency and excellent selectivity in the reduction of a variety of heterocyclic N-oxides and tertiary amines. Our computational DFT investigation revealed that the reduction mechanism entails a direct interaction between iodide and the oxygen of the N-oxide within a Mg2+/formic acid framework, resulting in the formation of the N-heterocycle and the release of a hypoiodite unit. Additionally, a molecular mechanism for the regeneration of iodide from hypoiodite, facilitated by formic acid, is suggested. This approach offers an environmentally friendly approach for the deoxygenation of N-oxides and related species.

Abstract:Regio‐ and chemo‐selective functionalization of electron deficient azaarenes often required their transformations to corresponding N‐oxides and subsequent removal of oxygen after functionalization to get back the desired substituted azaarenes. An efficient metal‐free visible‐light photo‐redox catalytic deoxygenation of N‐oxides of azaheterocyclic compounds has been developed using acridinium based organo‐photocatalyst in blue LED light. High efficiency and mildness of this methodology has been demonstrated through higher deoxygenation yield of wide variety of azaheterocyclic N‐oxides with reactive functional groups. Robustness of the photocatalytic reduction has been demonstrated through easy scaling‐up of the reaction to gram level without much change in the reaction yield.

Abstract:A rapid and practical protocol for the chemoselective deoxygenation of various aromatic ketones and aldehydes was described, which used a tandem catalyst composed of heterogeneous Pd/TiO2 + homogeneous FeCl3 with the green hydrogen source, polymethylhydrosiloxane (PMHS). The developed catalytic system was robust and scalable, as exemplified by the deoxygenation of acetophenone, which was performed on a gram scale in an atmospheric environment utilizing only 0.4 mol % Pd/TiO2 + 10 mol % FeCl3 catalyst to give the corresponding ethylbenzene in 96% yield within 10 min at room temperature. Furthermore, the Pd/TiO2 catalyst was shown to be recyclable up to three times without an observable decrease in efficiency and it exhibited low metal leaching under the reaction conditions. Insights toward the reaction mechanism of Pd-catalyzed reductive deoxygenation for aromatic ketones and aldehydes were investigated through operando IR, NMR, and GC-MS techniques.

Abstract:Hantzsch esters are used extensively in photoredox catalysis as terminal reductants, donating an electron and a hydrogen atom. They generally exert their reducing potential by means of single-electron transfer to a photoredox catalyst, which acts as an electron shuttle. Recently, they were found to effect single-electron transfer upon visible-light irradiation in the absence of a photocatalyst. We found that this ability could be used for debromination of vicinal dibromo compounds to afford alkenes, as well as for hydrodifluoroacetamidation reactions. We also found that substituted Hantzsch esters could be used as reductive alkylation reagents to build highly congested ketones.

Abstract:The development and implementation of innovative synthetic methods that incorporate sustainable reagents and solvents are crucial aspects in both academic and industrial settings. Here, we report a novel organic photoredox-based strategy for the chemoselective deoxygenation of various functionalised N-heterocyclic N -oxides with high efficiency. The most notable feature of this catalytic process is the utilisation of the isopropanol (IPA)-acetone system of the Meerwein-Ponndorf-Verley (MPV) reaction as the redox couple matching the photocatalytic cycle, generating easily separable byproducts, namely acetone and water. Exploiting these characteristics, we have successfully conducted gram-scale deoxygenation reactions, eliminating the need for expensive and time-consuming column chromatography by using recrystallisation and distillation for product separation and purification. Furthermore, we demonstrate the practicality and sustainability of the MPV-type deoxygenation protocol by efficiently recycling both the reductant and the solvent (i.e., IPA and acetone) up to 10 times through a simple distillation process.

Abstract:Aromatization-driven deconstruction and functionalization of spiro dihydroquinazolinones via dual photoredox/nickel catalysis is developed. The aromatization effect was introduced to synergistically drive unstrained cyclic C-C bond cleavage, with the aim of overcoming the ring-size limitation of nitrogen-centered radical induced deconstruction of carbocycles. Herein, we demonstrate the synergistic photoredox/nickel catalyzed deconstructive cross-coupling of spiro dihydroquinazolinones with organic halides. Remarkably, structurally diverse organic halides including aryl, alkenyl, alkynyl, and alkyl bromides were compatible for the coupling. In addition, this protocol is also characterized by its mild and redox-neutral conditions, excellent functional group compatibility, high atom economy, and easy scalability. A telescoped procedure involving condensation and ring-opening/coupling was found to be accessible. This work provides a complementary strategy to the existing radical-mediated C-C bond cleavage of unstrained carbocycles.

Abstract:The chemoselective catalytic reduction of nitro scaffolds to value-added amine functions is desirable but challenging. We developed a combined KOtBu/BEt3 catalyst for deoxygenative reduction of nitroarenes, nitro heteroarenes and even notoriously challenging nitroalkanes using pinacolborane under mild reaction conditions. The novel transition-metal-free strategy is readily applicable to the preparation of commercially available pharmaceuticals.

Abstract:Organic nitrates, as effective donors of the signaling molecule nitric oxide, are widely applied in the pharmaceutical industry. However, practical and efficient methods for accessing organic nitrates are still scarce, and achieving high regiocontrol in unactivated alkene difunctionalization remains challenging. Here we present a simple and practical method for highly regioselective halonitrooxylation of unactivated alkenes. The approach utilizes TMSX (X: Cl, Br, or I) and oxybis(aryl-λ3-iodanediyl) dinitrates (OAIDN) as sources of halogen and nitrooxy groups, with 0.5 mol % FeCl3 as the catalyst. Remarkably, high regioselectivity in the halonitrooxylation of aromatic alkenes can be achieved even without any catalyst. This protocol features easy scalability and excellent functional group compatibility, providing a range of β-halonitrates (127 examples, up to 99% yield, up to >20:1 rr). Notably, 2-iodoethyl nitrate, a potent synthon derived from ethylene, reacts smoothly with a variety of functional units to incorporate the nitrooxy group into the desired molecules.
Nitrogen‐doped carbon as a highly active metal‐free catalyst for the selective oxidative dehydrogenation of N‐heterocycles

Abstract:N ‐heteroarenes represents one of the most important chemicals in pharmaceuticals and other bio‐active molecules, which can be easily accessed from the oxidation of N ‐heterocycles over metal catalysts. Herein, the metal‐free oxidative dehydrogenation of N ‐heterocycles into N ‐heteroarenes was developed using molecular oxygen as the terminal oxidant. The nitrogen‐doped carbon materials were facilely prepared via the simple pyrolysis process using biomass (carboxymethyl cellulose sodium) and dicyandiamide as the carbon source and nitrogen source, respectively, and they were discovered to be robust for the oxidative dehydrogenation of N ‐heterocycles into N ‐heteroarenes under mild conditions (80 °C under 1 bar O 2 ) with water as the green solvent. Diverse N ‐heterocycles including 1,2,3,4‐tetrahydroisoquinolines, indolines and 1,2,3,4‐tetrahydroquinoxalines were smoothly converted into N ‐heteroarenes with high to excellent yields (76%~>99%). Superoxide radical (·O 2 ‐ ) and hydroxyl radical (·OH) were probed as the reactive oxygen species for the oxidation of N ‐heterocycles into N ‐heteroarenes. More importantly, the nitrogen‐doped carbon catalyst can be reused with a high stability. The method provides an environmentally friendly and economical route to access important N ‐hetero‐aromatic commodities.

Abstract:Hydroxyl is widely found in organic molecules as functional group and its deprivation plays an inevitable role in organic synthesis. However, the direct cleavage of Csp3-O bond in alcohols with high selectivity and efficiency, especially without the assistance of metal catalyst, has been a formidable challenge because of its strong bond dissociation energy and unfavorable thermodynamics. Herein, an efficient metal-free strategy that enables direct deoxygenation of alcohols has been developed for the first time, with hydrazine as the reductant induced by light. This protocol features mild reaction conditions, excellent functional group tolerance, and abundant and easily available starting materials, rendering selective deoxygenation of a variety of 1° and 2° alcohols, vicinal diols, and β-1 and even β-O-4 models of natural wood lignin. This strategy is also highlighted by its "traceless" and non-toxic by-products N2 and H2, as readily escapable gases. Mechanistic studies demonstrated dimethyl sulfide being a key intermediate in this transformation.

Abstract:A visible‐light‐induced site selective C‐H alkylation of heterocyclic N‐oxides with N‐hydroxyphthalimide esters was developed using Eosin Y as the photocatalyst in the presence of Cs2CO3 under redox‐neutral and mild conditions. Using N‐hydroxyphthalimide esters as the radical precursors, quinoline or pyridine N‐oxides were readily coupled with a wide range of primary, secondary, and tertiary radicals to afford the desired alkylated heterocyclic N‐oxides in moderated to excellent yields. Importantly, this reaction protocol also successfully demonstrated its applications for the construction of glycosyl or bioactive natural dehydroabietic acid containing heterocyclic N‐oxides as well as the pharmaceutical and agrochemical active alkylated quinine‐based functional molecules (potential antimalarial drug).

Abstract:Replacing hydrogen with deuterium raises the activation energy for C‒D bond cleavage. This approach has gained attention in drug design, especially to protect the ortho-position of pyridines, which are susceptible to enzymatic oxidation. Until now, direct hydrogen isotope exchange has been largely restricted to the use of reactive organolithium reagents or metal-catalysed deuteration methods. In this work, we present a metal-free, selective ortho-deuteration of N-heterocycles starting from their N-oxides, proceeding at room temperature in just 5 minutes. This method achieves high deuterium incorporation across a broad range of N-heterocycles, including bioactive compounds. Experimental and computational studies have elucidated the mechanism of the reaction, showing that regioselectivity is driven by a successful increase in acidity at the ortho-position, enabling deprotonation by the in-situ generated dimsyl anion.

Abstract:Hantzsch ester is a widely used organic reductant. We describe a new use of this classical reductant in visible-light-induced desulfurative coupling of alkyl benzothiazolyl sulfides with electron-deficient alkenes/alkynes through activation with base additives. The C(sp3)-S scission has been achieved in two independent ways, catalyst-free Hantzsch ester anion-mediated reaction and organo-photocatalysis. The synthetic utility is illustrated with several examples of the derivatization of natural products including monosaccharides. The mechanistic investigation has revealed that Hantzsch ester anion works as a photoreductant in the catalyst-free reaction, whereas it is included as a sacrificial reductant in the organo-photocatalysis.

Abstract:Organic diazo compounds are versatile reagents in chemical synthesis and would benefit from improved synthetic accessibility, especially for larger scale applications. Here, we report a mild method for the synthesis of diazo compounds from hydrazones using a heterogeneous Fe-N-C catalyst, which has Fe ions dispersed within a graphitic nitrogen-doped carbon support. The reactions proceed readily at room temperature using O2 (1 atm) as the oxidant. Aryl diazoesters, ketones, and amides are accessible, in addition to less stable diaryl diazo compounds. Initial-rate data show that the Fe-N-C catalyst achieves faster rates than a heterogeneous Pt/C catalyst. The oxidative dehydrogenation of hydrazones may be performed in tandem with Rh-catalyzed enantioselective C-H insertion and cyclopropanation of alkenes, without requiring isolation of the diazo intermediate. This sequence is showcased by using a flow reactor for continuous synthesis of diazo compounds.

Abstract:A simple, efficient and green protocol for chemoselective reduction of carbon-carbon double bond in conjugated alkenes with Hantzsch esters is described. Without any additional catalysts, a series of conjugated alkenes with strong electron-withdrawing groups were reduced in water with excellent yield. Functional groups such as nitrile, ester, nitro, fluoro, chloro, bromo, furanyl and benzyl are all tolerated by the reaction conditions employed.

Abstract:This method describes the iron-promoted amidation of dioxazolone under visible light to obtain aromatic/alkyl substituted amides. The nitrogen radicals formed during this process undergo hydrogen atom transfer (HAT) to form amides through the proton provided by HBpin. This reaction eliminates the need for external chemical oxidants and expensive photocatalysts, and exhibits compatibility with various aryl/alkyl substituted derivatives. This method has good prospects for application and development in organic synthesis and medicine.

Abstract:The tandem reaction of photoinduced double hydrogen-atom transfer and deoxygenative transborylation for chemo- and site-selective reduction of nitroarenes into aryl amines under catalyst-free, room temperature conditions was disclosed in excellent yields. In this reaction, isopropanol (iPrOH) was used as hydrogen donor and tetrahydroxydiboron [B2(OH)4] as deoxygenative reagent with green, cheap, and commercially available credentials. In particular, a wide range of reducible functional groups such as halogen (-Cl, -Br and even -I), alkenyl, alkynyl, aldehyde, ketone, carboxyl, and cyano are all tolerated. Moreover, the reaction preferentially reduces the nitro group at the electron-deficient site over another nitro group in the same molecule. A detailed mechanistic investigation in combination of experiments and theoretical calculations gave a reasonable explanation for the reaction pathway.

Abstract:Reductive dearomatization has been a broadly explored means for rapid generation of sp3 complexity from simple planar arenes. Breaking the electron rich, stable aromatic systems requires strong reduction conditions. It has been notoriously challenging to dearomatize electron even richer heteroarenes. Herein we report an umpolung strategy enabling dearomatization of such structures under mild conditions. The reversal of the reactivity of these electron rich aromatics via photoredox mediated single electron transfer (SET) oxidation generates electrophilic radical cations, which can react with nucleophiles and break the aromatic structure to form a Birch type radical species. A crucial hydrogen atom transfer (HAT) is successfully engineered into the process to efficiently trap the dearomatic radical and minimize the formation of the overwhelmingly favorable, irreversible aromatization products. Particularly, a non-canonical dearomative ring-cleavage of thiophene/furan through selective C(sp2)-S bond breaking was first discovered. The preparing power of the protocol has been demonstrated for selective dearomatization and functionalization of various electron rich heteroarenes including thiophenes, furans, benzothiophenes and indoles. Furthermore, the process offers an unrivaled capacity for simultaneously introducing C-N/O/P bonds on these structures as exemplified by various "N", "O" and "P" centered functional moieties with 96 examples.
