Abstract:A novel biscinchona alkaloid-catalyzed highly enantioselective desymmetrization reaction of bisphenol compounds with achiral Morita-Baylis-Hillman carbonate agents was developed. Through the asymmetric allylic alkylation strategy, a broad range of optically active P-stereogenic phosphine oxides were generated with excellent to good yields (up to 99%) and high enantioselectivities (up to 98.5:1.5 e.r.). The reaction was further investigated by the linear free energy relationship (LFER) analysis. A possible transition state was proposed and furthered verified by theoretical calculations.

Abstract:A novel chiral biamide–phosphine multifunctional catalyst has been developed that mediates the asymmetric intermolecular Michael addition of β-carbonyl esters to β-trifluoromethyl enones and 3-aroyl acrylates in the presence of competing methyl acrylate and the inorganic base. This method provides a facile access to structurally diverse trifluoromethyl and quaternary stereogenic centers with excellent enantioselectivity (up to 99% ee) and good diastereoselectivity (up to 13:1 dr). The addition of the inorganic base (K3PO4) does not cause the background racemic reaction and enhances the reactivity by serving as a co-catalyst.

Abstract:A highly enantioselective and regioselective substitution reaction of the Morita-Baylis-Hillman (MBH) carbonates with nitroalkanes catalyzed by a quinidine-derived tertiary amine-thiourea catalyst has been developed. Abstract:A highly efficient asymmetric allylic substitution of Morita-Baylis-Hillman (MBH) carbonates with phosphorus ylides under the catalysis of modified cinchona alkaloids was developed. The reaction delivered products in moderate to good yields with excellent enantioselectivities (up to 98%ee) for a wide range of MBH carbonates. The scope of the reaction was demonstrated by the synthesis of chiral 3-substituted 2,4-functionalized 1,4-pentadienes.

Abstract:An asymmetric 1,6-conjugate addition reaction of dialkyl diazomethylphosphonates to para-quinone methides promoted by phase-transfer catalysis has been developed. A series of chiral diarylmethylated diazomethylphosphonates were accessed with up to 85% yields and 99% ee enantioselectivities. The resulting products were further transformed into bioactive compounds, namely, a chiral dihydrocinnoline phosphonate and a chiral α-aminophosphonate, bearing diarylmethine stereogenic centers.

Abstract:A novel phosphine-catalyzed, highly enantioselective umpolung addition of trifluoromethyl ketimines to Morita-Baylis-Hillman carbonates was developed and it provides facile access to optically active trifluoromethyl amines with a chiral tertiary stereocenter under mild reaction conditions. The salient features of this reaction include general substrate scope, mild reaction conditions, good yields, high enantioselectivity, ease of scale-up to gram scale, and further transformations of the products.

Abstract:A series of chiral 1,1'-binaphthyl-2,2'-disulfonic acids was designed, synthesized, and applied in a highly enantioselective Ti-catalyzed intramolecular hydroalkoxylation of nonactivated alkenes. The catalyst is probably a complex between two chiral binaphthyldisulfonate ligands and a bis(mu-oxo)-dititanium(IV) core structure. The sulfonamide groups of the ligands and water are necessary for the catalysis, as they may stabilize the catalytically active complex through hydrogen bonding. Various 2-methylcoumarans were obtained in up to greater than 99% yields and up to 97% enantiomeric excess under mild conditions.

Abstract:Chiral dihydrobenzofuran units are frequently present in molecules with significant biological and pharmaceutical activities. Herein, we present the first enantioselective formal [4+1] annulation of Morita‐Baylis‐Hillman carbonates with o‐quinone methides (o‐QMs) catalyzed by a newly designed chiral phosphine catalyst. Under the mild and eco‐friendly conditions, a wide range of polysubstituted dihydrobenzofurans were obtained in good yields with excellent enantioselectivities.

Abstract:Chiral phosphine‐catalyzed asymmetric (4+2) annulation of the amide‐based Morita–Baylis–Hillman (MBH) carbonates with β,γ‐unsaturated butenolides has been developed to give enantiomer‐enriched bicyclic δ‐lactam γ‐butyrolactone compounds. In this reaction, the amide‐based MBH carbonates were first used as acceptor and aza‐C4 synthon in phosphine catalysis. Comprehensive Summary Chiral phosphine‐catalyzed asymmetric (4+2) annulation of the amide‐based Morita–Baylis–Hillman (MBH) carbonates with β,γ‐unsaturated butenolides has been developed to give enantiomer‐enriched bicyclic δ‐lactam γ‐butyrolactone compounds. The amide‐based MBH carbonates were first used as acceptor and aza‐C4 synthon in phosphine catalysis. Under mild reaction conditions, a variety of amide‐based MBH carbonates and unsaturated butenolides were well tolerated to provide chiral bicyclic δ‐lactam γ‐butyrolactone derivatives in high yields with excellent enantioselectivities as well as diastereoselectivities. A plausible reaction mechanism was also proposed based on control experiments and DFT calculations.

Abstract:A general, efficient, and highly diastereoselective method for the synthesis of structurally and sterically diverse P-chiral phosphine oxides was developed. The method relies on sequential nucleophilic substitution on the versatile chiral phosphinyl transfer agent 1,3,2-benzoxazaphosphinine-2-oxide, which features enhanced and differentiated P-N and P-O bond reactivity toward nucleophiles. The reactivities of both bonds are fine-tuned to allow cleavage to occur even with sterically hindered nucleophiles under mild conditions.

Abstract:A series of novel bifunctional phosphine-sulfonamide organic catalysts were designed and readily prepared from natural amino acids, and they were utilized to promote enantioselective aza-Morita-Baylis-Hillman (MBH) reactions. L-threonine-derived phosphine-sulfonamide 9b was found to be the most efficient catalyst, affording the desired aza-MBH adducts in high yields and with excellent enantioselectivities.

Abstract:The synthesis ofP-stereogenic building blocks is extremely difficult. Herein we report an efficient kinetic resolution of secondary phosphine oxidesviaa Le-Phos-catalyzed asymmetric allylation reaction with Morita-Baylis-Hillman carbonates. This method provides facile access to enantioenriched secondary and tertiaryP-chiral phosphine oxides with broad substrate scope, both of which could serve asP-stereogenic synthons, and can be rapidly incorporated into a given scaffold bearing aP-stereocenter. The highly desirable late stage modifications demonstrate the practicability of our method and can be a critical contribution to obtaining optimalP-chiral catalysts and ligands.
