Abstract:We developed an optimized Liebeskind-Srogl reaction in order to synthesize potentially biologically active 2-aryl-1,4-dihydropyrimidines. The pallado-catalyzed cross-coupling reaction between dihydropyrimidines and tributyltin derivatives appears particularly efficient (67-95% yields) when using CuBr.Me2S as the copper cofactor. Abstract:Copper(II)-catalyzed synthesis of sulfones from sulfinate salts and aryl halides was investigated by means of a combination of experiment and DFT calculation. Experimental results demonstrated the wide applicability of the title approach. The reaction mechanisms are revealed by in-situ IR and theoretical study. It reveals remarkable ligand effect the bidentate amine plays in the reaction, that is, it initially activats the C-I bond of iodobenzene so as to enhance the whole catalytic process. Abstract:We report a stereoretentive cross-coupling reaction of configurationally stable nucleophiles with disulfide and N-sulfenylsuccinimide donors promoted by Cu(I). We demonstrate the utility of this method in the synthesis of thioglycosides derived from simple alkyl and aryl thiols, thioglycosides, and in the glycodiversification of cysteine residues in peptides. These reactions operate well with carbohydrate substrates containing common protective groups and reagents with free hydroxyl and secondary amide functionalities under standardized conditions. Competition experiments in combination with computational DFT studies established that the putative anomeric intermediate is an organocopper species that is configurationally stable and resistant to epimerization due to its short lifetime. The subsequent reductive elimination from the Cu(III) intermediate is rapid and stereoretentive. Taken together, the glycosyl cross-coupling is ideally suited for late stage glycodiversification and bioconjugation under highly controlled installation of the aliphatic carbon-sulfur bonds.

Abstract:Access to 3,4-and 3,5-disubstituted piperidine derivatives has been achieved through [Cp*Ru]-catalyzed intermolecular coupling of allylic alcohols and propargylic amides. Tandem transformation was also possible via chemoselective [Cp*Ru]-catalyzed hydrogenation of the resulting homodienes.

Abstract:The potentially bioactive 2-aryl-thiazolo[4,5-b]pyridines were synthesized via palladium-catalyzed desulfitative cross-coupling reaction between multisubstituted thiazolo[4,5-b]pyridine thioethers and boronic acids. Yields of [2_TD$IF]48%–94% were obtained with copper(I) thiophene-2-carboxylate in the system.

Abstract:This work reports a Cu‐catalyzed coupling technique for synthesizing (E)‐dienyl sulfones and β‐chlorosulfones from 1‐arylbutadienes and styrenes, respectively. By employing easily available and inexpensive reagents, this method enables the synthesis of a diverse array of dienyl sulfones and β‐chlorosulfones with excellent functional group tolerance under base‐ and oxidant‐free conditions. It also proves versatile for the late‐stage functionalization of drug molecules and the scale‐up reactions. The method showcases notable selectivity, gentle reaction conditions, and compatibility with various functional groups as demonstrated through 66 examples with excellent display of regioselectivity.

Abstract:Abstract: A highly efficient and simple cross-coupling reaction of N-aryl aluminum reagents with Naryl thiosuccinimides for the synthesis of diarylsulide derivatives using CuCl (20 mol%) as a catalyst is reported. Under the optimum reaction conditions, the coupling reaction between aryl aluminum reagents with different substituents and N-aryl thiosuccinimides with different substituents can be carried out smoothly with 18-94% isolated yields of diarylsulides. The method avoids the use of expensive transition metals, such as Pd, Ir, or Rh, phosphine ligands, and has the advantages of simple operation and high reaction efficiency. The structures of all the target compounds are confirmed by 1 HNMR and 13CNMR.

Abstract:The coupling of 3-substituted 4-alkoxy or 4-aminocyclobutene-1,2-diones induced by TiCl4–Zn was studied. It is interesting to find that the double bonds involved in the coupling reaction and the unsymmetrical coupling compounds were obtained as major or sole products. A possible mechanism mediated by titanium coordination intermediates is proposed.

Abstract:Baes-free Cu(II)-catalyzed coupling reaction of arylboronic acid and thiols was described. After optimization of reaction conditions, such as solvents, reaction temperatures, copper salts and ligands, the results show that some biarylsulfides were synthesized in good yields using 1,10-phenanthroline-Cu(OAc)2 center dot H2O as catalyst, DMSO/H2O (V : V=1 : 1) as solvent, under air atmosphere at 60 degrees C.

Abstract:Direct coupling of benzotriazole to unsaturated substrates such as allenes represents an atom-efficient method for the construction of biologically and pharmaceutically interesting functional structures. In this work, the mechanism of the N2-selective Rh complex-catalyzed coupling of benzotriazoles to allenes was investigated in depth using a combination of experimental and theoretical techniques. Substrate coordination, inhibition, and catalyst deactivation was probed in reactions of the neutral and cationic catalyst precursors [Rh(μ-Cl)(DPEPhos)]2 and [Rh(DPEPhos)(MeOH)2]+ with benzotriazole and allene, giving coordination, or coupling of the substrates. Formation of a rhodacycle, formed by unprecedented 1,2-coupling of allenes, is responsible for catalyst deactivation. Experimental and computational data suggest that cationic species, formed either by abstraction of the chloride ligand or used directly, are relevant for catalysis. Isomerization of benzotriazole and cleavage of its N-H bond are suggested to occur by counteranion-assisted proton shuttling. This contrasts with a previously proposed scenario in which oxidative N-H addition at Rh is one of the key steps. Based on the mechanistic analysis, the catalytic coupling reaction could be optimized, leading to lower reaction temperature and shorter reaction times compared to the literature.

Abstract:A novel and highly efficient methodology for the synthesis of 2,4,6‐triaryl‐1,3,5‐triazines and 2,4,6‐trisubstituted pyrimidines from amidines and alcohols has been established. Using the Earth‐abundant and air‐stable chromium salt catalyst and the phosphine‐free ligand, the developed Cr‐catalyzed acceptorless dehydrogenative coupling provides 1,3,5‐triazines and pyrimidines with high yields and good functional group tolerance. Moreover, the synthetic value of this operationally simple protocol was demonstrated by gram‐scale experiment and the synthesis of the linker to construct various supramolecular complexes.

Abstract:SuzukiMiyaura coupling of heteroaryls is an important method for the preparation of compound libraries for medicinal chemistry and materials research. Although many catalysts have been developed, none of them have been generally applicable to the coupling reactions of heteroaryl chlorides and tosylates at room temperature. We discovered that a catalyst combination of Pd(OAc)2 and XPhos (2-dicyclohexylphosphanyl-2',4',6'-triisopropylbiphenyl) could efficiently catalyze these couplings. Besides the choice of catalyst, the use of hydroxide bases in an aqueous alcoholic solvent was essential for fast couplings. These conditions promoted fast release of active catalyst (XPhos)Pd0, and accelerated the transmetalation in the catalytic cycle. Most of the major families of heteroaryl chlorides (31 examples) and tosylates (17 examples) reached full conversion within minutes to hours at room temperature. The method could be easily scaled up for gram-scale synthesis. Furthermore, we examined the relative reactivity of coupling partners in whole reactions. Electron-rich heteroaryl chlorides and tosylates reacted more slowly than electron-deficient ones, in the order of indole, pyrrole < furan, thiophene < pyridine and other six-membered-ring azines. For heteroarylboronic acids, the reactivity ranking was reversed: indole, pyrrole > furan, thiophene > pyridine. Similarly, electron-deficient arylboronic acids were less reactive than electron-neutral and electron-rich ones. The reactivity trends from this study can help to choose appropriate coupling partners for Suzuki reactions.
