Abstract:A unified strategy involving visible-light-induced iminyl-radical formation has been established for the construction of pyridines, quinolines, and phenanthridines from acyl oximes. With fac[Ir(ppy)(3)] as a photoredox catalyst, the acyl oximes were converted by 1e(-) reduction into iminyl radical intermediates, which then underwent intramolecular homolytic aromatic substitution (HAS) to give the N-containing arenes. These reactions proceeded with a broad range of substrates at room temperature in high yield. This strategy of visible-lightinduced iminyl-radical formation was successfully applied to a five-step concise synthesis of benzo[c]-phenanthridine alkaloids.

Abstract:Under visible light irradiation, phenanthridines were synthesized directly from 2-biphenylaldehyde and inorganic ammonium salts in a mild manner. This new protocol employs inorganic ammonium salt as ammonia source, which is readily available in nature, to couple with 2-biphenylaldehyde to afford imine intermediates, and then the cascade 6π cyclization and dehydrogenation take place to afford the targeting phenanthridine product. The mild reaction conditions together with catalyst-free and additives-free mode, make this protocol benign and practical.

Abstract:A visible light-induced cyclization of arylimines using trifluoromethanesulfonic acid (TfOH) as the catalyst is reported. The transformation, conducted under air and mild conditions utilizes O 2 as the oxidant, eliminating the need for leaving groups or excess acid. Optimization studies reveal the pivotal role of TfOH and visible light in the reaction. The methodology allows a broad substrate scope, accommodating various electron-donating and electron-withdrawing groups. The reaction conditions enable gram-scale synthesis and show inhibitory effects on cancer cell lines and low toxicity towards normal cells. Mechanistic insights indicate a visible light-induced 6π-electrocyclization. This method provides a practical and efficient approach for the synthesis of phenanthridine derivatives.

Abstract:Simple and practical strategies for visible-light-induced C-H alkylation of 2-amino-1,4-naphthoquinones with cyclobutanone oxime esters and hydroxamic acid derivatives have been developed under mild and redox-neutral conditions. These two reactions can be carried out at room temperature and obtain a variety of 2-amino-1,4-naphthoquinone derivatives with cyano and amide groups. Moreover, the cyanoalkylation reaction of 2-amino-1,4-naphthoquinones can proceed smoothly in the absence of photocatalysts.

Abstract:An isocyanide insertion reaction promoted by the combination of an amide and a photoredox is now presented. By using visible light, 6-amidophenanthridine derivatives can be prepared in good chemical yields under mild and eco-friendly reaction conditions. The reaction was shown to proceed through an electron transfer sequence by mechanistic experiments, and thus enabled the efficient production of 24 compounds, each with one new intramolecular aromatic ring obtained. The synthesized compounds were tested to embody good antitumor, antimicrobial and neuraminidase inhibitory activities.
