Abstract:Compounds containing aryl structures are important organic synthesis intermediates,which are widely present in a large family of natural products and bioactive molecules.In recent years,direct asymmetric functionalization of benzylic C(sp3)-H bond for the efficient construction of arene and hetero-arene motifs with high stereoselectivity has drawn widespread concern from chemical community.Among the various strategies,small molecule-catalyzed metal-free functionalization of benzylic C(sp3)-H bond represents a more challenging but promising transformations.This review is intended to summarize and discuss the most recent advances in this area.

Abstract:A late-stage, green and practical preparation of 6-( ortho -nitroaryl) amino purine derivatives was reported by using t -BuONO as the nitrating agent without transition-metal catalysts under air conditions. This method tolerates a wide variety of functional groups and is applicable for the gram scale synthesis, which may prove instrumental for further application in biochemistry. Preliminary mechanism was proposed through a series of experiments.

Abstract:The chemoselective catalytic reduction of nitro scaffolds to value-added amine functions is desirable but challenging. We developed a combined KOtBu/BEt3 catalyst for deoxygenative reduction of nitroarenes, nitro heteroarenes and even notoriously challenging nitroalkanes using pinacolborane under mild reaction conditions. The novel transition-metal-free strategy is readily applicable to the preparation of commercially available pharmaceuticals.

Abstract:A simple, general and efficient method is developed for the metal-free iodination of arylboronic acids. The protocol uses very cheap molecular iodine as the halide source and potassium carbonate as the base. The method is highly tolerant of various functional groups present in the substrates. Importantly, the iodination strategy can also be applied very effectively in the one-pot, two-step synthesis of biaryl derivatives.

Abstract:A sustainable, cyanide-free synthesis of alkyl nitriles via the aerobic oxidative deconstruction of unstrained cycloalkanones with ammonium salts has been developed. Using inexpensive and stable ammonium salts as the nitrogen source, a variety of alkyl nitriles containing a distal carbonyl group were obtained in good yields under visible-light-promoted iron catalysis. This protocol is characterized by mild conditions, abundant and environmentally benign materials, and high atom and step economy with minimal waste generation. The primary mechanism study revealed that 1O2 is likely to be involved in this reaction.

Abstract:A metal-free and sustainable visible-light-mediated method for the preparation of organic nitrate esters has been developed through the aerobic nitrooxylation reaction of α-diazoesters and cyclic ethers with t-BuONO in the presence of dioxygen. This protocol provides an efficient approach to access nitrate esters with the advantages of clean energy, broad substrate scope, green oxidants, operational simplicity, and mild conditions.

Abstract:We have developed a simple and mild photochemical process for the reductive denitration of nitroarenes under transition-metal-free conditions. This method is compatible with a broad range of functional groups, providing a practical and efficient approach for converting nitroarenes into denitrated arenes. The utility of this protocol is demonstrated through the prompt synthesis of dibenzoxepane.

Abstract:A novel, simple and efficient ipso-nitration of arylboronic acids with iron nitrate has been developed. The protocol uses readily available arylboronic acids as the starting materials, inexpensive iron nitrate (0.5 equiv.) as the nitro source without addition of an additive, and the corresponding nitroarenes were obtained in good to excellent yields.

Abstract:Chiral tertiary alcohols are an important structural motif, however, the general and efficient methodologies for their synthesis are less reported. Herein, we report a Ni(ІІ)-catalyzed asymmetric alkenylation and arylation of aryl ketones with organoborons under air via a 1,5-metalate shift strategy to obtain chiral tertiary allylic alcohols and diaryl alcohols. The reaction demonstrates good functional group tolerance and delivers chiral tertiary alcohols with good to excellent results. Furthermore, this method can be applied to the late-stage modification of drugs and the efficient synthesis of natural products. Notably, the reaction proceeds through an outer-sphere mechanism. The Ni(II) complex functions both as a Lewis acid to activate the ketone and create a chiral environment, and as coordination bridge linking the ketone and the organoboron-derived "ate" complex, facilitating the 1,5-metalate shift without forming a C-Ni bond. This approach contrasts with traditional transition metal-catalyzed nucleophilic addition reactions that involve carbon-metal bond formation.

Abstract:Transition-metal-free synthesis of α-aryl esters and nitriles using arylboronic acids with α-aminoesters and α-aminoacetonitriles, respectively, as the starting materials has been developed. The reaction represents a rare case of converting C(sp(3))-N bonds into C(sp(3))-C(sp(2)) bonds. The reaction conditions are mild, demonstrate good functional-group tolerance, and can be scaled up.

Abstract:A practical denitrative cyanation of feedstock nitroarenes under mild and transition metal-free reaction conditions has been developed. The key to success lies in the use of electrochemically driven, inexpensive ionic liquid N-methylimidazolium p-toluenesulfonate-promoted selective cathode reduction of nitroarenes to anilines, followed by diazoation, cathode reduction to form the aryl radical, and the essential radical cyanation process in one pot. Our protocol shows broad functional group tolerance and can be applied for the modification of bioactive targets.

Abstract:The development of sustainable late-stage functionalization (LSF) methods has become a focal point of research in organic synthesis field. Boronic acids and their derivatives are some of the most useful reagents and are suitable triggers for LSF. This study reports the production of diverse alkylated/nitrosoalkylated N-heteroaromatics via the photo-mediated nucleohomolytic substitution of alkylboronic esters with N-nitrosamines in the absence of a photocatalyst. This green approach facilitates LSF through various protodeboronation sequences for natural products and drug molecules, expanding the biologically relevant chemical space.

Abstract:Aryldiazonium salts remain a staple in organic synthesis and are still prepared largely in accord with the protocol developed in the 19th century. Because of the favorable reactivity that often cannot be achieved with other aryl(pseudo)halides, diazonium chemistry continues to grow. Facile extrusion of dinitrogen contributes to the desired reactivity but is also reason for safety concerns. Explosions have occurred since the discovery of these reagents and still result in accidents. In this study, we report a diazonium chemistry paradigm shift based on nitrate reduction using thiosulfate or dihalocuprates as electron donors that avoids diazonium accumulation. Because nitrate reduction is rate-limiting, aryldiazoniums are produced as fleeting intermediates, which results in a safer and often more efficient deaminative halogenation in a single step from anilines.
