Abstract:Nitriles unveil widespread applications in pharmaceuticals, agrochemicals, textiles, rubber, polymers, and constitute a significant intermediate in several organic transformations, necessitating the design of simple and environmentally benign pathways for their synthesis. Over the recent years, electro‐organic reactions have found widespread attention in developing effective and selective organic synthesis. They possess several advantages: high atom economy, selectivity, minimal waste production, and shorter routes to multistep traditional organic reactions. The development of novel strategies for greener and sustainable electro‐organic synthesis of nitriles is therefore commendable. This review focuses on analyzing various methods and strategies used in the electrochemical synthesis of nitriles using phase transfer catalyst, N‐oxoammonium salts mediated electrocatalysis, iodine‐mediated electrocatalysis, and anodic oxidations of aldoximes. In addition, the recent trends including the synthesis of nitriles via C−H cyanation, domino oxidation, bio electrocatalysis, and metal‐ligand cooperative synthesis have been discussed. Electrochemical synthesis of Nitriles: Nitriles play a major role in different chemical industries and are a key intermediate in several organic transformations. Electro‐organic reactions have received a lot of interest in recent years as a way to produce more effective synthesis. This article summarizes the electrochemical synthesis of nitriles from substrates as well as the numerous methods and advances in the electrochemical synthesis of nitriles.

Abstract:A sustainable, cyanide-free synthesis of alkyl nitriles via the aerobic oxidative deconstruction of unstrained cycloalkanones with ammonium salts has been developed. Using inexpensive and stable ammonium salts as the nitrogen source, a variety of alkyl nitriles containing a distal carbonyl group were obtained in good yields under visible-light-promoted iron catalysis. This protocol is characterized by mild conditions, abundant and environmentally benign materials, and high atom and step economy with minimal waste generation. The primary mechanism study revealed that 1O2 is likely to be involved in this reaction.

Abstract:Nitrile compounds are a class of high-value chemicals and versatile intermediates which can easily be transformed into a variety of useful products bearing functional groups such as carboxyl, carbamoyl, aminomethyl, ketyl and heterocyclic derivatives. Various thermal catalytic cyanation procedures have been devised and scaled up industrially while developing alternative methods are actively pursued. The access to these classes of molecules electrochemically offers greener alternatives to their preparation. The development of electrochemical synthesis of cyano-containing compounds under mild conditions with low energy consumption will imminently become indispensable approaches for industrial production of nitriles. The electrochemical cyanation presents many challenges from the toxicity of cyanide to the development of catalysts and the design of electrochemical cells. Electrochemical cyanation reaction offers promise to conveniently accessing nitriles but still requires efficient electro-catalysts, safe protocols and scale up considerations. This review discusses recent progress in the field of electrochemical synthesis of nitrile compounds placing emphasis on electro-synthetic and electro-catalytic mechanism aspects while making reference to original works to highlight the progress in this area.

Abstract:Conventional methods for nitrile synthesis bring inherent environmental risks due to their reliance on oxidants and harsh reaction conditions. Meanwhile, direct electrooxidation of amines to nitriles suffers from low current density. In this study, we propose an innovative indirect electrooxidation strategy for nitrile formation, mediated by Br-/Br2, utilizing a highly efficient CoS2/CoS@Graphite Felt (GF) electrode. Notably, the anodic nitrile generation can be synergistically coupled with the cathodic hydrogen evolution reaction (HER). Through meticulous optimization of reaction parameters, we achieve an impressive 98% selectivity for octanenitrile at a current density of 60 mA cm-2 with a remarkable faradaic efficiency (FE) of 87%. Furthermore, our approach demonstrates excellent versatility, as we successfully evaluate both aliphatic and aromatic primary amines, highlighting its promising potential for practical applications in the field.

Abstract:Aryl nitriles are widely found in natural products, pharmaceuticals, agrochemicals, dyes, and herbicides. Moreover, because the cyano group can be easily converted into various functional groups, aryl nitriles are important intermediates in synthetic organic chemistry. Consequently, the development of new methodologies for the synthesis of aryl nitriles is of significant interest. The traditional methods for their synthesis include Sandmeyer reactions, Rosenmund–von Braun reactions, as well as dehydration reactions of aldoximes and amides. In recent years, numerous efforts have been devoted to transition metal‐catalyzed cyanation reactions of aryl (pseudo)halides, organometallic reagents and aryl C–H bonds. A number of cyanating reagents have been explored for such transformations, including metal cyanides and organic cyano group sources. In addition, styrenes and aryl‐substituted alkynes are successfully converted into aryl nitriles through carbon‐carbon double or triple bond cleavage. Various benzylic compounds have also been used for the synthesis of aryl nitriles, such as benzylic halides, alcohols, amines and azides. Furthermore, the direct conversion of the methyl group of toluene derivatives into a cyano group has also drawn attention. In this critical review, we summarize the recent developments and useful applications in this field and reaction mechanisms are also briefly discussed.

Abstract:An iodine-promoted, metal-, base-, and solvent-free cross-coupling reaction was developed for the synthesis of various useful secondary amides via an aryl N-addition reaction of aryl groups to cyano groups. This aryl transfer reaction proceeds with arylhydrazine hydrochlorides serving as the aryl donors. A labelling experiment shows that the N atom in the product comes from the cyano group of the nitriles, which are low in cost. A plausible radical-driven mechanism is also proposed.

Abstract:Developing green strategies for the synthesis of nitriles from cheap starting materials, without toxic and hazardous reagents under mild conditions is important for the sustainable development of the chemical industry. Herein, we report an oxidant- and cyanide-free catalytic system for the synthesis of various nitriles via electro-oxidative coupling of corresponding primary alcohol and ammonia in an aqueous solution with Pd nanoparticle-modified CuO nanowires as the catalyst. Produce benzonitrile from benzyl alcohol achieve a conversion of > 99.0 % and a selectivity of 83.2 %. Mechanism studies demonstrate that the coupling of surface-adsorbed active species NH 2 * and Ph-CHO* is the rate-limiting step, and the Pd/CuO heterojunction is the active center. The electrogenerated Cu III species provide the driving force for oxidation, and Pd contributes to improving the dehydrogen ability and facilitating the subsequent coupling reaction. Our research provides an environment-friendly oxidative C-N coupling method for efficient nitriles electrosynthesis.

Abstract:A novel and environmentally friendly electrochemical process is described for the direct C-3 arylation of quinoxalin-2(1 H )-ones using readily available cyanoaromatics through paired electrolysis. The reaction features excellent atom economy, mild reaction conditions and simple operation (using undivided electrolytic cells at ambient temperature), without the need for oxidants, catalysts, metals, or sacrificial reagents. It offers a practical method of synthesizing 3-arylquinoxalin-2(1 H )-one derivatives.

Abstract:The work employs aliphatic and aromatic isothiocyanates and utilizes electrochemical techniques for the reaction. The process involves a two-electrode system with a conductive carbon working electrode and a counter electrode. An appropriate electrolyte, such as an organic solvent, is used, and upon applying an electric potential, electrochemical oxidation and nucleophilic substitution reactions occur, leading to the formation of the desired 1,3,4-oxadiazole-2-amines. Key advantages of this electrochemical method include its high functional group tolerance, enabling the synthesis of a wide range of compounds with different substituents. The reaction conditions can be fine-tuned for high conversion and selectivity, ensuring good overall yields of the target products. Additionally, the electrochemical method offers sustainability and convenience, making it a promising alternative to traditional synthesis. The use of electricity as an energy source is more environmentally friendly compared to traditional heating methods, contributing to greener and eco-friendly chemical processes.

Abstract:Under very mild conditions, biotransformations of racemic azido nitriles using Rhodococcus erythropolis AJ270, a nitrile hydratase/amidase-containing microbial whole-cell catalyst, afforded highly enantiopure, (R)-α-arylmethyl- and (+)-α-cyclohexylmethyl-β-azidopropanoic acids and their (S)- and (−)-carboxamide derivatives in excellent yields. The resulting functionalized chiral organoazides were converted in a straightforward fashion to a pair of antipodes of α-benzyl-β-amino acids (R)-13 and (S)-13. Azido carboxamide (S)-11a and azido carboxylic acid (R)-12a underwent ‘click’ reactions with diethyl acetylenedicarboxylate and phenylacetylene to produce functionalized chiral triazoles 14 and 15, respectively. The easy preparation of the starting nitrile substrates, highly efficient and enantioselective biotransformation reactions, and versatile utility of the resulting functionalized azido carboxylic acids and amide derivatives, render this method very attractive and practical in organic synthesis.

Abstract:Asymmetric electrochemical synthesis is recognized as a powerful and scalable methodology for organic synthesis. This study presents the first example of electrochemical enantioselective Ni‐catalyzed reductive conjugate hydro‐arylation/alkenylation of enones in an undivided cell, affording more than 56 valuable functionalized β‐arylated ketones in good to excellent enantioselectivity. Furthermore, various bio‐relevant compounds are successfully synthesized through this scalable protocol. Herein, the study reports the first electrochemical nickel‐catalyzed enantioselective hydro‐arylation/alkenylation of enones in an undivided cell with low‐cost electrodes in the absence of external reductants and supporting electrolytes. Aryl bromides/iodides/triflates or alkenyl bromides are employed as electrophiles for the efficient preparation of more than 56 valuable β‐arylated/alkenylated ketones in a simple manner (up to 97% yield, 97% ee). With the advantages of electrochemistry, excellent functional group tolerance and late‐stage modification of complex natural products and pharmaceuticals made the established protocol greener and more economic. Mechanism investigation suggests that a NiI/NiIII cycle may be involved in this electro‐reductive reaction rather than metal reductant driven Ni0/NiII cycle. Overall, the efficient electrochemical activation and turnover of the nickel catalyst avoid the drawbacks posed by the employment of stoichiometric amount of sensitive metal powder reductants.

Abstract:Given the natural abundance and industrial accessibility, inert amides have gained considerable attention in homologous amine synthesis and the development of novel catalytic strategies for their deoxygenative functionalization is highly desirable. Although organic electrosynthesis has become an uprising research area by virtue of its sustainability and atom economy, the combination of amide reduction with an electrochemical process in value-added amine synthesis is still undeveloped. Herein, a strategy merging iridium-catalyzed activation of amides with electrochemical radical generation to access deoxygenative arylation of secondary amides is developed. This methodology is featured by broad substrate scope, mild reaction conditions, and operational simplicity, giving corresponding benzylic amines with high efficiency. Notably, the successful application of electrochemical method in the deoxygenative functionalization of amides would provide an efficient strategy for quick access to value-added amines from inert amides.

Abstract:A practical denitrative cyanation of feedstock nitroarenes under mild and transition metal-free reaction conditions has been developed. The key to success lies in the use of electrochemically driven, inexpensive ionic liquid N-methylimidazolium p-toluenesulfonate-promoted selective cathode reduction of nitroarenes to anilines, followed by diazoation, cathode reduction to form the aryl radical, and the essential radical cyanation process in one pot. Our protocol shows broad functional group tolerance and can be applied for the modification of bioactive targets.
