Abstract:Twenty‐one novel 2‐aryloxy‐3‐(4‐chlorophenyl)‐8‐substituted‐5‐aryl‐8,9‐dihydro‐3H‐chromeno[2,3‐d]pyrimidine‐4,6(5H,7H)‐diones 5a, 5b, 5c, 5d, 5e, 5f, 5g, 5h, 5i, 5j, 5k, 5l, 5m, 5n, 5o, 5p, 5q, 5r, 5s, 5t, 5u were designed and easily synthesized via a tandem aza‐Wittig reaction. Treatment of iminophosphorane 3 with 4‐chlorophenyl iso‐cyanate gave carbodiimide 4, which reacted with phenols to provide the title compounds in 50–73% isolated yields. All compounds 3 and 5 were confirmed by infrared, 1H‐NMR, mass spectra, and elemental analysis, and compound 5a was further analyzed by single‐crystal X‐ray diffraction, and the title compounds were synthesized with the purpose of bringing in some new chemical and biological interests.

Abstract:A series of novel 5-(3-aryl-1H-pyrazol-5-yl)-2-(3-butyl-1-chloroimidazo[1,5-a]- pyridin-7-yl)-1,3,4-oxadiazole derivatives has been synthesized from 3-butyl-1-chloroimidazo[1,5-a]pyridine-7-carboxylic acid and ethyl 3-aryl-1H-pyrazole-5-carboxylate. The compounds were characterized using IR, (1)H NMR, HRMS and UV-vis absorption. The fluorescence spectral characteristics of the compounds in dichloromethane were investigated. The results showed that absorption λmax and emission λmax was less correlated with substituent groups on N-1 position of pyrazole moiety and para position of benzene moiety. The calculated molecular orbital correlates well with their absorption.

Abstract:A new class of thiazolo[3,2-a]pyrimidine compounds were synthesized by the reaction of 5-carbethoxyl-4-aryl-6-methyl-1,2,3,4-tetrahydropyrimidin-2-ones with dimethyl acetylenedicarboxylate. The reaction has the advantages of excellent yields and simple workup procedures. The products were characterized thoroughly by IR and NMR (H-1, C-13, COSY, HSQC and HMBC) together with elemental analysis. The X-ray crystallographic analysis of the 1,3-dipolar cycloaddition derivative of the title product provided the direct information for the structure of the product.

Abstract:Aim and objective: According to the literature survey, pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the present work, pyrazol- 3-one 1 was reacted with cyanoacetic acid hydrazide and elemental sulfur to afford the corresponding thieno[3,2-c]pyrazol-6-carbohydrazide 3 derivatives. The latter compound reacted with some electrophilic reagents such as DMF-DMA, triethylorthoformate, arylidenemalononitriles and chalcones under neat conditions to give substituted oxadiazole and pyrazole, respectively. The treatment of compound 3 with active methylene reagents such as acetylacetone, diethylmalonate, ethyl acetoacetate and ethyl cyanoacetate under suitable conditions afforded pyrazole derivatives 10, 11, 13, and 15, respectively. Novel pyrazolothienopyrimidine 27 and 30 were prepared from precursor 26 with carbon disulfide and triethylorthoformate, respectively. The chemical structures of the newly synthesized compounds were established by elemental and spectral analyses including IR, and 1HNMR in addition to 13C-NMR and mass spectra. Materials and methods: A novel substituted pyrazole, pyrimidine and pyrazolothienopyrimidine were obtained via Gewald synthesis of thiophene and fused thiophene and Mannich reactions of 5-amino-3-phenyl-1Hthieno[ 3,2-c]pyrazole-6-carbohydrazide. Results and discussion: A series of some newly azoles and azines were prepared via reaction of thieno[3,2- c]pyrazol-6-carbohydrazide derivative 3 as starting material with some electrophilic and nucleophilic reagents. The structures of target compounds were established by elemental analyses and spectral data. Conclusion: Pyrazole is a unique template that is associated with several biological activities. This article highlighted the research work of many researchers reported in the literature for synthesis and different pharmacological activities of the pyrazole nucleus. In the current investigation, we have developed new and efficient methods for the synthesis of thieno[3,2-c]pyrazol-6-carbohydrazide derivatives. In addition, we have explored the preparative potential of these substances as intermediates for the synthesis of substituted pyrazoles and fused pyrazoles 10-30, respectively.

Abstract:A new synthesis of 4-methoxy (or ethoxy)-1,6-diaryl-1H-pyrazolo[3,4-d]pyrimidine derivatives is described. An efficient one-step methodology for the title compounds was achieved by using 5-amino-1-aryl-1H-pyrazole-4-carbonitriles with arylaldehydes in CH3OH or C2H5OH and NaOH at 60 degrees C. The new compounds were obtained in moderate to good yields as stable solids and easily purified by filtration.

Abstract:A new series of imidazo[1,2-a]pyridin-2-one derivatives were synthesized by the reaction of 4-arylidene-2-phenyl-5(4H)-oxazolones and pyridin-2-amine under microwave irradiation in ethylene glycol. The starting materials were easily obtained and the operation was very convenient.

Abstract:Pyrazolo [1,5-a] pyrimidinies have shown many excellent biological activities in the areas of medicine.In this paper,we report the synthesis and characterization of three new 6-arylsubstituted pyrazolo [1,5-a] pyrimidine derivatives.The intermediate 4-(2,4-dichlorophenyl)-3-methyl-1H-pyrazol-5-amine was synthesized from 2,4-dichlorobenzyl Cyanide,through acetylation with acetyl chloride,followed by condension with hydrazine hydrate;Methyl 4-(trifluoromethyl)benzoate were condensed with acetonitrile to yield 3-oxopropanenitrile that reacted with hydrazine hydrate to yield 3-(4-(trifluoromethyl)phenyl)-1H-pyrazol-5-amine.The desired 6-arylsubstituted pyrazolo [1,5-a] pyrimidines are prepared by the reaction of diethyl 2-phenylmalonates with 5-amino-1H-pyrazoles in good yields.The structure of two pyrazolo [1,5-a] pyrimidines was characterized by IR and 1HNMR.

Abstract:Pyrazolo[4,5-a]pyrimidines have shown many excellent biological activities in the areas of medicine.In this paper,three novel trifluoromethylated pyrazolopyrimidines(5a,5b and 7) were synthesized.The intermediate ethyl 5-amino-1H-pyrazole-4-carboxylate(2) was prepared via the condensation of ethyl 2-cyano-3-ethoxyacrylate and hydrazine hydrate.4,4,4-trifluoro-1-(4-substitutedphenyl)butane-1,3-dione(4) were obtained by the reaction of acetophenone with ethyl trifluoroacetate.The desired pyrazolopyrimidines(5a,5b) were prepared via the condensation of ethyl 5-amino-1H-pyrazole-4-carboxylate(2) and 4,4,4-trifluoro-1-(4-substitutedphenyl)butane-1,3-dione(4) in good yields.The compound 7 was obtained by the reaction of 5a with NBS,and then reacted with morpholine in N,N-dimethylformamide.The structure were characterized by IR and 1HNMR.
