Abstract:A new method was developed for the synthesis of 6,7-dihydro-5H-pyrimido[4,5-e][1,4]diazepin-8(9H)-one derivatives. The key to construct the pyrimido[4,5-e][1,4]diazepine core is the intramolecular amidation of N-((4-amino-6-chloropyrimidin-5-yl) methyl)-substituted amino acid esters. This methodology was validated through the preparation of 13 representative 6,7-dihydro-5H-pyrimido[4,5-e][1,4]diazepin-8(9H)-ones in moderate to good yields.

Abstract:Practical and efficient methods have been developed for the synthesis of 4,6,8,9-tetrasubstituted 8,9-dihydro-5H-pyrimido[4,5-e][1,4]diazepin-7(6H)-ones from 4,6-dicholoropyrimidine aldehyde, N-substituted amino acid esters, and amines in five steps. This synthetic strategy is based on suitably substituted pyrimidines as bis-electrophilic species reacting with various amines to construct the pyrimido[4,5-e][1,4]diazepine core with a strategically anchored functional group for further derivatization. The utility of this methodology was demonstrated through the preparation of a 33-membered library of representative 8,9-dihydro-5H-pyrimido[4,5-e][1,4]diazepin-7(6H)-ones in good to excellent yields.

Abstract:A series of tricyclic 7,8,10,11-tetrahydro-5H-benzo[e]pyrimido[4,5-b][1,4]diazepin-9(6H)-ones were prepared in moderate to high yields using TFA-promoted iminium-cyclization reactions of 3-(6-(butylamino)-4-chloropyrimidin-5-ylamino)cyclohex-2-enones and various aldehydes.

Abstract:A new class of thiazolo[3,2-a]pyrimidine compounds were synthesized by the reaction of 5-carbethoxyl-4-aryl-6-methyl-1,2,3,4-tetrahydropyrimidin-2-ones with dimethyl acetylenedicarboxylate. The reaction has the advantages of excellent yields and simple workup procedures. The products were characterized thoroughly by IR and NMR (H-1, C-13, COSY, HSQC and HMBC) together with elemental analysis. The X-ray crystallographic analysis of the 1,3-dipolar cycloaddition derivative of the title product provided the direct information for the structure of the product.

Abstract:We have previously reported the preliminary fungicidal activityof 3-anilinemethylene-5,6-dihydro-6-phenyl-2H-pyran-2,4-diones and their 3D-QSAR study. In this paper, sixteen novel compounds of this series are obtained in satisfactory yields (87%～97%) by using a new “one-pot” synthetic method. The Z/E isomer relation of compounds (2) is discussed briefly.

Abstract:A copper-catalyzed sequential arylation and intramolecular annulation of 2-(2-bromophenyl)-2,3-dihydroquinazolin-4(1H)-ones with alkyl amidines has been developed for the synthesis of 6-alkyl 11bHquinazolino[ 3,4-a] quinazolin-13(12H)-ones (3) in moderate to good yields. However, the arylated products N-(2-(4-oxo-1,2,3,4-tetrahydroquinazolin- 2-yl) phenyl) benzimidamides (4) are isolated when aryl amidines are used in the transformation.
